        To evaluate antibiotic-induced endotoxin release(AIER) and its correlation with some cytokines, we measured endotoxin level and tumor necrosis factor alpha(TNF-α) and interleukin6(IL-6) production in mononuclear cells in vitro after exposure of Pseudomonas aeruginosa to antibiotics belonging to different class with two extreme concentrations. The tested concetration of antibiotics were set up according to peak serum level. The low concetration of ceftazidirne and low concentration of imiperiem increased AIER, but high concentration of ceftazideme, high concentration of ciprofloxacin, high concentration of cefoperazone/sulbactam, high concentration of amikacin, and high concentration of meropenem reduced AIER.Interestingly, combined treatment of these antibiotics markedly reduced AIER, But the major cyotkines, TNF-α and IL-6 were not affect by type and concettration of antibiotics, combined treatment of antibiotics, and level of endotoxin released by antiboitics. In this study, we observed AIER was different according to type of antibiotics, concentration of antibiotics, and combination of antibiotics, But AIER had poor correlation with TNF-α and IL-6 in Pseudomonas aeruginosa. It suggests that cytokine release is not solely dependent to endotoxin, but more complex cascade is needed. More invesfigations, such as endotoxin induced cytokine mRNA expression, relationship with penicillin-binding proteins and endotoxin-neutralizing effect of antibiotic itself, must be performed.

        According to the investigation made by analyzing cause of the detail about sports injury. We make this things an object of all the male representative players 254 out of 286 (88.81%) that took part in the whole event from April 15, 1988 to September 16, 1988 in Seoul Olympic Games. The results are as follows. 1. The representative players in Korea started in the athlete life at age 13.1 on the average. Gymnast early started at age 8.4 and yachtist was later at age 18 in the athlete life. 2. There are about 5.7 cases on the outbreak of the sports trauma per person of player a year. This came into existence near the ankle at the high rate of damage formed 11.90%. 3. A sort of sports injury in analysis muscular bruise broke out at the highest rate of occurrence formed 15.70%. 4. The greater part of representative players themselves the injury of trauma by massage (12.53%). Some of them undergo medical treatment to the specialist hospital once a year on the average. Most of hockey players have had experience in this kind of injury treatment three times one year to per person of players. A few of them showed 0.15 times per person a year. 5. Most of representative players have been periodically to the orthopedist formed 27.59 (732) out of all 2653 cases at the high rate of fact. Especially a sort of injury in analysis, the injury of ankle region showed the percentage 11.86 (704) out of all 5,937 cases at the highest rate. 6. The period of medical treatment of the representative players in Korea came out about 14.7 days. Besides period of the other player's treatment came out mostly from one to five days formed 44.82%(935 cases) 7. Manyof them have been roughly treated with muscular bruise in analysis of sports injury. They were taking pains with muscular bruise formed 15.65% (1003) out of all 6408 case. 8. In major cause of sports injury came into existence during the training with waste of physical stamina formed 35.89% and were beyond their power with practicing the game formed 48.09%. Also, a sort of those were brought about the cause of sports injury with desire of outcome too much during in the game formed 44.20%

        Background : Loss of bone mass is usually detected after BMT. The causes of bone loss are related with gonadal dysfunction and immunosuppressants. Cytokines, especially IL-6, play an important role in the pathogenesis of postmenopausal osteoporosis. However, the pathogenetic role of cytokines in post-BMT bone loss is unknown and data on the changes of cytokines in accordance with bone turnover markers are scarce. The aim of this study is to assess the relationship of bone turnover markers and cytokines of peripheral blood and bone marrow before and after allogeneic BMT. Methods : This prospective study included two analyses. The first was a study of 46 BMT recipients, examining the relationship between bone turnover markers and cytokines of serum which were measured before and 1, 2, 3, 4 week and 3 months after BMT. The second was a study of 14 BMT patients, measuring bone marrow plasma cytokines such as IL-6 and TNF-? at post-BMT 3 week and bone turnover marker at the same time to assess the relationship beween two parameters. Results : Serum ICTP, bone resorption marker, increased progressively until 4 weeks (peak) after BMT and then decreased thereafter. Serum osteocalcin, bone formation marker, decreased progressively until 3 weeks after BMT and then increased thereafter. There was positive correlation between serum ICTP and bone marrow IL-6 levels at the post-BMT 3 week with a statistical significance, but the correlation between bone turnover markers and bone marrow TNF-? or peripheral blood cytokines was not found. Conclusion : Our data suggest that the progressive increase of bone resorption after BMT is related with the increase of bone marrow IL-6, which is a potent stimulator of bone resorption in vivo(J Kor Soc Endocrinol 15:85-96, 2000).

