        진단병리학 분야에서의 전자현미경의 이용

        The eletron microscope has become an established and indispensable tool in diagnostic pathology. There is an increasing demand of ultrastructural studies for the cases that pathologists in making definite diagnosis. In this paper the author described representative cases that were experienced at the Seoul National University Hospital since 1985. Characteristic ultrastructural features of these diseases are presented along with electron micrographs. Among diseases involving parenchymal cells, several examples of abnormalities of organelles, metabolic disturbance, hair and ciliary anomaly and red cell anomaly are described. Among infectious agents 4 viruses, 2 fungi, 2 protozoa and 2 metazoa are described.

      • 중첩 방전을 이용한 CO₂레이저의 동작 특성 : 다중펄스 중첩을 이용한 long 펄스형 CO₂레이저의 펄스 형상 제어 Pulse shape control of pulse CO₂laser using multiple-pulse superposition

        본 연구에서는 PIC 원 칩 마이크로 프로세서 기술을 접목시켜, 저전압 측에서 2단 또는 3단의 회로를 능동적으로 중첩시킴으로써, 다양한 펄스폭을 가지는 고전압의 펄스형상을 만들었다. 이렇게 만들어진 다양한 중첩펄스를 현재 산업용 및 의료용으로 많이 사용되고 있는 CO2 레이저에 적용하여 각 단의 스위치의 트리거 지연시간에 따른 레이저 빔의 출력 특성을 고찰하였다. 그 결과 다양한 펄스폭을 가지는 레이저 빔을 얻을수가 있었고 특히, 3단 중첩회로에서 스위칭 지연 시간을 변화시킴에 따라 얻어진 레이저 펄스폭은 최소 약 4ms였고 최대 약 10ms였다. In this study, a variable pulse length, high duty cycle Pulse Forming Network(PFN) is constructed by time sequence. The power supply for this experiment consists of three switching circuits. Each switching circuit uses 2 mesh PFN, a charging circuit, a HV leakage transformer, and bridge rectifier on each transformer secondary. The PFN elements operate at low voltage and drive the primary of HV leakage transformer. The secondary of the transformer has a full-wave rectifier, which passes the pulse energy to the load in a continuous sequence. We investigated laser pulse width of pulsed CO2 laser as various delay time among three switching circuits. As a result, we can obtain various laser pulse width from about 4 to 10ms.

      • 광주 신창동유적 : 제1차조사개보

        이 글은 국립광주박물관이 광주시의 의뢰를 받아 실시한 광주 신창동 소재 유적에 대한 조사 약보(略報)이다. 유적은 광주-장성을 잇는 국도확포장구간에 포함되었고 공사의 진척에 따라 유적 일부가 잘려나가는 상황에 놓이게 되었다. 특히 유적 일대의 충적 저지대에서는 공사로 인해 패인 지표하 2m 정도의 깊이에 있는 황갈색 사질층과 흑갈색 진흙층에서 볍씨와 탄화미(炭化米) 등이 곡물자료와 함께 일부 가공흔이 나타나는 목재류 등이 채집되었다. 이에 광주박물관은 광주시 등 관계기관과 협의하여 1차로 공사구간에 대한 유구확인을 위한 발굴조사를 실시하게 되었다. 조사는 1992년 6월 26일부터 7월 25일까지 이루어졌다. 그 결과 국내에서는 처음으로 목제 농기구가 출토된 소택지(沼澤地)의 존재가 알려졌으며 이와 함께 초기철기시대에서 원삼국시대에 이르는 것으로 생각되는 토기요지·구상유구(溝狀遺構)·조상유구(條狀遺構)·주거지 등이 확인되었다. 소위 소택지에서는 흑도(黑陶)·고배(高杯)·점토대토기(粘土帶土器) 등 각종 토기류와 함께 칠기고배, 빗(櫛), 검파(劍把), 통발 등 다양한 목제유물과 볍씨 등 탄화미를 비롯한 곡물자료가 발견되었다. 특히 칠기고배를 비롯한 칠기 등과 목제유물은 지금까지 창원 다호리를 비롯한 영남지방에서는 드물게나마 출토된 바 있으나 한반도 서남부지역에서는 초출(初出)의 자료로서 시사되는 바 크다. 또한, 주거지를 제외한 각 유구에서 출토된 토기류의 형태적 특징에서 요지 출토품과 유사성이 인정됨으로써 당시의 생산과 유통에 관한 새로운 인식의 계기를 마련해 주고 있다. 이러한 물질자료들은 유적의 극히 일부에서 출토된 것이지만 아직 밝혀지지 않은 이 시기의 생산양식과 농경문화의 구체적인 실체에 접근하는 데 필요한 하나의 수확이며 고환경의 복원 및 문화발전과 전파문제의 해석에도 많은 도움이 될 것이다. The Shinchang-dong site in Gwangju, Jeollanam-do Province, was excavated by the Gwangju National Museum in 1992, prior to the construction of the national road. The excavation revealed a peat bog, a pottery kiln, a ditch-shaped feature, a dry field-like feature and a dwelling house. Among the materials recovered from the peat bog were 1) lacquer wares and wooden artefacts including a hoe, mounted dishes, a comb, a dagger handle, basketry fish traps, bird's head-shaped scuplture, and various tools for woodworking, 2) potteries including plain-coarse pottery, black-burnished pottery, and pottery with clay stripe along the rim, the shapes of which are mounted dishes, long-necked jars, wide-mouthed bowls, dish-shaped covers and steamers, 3) organic remains including grains and seeds of rice, barley, wheats, cucumber, legumes and apricots, walnut shells, fish bones and snails, 4) accumulations of large quantities of rice husks. Many wooden pieces, planks and tools with some indications of woodworking as well as the existence of posts for hanging wooden implements indicate the use of swamp as the place for woodworking. The pottery kiln was dug in the direction of contour. Its size is 8m in length by 2m in width, and the slopeness of the floor is 5~8°. It is a open kiln without any additional structure such as roof, wall and combustion chamber. The potteries found from the kiln are similar to those from the bog site. The floor plan of dwelling site is a rectangle about 430cm by 320cm, in the depth of 20cm. It has a central hearth in an oval shape, 50cm in diameter. The peat bog, the kiln and other features date to about the late 2nd century B.C. or the 1st century B.C. while the dwelling site dates to the 3rd century AD. Recognizing the importance of the site, it is designated as the NATIONAL HISTORIC SITE NO. 375.

