      • Direct Reprogramming of Fibroblasts into Neural Stem Cells by Defined Factors

        Recent studies have shown that defined sets of transcription factors can directly reprogram differentiated somatic cells to a different differentiated cell type without passing through a pluripotent state, but the restricted proliferative and lineage potential of the resulting cells limits the scope of their potential applications. Here we show that a combination of transcription factors (Brn4/Pou3f4, Sox2, Klf4, c-Myc, plus E47/Tcf3) induces mouse fibroblasts to directly acquire a neural stem cell identity-which we term as induced neural stem cells (iNSCs). Direct reprogramming of fibroblasts into iNSCs is a gradual process in which the donor transcriptional program is silenced over time. iNSCs exhibit cell morphology, gene expression, epigenetic features, differentiation potential, and self-renewing capacity, as well as in vitro and in vivo functionality similar to those of wild-type NSCs. We conclude that differentiated cells can be reprogrammed directly into specific somatic stem cell types by defined sets of specific transcription factors.

        Infra-colic gastrostomy: Technique and anatomical considerations

        Background: Infra-colic radiologically inserted gastrostomy is not well documented, and the presence of an insertion window solely below the transverse colon is generally regarded as a contraindication to gastrostomy insertion. A perceived increased risk is due to the presence of vessels and lymphatics within the omental and peritoneal structures, such as the epiploic arteries, the arterial arcade of Barkow, and middle colic artery branches from the superior mesenteric artery. Colonic obstruction is also an additional theorised risk. We provide evidence that infra-colic insertion of a feeding tube through the greater omentum can be performed safely. Methods: A total of 1,156 gastrostomies were inserted over an 8-year period. A retrospective review of the 5 cases was conducted. Electronic patient records were reviewed including clinical consultations, procedure reports and images. Results: In all cases, barium was administered orally/per nasogastric tube the day before to delineate the colon. All patients underwent sedo-analgesia with insufflation of the stomach achieved by a temporising nasogastric/orogastric tube. Infra-colic gastropexy with three SafeTpexy T-fasteners was undertaken. Standard 12 Fr balloon retained tubes were inserted through the greater omentum with no post-procedural complications or tube malfunctions in four cases. A 16 Fr disc retained tube was inserted in a fifth case. Conclusion: Despite the perceived difficulties, we suggest that infra-colic gastrostomies can be performed with confidence, and with little deviation from standard insertion techniques. They can be inserted without an apparent increase in complications, although operators need to be aware of the anatomical differences and additional structures traversed when performing infra-colic gastrostomies.

        Nanotribological behavior of diamond surfaces using molecular dynamics with fractal theory and experiments

        The frictional and indented behavior of a diamond asperity on a diamond plate was carried out using a molecular dynamics (MD) and experiments. The contact load, contact area, dynamic frictional force,and dynamic frictional coefficient increased as the contact interference increased at a constant loading velocity. The microcontact and frictional behavior can be evaluated between a rigid smooth hemisphere to a deformable rough flat plane by combined the deformed behavior of the asperity obtained from MD results with the fractal and statistic parameters. The comparison and the discrepancy of simulated results and nanoindentation and scratching experimental results will be discussed.

        Collision Tumor Composed of Meningioma and Cavernoma

        A true collision tumor is a rare entity composed of two histologically distinct neoplasms coinciding in the same organ. This paper reports a unique case of cerebral collision tumor consisting of two benign components. On the first hand, meningioma which is usually a benign lesion arising from the meningothelial cell in the arachnoidal membrane. On the other, cerebral cavernoma which is a well-circumscribed, benign vascular hamartoma within the brain. To our knowledge, there is no previously documented case of cerebral collision tumor consisting of two benign components. A 56-year-old Caucasian male suffered in 2002 from an atypical meningioma WHO $II^{\circ}$ located in the left lateral ventricle. Three years after the tumor extirpation, the patient suffered from a hematoma in the fourth ventricle due to a recurrently haemorrhaged cavernoma. In 2008, a recurrence of the tumor in the left lateral ventricle was discovered. Additionally, another tumor located in the quadrigeminal lamina was detected. After surgical resection of the tumor in the left lateral ventricle, the pathological examination confirmed the diagnosis of a collision tumor consisting of components of a meningioma WHO $II^{\circ}$ and a cavernoma. Postoperatively, no adjuvant treatment was needed and no tumor recurrence is discovered up to the present. A possible explanation for the collision of those two different tumors may be migration of tumor cells mediated by the cerebrospinal fluid. After 5-years of follow-up, there is no sign of any tumor recurrence; therefore, surgical tumor removal without adjuvant therapy seems to be the treatment of choice.

