노랑초파리 발생과정에서 sta 유전자의 발현 양상에 관한 연구

Drosophila stuvarista(sta) gene encodes the ribosomal protein D-p40, consisting of 270 amino acids. The D-p40 is highly conserved between such evolutionarily distinct organisms as human(63% identity) and hydra(58% identity) and has the effect of Minute phenotype such as other ribosomal proteins. We identified the sta gene in the process of Drosophila cDNA library screening and analyzed the developmental expression pattern of sta gene in the transcription level using in situ hybridization. Each animal stage and tissues of 3rd instar larva and adults were showed ubiquitously expression pattern. These results were similar to other ribosomal proteins that represented Minute phenotype. Also, we expressed the 31kDa recombinant proteins of D-p40 using his-tag expression system in E. coli and purified by affinity chromatography. Immunoblot result of the developmental stages was showedthat strongly detection only the stage of embryo and pupa. This result suggested that the expression of sta gene were regulated in the level of post-translation modification ailed considered that sta gene had to other function during development as well as to function as a core ribosomal component.
노랑초파리 ecdysone 결핍 돌연변이(ecd1ts)에서 기능적 ecdysone수용체 구성분의 발현

In Drosophila, the steroid hormone ecdysone triggers the key regulatory cascades controlling the coordinated changes in the developmental pathway of molting and metamorphosis. Ecdysone action is mediated by heterodimer consisting of the ecdysone receptor(EcR) and ultraspiracle proteins(Usp). Heterodimers of these proteins bind to ecdysone response element and ecdsone to modulate gene transcription. In this study, the transcripts of functional ecdsone receptors and ecdysone response genes(Fbp1 and Uro) in ecdysoneless(ecd-1ts) mutation were analyzed by using a RT-PCR and competitive RT-PCR method. The expression of functional ecdysone receptor components were not correlated with tissue-specific ecdysone response genes(Fbp1 and Uro), Suggesting that several other factors involved in the complexity of various ecdysone responses in Drosphila melanogaster. But we showed that the transcripts of EcR-B1 isoforms were expressed with higher level than those of EcR-A in the late third instar larvae of ecdysoneless(ecd-1ts) mutation.

Chitosan is the deacetylated product of chitin. Chitosan and its derivatives have many properties that make them attractive for a wide variety of health applications. We reported recently that water soluble chitooligosaccharides have hepatoprotective effect on ethanol induced liver injury in the mouse. The current study examined the effect of water soluble chitooligosaccharides on CC1₄-induced liver injury in rats. Male Sprague-Dawley rats were orally treated with CC1₄without or with administration of 5% chitooligosaccharides. After 5 weeks after CC1₄treatment, the histological sections and the expression of mRNA of interleukin-12(IL-12), hepatocyte growth factor(HGF), and tumor necrosis factor(TNF- ) were examined. The liver section of rats supplemented with chitooligosaccharides showed attenuated effects of the necrosis, steatosis, cirrhosis compared with those of non-supplemented rats. The level of TNF- mRNA in CC1₄-treated rats were higher than those in the rats supplemented with chitooligosaccharides, while the levels of IL-12 and HGF mRNA were higher in the rats supplemented with chitooligosaccharides than those of in CC1₄-treated rats. These findings suggested that water soluble chitooligosaccharides may have a hepatoprotective activity against CCl₄-induced liver injury in rats.

목적: The aim of present study was to evaluate the perinatal outcomes of selective intrauterine growth restriction (sIUGR) and non-selective IUGR (non-sIUGR) in twin pregnancy. 방법: This was a retrospective study of twin pregnancies from 2007 to 2014. A total number of 385 pairs of twin born between 2007 and 2014 at Ajou University Hospital were enrolled in the study. According to the birth weight, the 385 pairs of twins were divided into sIUGR, non-sIUGR, and normal growth groups. Maternal characteristics and perinatal outcomes were compared among three groups. 결과: Among 385 twin pregnancies, 80 (20.8%) were sIUGR, 15 (3.9%) were non-sIUGR, and 290 (75.3%) were normal growth group. There was no significant difference between IUGR and sIUGR groups according to chorionicity (p=0.417). Vaginal delivery was significantly higher in non-sIUGR group compared to sIUGR and normal growth groups (46.7% vs. 16.2% vs. 14.5%, p=0.014). The incidence of fetal distress and chorioamnionitis were higher in the non-sIUGR group than in sIUGR and normal growth groups (fetal distress: 20.0% vs. 2.5% vs. 1.4% , p<0.001, chorioamnionitis: 53.0% vs. 30.0% vs. 21.7%, p=0.004). Maternal preeclampsia was higher in IUGR group (sIUGR and non-sIUGR) than in normal growth group (18.8% vs. 26.7% vs. 9.3%, p=0.004). With respect to perinatal outcomes, perinatal death was higher in sIUGR group than in normal growth group (10.6% vs. 4.8%, p=0.006). The incidence of poor perinatal outcome (any of the followings: NEC, BPD, RDS, and brain hemorrhage) was higher in both s-IUGR and non-sIUGR groups than in normal growth group (17.6% vs. 23.3% vs. 11.3%, p=0.007). 결론: There was significant difference between sIUGR and non-sIUGR groups in terms of vaginal delivery, fetal distress, and chorioamnionitis. Poor perinatal outcome was higher in both non-sIUGR and sIUGR groups compared to normal growth group.