        The objective of this present study is to solve problems in PC tile pre-fixing method using ordinary concrete by adopting the liquefaction method in manufacturing PC to which the tile pre-fixing method is applied, and to provide basic information for the practical use of the PC tile pre-fixing method, which uses high workable concrete, through manufacturing a complex body based on the optimal mix proportion.

      • 태아 피부 에크린한선의 발육에 관한 전자현미경적 연구

        To observe the developing process of human fetal eccrine sweat gland during the intrauterine life, an electron microscopic study was carried out based on 12 fetuses ranging from 20 to 42 weeks of gestation. These fetuses were the products of induced deliveries found to have not associated disease or congenital anomalies at the autopsy. Following observations were made. The various types of cell consisting coiled portion of eccrine sweat gland were recognized at the 20th weed of gestation and they were clear cells, undifferentiated dard cells, myoepithelial cells. clear cells were located in central central portion and dark or myoepithelial cells were in peripheral portion. While undifferentiated dark cells were randomly distributed in the straight portion of eccrine sweat gland. The differential points between clear cell and undifferentiated dark cell were its round nucleus and glycogen contents. Marked hydropic changes of clear cells were noted at the 30th week of gestation. Secretory granules were noted at the 31st week of gestation. Secretory cells in the coiled portion of eccrine sweat gland was not fully mature until the 42nd week of gestation. Clear cells showed clear cytoplasm, abundant free ribosomes and scanty organelles. Undifferentiated dark cells showed many mitochondria and few glycogen. Secretory granules were rarely seen. Myoepithelial cells showed a few mitochondria, glycogen and microfilaments. Among three portions of eccrine gland, duct lumen in the acral and coiled portion were formed through the intracellular canaliculus and in the straight portion were made through the intercellular canaliculus. The main basic morphological mechanism of the lumen formation were microvesicle formation and pinched off phenomenon.

      • 들잔디 성숙종자로부터 캘러스배양 및 식물체 재분화에 미치는 몇 가지 요인의 영향

        In an effort to optimize tissue culture responses of zoysiagrass (Zoysia japonica Steud.) for genetic transformation, factors affecting callus induction and plant regeneration were investigated. MS medium containing 3 ㎎/L 2,4-D was optimal for embryogenic callus induction from mature seed. The plant regeneration frequency of 73.3% was observed when embryogenic calli induced in this medium were transferred to N6 medium supplemented with 0.1 ㎎/L 2,4-D and 5 ㎎/L BA. Among several basic media, MS and N6 medium were optimal for callus induction and plant regeneration, respectively. Regenerated plants were grown normally when shoots transplanted to the soil. A rapid and efficient plant regeneration system established in this study will be useful for molecular breeding of turfgrass through genetic transformation.

