형광검출 역상 액체크로마토그래피에 의한 Histamine의 정량

Histamine을 정확하고 신속하게 정량하기 위해 9.Fiuorenylmethyl chloroformate를 형광유도체화제로 하여 역상 HPLC법으로 정량하였다. 히스타민을 형광유도체화할 때 반응액의 pH, 반응시간, 형광유도체화제의 농도 등 최적 반응 조건을 검토하였다. 이 방법으로 히스타민을 분석한 결과 0.1~0.5ug/ml의 농도범위에서 상관계수가 0.922인 양호한 직선성을 나타내었으며 검출한계는 0.01??/ml였다. A rapid and simple method for the determination of histamine by reverse-phase high performance liquid chromatography with fluorescence detection was established. 9-Fluorenylmethyl chloroformate(FMOC) was used as fluorescent derivative reagent. The optimum conditions for the derivatiation such as pH, reaction time and he concentration of FMOC were investigated. Linearity of calibration curve was obtained between 0.1ug/ml/and 0.5ug/ml(r=0.922) and the limit of detection was 0.01ug/ml.

The current study evaluates the capacity of eugenol to regulate immediate allergic reaction by control of histamine release. Administrations of eugenol (1 M/kg. i.p.) at 60 min before and 5.10 min after the compound 48/80 treatment (8 mg/kg.i.p.) were shown the mortality rates as 0.44.4. and 77.8%. respectively. A 60 min before administered group revealed a singificant inhibition of serum his-tamine release compared with those of 5 and 10 min after the compound 48/80 injection. Eugenol (6-48mM) was also showed a dose-dependent activity on the compound 48/80-induced histamine release from the highly purified population of Alcian Blue-positive peritoneal mast cells. These results indicate that in vitro treatment with exogenous eugenol inhibited the active response of mast cell populations and modulated its characteristics.

Samsoeum (SSE) is used in traditional oriental medicine for various medicinal purposes. However, the exact mechanism that accounts for the anti-allergy and anti-inflammatory effects of the SSE is still not fully understood. The aim of the present study is to elucidate whether and how SSE modulates the allergic reactions in vivo, and inflammatory reaction in vitro. In this study, we showed that SSE significantly decreased compound 48/80-induced systemic anaphylaxis, ear-swelling response, histamine release from preparation of rat peritoneal mast cells and anti-dinitropheny IgE-induced passive cutaneous reaction. Also, SSE inhibited the expression of inflammatory cytokine and cyclooxygenase-2 in PMA plus A23187-stimulated human mast cells (HMC-1). In addition, we showed that anti-inflammatory mechanism of SSE is through suppression of nuclear factor-${\kappa}B$ activation and $I{\kappa}B-{\alpha}$ phosphorylation/degradation in HMC-1. These results provided new insight into the pharmacological actions of SSE as a potential molecule for therapy of inflammatory allergic diseases.

변화되는 위 측면 기울기를 가진 비대칭 사다리꼴 핀의 온도분포가 2차원 해석적 방법을 사용하여 조사되어 진다. 이 비대칭 핀을 위해, 내부유체로부터 내벽까지의 대류, 내벽으로부터 핀 바닥까지의 전도, 그리고 핀 바닥을 통한 전도가 동시에 고려된다. 무차원 핀 길이와 높이좌표 변화에 따른 온도 형상이 보여진다. 또한 핀 아래 끝에서 온도 변화가 핀 형상계수의 함수로 나타내어진다. 핀 길이의 변화에 따라 핀 바닥을 통한 열손실과 각 면으로부터의 열손실들이 비교된다. 결과들 중 하나는 핀 형상계수가 증가함에 따라 핀 아래 끝에서 온도는 선형적으로 감소함을 보여준다. The temperature distribution of an asymmetric trapezoidal fin with various upper lateral surface slopes is investigated by using the two-dimensional analytic method. For this asymmetric fin, convection from the inner fluid to the inner wall, conduction from the inner wall to the fin base and conduction through the fin base are considered simultaneously. The temperature profile with the variation of dimensionless fin length and height coordinates is shown. Also, the temperature variation at the bottom tip of the fin is presented as a function of the fin shape factor. Heat losses through the fin base and from each side are compared for variations in fin length. One of the results shows that temperature at the fin bottom tip decreases linearly as the fin shape factor increases.

