        형광직접접합법에 의한 인간정자의 연령별 염색체이상에 관한 연구

        N/A Determmation of the chomosomal constitution of human spermatozoa has been camed out though the human-hamster interspecific in vitro fertilization(IVF) system. In recent years, the introduction of fluorescence in-situ hybridization(FISH) technique has provided an alternative approach to evaluate the cbmmosomal constitution of human spermatozoa. The nuclei of mature spermatozoa are highly condensed with interpmtamine disulphide bridges, therefore the success of FISH on interphase human spermatozoa relies on partial decondensation of the sperm chromatin. In early studies, dithioothreitol(DTT) has been known as an efficient decondensation agent. Since then, several different decondensation methods using D1T have been establisdhed, and in terms of decondensation, we were tried to fix the optimal decondensation protocol using DlT. In our study, the optimal concentration and treatment time were 1-mM and 30 min, respectively. We examined chromosome complements of human sperm to investigate the effect of paternal age on the hequency of nondisjunction in human sperm. We investgated sperm karyotypes ftom two diffaent age groups)28+-0.5, 46+-6), A minimum of 1000 spermatozoa for one patient were analyzed. The mean frequencies of YY, XX, XY, 21-disamy spermatozoa ware 0.04%, 0.45%, 0.40%, 0.45% respectively in young age group and 1.06%, 0.62%, 1.06%, 0.76% in old ages. The mean frequency of disomy spermatozoa was higher in old age poup compare with those of young age group.

        Multi-Homologous Recombination-Based Gene Manipulation in the Rice Pathogen Fusarium fujikuroi

        Gene disruption by homologous recombination is widely used to investigate and analyze the function of genes in Fusarium fujikuroi, a fungus that causes bakanae disease and root rot symptoms in rice. To generate gene deletion constructs, the use of conventional cloning methods, which rely on restriction enzymes and ligases, has had limited success due to a lack of unique restriction enzyme sites. Although strategies that avoid the use of restriction enzymes have been employed to overcome this issue, these methods require complicated PCR steps or are frequently inefficient. Here, we introduce a cloning system that utilizes multi-fragment assembly by In-Fusion to generate a gene disruption construct. This method utilizes DNA fragment fusion and requires only one PCR step and one reaction for construction. Using this strategy, a gene disruption construct for Fusarium cyclin C1 (FCC1), which is associated with fumonisin B1 bio-synthesis, was successfully created and used for fungal transformation. In vivo and in vitro experiments using confirmed fcc1 mutants suggest that fumonisin production is closely related to disease symptoms exhibited by F. fujikuroi strain B14. Taken together, this multi-fragment assembly method represents a simpler and a more convenient process for targeted gene disruption in fungi.

        Primary candidate for pancreas transplantation is patients with type Ⅰ diabetes mellitus of juvenile oneset. At present the patients selected for transplantation often have end stage renal disease, far advanced vascular disease and diabetic neuropathy. In these days, the survival of pancreas transplantation for treatments of patients with type Ⅰ diabetes mellitus has been improved especially in simultaneous pancreas and kidney transplantation. Various innovations in immunosuppression, management of the pancreatic ductal exocrine secretion, recipient selection and HLA matching have been contributed to achieve a reasonable outcome in pancreas transplantation compared with other organ transplantation. Current improved graft survival has stimulated many centers in the world to start pancreas transplantation program. Recently we successfully performed the first case of simultaneous pancreas and kidney transplantation in Korea. After 5 cases of pancreas transplantation in our department in type Ⅰ DM, we think that it is the time to review the pancreas transplantation to give an insight of this new field to our colleagues.

        An investigation of the water quality for the streams of Daejon area in Chung-nam province, korea, was conducted from January to December 1981. By this analysis of water quality for the streams of Daejon area, annual mean values of BOD: 49.3㎎/l, COD: 33.4㎎/l, DO: 7.5㎎/l, SS: 39.1㎎/l were determined respectively and those values of parameters were comparatively high. The increasing order of the water pollution for the streams of Daejon area was as follows; confluence place Daewha-Dang (D)> Yudeung Stream (B)> Daejon Stream (A)> Gab Stream (C). Therefore, it suggest that the water pollution of the streams of Daejon area have greatly influenced upon the water quality of Kum River in Chungnam and Jon-buk province.