목적: The purpose of this study was to evaluate the predicting model for lymph node metastasis using preoperative tumor grade, transvaginal ultrasound (TVS), and serum CA-125 level in patients with endometrial cancer. 방법: Between January 2000 and February 2013, we identified 172 consecutive patients with surgically staged endometrial cancer. TVS was performed by an expert gynecologic radiologist in all patients. All patients had complete staging surgery including bilateral pelvic and para-aortic lymphadenectomy and were staged according to the 2009 FIGO classification. Various clinico-pathological data were obtained from medical records and retrospectively analyzed. 결과: Of 172 patients, 138 patients presented with stage I (118 IA, 20 IB), 12 had stage II, 18 had stage III (2 IIIA, 1 IIIB, 8 IIIC1, 7 IIIC2), and 2 had stage IV diseases. The majority of patients had endometrioid adenocarcinoma (88.4%) and others (12.6%) had non- endometrioid histology. Eighteen patients (10.5%) were found to have lymph node metastasis. Deep (≥50%) myometrial invasion on preoperative TVS, high serum CA-125 level (≥ 35 IU/mL), preoperative grade 3 tumors were significant preoperative factors predicting lymph node metastasis. There was no significant association between preoperative histology and lymph node metastasis. We calculated the simple model predicting lymph node metastasis based on preoperative tumor grade, TVS findings, and CA-125 level using logistic regression analysis. With the cut-off point of 1.5, the sensitivity and specificity of this model were 94% and 57%, respectively (AUC = 0.84, [95% CI, 0.74-0.93], p< 0.01). 결론: Preoperative tumor grade, myometrial invasion on preoperative TVS, and CA-125 can accurately predict lymph node metastasis in endometrial cancer patients. The current study suggests the possibility that TVS could be positively used for preoperative evaluation strategy in the low-resource countries instead of expensive imaging modalities such as MRI or PET-CT.
Expression of rpr, grim, dcp-1, diapl, and diap2 during Drosophila Development

초파리 발생과정에서 세포사멸에 관여하는 유전자인 reaper(rpr), grim, dcp-1, diap1, diap1, diap2의 발현양상을 경제적 RT-PCR 방법으로 분석하였다. 세포사멸 유도 유전자인 rpr, grim의 발현양상은 발생단계에 따른 ecdysone titer 변화 양상과 매우 유사하였다. Effector caspase인 dcp-1 전사체는 초기 배와 암컷 성체에서 높은 발현을 보였다. 반면에 세포사멸 억제인자인 diap1과 diap The developmental profiles of rpr, grim, dcp-1, diapl, diap2 transcripts, which were involved in programmed cell death, were analyzed using competitive RT-PCR in whole animals during Drosophila development. The fluctuation patterns of transcript levels of the apoptotic initiators(rpr and grim) were similar to those of the ecdysone titer in Drosophila life cycle. The transcript of dcp-1, which is considered as effector caspase, was expressed strong1y at early embryo and female adult stages. However, the transcript levels of anti-apoptotic factors diap1 and diap2, showed the reverse pattern comparing with those of apoptotic factors(rpr and grim). Also, the transcript levels of rpr, diap2 and dcp-1 were quantified in the salivary glands and wing discs dissected from the wandering late third instar larva. The transcript levels of rpr and diap2 were changed reversely each other in both tissues from wandering stage to puparium formation. These results suggest that the expressions of cell death related genes are regulated by the ecdysone signals during normal development.