        급성 림프구성 백혈병 환자에서 발생한 침습성 국균 심내막염 2예

        국균은 호흡기를 통해 유입되며 심한 면역기능저하 환자에서 기회감염을 일으키는 진균이며 주로 혈관을 따라 폐를 침범하고 심장과 골침범은 매우 드문 것으로 알려져 있다. 저자들은 급성 림프구성 백혈병 환자에서 발생한 침습성 국균 심내막염을 2예을 경험하였다. 19세 남자환자로 동종 조혈모세포이식 후 발생한 고열과 요통의 원인으로 감염성 심내막염 진단하에 수술결과 국균에 의한 감염으로 확증되어 수술적 절제술과 liposomal amphotericin B로 치료하였으나 국균 심내막염의 재발과 국균 척추염의 악화, 뇌 출혈로 사망하였다. 23세 남자환자로 관해요법 후 발생한 고열과 요통의 원인으로 국균 심내막염과 국균 척추염 진단하에 수술적 절제술과 amphotericin B로 치료하여 1년 2개월 동안 국균 심내막염의 재발은 없었으나 지속적인 백혈병의 재발과 국균 척추염의 악화, 침습성 폐 국균증으로 사망하였다. Although Aspergillus endocarditis has rarely been reported, it can cause fatal complications in hematologic malignancy patients and allogeneic stem cell transplant recipients. We experienced two cases of aspergillus endocarditis developed in acute lymphoblastic leukemia patients. Case; A 19-year-old patient developed Aspergillus endocarditis after allogenic hemopoietic stem cell transplantation. He was treated with surgical intervention and liposomal amphotericin B. He died of recurred Aspergillus endocarditis and cerebral hemorrhage probably related with aspergillosis of central nervous system. Case 2; A 23-year-old patient developed invasive Aspergillus endocarditis after induction chemotherapy. Aspergillus endocarditis was successfully treated by surgical intervention and amphotericin B. He died of refractory neutropenic fever and sepsis after the third relapse of leukemia and repetitive chemotherapy. He probably had invasive pulmonary aspergillosis without evidence of endocarditis recurrence. Because the mortality of Aspergillus endocarditis is very high, early diagnosis and surgical intervention are very important for better outcome.

      • 국내육성 오차드그라스 품종들의 캘러스 형성율 및 식물체 재분화 효율

        Comparisons of callus formation ratios from seed explants, callus sizes, regeneration ratios from callus and regeneration efficiency for 4 orchardgrass (Dactylis glomerata L.) varieties (Three were developed in Korea and one was imported from foreign nation) are as follows; Jangbeol 102 (67.0%) has the highest callus formation ratio in 4 weeks incubated callus after bedding the seed explants, but Potomac (68.4%) has the highest ratio in 6-seek callus. Potomac (3.93cm) has the highest callus size in 4-week callus, but Jangbeol 101 (4.32cm) has the highest size in 6-week callus. Jangbeol 101 (17.7%) has the highest plant regeneration ratio in 4-week callus, but Potomac (37.4%) has the highest raion in 6-week callus. Jangbeol 102 (11.5%) has the highest plant regeneration efficiency in 4-week callus, but Potomac (25.6%) has the highest efficiency in 6-week callus.

        Electric current control of creation and annihilation of sub-100 nm magnetic bubbles examined by full-field transmission soft X-ray microscopy

        <P>The effect of electric current pulses on a sub-100 nm magnetic bubble state in a symmetric Pt/Co multilayer was directly observed using a full-field transmission soft X-ray microscope (MTXM). Field-induced evolution of the magnetic stripe domains into isolated bubbles with their sizes down to 100 nm was imaged under varying external magnetic fields. Electric current pulses were then applied to the created magnetic bubbles, and it was observed that the bubbles could be either created or annihilated by the current pulse depending on the strength of applied magnetic field. The results suggest that the Joule heating plays a critical role in the formation and/or elimination of the bubbles and skyrmions. Finally, the schematic phase diagram for the creation and annihilation of bubbles is presented, suggesting an optimized scheme with the combination of magnetic field and electric current necessary to utilize skyrmions in the practical devices.</P>
