Isolation of a Calmodulin-binding Transcription Factor from Rice (Oryza sativa L.)

Calmodulin (CaM) regulates diverse cellular functions by modulating the activities of a variety of enzymes and proteins. However, direct modulation of transcription factors by CaM has been poorly understood. In this study, we isolated a putative transcription factor by screening a rice cDNA expression library by using CaM:horseradish peroxidase as a probe. This factor, which we have designated OsCBT (Oryza sativa CaM-binding transcription factor), has structural features similar to Arabidopsis AtSRs/AtCAMTAs and encodes a 103-kDa protein because it contains a CG-1 homology DNA-binding domain, three ankyrin repeats, a putative transcriptional activation domain, and five putative CaM-binding motifs. By using a gel overlay assay, gel mobility shift assays, and site-directed mutagenesis, we showed that OsCBT has two different types of functional CaM-binding domains, an IQ motif, and a Ca^(2+)-dependent motif. To determine the DNA binding specificity of OsCBT, we employed a random binding site selection method. This analysis showed that OsCBT preferentially binds to the sequence 5'-TWCG(C/T)GTKKKKTKCG-3' (W and K represent A or C and T or G, respectively). OsCBT was able to bind this sequence and activate β-glucuronidase reporter gene expression driven by a minimal promoter containing tandem repeats of these sequences in Arabidopsis leaf protoplasts. Green fluorescent protein fusions of two putative nuclear localization signals of OsCBT, a bipartite and a SV40 type, were predominantly localized in the nucleus. Most interestingly, the transcriptional activation mediated by OsCBT was inhibited by co-transfection with a CaM gene. Taken together, our results suggest that OsCBT is a transcription activator modulated by CaM.
독거노인 응급안전안심서비스의 주요 쟁점과 방향성: 현장실무자 관점을 중심으로

본 연구는 복지기술을 이용한 공공사회서비스의 일환인 독거노인대상 응급안전안심서비스 사례를 중심으로 현장 실무자 관점에서 주요 쟁점과 방향성을 파악하는 것을 주요목적으로 하고 있다. 본 연구를 수행하기 위해 개별인터뷰와 초점집단인터뷰를 포함해서 총 7명의 현장 관리자 및 실무자를 대상으로 질적 연구를 수행하였다. 본 연구에서는 크게 4개 주요 범주, 9개의 하위 범주를 포함하여 총 35개의 개념이 도출되었다. 특히, 장기간 사업표류, 댁내장비의 높은 오작동율, 업무성격과 사회복지전문직과의 관계, 사업간 통합 방향 등 4개 주요 범주를 통해 응급안전알림서비스의 주요 현안을 확인할 수 있었고, 사업의 책임성과 기기오작동에 대한 환류, 서비스 간 통합 등의 핵심문제와 쟁점들을 살펴볼 수 있었다. 응급안전안심서비스가 복지기술 기반의 새로운 사회서비스로서 공공성 하에 보다 효율적으로 서비스를 제공할 수 있기 위해서는 정부 정책추진 의지와 책임성, 댁내 장비 개선의 혁신성, 사회통합 이후의 응급관리요원의 존재, 지방관리 체제 vs. 중앙관리체제 등에 대한 방향성이 수립되어야 할 것으로 보인다. The current study aims to identify major issues and direction for Korean social services provided by the public sector based on the perspective of practitioners, focusing on Emergency Safety Relax Service for older adults living alone. A total of seven practitioners participated in either focus group or individual interviews. As an analytical method, the grounded theory was utilized to reflect practical experiences and perspectives for practitioners. Findings indicated that a total of 35 concepts, including four major themes and 9 sub-themes were derived. Specifically, core problems and issues were identified through four themes including the long-term business drift, high rate of mal-function in the home equipment, the relationships between business characteristics and social welfare professionals, and the integration direction among businesses. For delivering social services efficiently in public, willingness to promote government policy and responsibility, innovation in improving home equipment, the existence of emergency management personnel, and the direction of local management system vs. central management system should be established.
항암화학요법 중 호중구감소증이 발생한 저위험군 발열 환자들을 대상으로 한 경구 항균제 요법의 임상적 유용성 및 안정성에 대한 연구