      • A zeolite membrane module composed of SAPO-34 hollow fibers for use in fluorinated gas enrichment

        <P><B>Abstract</B></P> <P>A membrane module composed of SAPO-34 hollow fibers was constructed for the enrichment of NF<SUB>3</SUB>, a strong greenhouse gas with a 100-year global warming potential (GWP)<ce:cross-ref refid='fn1'> <SUP>1</SUP> </ce:cross-ref> of > 17,000. Growth of the SAPO-34 membrane was investigated on the alumina hollow fibers using in-situ growth and secondary growth methods. Heterogeneous nucleation of SAPO-34 crystal took place on the surface because of hetero-atoms existing on the support, contributing to early development of continuous zeolite film. Zeolite membranes were successfully prepared from both in-situ and secondary growth. The membrane prepared by secondary growth presented N<SUB>2</SUB> permeance as high as 168.4 GPU with an N<SUB>2</SUB>/NF<SUB>3</SUB> gas selectivity of 580.7. A SAPO-34 membrane module was assembled using the hollow fiber membranes produced by secondary growth, but separation performance of the module was reduced to N<SUB>2</SUB> permeance of 136.4 GPU with an N<SUB>2</SUB>/NF<SUB>3</SUB> gas selectivity of 133.7. When used for the enrichment of NF<SUB>3</SUB> gas, a superior yield (98%) and enrichment ratio (20) were obtained from the module at a stage-cut of 0.95.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Thin SAPO-34 membranes (< 2µm) were synthesized for the enrichment of NF<SUB>3</SUB> gas. </LI> <LI> Alumina hollow fibers were used as a porous support for the SAPO-34 membrane. </LI> <LI> Both in-situ growth and secondary growth methods were studied for SAPO-34 membrane. </LI> <LI> A membrane module consisting of SAPO-34 hollow fibers was constructed. </LI> <LI> The module enriched NF<SUB>3</SUB> gas from a N<SUB>2</SUB>/NF<SUB>3</SUB> mixture with high separation efficiency. </LI> </UL> </P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>

        Air-Lubricated tilting pad journal bearing that has high stability is analyzed by using the Direct analysis, and this bearing is usually sued to need high precision. The pressure that supports the shaft is occured by the differences between the shaft and pads radii of curvatures. So the characteristics of load capacity for their variable values is important. In this paper the load capacity is compared with some of the eccentricity ratio values of 0.1, 0.2, 0.5, 0.8. The large load capacity comes form large eccentricity ratio, high bearing number and high preload. But if the preload becomes too high, the shaft comes into contact with the pads.

        The purpose of this study was to develop the computer program to compare physical fitness of Hanyang Academy Employes with it's mean of korean people, and to promote physical fitness with motivation and interest in the physical fit- ness of them. But if user should be tested self-level of physical fitness, this program can't be use for physical fitness by assisting tile research institute for sport science on Hanyang University. This program were divided into 4 domains, tile first is area for comparison ac- cording to the way of attendance to office, the second is area for comparison physique and obesity, the third is area for comparison physical fitness with korean people's mean. The final is area for exercise prescription. As added, we want to serve the physical fitness promotion of Hanyang academy employes using this program.

      • 할리우드의 映畵産業 戰略과 韓國映畵産業의 新 進路

        This paper discusses the American movie industry, represented by Hollywood. and its foreign market strategy and its overall impact on the korean movie industry. Especially, this paper focuses on analyzing the current crisis of the Korea movie industry and developing the possible alternatives or new path for its reinvigoration by comparing the Production, distribution, and promotion system between the movie industries of the two countries. This paper argues that the current world dominance of the American movie industry can be attributed to its economies of scale, mass production, windowing effect through the horizontal and vertical integration among the relevant industries, strong marketing and distribution system, successful creation of derivative industries through the utilization of mass communication, and the export push and price differentiation policy of the American government. In contrast, this paper argues, the Korean movie industry is handicapped by insufficient movie infrastructure, emphasis on import of foreign movies rather than on national production of its own movies, meagerness of movie production capital, absence of the cooperation system or structure among the movie-relevant industries, and non-transparency of capital flowing for movie production. Following these arguments, this paper suggests that establishment of the strong infrastructure, creation of its own new image and brand for Korean movies, aggressive and efficient marketing strategy, and appropriate policy support of the government are necessary for the reinvigoration and development of the Korean movie industry.
