        Employing rotating vaneless diffuser to enhance the performance of plenum fan

        Numerical simulation is carried out for flow characteristics in a plenum fan and the influence of the diameter ratio of the rotating vaneless diffuser on the performance of plenum fan is analyzed. The diameter ratio of the rotating vaneless diffuser employed is from 1.03 to 1.3. The research results show that the rotating vaneless diffuser is able to enhance the performance of plenum fan. It is found that there is significant improvement in static pressure and efficiency at the diameter ratio of 1.05 at high flow coefficients, while the optimal diameter ratio is 1.2 at rated and low flow coefficient.

        The white gene in Nilaparvata lugens and its expression pattern under two different survival stresses

        The brown planthopper (BPH), Nilaparvata lugens (Stål), is a serious pest of rice. Chemical control and pestresistant rice breeding are always two major management strategies of BPH. In order to keep effective control, understanding the detoxification mechanism of this insect pest appears to be high important. In addition to be an eye pigment transporter, White protein is also important for its non-eye roles, such as a potential target for insecticides and Bt toxin. In this study, we characterized the full-length white orthologues gene in N. lugens (Nlwhite). NlWhite, the deduced protein of Nlwhite, has feature motifs of the eye pigment transporter. Knocking down Nlwhite transcript, the compound eye color partially changed to white. Nlwhite can be detected in all developmental stages and body parts, indicating that it is not a developmental or tissue-specific expression gene. The expression levels of Nlwhite were all significantly upregulated in both pymetrozine-selection and Mudgoselection BPH strains, which were selected for several generations. These results indicated that NlWhite participated in the detoxification not only for the exogenous insecticides but also for the endogenous toxic substance.

        Comparison of Transforming Growth Factor-Beta1 and Lovastatin on Differentiating Mesenchymal Stem Cells toward Nucleus Pulposus-like Phenotype: An In Vitro Cell Culture Study

        Study Design: In vitro cell culture study. Purpose: This study aims to investigate the impact of transforming growth factor-beta1 (TGF-β1) and lovastatin on differentiating human mesenchymal stem cells (MSCs) toward nucleus pulposus (NP)-like phenotype. Overview of Literature: MSCs offer a cell source to the cell-based therapy for intervertebral disc degeneration. TGF-β1 is used to induce MSCs to differentiate into NP-like cells; however, an undesired expression of collagen type I has been reported. Statins reportedly stimulate expression of bone morphogenetic protein-2 (BMP-2) and promote the chondrogenic phenotype to NP cells. However, the effects of statins with or without TGF-β1 on the differentiation of MSCs into NP-like cells remain unclear. Methods: Human MSCs were treated with TGF-β1 alone, lovastatin alone, and simultaneous or sequential treatment with TGF-β1 and lovastatin. After the proposed stimulation, the total RNA was extracted to assess the expression profile of NP cells-specific genes. Hematoxylin–eosin staining was used for examining the microscopic morphology. Furthermore, we detected the syntheses of S-100 protein, aggrecan , and collagen type II in the extracellular matrix using immunohistochemical staining. Results: Simultaneous or sequential treatment of TGF-β1 and lovastatin could further augment the BMP-2 overexpression compared with lovastatin-alone treatment. However, the mRNA expression of aggrecan and collagen type II was not compatible with the expression level of BMP-2 . Immunohistochemical studies revealed compatible production of aggrecan, collagen type II , and S-100 protein in all three groups treated with lovastatin. Cells in groups treated with lovastatin were less populated than that in the group treated with TGF-β1 alone. Conclusions: This study demonstrates a promising role of lovastatin in inducing human MSCs into NP-like cells. However, further optimization of cell density before lovastatin treatment, treatment duration, and combination with TGF-β1 are warranted to attain better stimulatory effects.

