        Photoreflectance 측정에 의한 InxGa₁-xAs(0.03≤X≤0.11) 에피층의 특성 연구

        Molecular Beam Epitaxy(MBE)법으로 성장된 In_xGa_(1-x)As/GaAs 에피층에 대해 photoreflectance (PR)실험을 통해 특성을 조사하였다. PR 측정결과 성장된 In_xGa_(1-x)As 에피층의 띠간격 에너지(E。) 신호가 시료의 변형(strain)에 의해 heavy-hole(E。(HH))과 light-hole(E。(LH))로 분리되어 관측되었다. 에피층의 조성과 변형은 각각 시료에서의 E。(HH) 및 Eo(HH)와 Eo(LH)신호의 에너지 차이를 이용하여 구하였다. 또 160 K이하의 온도에서는 Eo(LH)의 신호가 사라짐을 볼 수 있었다. Franz-Keldysh oscillation(FKO) 피크로부터 계산되어진 InGaAs/GaAs 계면전장 (E)은 In 조성의 증가에 따라 0.75×10^5 V/㎝에서 2.66×10^5 V/㎝로 증가하였다. In 조성이 x=0.09인 시료에 대한 PR신호의 온도의존성 실험에서 Varshni 계수와 Bose-Einstein 계수들을 각각 구하였다. Photoreflectance (PR) measurents have been performed on In_xGa_(1-x)As/GaAs grown by molecular beam epitaxy (MBE). Bandgap (E。) of In_xGa_(1-x)As epilayer measured from PR was separated as heavy-hole (E。(HH)) and light-hole (E。(LH)) by strain effect. The compositions and the strains of epilayer were obtained from the energy value of E。(HH) and from enegy difference of E。(HH) and E。(LH), respectively. In addition, the PR signal of E。(LH) was diminished below 160 K. The interface electric field (E) of InGaAs/GaAs was increased from 0.75×10^5 V/㎝ to 2.66×10^5 V/㎝ as In composition increased, which was calculated from Franz-Keldysh oscillation (FKO) peaks. As the temperature dependence of the PR signal at x=0.09 sample, we obtained Varshni and Bose-Einstein coefficients.

        치아회분과 석고혼합제재 매식과 자가골 동시 이식후 치유과정에 관한 실험적 연구

        The purpose of this study was to evaluate the efficacy of adding autogenous bone to the toothash-plaster mixture in the healing process of bone. Full-thickness round osseous defects with the diameter of 20mm were made at the calvarial bone of adult dogs (n=19) bilaterally, which were thought to be critical size defect. The right defects were repaired with the toothash-plaster mixture plus autogenous bone (compressed volume 0.3cc) and the left defects with only toothash-plaster mixture. At 2-, 4-, 8-, 12- and 20- week after implantation, dogs were sacrificed and evaluated the osseous healing of bony defects clinically, radiographically, and microscopically. The results were as follows : 1. At the clinical observation, the wound healed very well without any problem except severe swelling in the early period after operation. Slight depression was recognized at the both sides when the portions of cranial defect were palpated. 2. There were statistically significant differences between toothash-plaster mixture groups and autogenous bone added groups at the same period, and among the groups in the bone density of the digital radiograms (P<0.001).There was a tendency that bone density was increasing with time. 3. In light microscopic examination, new bone formation was more active in the autogenous bone added groups than toothash-plaster mixture groups at the early period after mplantation but there is little difference at 20-week after implantation. 4. In fluorescent microscopic examination, the fluorescent band could be observed at the area of active bone formation and the band was more distinct in the autogenous bone added groups then toothash-plaster mixture groups. 5. In transmitted electron microscopic examination, organelles such as rER, Golgi complex and secretory granule and osteoblast were observed. In summary higher volume ratio of autogenous bone is needed to improve the bone healing in that there is little difference between toothash-plaster mixture group and autogenous bone added group at the 20-week after implantation in spite of new bone formation was more active in the autogenous bone added groups than toothash-plaster mixture groups at the early period after operation.

