        <P>The prototype of the cdc2 protein kinase in mammalian cells regulates its entry into mitosis by phosphorylating a group of key proteins in the major cell cycle transitions. In this study, using the mep45 gene encoding the 45 kDa major envelope protein (Mep45) of Selenomonas ruminantium, a rumen bacteria, a Mep45-fused substrate (PKTPKKAKKL-Mep45, MFS-cdc2) was cloned to detect the activity of cdc2 protein kinase. We report here on a strategy for the detection of a phosphorylation of a substrate catalyzed by cdc2 protein kinase by using a radioisotope detection technique. It is possible to constantly obtain a reasonable quantity of MFS-cdc2 for the cdc2 protein kinase assay and its cost can be as low as a synthesized peptide. Results of the study indicate that the Mep45-fused protein can be used effectively as a substrate for detecting the activity of cdc2 protein kinase and it can be used in developing a protein biochip for a high-throughput screening and also for studying protein–protein interactions.         발열을 동반한 호중구감소증에서 Cefepime 단독투여와 Ceftazidime 및 Tobramycin 병합투여의 효능비교

        목적 : 국내에서 발열을 동반한 호중구감소증이 있는 악성 종양환자에게 경험적 항생제로 베타락탐계 항생제와 아미노배당체의 병합요법의 사용이 일반적이다. Cefepime은 광범위 항균 작용을 가지고 있어, 그람 음성균 뿐만 아니라 그람 양성균에 대해서도 우수한 효과를 나타낸다. 재료 및 방법: 발열을 동반한 호중구감소증이 있는 악성 종양환자를 대상으로 무작위, 공개, 비교 연구를 시행하였다. 대상 환자를 무작위로 cefepime 단독요법군과 ceftazidime 및 tobramycin 병합요법군으로 나누어 투여하고 각각의 임상적 효능과 안전성을 비교하였다. 구강및 인후 점막염이 있는 환자에서 분리된 녹색 연쇄알 구균에 대한 항생제 내성 정도를 조사하였다. 결과 : 대상환자 89명 중 CA 투여군이 48예(53.9%), CT 투여군이 41예(46.1%)이었다. 발열의 유형별로 MDI는 18예(20.2%), CDI는 9예(10.1%), UF는 62예(69.7%)로 두 군 간에 차이가 없었다. CA 투여군과 CT 투여군의 임상적 호전률은 시험약 투여 후 2-4일째 각각 91.7%, 85.4% (P=0.31), 치료 종료 시 각각 91.7%, 100% (P=0.15)로 두 군간에 유의한 차이가 없었다. 치료 종료 시 CA 투여군과 CT 투여군의 세균학적 소실률은 모두 100%로 두 군간의 유의한 차이가 없었다(P=0.78). 점막염이 있는 환자로부터 녹색 연쇄알 구균이 분리된 경우는 25예(28.1%)이었으며, 분리된 녹색 연쇄알 구균은 penicillin, ceftriaxone, cefepime, vancomycin에 모두 감수성을 보였다. 약제 관련 이상 반응의 발생 빈도도 두 군간에 유의한 차이가 없었다. 결론 : 발열을 동반한 호중구감소증이 있는 악성 종양환자의 경험적 항생제로서 cefepime 단독요법은 ceftazidime 및 아미노배당체의 병합요법만큼 효과적이고 안전하였다. Background : Broad-spectrum antibiotic therapy has been recommended as an empirical regimen in cancer patients with febrile neutropenia. Cefepime is a fourth generation cephalosporin with good activity against both gram-positive cocci and gram-negative bacilli. Materials and Methods : To compare the efficacy and safety of cefepime alone with ceftazidime plus tobramycin as empirical regimen for adult cancer patients with febrile neutropenia, a randomized, open label, comparative trial was performed. If the patient showed clinical improvent 72 hours, antibiotic could be changed to oral ciprofloxacin. Clinical and microbiological responses were determined at 72 hours and at the end of therapy. To investigate the antimicrobial resistance of viridans streptococci, swab cultures were obtained from throat in all enrolled patients and antimicrobial susceptibility tests were performed by using microdilution method according to the NCCLS. Results : A total of 89 patients were enrolled. Forty-eight patients received cefepime alone (CA), and 41 patients received ceftazidime plus tobramycin (CT). Demographic and baseline clinical characteristics were similar in both groups (P>0.05). The initial clinical success rate at day 2-4 in group CA (91.7%) was similar with that in CT group (85.4%) (P=0.31). At the end of therapy, the final clinical success rate in CA group (91.7%) was similar to that in CT group (100%) (P=0.15). In 18 patients, with microbiologically defined infections, the eradication rate was 100% in both groups. Adverse events including liver dysfunction (21.3%) and renal dysfunction (2.2%), were similar in both groups (P=0.87). Viridans streptococci were isolated from the throat cultures in 25 cases, and all of these strains were susceptible to penicillin (MIC_(90) 0.12 ㎍/mL), cefepime (1 ㎍/mL), and vancomycin (0.12 ㎍/mL). Conclusion : Efficacy and safety of cefepime monotherapy was comparable to the combination of ceftazidime and tobramycin. It could be used as an alternative empirical regimen for treating cancer patients with febrile neutropenia.

