      • Study on the Linkage Effect between China and South Korea Stock Market

        As the trend of the world financial integration gradually strengthen, the research of the relationship between different stock markets mean a lot to improve the capital markets. The purpose of this article is to explore the linkage effect of market between China and South Korea. There is no evidence to prove that the five variables on behalf of the Chinese stock market have any relationship with KOSPI200 index by the granger causality test. There is no co-integration relationship between two markets by using co integration test to explore the long-term stable equilibrium relationship between China and South Korea stock markets. Therefore there is no significant relationship between China and South Korea market.

        Excessive mechanical strain accelerates intervertebral disc degeneration by disrupting intrinsic circadian rhythm

        Night shift workers with disordered rhythmic mechanical loading are more prone to intervertebral disc degeneration (IDD). Our results showed that circadian rhythm (CR) was dampened in degenerated and aged NP cells. Long-term environmental CR disruption promoted IDD in rats. Excessive mechanical strain disrupted the CR and inhibited the expression of core clock proteins. The inhibitory effect of mechanical loading on the expression of extracellular matrix genes could be reversed by BMAL1 overexpression in NP cells. The Rho/ROCK pathway was demonstrated to mediate the effect of mechanical stimulation on CR. Prolonged mechanical loading for 12 months affected intrinsic CR genes and induced IDD in a model of upright posture in a normal environment. Unexpectedly, mechanical loading further accelerated the IDD in an Light-Dark (LD) cycle-disrupted environment. These results indicated that intrinsic CR disruption might be a mechanism involved in overloading-induced IDD and a potential drug target for night shift workers.

        Assessment of nephrotoxicity of herbal medicine containing aristolochic acid in mice

        Background/Aims: It is undetermined if herbal medicines (HM) containing aristolochic acid (AA)-containing have similar nephrotoxicity to AA itself. Methods: We administered HM containing a high concentration of AA for 5 days (short-term study) or a low concentration of AA for 30 days (long-term study) to C57BL/6 mice; for comparison, same dose of AA compound was used as controls. Results: The nephrotoxicity in the HM- and AA-treated mice was compared in terms of renal function, histopathology, oxidative stress, apoptotic cell death, and mitochondrial damage. Short-term HM treatment resulted in acute kidney injury (marked renal dysfunction, acute tubular necrosis, and neutrophil gelatinase-associated lipocalin [NGAL] expression) in which the severity of renal dysfunction and histopathology was comparable with that induced by the administration of AA alone. Long-term HM treatment resulted in features of chronic kidney disease (CKD, mild renal dysfunction and tubular atrophy and dilatation). No significant differences in these parameters were observed between the HM- and AA-treated mice. HM-induced oxidative stress (8-hydroxy-2’-deoxyguanosine and manganese- dependent superoxide dismutase expression) and apoptotic cell death (terminal deoxynucleotidyl transferase dUTP nick end labelling [TUNEL]-positive cells and active caspase-3 expression) were similar in HM- and AA-treated mice in the short-term and long-term studies. Mitochondrial injury, evaluated by electron microscopy, was also similar in HM- and AA-treated mice in the short-term and long-term studies. Conclusions: The nephrotoxic potential of HM containing AA was similar to that of AA itself.

      • Ginseng extract reduces tacrolimus-induced oxidative stress by modulating autophagy in pancreatic beta cells

        <P>We previously reported that long-term treatment with a calcineurin inhibitor impairs autophagy process in pancreatic beta cells. This study investigated the effect of Korean red ginseng extract (KRGE) on autophagy modulated by oxidative stress. In mice with tacrolimus (Tac)-induced diabetes mellitus, KRGE alleviated islet dysfunction and decreased oxidative stress and autophagic vacuoles. In vitro, KRGE decreased autophagosome formation and attenuated lysosomal degradation, accompanied by improved beta cell viability and insulin secretion. Addition of 3-methyladenine (3-MA), an inhibitor of autophagosomes, to KRGE further improved cell viability and insulin secretion, and bafilomycin A (BA), an inhibitor of lysosomal function, reduced the effects of KRGE. At the subcellular level, Tac caused mitochondrial dysfunction (impaired mitochondrial oxygen consumption, ATP production, and increased reactive oxygen species production). But KRGE improved these parameters. The effect of KRGE on mitochondrial function enhanced by 3-MA but decreased by BA, suggesting a causal relationship between KRGE effect and autophagy modulation in Tac-induced mitochondrial dysfunction. These findings indicate that KRGE modulates autophagy favorably by reducing Tac-induced oxidative stress, and this effect is closely associated with improvement of mitochondrial function.</P>

