        DNA methylation and mRNA expression of COL6A3 in antler mesenchyme of female and male reindeer

        Backgroud Reindeer is the only deer species that both male and female produce antlers, which provides a particularly interesting case in studying the differences between antlers of the two sexes. Alpha 3(VI) Collagen Gene (COL6A3), forms a microfibrillar network associated with the structural integrity and biomechanical properties, has been found to be one of the differentially expressed genes in antler mesenchyme of female and male reindeer. Objective and Methods The promoter sequence of reindeer COL6A3 gene was obtained using the cloning technology and analyzed by the bioinformatics methods. Bisulfite sequencing PCR (BSP) was used to detect the methylation status of the COL6A3 promoter in reindeer antler mesenchyme. Real-time quantitative PCR was used to detect COL6A3 expression in the antler mesenchyme of female and male reindeer. Results Sequence analysis revealed that the reindeer COL6A3 partial promoter sequence was 983 bp including the possible promoter region at + 105 bp to + 155 bp. Homology and phylogenetic analysis indicated that the COL6A3 promoter of reindeer had the closest genetic distance with Bos taurus, Capra hircus and Ovis aries. BSP results indicated that the methylation level of COL6A3 promoter in the female reindeer antler mesenchyme was significantly higher than in the male. Correlating with increased methylation status, we also found that COL6A3 mRNA expression in female reindeer antler mesenchyme was significantly lower than in the male. Conclusion The higher methylation level of the COL6A3 gene in female reindeer antler mesenchyme coincides with decreased COL6A3 mRNA expression, thereby affecting the transposon silencing mechanism and possibly contributing to apparent differences of antlers in female and male reindeer.

        Effects of temperature and humidity on immature development of Lygus pratensis (L.) (Hemiptera: Miridae)

        Lygus pratensis distributes worldwide and is one of the common insect pests on cotton in Xinjiang Uygur Autonomous Region, the largest cotton growing region of China. This study assessed the effects of six constant temperatures (10, 15, 20, 25, 30 and 35 °C) at 60% relative humidity (RH) on egg and nymphal development of L. pratensis. Eggs failed to hatch and the newly-hatched nymphs could not normally develop at 10 °C. The developmental duration of egg and nymph decreased as temperature increased from 15 to 30 except 35 °C for egg. Based on the linear model, the lower developmental threshold and effective accumulated temperature were 10.68 °C and 150.2 DD for egg, and 12.08 °C and 208.3 DD for nymph, respectively. Among three non-linear models (Briere-1, Logan-6 and Lactin), Logan-6 provided the most accurate estimate for the mean optimum and lethal maximum temperatures (33.6 and 40.9 °C for egg, and 34.0 and 37.4 °C for nymphs). The interactive effects of three RH levels (45, 60 and 75%) and two temperatures (25 and 35 °C) on the immature developmental stages were tested. Temperature, RH and their interaction showed significant effects on egg and nymphal development. High relative humidity (75% RH) shortened the developmental duration of egg at 25 °C and nymph at 25 and 35 °C,whereas no significant difference was found for egg duration between different RH levels at 35 °C. The present study is useful for further predicting the phenology of L. pratensis and developing forecast and management strategies for this emerging mirid bug in China.

      • Systematic Analysis of Icotinib Treatment for Patients with Non-Small Cell Lung Cancer

        Purpose: This analysis was conducted to evaluate the efficacy and safety of icotinib based regimens in treating patients with non-small cell lung cancer (NSCLC). Methods: Clinical studies evaluating the efficacy and safety of icotinib-based regimens with regard to response and safety for patients with NSCLC were identified using a predefined search strategy. Pooled response rates of treatment were calculated. Results: With icotinib-based regimens, 7 clinical studies which including 5,985 Chinese patients with NSCLC were considered eligible for inclusion. The pooled analysis suggested that, in all patients, the positive reponse rate was 30.1% (1,803/5,985) with icotinib-based regimens. Mild skin itching, rashes and diarrhea were the main side effects. No grade III or IV renal or liver toxicity was observed. No treatment-related death occurred in patients treated with icotinib-based regimens. Conclusions: This evidence based analysis suggests that icotinib based regimens are associated with mild response rate and acceptable toxicity for treating Chinese patients with NSCLC.