      • 遮光水準이 麥門冬의 生育 및 收量에 미치는 影響

        SummaryFive shading levels (0, 5, 50, 75 and 90%) were investigated for their effects on growth and yield of Liriope platyphylla Wang et Tang. The results obtained were summarized as follows;1. Plant height and leaf length increased with increasing shading levels up to 50% and then decreased with further increasing shading. Leaf width ranging from 0.49 to 0.52cm tended to linearly decrease as shading level was increased. 2. SPAD reading was linearly increased from 56.9 to 69.5 as shading level was increased from 0 to 90%. 3. There was no difference in fresh leaf weight among shading levels. However, fresh weight of roots, tuberous roots, and shoots + roots decreased with increasing shading level. 4. The number of tuberous roots per plant decreased from 21.4 at 0% shading to 11.8 at 90% shading. Dry weight per tuberous root was heaviest (0.4g) at 35% shading and then declined with increasing shading level. 5. Dry tuberous root yield per plant ranged from 6.5g at 0% shading to 3.1g at 90% shading showing linear relationship between yield and shading level. Yield per plant highly correlated wish the number of tuberous roots per plant. 6. The results indicate that growing Liriope platyphylla Wang et Tang under shading has no advantage in Cheju province.

        외나로도 주변해역 퇴적물의 지화학적 특성과 저서 생물상

        In order to evaluate the relationship between geochemical characteristics and benthic facies of the sediments from the Oenaro Island where red tide proliferation is first observed every year including this year, surface and short multiple core sediment samples were analyzed in terms of geochemical and benthic facies variation. The contents of organic carbon, carbonate, and sulfide gas were relatively low. The variation in C/N ratios, which indicate nature of organic carbon, suggested that the organic carbon recorded in the study area is composed of mixtures of marine and terrigenous organic matters. The concentration of minor elements found at the surface and multiple core sediment samples were also low as well as the enrichment factors(Ef) for the seven heavy metals indicated that the sediment of this area is not polluted significantly. The macrobenthic faunal community comprised 61 species, and their mean density was 708 ind./m2. Polychaete worms were major taxa of this benthic community. A crustacean amphipod, Melita sp. was the most abundant species accounted for 20.7% of total abundance, and the small polychaetes such as Heteromastus filiformis, Paralacydonia poradoxa, Magelona japonica, and Sigambra tentaculata were the next dominant species. The macrobenthos around the Oenaro Island were more diverse and abundant than that in Gamak Bay. The benthic communities in the study area sustained somewhat different species composition based on the cluster analysis and the MDS ordination. The benthic community health condition at three stations seemed to be unbalanced, and slightly polluted based on the biological index such as BPI and BC. There was no clear relationship between the geochemistry characteristic and the benthic faunal facies attributed by the micro-algal blooms in this coastal area.