        In Vitro 고삼투압이 정자 원형질막의 Protein Tyrosine Phosphorylation에 미치는 영향

        정자의 원형질막은 삼투압에 의해서 영향을 받는다고 보고되고 있다. 이중 세포막내 분자구조의 변화 특히 막지질 구조의 변화와 동반되는 이온채널의 변화 그리고 Ca2+과 HCO-3의 유동성과도 밀접한 관련이 있는 것으로 보고되고 있다. 지금까지의 연구보고에 의하면, 정자의 첨체반응 (acrosome reaction)이 일어날 경우 protein tyrosine phosphorylation이 증가되는데 이것은 cAMP, protein kinase A 등을 통하여 작용되는 것으로 보고되고 있다. 막의 지질변화를 유도하는 물질로 일종의 sterol acceptor인 BSA가 알려져 있는 바, 실제로 BSA가 막지질 성분에 미치는 영향을 관찰한 결과 cholesterol이 유출되고 이온 등의 유동성 변화가 일어나, 이 유동성 변화가 정자의 adenylyl cyclase를 활성화시켜 cAMP를 증가시키고, PKA가 활성화되어 결과적으로 Protein tyosine phosphorycation이 유도된다고 보는 것이다. 침체반응과 protein tyrosine phosphorylation과는 밀접한 관계가 있는 것으로 사료되고 있다. 본 연구는 정자 원형질막에서 cholesterol이 유출되어 protein tyrosine phosphorylation이 유도될 때, BSA와 같은 sterol acceptor가 작용할 것이라는 전제하에, 고삼투압 하에서 탈수로 인해 원형질막이 위축되더라도 sterol acceptor가 존재한다면 막지질 성분의 구조적 변화가 억제 될 수 있을 것이라는 가설을 설정하였다. 실험결과, 저온 및 고삼투압 하에서 정자운동은 감소되지만 원형질막의 구조적 변화는 없고, 삼투압에 대한 반응정도는 원형질막을 통한 수분이동과 세포용적 변화에 따라 비례적으로 일어난다고 하는 사실을 발견하였다. 이 결과는 정자보존에 있어서 저온변화에 영향을 미치는 여러 인자들 특히 protein fgrosine phosphorylation의 증가와 밀접한 관계가 있음을 시시해 준다. 또한 sterol acceptor로 알려진 BSA는 삼투압이 변화되더라도 역시 중요한 인자로 작용할 수 있음을 알 수 있었으며, 특히 고삼투압으로의 변화는 cAMP와 protein kiname A를 거치는 신호전달과정에 있어서 중요한 요인이라는 사실을 확인할 수 있었다. It has been reported that plasma membrane activity of the spermatozoa may be susceptible to be influenced by extracellular osmolality and such membranous changes involve intracellular molecular changes, special regard to the structure of membranous lipids, and the accompanying ion-channel of which are closely related with their fluidity of Ca2+ and HCO-3. It is of common recognition that a certain kind of sterol acceptor player an important to induce lipid fluctuation of the sperm plasma membrane which have been influenced by BSA administration and came in effect to outflow of cholesterol from the spermatozoa and resulted in changes of ionic fluidity to facilitate adenylyl cyclase, and to induce protein tyrosine phosphorylation by increase of cAMP and activation of PKA. Thus it seems likely that an augmentation of the acrosomal reaction is closely related with protein tyrosine phosphorylation. The following experimental results were obtained in the present study; Under the high osmolality conditions, the spermatozoa motility declined significantly and the structural change of the plasma membrane diminished to confirm that the response degrees to the osmolality depended upon the water transfer volume through the plasma membrane and the changes of cellular volume. Those experimental results suggest that a physiological parameter such as low temperature condition played an important role for presentation of spermatozoa and that inducement of spermatozoa activation for reinforcement of protein tyrosine phosphorylation. On the other hand, it seemed likely that the BSA administration as one of sterol acceptors might represent a key role also under the high osmolality condition and their result also suggests that osmolality change, special regard to high osmolality condition may play an important role also in the processes of signal transmission

        Experience of Pain From Patients With Non-Acute Pain After Back Surgery

        Purpose: The aim of this study was to describe and understand the experiences of pain and the perceptions of treatment through a traditional Korean medicine clinical trial in patients with non-acute pain after back surgery. Methods: The data collection was recorded through in-depth interviews with 20 participants and transcribed with verbatim. The data were analyzed with qualitative contents analysis. Results: Patients with persistent or recurring pain after back surgery experienced physical and psychological symptoms that resulted in severe difficulties in daily and working life. Their persistent or recurring pain after surgery was a burden in their life and a major source of depression. Because of the difference in the perception of pain between doctors and patients, the patients continuously sought and received various treatments. The patients also wanted to receive customized self-management in exercise so that they could control pain themselves in daily life. Conclusion: A multidisciplinary approach that includes psychological counseling, pain management, individualized exercise, and education for proper posture should be needed to reduce low back pain in patients with pain after back surgery.

        In the present study, we aimed. to consider the name of twelve main Meridian in order to understand the concept of the Meridian system and to applicate acupuncture theory in the clinical field properly and fundamentally. We referred to several literatures on the matter and we found out that the name of present twelve Meridian was not settled until the period of Yellow Emperor's Classic. In the time of Yellow Emperor's Classic, the two theory of Internal Organ and Meridian system were combined and added new theory of 'WoonGi'. The general rule of combination is that the component of name of Meridian consist of 'Hand' or 'Foot', 'three Eum and Yang' and internal organ. The internal organ coupled with the root ki or middle ki of 'three Eum and Yang' and the 'Hand' or 'Foot' depend on the location of internal organ that belongs to meridian. The name of Twelve Meridian is a kind of system formed to analyze the two concepts organically and dynamically. One is material internal organ and the other is immaterial meridian system.