        Immune Enhancement Effect of Asterias amurensis Fatty Acids through NF-κB and MAPK Pathways on RAW 264.7 Cells

        Asterias amurensis is a marine organism that causes damage to the fishing industry worldwide; however, it has been considered a promising source of functional components. The present study aimed to investigate the immune-enhancing effects of fatty acids from three organs of A. amurensis on murine macrophages (RAW 264.7 cells). A. amurensis fatty acids boosted production of immune-associated factors such as nitric oxide (NO) and prostaglandin E2 in RAW 264.7 cells. A. amurensis fatty acids also enhanced the expression of critical immuneassociated genes, including iNOS, TNF-α, IL-1β, and IL-6, as well as COX-2. Western blotting showed that A. amurensis fatty acids stimulated the NF-κB and MAPK pathways by phosphorylation of NF-κB p-65, p38, ERK1/2, and JNK. A. amurensis fatty acids from different tissues resulted in different levels of NF-κB and MAPK phosphorylation in RAW 264.7 cells. The results increase our understanding of how A. amurensis fatty acids boost immunity in a physiological system, as a potential functional material.

      • Antiviral and anti-inflammatory activity of budesonide against human rhinovirus infection mediated via autophagy activation

        <P>Human rhinovirus (HRV) infection causes more than 80% of all common colds and is associated with severe complications in patients with asthma and chronic obstructive pulmonary disease. To identify antiviral drug against HRV infection, we screened 800 FDA-approved drugs and found budesonide as one of the possible drug candidates. Budesonide is a corticosteroid, which is commonly used to prevent exacerbation of asthma and symptoms of common cold. Budesonide specifically protects host cells from cytotoxicity following HRV infection, which depend on the expression of glucocorticoid receptor. Intranasal administration of budesonide lowered the pulmonary HRV load and the levels of IL-1 beta cytokine leading to decreased lung inflammation. Budesonide regulates IL-1 beta production following HRV infection independent of inflammasome activation. Instead, budesonide induces mitochondrial reactive oxygen species followed by activation of autophagy. Further, the inhibition of autophagy following chloroquine or bafilomycin Al treatment reduced the anti-viral effect of budesonide against HRV, suggesting that the antiviral activity of budesonide was mediated via autophagy. The results suggest that budesonide represents a promising antiviral and anti-inflammatory drug candidate for the treatment of human rhinovirus infection.</P>

        Immune Enhancement Effects of Neutral Lipids, Glycolipids, Phospholipids from Halocynthia aurantium Tunic on RAW264.7 Macrophages

        Fractionated lipids of Halocynthia aurantium (Pyuridae) have been demonstrated to possess antiinflammatory properties. However, their modulatory properties have not been reported yet. Thus, the objective of this study was to determine immune enhancing effects of fractionated lipids from H. aurantium tunic on macrophage cells. The tunic of H. aurantium was used to isolate total lipids, which were then subsequently separated into neutral lipids, glycolipids, and phospholipids. RAW264.7 cells were stimulated with different concentrations (0.5, 1.0, 2.0, and 4.0%) of each fractionated lipid. Cytotoxicity, production of NO, expression levels of immune-associated genes, and signaling pathways were then determined. Neutral lipids and glycolipids significantly stimulated NO and PGE<sub>2</sub> production and expression levels of IL-1β, IL-6, TNF-α, and COX-2 in a dose-dependent manner, while phospholipids ineffectively induced NO production and mRNA expression. Furthermore, it was found that both neutral lipids and glycolipids increased NF-κB p-65, p38, ERK1/2, and JNK phosphorylation, suggesting that these lipids might enhance immunity by activating NF-κB and MAPK signaling pathways. In addition, H. aurantium lipids-induced TNF-α expression was decreased by blocking MAPK or NF-κB signaling pathways. Phagocytic activity of RAW 264.7 cells was also significantly enhanced by neutral lipids and glycolipids. These results suggest that neutral lipids and glycolipids from H. aurantium tunic have potential as immune-enhancing materials.