        사람의 생식과 발생에 대한 애니메이션 학습 프로그램의 개발 및 적용

        중학교 3학년 생명 영역의 생식과 발생 단원에 대한 애니메이션 학습 프로그램(Animation Learning Programs)을 개발하였다. 애니메이션 학습 프로그램의 교육적 효과는 지식 성취도와 개념 변화를 통해 조사하였다. 본 프로그램의 구성은 (1) 남, 여성의 생식기 구조와 기능, (2) 수정과 임신, (3) 태아 발생 등의 주제로 이루어져있다. 지식 성취도의 결과 여성의 생식구조와 수정과 임신에 대해선 매우 유의미한 효과(p=0.009)를 나타났으며 남성의 생식기 구조와 기능에서는 유의미하지는 않지만 효과적(p=0.061)으로 나타났다. 학습자 수준에 의한 지식성쥐도 분석결과 중수준의 학생에게 유의미한 효과가 나타나는 것으로 보아 본 프로그램의 활용도 매우 보편적이라 사료 된다. 특히 면담에 의한 개념 변화 조사 결과 학생들은 수정과 임신의 초기발생에 대해 본 프로그램이 매우 영향을 미쳤으며 실험 집단의 학생들은 사전보다 사후에 향상된 개념을 갖는 것으로 조사 되었다. 따라서 본 애니메이션 학습 프로그램은 생물학에서 현상위주의 과정을 설명하는데 매우 효과적이라 사료된다. The effect of animation learning programs (ALPs) upon achievement and conceptual change of the topics on reproduction and development was investigated in the middle school (9th grade) students. The topics covered by the ALPs were as follows: (1) reproductive structure and function of male and female in human; (2) process of fertilization and pregnancy in human; and (3) development of fetus. Two groups of student participated in this experiment; (1) treatment group subjected to the use of ALPs, and (2) control group. The results showed significant effect (p<0.05) on the achievement in learning of reproductive structure and function of female and process of fertilization and pregnancy. The results mostly showed significant improvement on the achievement in students of the middle level of learning. Thus, the ALPs may be applied universally in other schools. The conceptual change on the process of fertilization and pregnancy in human showed significant improvement in treatment group in comparison to control group. Therefore, the use of ALPs was effective in understanding and perception of abstract or difficult to direct experience learning contents.

        웹마이닝을 이용한 M-Commerce 추천시스템 설계 및 구현

        추천 시스템들은 고객들이 E-Commerce 사이트에서 상품들을 사는 것을 도와주기 위해서 지속적인 증가추세로 사용되었다. 추천시스템들은 다양한 고객들의 선호도에 따라 유사성과 비유사성에 대한 정보의 기초위에서 고객들의 잠재적인 관심 항목들에 대해 개인의 취향에 맞게 추천하는 기술들은 제공한다. 그러나 추천시스템에 많은 관심을 가짐에도 불구하고 그들의 성능에 대한 공개된 기술이나 정보는 매우 제한적이다. 본 논문에서는, 과거 고객들의 구매행동, 고객정보, 데이터마이닝의 연관규칙을 이용한 M-Commerce 추천시스템을 설계하고 구현하였다. Recommender systems are being used by an ever-increasing number of E-Commerce sites to help consumers find products to purchase. Recommender Systems offer a technology that allows personalized recommendation, of items of potential interest to users based on information about similarities and dissimilarities among different user’ tastes. However, despite enormous interest in recommender systems, both the number of available published techniques and information about their performance arc limited. In this paper, we design and implement an M-Commerce recommendation systems using the past buying behavior of the consumer, consumer information, and association rule mining.