        Predatory efficacy of Orius strigicollis (Hemiptera: Anthocoridae) against Tetranychus urticae (Acarina: Tetranychidae) on strawberry

        In this study, the predatory efficacy of Orius strigicollis (Poppius), a predacious flower bug, against the twospotted spider mite (TSSM), Tetranychus urticae Koch, on strawberry (Fragaria ananassa) was investigated. Our results showed that release of 2, 4, 6, and 12 neonates of O. strigicollis per strawberry plant could suppress 58.9, 72.6, 88.6, and 92.8% of T. urticae, respectively, at 14 days after treatment. A single release of 12 neonate bugs per plant achieved and maintained, over a period of 3 months, 82% to 98% control of T. urticae population. A biweekly release of 6 neonate bugs per plant; likewise, suppressed ca. 90% of T. urticae population throughout the same period. However, predatory efficacy was reduced by an over-release of predators which may cause cannibalism as well as lower possibility of finding prey. Since the released neonates survived well to adults with offspring that retained their predatory ability, this study has demonstrated that O. strigicollis is a predatorwith good potential for sustainable suppression of T. urticae on strawberry, and one application of O. strigicollis neonates at a predator:prey ratio of 1:200 egg plus motile TSSM or 1:60 motiles per strawberry plant promises a season-long control and substantial management of spider mite infestation.

        Antioxidation and Antiglycation of 95% Ethanolic Extracts Prepared from the Leaves of Black Nightshade (Solanum nigrum)

        Hyperglycemia results in the formation of advanced glycation end-products (AGE), resulting in an inflammatory response that induces insulin resistance. Evidence indicates that antioxidants can suppress the formation of reactive oxygen species, decrease levels of AGEs by inhibiting glycation. Black nightshade (Solanum nigrum) can be used as a medicinal food for improving blood glucose; however, the identities of the active compounds and how they counteract diabetes remain unknown. This study demonstrate that 95% ethanolic extracts of black nightshade exerted significant antioxidative activity compared with 50% ethanolic extracts and aqueous extracts. Moreover,95% ethanolic extracts of black nightshade produced antiglycative activity, which contributed to the inhibition of fructosamine and generation of α-dicarbonyl compounds. The concentrations of solasonine and solamargine in the 95% ethanolic extracts were 0.484 and 0.183 mg/mg,respectively. These results suggest that black nightshade might serve as a novel source of functional ingredients that exert antiglycation and anti-diabetes activities.

      • Numerical Simulation of Methane Slip in Dual Fuel Marine Engines

        The methane slip is the problematic issue for the engines using natural gas(NG). Because methane is more powerful greenhouse gas (GHG) than CO₂, understanding of the methane slip during gas exchange process of the engines is essential. In this study, the influence of the gas pipe geometry and the valve timings on the methane slip was investigated. MAN L28/32DF engine was modeled to simulate the gas exchange process of the four stroke NG-diesel dual fuel engines. The mesh size of the model was decided based on the sensitivity study on the peak pressure of the cylinder and the fuel mass estimations. The simulations with various gas pipe geometries were conducted. It seemed that the effect of the change in injection direction is more dominant than the change in the gas hole configuration. The favorable injection direction for minimum amount of methane slip was discovered as the direction which helps developing the flow of methane far from the exhaust ports. The effects of various valve tuning settings were also simulated. The advancement of the exhaust valve closing was more efficient than the retardation of the intake valve opening. A little retardation of the intake valve opening even resulted in the increase of the amount of methane slip.