      • Comprehensive Bioinformation Analysis of the MRNA Profile of Fascin Knockdown in Esophageal Squamous Cell Carcinoma

        Background: Fascin, an actin-bundling protein forming actin bundles including filopodia and stress fibers, is overexpressed in multiple human epithelial cancers including esophageal squamous cell carcinoma (ESCC). Previously we conducted a microarray experiment to analyze fascin knockdown by RNAi in ESCC. Method: In this study, the differentially expressed genes from mRNA expression profilomg of fascin knockdown were analyzed by multiple bioinformatics methods for a comprehensive understanding of the role of fascin. Results: Gene Ontology enrichment found terms associated with cytoskeleton organization, including cell adhesion, actin filament binding and actin cytoskeleton, which might be related to fascin function. Except GO categories, the differentially expressed genes were annotated by 45 functional categories from the Functional Annotation Chart of DAVID. Subpathway analysis showed thirty-nine pathways were disturbed by the differentially expressed genes, providing more detailed information than traditional pathway enrichment analysis. Two subpathways derivated from regulation of the actin cytoskeleton were shown. Promoter analysis results indicated distinguishing sequence patterns and transcription factors in response to the co-expression of downregulated or upregulated differentially expressed genes. MNB1A, c-ETS, GATA2 and Prrx2 potentially regulate the transcription of the downregulated gene set, while Arnt-Ahr, ZNF42, Ubx and TCF11-MafG might co-regulate the upregulated genes. Conclusions: This multiple bioinformatic analysis helps provide a comprehensive understanding of the roles of fascin after its knockdown in ESCC.

      • The Blood Neutrophil-to-lymphocyte Ratio Predicts Survival in Patients with Advanced Hepatocellular Carcinoma Receiving Sorafenib

        Background and Aim: Increasing evidence correlates the presence of systemic inflammation with poor survival in patients with hepatocellular carcinoma (HCC). The aim of this study was to investigate the prognostic significance of the blood neutrophil-to-lymphocyte ratio (NLR) in patients with advanced HCC who received sorafenib monotherapy. Methods: A total of sixty-five patients with advanced HCC, not eligible for locoregional therapy, treated with sorafenib were enrolled. Potential prognostic factors such as age, gender, tumoral characteristics, performance status and NLR were analyzed. Results: Median OS and TTP for the entire cohort were 10.0 months (95%CI, 7.6-12.3 months) and 4.5 months (95% CI, 4.0-4.9 months). The mean NLR at baseline was 2.89. The median OS of patients with a high NLR (>4) was 6.5 months (95%CI, 5.2-7.7 months) compared with 12.5 months (95%CI, 9.9-15.0) for patients with a normal NLR (${\leq}4$) (P=0.01). Age ${\leq}65$, NLR>4, extrahepatic metastases and vascular invasion were all predictors of poorer overall survival. Multivariate analysis showed that NLR > 4, vascular invasion and extrahepatic metastases were independent predictors of poorer overall survival. The median TTP of patients with a high NLR was 2.5 months (95%CI, 1.4-3.6 months) compared with 4.5 months (95%CI, 3.9-5.1 months) for patients with a normal NLR (P=0.012). Conclusions: High baseline NLR was associated with worse OS and TTP for patients with advanced HCC treated with sorafenib.

      • miR-124 Inhibits Growth and Invasion of Gastric Cancer by Targeting ROCK1

        MicroRNAs (miRNAs) act as critical regulators of genes involved in many biological processes. Aberrant alteration of miRNAs have been found in many cancers, including gastric cancer (GC), but the molecular mechanisms are not well understood. Herein, we investigated the role of miR-124 in GC. We found that its expression was significantly reduced in both GC tissue samples and cell lines. Forced expression of miR-124 suppressed GC cell proliferation, migration, and invasion. Furthermore, the Rho-associated protein kinase (ROCK1) was identified as a direct target of miR-124 in GC cells. Finally, silencing of ROCK1 showed similar effects as miR-124 overexpression, while supplementation of ROCK1 remarkably restored the cell growth and invasion inhibited by miR-124. Together, our data demonstrate that miR-124 acts as a tumor suppressor by targeting ROCK1, and posit miR-124 as a novel strategy for GC treatment.

        Metastasis associated genomic aberrations in stage II rectal cancer

        Genomic aberrations of rectal carcinoma, especially DNA copy number changes associated with metastasis were largely unclear. We aim to identify the metastasis associated biomarkers in stage II rectal cancer. Formalin-fixed, paraffin-embedded primary tumor tissues of stage II rectal carcinoma were analyzed by array-based comparative genomic hybridization, and genomic aberrations were identified by Genomic Workbench and SAM software. Copy number changes and mRNA expressions were validated by Real-time PCR in an independent rectal cancer samples. The results showed that the most frequent gains in stage II rectal cancer were at 1q21.2-q23.1, 3p21.31, 11q12.2-q23.3, 12q24.11-q24.31, 12q13.11-q14.1 and losses in 18q11.2-q23, 17q21.33-q22, 13q31.1-q31.3, 21q21.1-q21.3, 8p23.3-p23.1 and 4q22.1-q23. Twenty-two amplifications and five homozygous deletions were also identified. We further found that S100A1 (1q21.3-q23.1), MCM7 (7q22.1) and JUND (19p13.11) were amplified and overexpressed in stage II rectal cancer. Interestingly, the genomic aberrations affected 14 signaling pathways including VEGF signaling pathway and fatty acid metabolism. Most importantly, loss of 13q31.1-q34 and gain of 1q44 were associated with distant metastasis. Our results indicated that these metastasis associated genomic changes may be useful to reveal the pathogenesis of rectal cancer metastasis and identify candidate biomarkers.