      • 일부 종합병원 간호원의 이직의사 요인에 대한 조사 연구

        This study was attempted to determine the main factors on the turnover intention of the nurses in Taejeon area. The questionaires were given to 170 nurses were working at hospital in Taejeon from 2 to 8 June, 1984. The scales used for this study were the job satisfaction scale and the turnover intention scale developed. by the investigator. The results of the study were as follows : 1. The relationships between turnover intention and general characteristics-age, marital status, and level of education showed no statistical significance(P>0.05). The length of service, a˙ptitude to the job, concern of supporting family and the kind of hospital(i, e. , public or private) were proved to be significantly related to the turnover intention (P<0.05). 2. The relationships between turnover intention and job factors-pay, promotion, the type of supervision, co-worker, hospital administration, recognition, benefits, work itself were significant statistically(P<0.05). 3. Factors including job factors and general characteristics seem to be responsible to the turnover intention by 33.54%(R^2=0.3354). Among them hospital administration was the most significant .

        본 연구는 제 7차 교육과정에 따른 고등학교 생물Ⅱ 교과서에 수록된 생명공학 관련 단원의 내용과 구성의 체제를 분석하고,6차 교과서와의 연계성과 탐구영역 등을 비교 하였다. 또한 단원의 내용의 분석을 통해서 생명공학의 응용분야, 문제점과 차후 발전상, 연계성, STS 내용면,기타 오개념 등의 문제점들을 논의 하였다. This study was carried out to analyze the organization and contents of the biotechnology related chapter in high school Biology II of 7th Science Educational Curriculum. Especially the characteristics of connection, the inquiry activities between sixth and seventh textbook were comparatively analyzed. And also based on the analysis of this chapter, it introduces an applications and development of biotechnology in the future, the problems of biotechnology, the connections, STS contents and the problems of misconception were discussed.

      • Rifampicin에 의한 것으로 추정되는 위막성 대장염1예

        Pseudomembranous colitis, caused by altering the normal colonic flora and allowing the multiplication of Clostridium difficile, is an deleterious adverse effect of antibiotics. But it is rarely reported by rifampicin. Rifampicin is one of the first line drug in the treatment of tuberculosis and many patients are exposed to its potential adverse effects. We experienced a patient that had abdominal discomfort and hematochezia due to pseudomembranous colitis after receiving antituberculous medication, and which was probably caused by rifampicin. A 82 years old man was admitted with abdominal discomfort and hematochezia for one week. On the past history he had been diagnosed as endobronchial tuberculosis about 4 months ago. Colonoscopy revealed multiple discrete whitish mucosal lesion on rectosigmoid colon, and histologic findings were consistent with pseudomembranous colitis. The antituberculous agents were discontinued and vancomycin was administered. The patient's symptoms were resolved within several days. There was no recurrence after reinstitution of the antituberculous agents excluding rifampicin. We report here on a case of pseudomembranous colitis probably due to rifampicin.

        In this study, examined the cutting characteristics of alumuminum alloy AC8A-T6 that is used to present car piston materials. And in been holding materials machining empirically escape as result that experiment comparison changing the cutting speed and feed on various condition to choose efficient machining condition. The following results can be summarized from this research. 1.As the cutting speed decreased, principal cutting force and thrust cutting force is increased, and reason that cutting force interacts greatly in the low cutting speed is thought by result by BUE's stabilization. 2.The feed speed and cutting speed increase, friction factor is decrescent and the cause appeared the thrust cutting force is fallen than cutting force relatively because chip flow according to increase of the feed rate is constraint. 3.Though specific cutting resistance grows cutting area and the feed rate are few, the cause was expose that shear angle decreases by rake face of tool gets into negative angle remarkably as wear of a cutting tool or defect part of workpiece is cut. 4.Cutting speed do greatly depth of cut is slow, surface roughness examined closely through an experiment that becomes bad, and know that it can get good surface that process cutting speed because do feed rate by 0.1㎜/rev low more than 250m/min to get good surface roughness can.