In this study, we have developed a mobile-based visible/NIR(Near InfraRed) imaging equipment for the animal testing. This equipment can provide visible, NIR and merged image through the viewer program. Especially, merged image help researcher to understand visual messages at animal in-vivo test. Also, it is available to send real-time images through the smart phone. Researcher can communicate with another researcher who is a long distance away. Also, the equipment was made with portable small size to enable it to commercialize.

For the block transpotation and loading, the D-type lug is widely used in the heavy industry compaies. Therefore, it is demanded to develop the method of structural safety assessment for lug sysetem. In this study, the strength analyses were performed using elasto-plastic FE model and accomplished a test using the loading actuator. And then, the analysis results and the test results are compared. FE model is composed of lug, welding bead, shackle and shackle pin. FE analyses considering material/geometrical non-linearity and contact conditions were carried out. Following in the field work environments, the ultimate strength were presumed in in-phase and out-phase loading directions. As the loading and unloading test results, elasto-plastic displacements of the lug were measured, and compared with analysis results. From these processes, safety assessment is predictable considering the residual plastic deformation.

Animal welfare has been a growing concern in the livestock industry. Korea introduced the animal welfare labeling system in 2012. Consumers`s preferences for pork products (belly) with a focus on use of gestation stalls are analyzed using choice experiments in this paper. Volunatry and legislative schemes in producing gestation stalls-free pork are also examined. The results show consumers are more likely to prefer gestation stalls-free pork. However, the null hypothesis that the willingness to pay for gestation stalls-free pork under legislative scheme is higher than that under voluntary scheme is not rejected. This suggests that prohibiting producers from using gestation stalls will not improve consumer welfare in the presence of a labeling scheme. The results of this paper could contribute to govdernment`s policies for animal welfare.

버섯 균사체로부터 분리한 단백다당체의 Th1 cell 증식효과 및 각종 암세포에 대한 세포독성을 조사하였다. 단백다당체 시료는 영지버섯, 아가리쿠스, 표고버섯, 운지버섯, 그리고 상황버섯을 100℃에서 3시간 열수 수출하는 방법으로 조제하였다. Th1 cell의 세포 증식을 조사한 결과, 10mg/ml 농도에서 모든 시료가 40% 이상 억제하는 효과가 있는 것으로 나타났다 7종류의 암세포를 이용한 암세포 생존율을 조사한 결과, 1mg/ml 농도에서 P388D1와 L1210에서 아가리쿠스는 2.4% 와 39.7%, 표고버섯은 48.4%와 52.5% 의 생존율을 보였다. Sarcoma 180 으로 복수암을 유발시킨 마우스에게 아가리쿠스와 표고버섯으로부터 추출한 단백다당체를 섭취시킬 경우 생존율이 27 ~ 40%까지 유의적으로 증가하는 것으로 나타났다. The purpose of this study is to observe the effect of protein-bound polysaccharide (PBP) on proliferation of Th1 cells and cytotoxicity of cancer cell. Mushrooms (Ganoderma lucidum, Agaricus blazei, Lentinus edodes, Coriolus versicolor and Phellinus linteus) were fractionated by 100℃ hot water for 3hr. PBP was stimulated and proliferated Th1 cells most at 10 mg/ml concentration and the percentage of cell proliferation was 40%. It was estimated cytotoxicity of PBP against 7 kinds of cancer cell lines. Antitumor activities of Agaricus blazei against P388D1 and L1210 (tumor cell lines) were 2.4% and 39.7% survival rate, and Lentinus edodes was 48.4$ and 52.5% survival rate, respectively. PBP mixtures of Agaricus and Lentinus edodes prolonged (27 ~ 40%) significantly the survival rate of mice intraperitoneally implanted with sarcoma 180.