목적 : 항암화학요법 중 호중구감소증을 동반한 발열이 발생하는 암환자들을 치료하기 위한 다양한 항균제와 여러 가지 방법들이 시도되고 있는 가운데, 합병증과 사망률의 발생가능성이 적은 저위험군 환자들을 대상으로 초기 72시간동안 정주 항균제를 투여한 이후 경구 항균제로 전환하는 요법의 유용성과 안정성을 평가해보고자 본 연구를 시행하였다. 방법 : 1998년 2월부터 1999년 9월까지 본원에서 항암화학요법 중 호중구감소증과 발열이 발생한 환자들 가운데 기저 암질환이 고형암이거나 림프종이고, 입원당시 패혈증의 증후가 없으며 입원 72시간이내에 해열되고 백혈구수치가 증가 추세인 환자들을 대상으로 하여 72시간 동안 정주 항균제를 투여한 이후 경우 ciprofloxacin 750㎎을 하루 2회씩 투여하여 총 4일간 투여하였다. 모든 환자들은 열이 떨어지고 호중구감소증이 회복될 때까지 입원하도록 하였다. 결과 : 총 38명 환자의 40예가 등록이 되었고, 환자들의 기저암 질환은 고형함이 72.5%, 림프종이 27.5%였다. 입원당시 평균 호중구치수는 156/㎕였고, 호중구수치가 100/㎕미만인 경우는 65%였으며, 호중구감소증이 지속된 기간의 평균은 2.4일이었다. 40예 중 39예가 항균제의 변형이나 추가 없이 호중구감소증과 발열로부터 회복이 되어 97.5%의 성공율(95% 신뢰구간: 86.8-99.9%)을 보였다. 부작용으로 피부발진이 있었던 경우가 한 예 있었는데, 증상이 경하여 경구 항균제를 지속할 수 있었다. 심와부의 동통으로 복용을 지속할 수 없어서 대상에서 제외된 예가 또 한 예 있었다. 결론 : 항암요법 중 호중구감소증과 발열을 동반한 환자들 가운데 저위험군 환자들에서 항균제 72시간정주 이후 경구 항균제로의 전환요법은 효과적이고도 안전한 치료방법이라고 할 수 있다. Background : Oral antibiotic therapy following empirical intravenous antibiotics may be effective and safe for febrile neutropenic patients with lowrisk for complications. Methods : We conducted a prospective clinical trial of oral antibiotic therapy in the patients with neutropenia and fever during chemotherapy for cancer. Underlying malignancies were solid tumor or lymphoma with short duration of neurtropenia and the patients had no evidence of clinically or microbiologically documented infections. Oral ciprofloxacin was given to the patients who lacked signs of sepsis on admission, had a rising tendency of neutrophil count (ANC >100 /㎕ ) at 72 hours, and were afebrile at 72 hours. All patients were hospitalized until neutropenia and fever resolved. Results : A total of 40 episodes of 38 patients were enrolled from February 1998 to September 1999. The mean neutrophil counts on admission were 156/㎕ and the mean duration of neutropenia was 2.4 days. The episodes which had neutrophil count below 100 /㎕ were 26 (65%). Treatment was successful in 39 of 40 episodes (97.5% : 95 % confidence interval, 86.8% to 99.9%). Adverse reactions of oral ciprofloxacin were skin rash and epigastric soreness in two cases, respectively. There were no deaths during the study. Conclusions : For low-risk febrile patients with neutropenia during cancer chemotherapy, switch therapy to oral ciprofloxacin at 72 hours following intravenous broad-spectrum antibiotics is effective and safe,
Facet이론을 이용한 지진발생시 피난 행동심리에 관한 연구

The purpose of this study is to examine and analyze the level of awareness in the ability to evacuate in the case of collapse of a building and the resulting long term damage by using the behavioral and psychological system that can occur in a building in an earthquake that takes place when least expected, and to present a solution. In the main body of this study, the Facet theory used to limit, collect, and simplify the multilateral scope of human psychological system shown through results of a questionnaire. The values that were logged and extracted were interpreted using three analysis techniques. 1. Integrative behavior and psychological analysis using the Facet Theory 2. Sequential behavior and psychological analysis using the Fact Theory 3. MDS analysis based on factor analysis of the Facet Theory By using the three methods of analysis above, multilateral behavior and psychology toward earthquake was analyzed from multiple angles, and reached the final conclusion. The tendency of relying on the first order instinct was found to be very high. Such psychological behavior can sufficiently be prevented and solved through training and simulation of earthquakes and the conclusion shows that national education system and training for personal evacuation is much needed.

Calmodulin (CaM) is a ubiquitous Ca^(2+)-binding protein known to regulate diverse cellular functions by modulating the activity of various target proteins. We isolated a cDNA encoding AtWRKY7, a novel CaM-binding transcription factor, from an Arabidopsis expression library with horseradish peroxidase-conjugated CaM. CaM binds specifically to the Ca^(2+)-dependent CaM-binding domain (CaMBD) of AtWRKY7, as shown by site-directed mutagenesis, a gel mobility shift assay, a split-ubiquitin assay, and a competition assay using a Ca^(2+)/CaM-dependent enzyme. Furthermore, we show that the CaMBD of AtWRKY7 is a conserved structural motif (C-motif) found in group Ⅱd of the WRKY protein family. ⓒ 2005 Federation of European Biochemical Societies.
