Plk 1 is overexpressed in many human malignancies including laryngeal carcinoma. However, its therapeutic potential has been never examined in laryngeal carcinoma. In the present study, a simple cellular morphology-based strategy was firstly proposed for rapidly screening the effective siRNAs against Plk1. Furthermore, we investigated the effects of Plk1 depletion via a novel identified effective siRNA against Plk1, Plk1 siRNA-607, on human laryngeal carcinoma Hep-2 cells. The results indicated that Plk1 siRNA-607 transfection resulted in a significant inhibition in Plk1 expression in cells, and subsequently caused a dramatic mitotic cell cycle arrest followed by massive apoptotic cell death, and eventually resulted in a significant decrease in growth and viability of the laryngeal carcinoma cells. Taken together, our present study not only suggests a simple strategy for rapidly screening effective siRNAs against Plk1 but also implicates that Plk1 may serve as a potential therapeutic target in human laryngeal carcinoma. In this study, we investigated the antifibrotic effects of Rha on letrozole-induced PCOS in rats. Rha treatment restored letrozole-induced alterations in the body weight (BW), relative weights of the ovaries, and relative weights of uterine and visceral adipose tissues. Histological observation showed that Rha reduced the number of atretic follicles, cystic follicles, and fibrosis of the ovaries in the PCOS rats. Administration of Rha reduced letrozole-induced metabolic dysfunction by decreasing the serum levels of TG and TC and increasing the levels of HDL-C in the PCOS rats. Rha treatment also modulated the serum levels of sex hormones, which decreased the LH, E2 and T and increased the FSH in PCOS rats. Finally, Rha treatment appeared to increase the activity of PPAR-γ, reduce the activation of the TGF-β1/Smad pathway, and reduce the collagen I, α-SMA and CTGF levels in the ovaries of the PCOS rats. Our results demonstrated that Rha significantly ameliorated metabolic disturbances and ovarian fibrosis in the letrozole-induced PCOS rats. Rha may be an effective compound in preventing ovarian fibrosis in the future.

Finally, SA treatment appeared to increase the activity of PPAR-γ, reduce the activation of the TGF-β1/Smad pathway, and reduce the collagen I, α-SMA and CTGF levels in the ovaries of the PCOS rats. Our results demonstrated that SA significantly ameliorated ovarian fibrosis and abolic disturbances in the letrozole-induced PCOS rats.

B-32 is one of a panel of monoclonal anti-idiotypic antibodies to growth hormone (GH) that we developed. To characterize and identify its potential role as a novel growth hormone receptor (GHR) agonist, we determined that B-32 behaved as a typical $Ab2{\beta}$ based on a series of enzyme-linked immunosorbent assay assays. The results of fluorescence-activated cell sorting, indirect immunofluorescence and competitive receptor binding assays demonstrated that B-32 specifically binds to the GHR expressed on target cells. Next, we examined the resulting signal transduction pathways triggered by this antibody in primary porcine hepatocytes. We found that B-32 can activate the GHR and Janus kinase (2)/signal transducers and activators of transcription (JAK2/STAT5) signalling pathways. The phosphorylation kinetics of JAK2/STAT5 induced by either GH or B-32 were analysed in dose-response and time course experiments. In addition, B32 could also stimulate porcine hepatocytes to secrete insulin-like growth factors-1. Our work indicates that a monoclonal anti-idiotypic antibody to GH (B-32) can serve as a GHR agonist or GH mimic and has application potential in domestic animal (pig) production.

Input signals originating from baroreceptors and vestibular receptors are integrated in the rostral ventrolateral medulla (RVLM) to maintain blood pressure during postural movement. The contribution of baroreceptors and vestibular receptors in the maintenance of blood pressure following hypotension were quantitatively analyzed by measuring phosphorylated extracellular regulated protein kinase (pERK) expression and glutamate release in the RVLM. The expression of pERK and glutamate release in the RVLM were measured in conscious rats that had undergone bilateral labyrinthectomy (BL) and/or sinoaortic denervation (SAD) following hypotension induced by a sodium nitroprusside (SNP) infusion. The expression of pERK was significantly increased in the RVLM in the control group following SNP infusion, and expression peaked 10 min after SNP infusion. The number of pERK positive neurons increased following SNP infusion in BL, SAD, and BL+SAD groups, although the increase was smaller than seen in the control group. The SAD group showed a relatively higher reduction in pERK expression when compared with the BL group. The level of glutamate release was significantly increased in the RVLM in control, BL, SAD groups following SNP infusion, and this peaked 10 min after SNP infusion. The SAD group showed a relatively higher reduction in glutamate release when compared with the BL group. These results suggest that the baroreceptors are more powerful in pERK expression and glutamate release in the RVLM following hypotension than the vestibular receptors, but the vestibular receptors still have an important role in the RVLM.

The clay-based Fe-bearing catalyst was successfully prepared through ion-exchange reaction and applied as heterogeneous catalyst for discoloration of acid fuchsine (AF) in an aqueous solution by Fenton-like action.Experimental results demonstrated that the AF discoloration ratios increased by increasing Fe-loaded clay dosage and initial H2O2 concentration, and by decreasing the pH, respectively. The lower the initial AF concentration, the shorter the reaction time needed to achieve complete discoloration of AF. Comparative studies indicated that AF discoloration ratios were much higher in presence of Fe-loaded clay and H2O2 than those in presence of H2O2, raw natural clay or Fe-loaded clay only and raw natural clay and H2O2 jointly. After AF discoloration, there existed no new phases in the clay samples detected by XRD and no change in the clay crystal morphology observed by SEM. A mechanism proposed suggested adsorption and Fenton-like reaction were responsible for discoloration of AF.

Antioxidant and anti-inflammatory activities of methanolic extract of Clerodendrum cyrtophyllum Turcz (ECT) were investigated in the present study. ECT showed high phenolic content (223 μg・GAE-1 equivalent) and strong 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging (IC50 = 52.74 μg・mL-1) and reduced power ability. Furthermore, ECT exhibited significant protective ability in H2O2/Fe3+/ascorbic acid-induced DNA and protein damage. Moreover, ECT inhibited NO production by 47.15% at 0.8 mg・mL-1 in lipopolysaccharides-stimulated RAW264.7 macrophages. Semi-quantitative reverse transcription polymerase chain reaction (RT-PCR) analysis showed that the expression of iNOS and COX-2 in RAW264.7 cells was down-regulated after treatment with increasing concentration of ECT. In conclusion, ECT has shown potent antioxidant and anti-inflammatory activities could be considered as a source of health promoting antioxidant and complementary medicinal supplement.

Open Innovation gains a wide spread attention in technology management research in recent years. Because existing research about the relationship between outbound open innovation and innovation performance is limited and context factors are ignored in existing research and based on Chesbrough’s open innovation model and theory of enterprise competence, this study introduces enterprise competence and knowledge spillovers into the field of outbound open innovation, explores their moderated roles between open innovation and innovation performance. We use high-tech firms as research sample to test the hypothesis that we proposed and find that: (1) outbound open innovation has a positive effect on innovation performance; (2) both enterprise competence and knowledge spillovers moderate the relationship between outbound open innovation and innovation performance. These conclusions have important implications for the development and application of open innovation theory.