        This study was focused to find out main effects influencing self citizens's yes or no of participation and levels of sport for all in leisure competence, that is, physical leisure competence, social leisure competence, and cognitive leisure competence and life satisfaction contrasting relationships and effects between participation of sport for all and life satisfaction of most prestudies in past were related to socio-economic variables The Questionnaire about leisure competence and life satisfaction is based on the scale of domestic previous studies using Harter's(1982) and LDB(The Leisure Diagnostic Battery of Witt and Ellis(1982, 1989), and used them after examine validity and reliability through pilot test. The methods of statistics used to analyze the collected dates are covariance analysis, correlation analysis, multiple regression analysis Results obtained from this research are as follows: Firstly, There were exposed to the significant relationship between physical leisure competence(2.42), social leisure competence(2.11) and cognitive leisure competence(2.14) and life satisfaction of participants of sport for all.

        액상정액을 이용한 인공수정시 품종, 계절, 인공수정 횟수 및 정자농도가 번식성적에 미치는 영향

        돼지에서 액상정액을 이용한 인공수정이 번식성적에 미치는 영향을 구명하기 위하여 축산기술연구소 종축개량부 (충남, 성환) 돼지 인공수정센터에서 사육중인 인공수정용 종모돈을 이용하여 1995년부터 2000년까지 액상정액을 이용한 인공수정시 종모돈의 품종, 인공수정횟수, 계절 및 정자농도가 번식성적에 미치는 영향을 조사하여 돼지 인공수정시 번식성적 향상과 실용화에 기여하고자 본 연구를 실시하였다. 액상정액 인공수정시 정액을 생산한 종모돈의 품종 (Landrace, Yorkshire, Duroc)간 번식성적은 유의차가 인정되지 않았다. 발정당 2회 또는 3회 인공수정 시에도 번식성적에 차이가 없었다. 인공수정을 실시한 계절별로 번식성적을 조사하였으나 계절간 차이점은 발견할 수 없었다. 액상정액 인공수정시 1회 주입정자의 최저농도를 구명코자 활력 70% 이상인 정자수를 기준으로 80 ml 병당 정자농도를 3.0, 2.5, 2.0 및 1.5×10^(9)로 조절하여 인공수정한 결과 수태율, 분만율, 총산자수 및 생존산자수에서 통계적인 유의차가 인정되지 않아 1회 주입정자농도는 운동성 정자 기준으로 1.5×10^(9)/80 ml 이상으로 조절하면 인공수정에 실용적으로 사용 할 수 있음을 보였다. This study was carried out to investigate the effects of liquid boar semen on reproductive performance in swine artificial insemination Many factors, which were breeds, time of insemination, breeding season, sperm per dose etc. have been tried to improve reproductive efficiency. Boars were raised at Swine Artificial Insemination Center in National Livestock Research Institute. Sunghwan, Chungnam, Korea. This experiment was carried out from 1995 to 2000. There were no differences in the fertility results compared with 3 breeds (Landrace, Yorkshire and Duroc), frequencies of artificial insemination (double and triple) per estrus cycle and different seasons by using liquid boar semen. There were no significant differences in conception rate, farrowing rate and litter size using 4 trials of 3.0, 2.5, 20 and 1.5×10^(9)/80㎖ in liquid boar semen with 70% of motile sperm cells. We confirmed that the sperm number per dose of 1.5×10^(9)/80㎖ could be used for commercial artificial insemination.