        Knockdown of an ABC transporter leads to bright red eyes in the brown planthopper, Nilaparvata lugens (Stål) (Hemiptera: Delphacidae)

        Compound eye color is an important biological character of insects,which is determined by the nature of eye pigments. Ommochrome is the solely source of eye color for some insects, while pteridines is also needed for the other insect species. However, little is known about the eye pigment composition for any planthopper. Scarlet is an ABC (ATP-binding cassette) transporter protein, which functions as the transmembrane transporter for ommochrome precursor. The failure of Scarlet function can cause bright red or white eyes in different species, which depending on the nature of eye pigments. Here, we identified a scarlet ortholog gene (Nlst) from the brown planthopper (BPH), Nilaparvata lugens, which is a destructive insect pest of rice. Nlst is the first characterized eye pigment transporter gene from Hemipteran. NlSt, the protein deduced from Nlst, has an open reading frame (ORF) of 629 amino acidswith the two sequence logos for eye pigment transporters. Expression profile revealed that Nlst expressed at all development stages and had the highest transcript level in head. Knockdown the Nlst transcript, the wild-type eye color partially changed to bright red, while not to white. Meantime, the ommochrome level in heads reduced to 73.4%. These results suggested that the eye coloration of BPH needs both ommochrome and pteridines pigments. Because nymphal RNAi with Nlst leading to a clearly distinguished phenotype from the control individuals, Nlst maybe a suitable genetic marker to exploit embryonic RNAi technique in this insect pest.

        Identification and Characterization of the Antifungal Substances of a Novel Streptomyces cavourensis NA4s

        A new actinomycete strain NA4 was isolated from a deep-sea sediment collected from the South China Sea and showed promising antifungal activities against soilborne fungal pathogens. It was identified as Streptomyces cavourensis by morphological, physiological, and phylogenetic analyses based on its 16S rRNA gene sequence. The main antifungal components were isolated and identified from the fermentation culture as bafilomycins B1 and C1. These compounds exhibited significant antifungal activities and a broad antifungal spectrum. The results suggest that the Streptomyces cavourensis NA4 and bafilomycins B1 and C1 could be used as potential biocontrol agents for soilborne fungal diseases of plants.

        An instability criterion for viscoelastic flow past a confined cylinder

        It has been known that there is a viscoelastic instability in the channel flow past a cylinder at high Deborah (De) number. Some of our numerical simulations and a boundary layer analysis indicated that this instability is related to the shear flow in the gap between the cylinder and the channel walls in our previous work. The critical condition for instability initiation may be related to an inflection velocity profile generated by the normal stress near the cylinder surface. At high De, the elastic normal stress coupling with the streamline curvature is responsible for the shear instability, which has been recognized by the community. In this study, an instability criterion for the flow problem is proposed based on the analysis on the pressure gradient and some supporting numerical simulations. The critical De number for various model fluids is given. It increases with the geometrical aspect ratio h/R (half channel width/cylinder radius) and depends on a viscosity ratio β (polymer viscosity/total viscosity) of the model. A shear thinning first normal stress coefficient will delay the instability. An excellent agreement between the predicted critical Deborah number and reported experiments is obtained.

        Theory and Prediction of Turbulent Transition

        We have proposed a new approach based on energy gradient concept for the study of flow instability and turbulent transition in parallel flows in our previous works. It was shown that the disturbance amplitude required for turbulent transition is inversely proportional to Re, which is in agreement with the experiments for imposed transverse disturbance. In present study, the energy gradient theory is extended to the generalized curved flows which have much application in turbomachinery and other fluid delivery devices. Within the frame of the new theory, basic theorems for flow instability in general cases are provided in details. Examples of applications of the theory are given from our previous studies which show comparison of the theory with available experimental data. It is shown that excellent agreement has been achieved for several configurations. Finally, various prediction methods for turbulent transition are reviewed and commented.

        Blend fibers of chitosan–agarose by electrospinning

        <P><B>Abstract</B></P><P>A novel fibrous system, chitosan/agarose, was successfully developed via the electrospinning of their blend solutions. The trifluoroacetic acid (TFA)/dichloromethane (DCM) mixture was used as a suitable solvent for both chitosan and agarose. It was found that smooth, continuous fibers were generated at an agarose content of less than 50%. Moreover, with the increase of the agarose content from zero to 50%, the average diameter of the fibers was significantly decreased from about 1.76 to 0.14μm. The FT-IR analysis revealed a gradual shift of the absorption band in the blend fibers, suggesting the strong interaction and good compatibility between chitosan and agarose.</P>