      • A SANDWICH-INJECTION METHOD FOR MICROCHIP ELECTROPHORESIS

        In microchip electrophoresis (μ-CE), sample injection is generally achieved through cross, double-T, or T-form injector structures. In these reported approaches, the separation efficiency and detection sensitivity of μ-CE is significantly influenced by the shape and size of the sample plug introduced into the separation channel or sample leakage in separation phase. Here, we present a sandwich-injection method for controlling discrete sample injection in μ-CE. This method involves four accessory arm channels in which symmetrical potentials are loaded to form a unique parallel electric field distribution at the intersection of sample and separation channels. The parallel electric field effectuate a virtual wall to confine the shape of a sample plug and depress the spreading of the sample plug at the junction of sample and separation channels, and also prevent sample leakage during separation step. The key features of this method are the ability to inject well-defined sample plugs at the original sample concentration and the ability to control the sample plug size. The virtues of the novel injection technique were demonstrated with numerical models and validated with fluorescence visualizations of electrophoretic experiments.

        RPK118, a PX Domain-containing Protein, Interacts with Peroxiredoxin-3 through Pseudo-Kinase Domains

        RPK118 is a sphingosine kinase-1-binding protein that has been implicated in sphingosine 1 phosphate-mediated signaling. It contains a PX (phox homology) domain and two pseudo-kinase domains, and co-localizes with sphingosine kinase-1 on early endosomes. In this study we identified a novel RPK118-binding protein, PRDX3 (peroxiredoxin-3), by yeast two-hybrid screening. The interaction between these proteins was confirmed by pull-down assays and co-immunoprecipitation experi-ments. Deletion studies showed that RPK118 inter-acted with PRDX3 through its pseudokinase domains, and with early endosomes through its PX domain. Dou-ble immunofluorescence experiments demonstrated that PRDX3 co-localized with RPK118 on early endosomes in COS7 cells. PRDX3 is a member of the antioxidant family of proteins synthesized in the cytoplasm and functioning in mitochondria. Our findings indicate that RPK118 is a PRDX3-binding protein that may be involved in transporting PRDX3 from the cytoplasm to its mitochondrial site of function or to other mem-brane structures via endosome trafficking

        Distributed Functional Observer-based Event-triggered Containment Control of Multi-agent Systems

        This paper studies the containment control problem of linear multi-agent systems (MASs) with a directed graph, where the states of the followers will asymptotically converge to the convex hull formed by those of the leaders. By introducing the functional observer, a novel distributed event-triggered control strategy is presented to schedule communications between agents, which depends on the relative input and output measurements with its neighbors. Furthermore, the minimum inter-event time is guaranteed to be strictly positive. An example is presented to illustrate the feasibility and efficiency of the theoretic results.

        New Event-based Control for Sampled-data Consensus of Multi-agent Systems

        This paper investigates the sampled-data consensus problem of general linear multi-agent systems vianew event-based control. Both the leaderless and leader-following consensus problems are considered. Differentfrom the existing results, the state information is assumed to be unknown for each following agent and only therelative output information between neighbouring agents can be measured. A new Kx-functional observer-basedoutput feedback event-based protocol is designed, which may have lower dimension than the full-order and reducedorderobserver. The improved event-based condition bounds each agent’s measurement error by a time-dependentthreshold. Furthermore, the Zeno behavior is excluded by showing that the inter-event time between any twotriggering events is lower bounded by a strictly positive value. Finally, a numerical simulation example is presentedto demonstrate the effectiveness of the proposed control protocols.

      • Growth, Clonability, and Radiation Resistance of Esophageal Carcinoma-derived Stem-like Cells

        Objective: To separate/enrich tumor stem-like cells from the human esophageal carcinoma cell line OE-19 by using serum-free suspension culture and to identify their biological characteristics and radiation resistance. Methods: OE-19 cells were cultivated using adherent and suspension culture methods. The tumor stem-like phenotype of CD44 expression was detected using flow cytometry. We examined growth characteristics, cloning capacity in soft agar, and radiation resistance of 2 groups of cells. Results: Suspended cells in serum-free medium formed spheres that were enriched for CD44 expression. CD44 was expressed in 62.5% of suspended cells, but only in 11.7% of adherent cells. The suspended cells had greater capacity for proliferation and colony formation in soft agar than the adherent cells. When the suspended and adherent cells were irradiated at 5 Gy, 10 Gy, or 15 Gy, the proportion of CD44+ suspended cells strongly and weakly positive for CD44 was 77.8%, 66.5%, 57.5%; and 21.7%, 31.6%, 41.4%, respectively. In contrast, the proportion of CD44+ adherent cells strongly positive for CD44 was 18.9%, 14.%, and 9.95%, respectively. When the irradiation dose was increased to 30 Gy, the survival of the suspended and adherent cells was significantly reduced, and viable CD44+ cells were not detected. Conclusion: Suspended cell spheres generated from OE-19 esophageal carcinoma cells in serum-free stem medium are enriched in tumor stem-like cells. CD44 may be a marker for these cells.