        Enzyme Hydrolysates of Ginseng Marc Polysaccharides Promote the Phagocytic Activity of Macrophages Via Activation of TLR2 and Mer Tyrosine Kinase

        Although ginseng marc is a by-product obtained during manufacturing of various commercial ginseng products and has been routinely discarded as a waste, it still contains considerable amounts of potential bioactive compounds, including saponins and polysaccharides. Previously, we reported that ginseng oligosaccharides derived from ginseng marc polysaccharides by enzymatic hydrolysis exert immunostimulatory activities in macrophages and these activated macrophages are in turn able to inhibit the growth of skin melanoma cells by inducing apoptosis. In the present study, a more detailed investigation of the immunostimulatory activity and underlying action mechanisms of an enzymatic hydrolysate (GEH) containing these oligosaccharides derived from ginseng marc polysaccharides was performed. The levels of proinflammatory cytokines and anti-inflammatory cytokines were measured in GEH-stimulated RAW264.7 macrophages using RT-PCR analysis and ELISA. The expression levels of Toll-like receptor 2 (TLR2) and TLR4, Dectin-1, and MerTK were measured by RT-PCR analysis or western blot analysis, and the phagocytic activities of GEH-challenged bone marrow-derived macrophages toward apoptotic Jurkat cells were assayed using fluorescence microscopy. GEH induced the production of both proinflammatory cytokines TNF-α and IL-6, and anti-inflammatory cytokine IL-10 in RAW 264.7 cells. The expression of the TLR2 and MerTK mRNAs was increased upon GEH treatment. Phagocytosis of apoptotic Jurkat cells was enhanced in GEH-treated macrophages. Based on the results, this enzymatic hydrolysate (GEH) containing oligosaccharides exerts immunostimulatory effects by maintaining the balance between M1 and M2 cytokines, facilitating macrophage activation and contributing to the efficient phagocytosis of apoptotic cells. Therefore, the GEH could be developed as value-added, health-beneficial food materials with immunostimulatory effects.

      • 열병합발전용 Dual Fuel Engine의 질소산화물 배출저감에 관한 연구

        This study shows the correlation between ?? emission in the exhaust gas and various operation factors of dual fuel engine for Co-generation system. General tendency was shown that the thermal efficiency was lowered by the change of operation factors. However these were not confirmed on this experiment. Increasing T4 temperature (exhaust gas temperature at turbo-charger inlet)reduces ?? emission rate. The higher T4 temperature requires lower excess air as the excess air ratio is controlled by T4 temperature on gas mode operation. Another tendency was that ?? emission rate is reduced in case of increasing boost air temperature, quantity of pilot oil bypassing flue gas through the through the exhaust gas boiler. The diameter of the fuel injection nozzle was changed smaller than design value and the injection timing was readjusted. Thus ?? enission rate could be reduced as retarding injection timing and changing hole diameter of fuel injection nozzle, however maxium engine out-put was decreased by changing fuel nozzle on the diesel mode operation.

        The purpose of this study is to find out the effect group jogging(the 600-meter run)on the improvement of student's health when it is applied to them. Students would jog 5-7 minutes out of 50 minutes in physical education class as a warming-up exercise in introduction stage. The third class of the second grade was chose as an experimental class, the fourth class or the second grade as a comparative class. The study shows that group jogging not only makes students pay good attention to the warming-up exercise and their physical fitness but also improves students' bodily powers, such as muscular staying power, whole body staying power and agility.

The International Organization was established by a large number of countries through various forms of agreement in order to accomplish their own goals and interests. The existence of the I.O. (International Organization) holds good only for those countries which have a legally authorized member-ship position in the organization ; there fore, it can be logically inferred that the non-member countries bear no obligation to approve its ways of performance. A country meat respect other counterpart's sovereignty in accordance with existing international lows, acknowledge its right to exist, and ultimately be willing to take comprehensive responsibilities. If any violation in executing suck obligations should take place, she must be held responsible on the international level. As for example, any of its member countries is given the right to send a compensation-claim slit 4irectly to the U. N. for damages and tosses incurred because of other countries. The major function of the I.O. is to achieve its common goals and ideals Bet up by the member countries through overall agreement. Two good examples for its functions are the maintenance of the global peace, and the adjustment of the related affairs in domestic administration. In conclusion, while working on this thesis on the study of the I.O, I attempted not only to classify the major conctions of the organization, hut also considered itsimpact on the domestic laws and public administration of a member-country.