      • 복숭아의 PCR 최적 조건 구명

        To optimize PCR condition is the most important and fundamental step for experiments using PCR. The purpose of this study was to optimize PCR condition for RAPD analysis in peach. 3×3×3 factorial experiment for template DNA concentration, MgCl2 concentration, and amount of taq polymerase was conducted. Another factorial experiment for denature, annealing, extension temperature on the optimal PCR condition was also conducted. The best template DNA concentration was 20ng(0.5unit taq polymerase and 2.5mM MgCl2), 40ng(0.5unit taq polymerase and 4.5mM MgCl2, and 1unit taq polymerase and 2.5mM MgCl2), and 60ng(0.5unit taq polymerase and 2.5mM MgCl2, 0.5unit taq polymerase and 4.5mM MgCl2, 1unit taq polymerase and 2.5mM MgCl2, 1unit taq polymerase and 4.5mM MgCl2, and 1unit taq polymerase and 7.0mM MgCl2). The best MgCl2 concentration was 2.5mM(40ng template DNA and 1unit taq polymerase, 60ng template DNA and 0.5unit taq polymerase, and 60ng template DNA and 1unit taq Folymerase), and 4.5mM(40ng template DNA and 0.5unit taq polymerase, 40ng template DNA and 1unit taq polymerase, 60ng template DNA and 0.5unit taq polymerase, and 60ng template DNA and 1unit taq polymerase), and 7.0mM (60ng template DNA and 1unit taq polymerase). The best taq polymerase concentration was 0.5unit(20ng template DNA and 2.5mM MgCl2, 40ng template DNA and 4.5mM MgCl2, 60ng template DNA and 2.5mM MgCl2, and 60ng template DNA and 4.5mM MgCl2), and 1unit(40ng template DNA and 2.5mM MgCl2, 60ng template DNA and 2.5mM MgCl2, 60ng template DNA and 4.5mM MgCl2, and 60ng template DNA and 7.0mM MgCl2). When we consider results from template DNA concentration, MgCl2 concentration, and amount of taq polymerase, 5 conditions(60ng of template DNA, 7.0mM MgCl2 and 1unit taq polymerase; 60ng of template DNA, 4.5mM MgCl2 and 1unit taq polymerase; 60ng of template DNA, 2.5mM MgCl2 and 0.5unit taq polymerase; 40ng of template DNA, 2.5mM MgCl2 and 1unit taq polymerase; and 40ng of template DNA, 4.5mM MgCl2 and 0.5unit taq polymerase;) were best combinations for the optimal PCR condition. Reaction temperatures for the optimal PCR condition were 90℃, 40℃, 72℃ and 92℃, 36℃, 72℃.

        The object of this study was to optimize PCR condition for RAPD analysis in adzuki bean. The best template DNA concentration was 20ng(0.25unit taq polymerase and 2.5mM MgCl2), 40ng(1unit taq polymerase and 2.5mM MgCl2, 1unit taq polymerase and 4.5mM MgCl2 and lunit taq polymerase and 7.0mM MgCl2), and 60ng(0.5unit taq polymerase and 2.5mM MgCl2, 0.5unit taq polymerase and 4.5mM MgCl2, 1unit taq polymerase and 2.5mM MgCl2, and 1unit taq polymerase and 4.5mM MgCl2) The best MgCl2 concentration was 2.5mM(40ng template DNA and 1unit taq polymerase, 60ng template DNA and 0.5unit taq polymerase, and 60ng template DNA and 1unit taq polymerase), 4.5mM(20ng template DNA and 0.25unit taq polymerase, 60ng template DNA and 0.5unit taq polymerase, and 60ng template DNA and 1unit taq polymerase), and 7.0mM(40ng template DNA and 1unit taq polymerase). Amount of taq polymerase was 0.25unit(20ng template DNA and 2.5mM MgCl2), 0.5unit(60ng template DNA and 2.5mM MgCl2, 60ng template DNA and 4.5mM MgCl2) and 1unit(40ng template DNA and 2.5mM MgCl2, 40ng template DNA and 4.5mM MgCl2, 40ng template DNA and 7.0mM MgCl2, 60ng template DNA and 2.5mM MgCl2, 60ng template DNA and 4.5mM MgCl2, and 60ng template DNA and 7.0mM MgCl2). When we consider results from template DNA concentration, MgCl2 concentration, and amount of taq polymerase, the best condition for PCR optimization was 60ng template DNA, 4.5mM MgCl2, and 1unit taq polymerase. Reaction temperatures for the optimal PCR condition were 84℃, 32℃, 62℃; 90℃, 40℃, 72℃; and 92℃, 36℃, 72℃.

      • 神闕의 穴位 특징과 鍼灸治療에 대한 문헌 고찰

        This study was designed to investigate the location of Sin-Guel(CV_(8), 神闕) through literature research. We extracted the part about the location of Sin-Guel(CV_(8), 神闕) from ancient and modern oriental medical literature which were used commonly in clinic. The results were summarized as follows; 1. The location of Sin-Guel(CV_(8), 神闕) were generally recorded DangJeJung. 2. Sin-Guel(CV_(8), 神闕) placed in middle of linea alba. 3. Treatment effect of moxibustion on Sin-Guel(CV_(8), 神闕) were recorded as diarrhea, dropsy, prolapse of the anus, stomachache, paralysis etc.. 4. Acupuncture therapy on Sin-Guel(CV_(8), 神闕) is dangerous because of inflammation, it is suggested that acupuncture therapy may be possible if acupuncture therapy do not induce inflammation on Sin-Guel(CV_(8), 神闕).