        A Design Approach for the Interior Anchorage Zone of Post-tensioned Concrete Structure

        For the interior anchorage zone in post-tensioned concrete structure, International Federation for Presterssing (FIP) proposed a design method based on the theory of Strut-and-Tie Model (STM) (called FIP-STM in the following context). However, the condition of FIP-STM is that the dimension ratio of anchor, b/h, is equal to 1/9. In this paper, the stress distribution in interior anchorage zone was analyzed via using Finite Element Method (FEM) at first. The relationship between ratio of internal force over post-tensioned load (F/P) and dimension ratio (b/h) (F/P-b/h curve) was obtained. FEM results show that in case of b/h<1/9, the interior anchorage zone can be simplified to the core region, in which b/h is equal to 1/9 and FIP-STM is still applicable. However, in case of b/h>1/9, the amount of non-prestressed rebar designed in interior anchorage zone by use of FIP-STM is more than that required, and in some cases it is difficult to allocate non-prestressed rebar in the crowded space. Based on the results of stress analysis, a modified approach for FIP-STM was presented, which can be applied for any dimension ratio. With the proposed modification on FIP-STM, the non-prestressed rebar can be reasonably allocated in the narrow space of interior anchorage zone via modifying the internal forces and geometry of FIP-STM. The analytical result shows that for interior anchorage zone about 30% of non-prestressed rebar can be reduced via using the proposed method compared with that using FIP-STM if b/h is equal to 3/10.

        Association between Toll-Like Receptor 9 -1237T/C Polymorphism and the Susceptibility of Inflammatory Bowel Diseases: A Meta-Analysis

        Purpose: The -1237T/C polymorphism of the Toll-like receptor 9 (TLR9) gene has been implicated in the susceptibility of inflammatorybowel diseases (IBDs), but the results remain conflicting. We further investigated this association via meta-analysis. Materials and Methods: Multiple electronic databases were extensively searched until February, 2015. The strength of associationwas evaluated by calculating the pooled odds ratios (ORs) and 95% confidence intervals (CIs). Results: A total of 2987 cases and 2388 controls from eight studies were analyzed. Overall, association was found between TLR9 -1237T/C polymorphism and the risk of IBDs when all the studies were pooled (recessive model, OR: 1.59, 95% CI: 1.02–2.47, p=0.04; homozygote comparison, OR: 1.62, 95% CI: 1.04–2.52, p=0.03; allele model, OR: 1.13, 95% CI: 1.00–1.27, p=0.05). Stratificationby ethnicity indicated an association between TLR9 -1237T/C polymorphism and IBDs risk in Caucasians (recessive model,OR: 1.59, 95% CI: 1.02–2.47, p=0.04; homozygote comparison, OR: 1.62, 95% CI: 1.04–2.52, p=0.03; allele model, OR: 1.12, 95% CI: 1.00–1.27, p=0.05). When stratified by disease type, significant correlation were only found in the Crohn’s disease subgroup (recessive model, OR: 1.69, 95% CI: 1.05–2.73, p=0.03; homozygote model, OR: 1.74, 95% CI: 1.07–2.82, p=0.02; allele model, OR: 1.15, 95% CI: 1.01–1.32, p=0.04). Conclusion: The present study suggested that the TLR9 -1237T/C polymorphism might act as a risk factor in the development of IBDs, particularly in Caucasians.

        Stability and L2-gain Analysis for Impulsive Delay Systems via Two-sided Looped-functional

        This paper investigates the problems of stability and L2-gain for impulsive systems with time-delay. Inspired by the two-sided looped-functional method, a Lyapunov-like functional is established. Distinct from the previous researches, the information of the whole intervals x(t+k) to x(t) and x(t) to x(tk+1) is completely exploited by the functional. Besides, a more advanced integral inequality is used and several integrals of the state are augmented, which are helpful to reduce the conservativeness. Based on the proposed method, sufficient criteria for stability and L2-gain are obtained. Numerical examples are provided to demonstrate the effect and the advantages of the proposed method.
