Metastasis associated genomic aberrations in stage II rectal cancer

Genomic aberrations of rectal carcinoma, especially DNA copy number changes associated with metastasis were largely unclear. We aim to identify the metastasis associated biomarkers in stage II rectal cancer. Formalin-fixed, paraffin-embedded primary tumor tissues of stage II rectal carcinoma were analyzed by array-based comparative genomic hybridization, and genomic aberrations were identified by Genomic Workbench and SAM software. Copy number changes and mRNA expressions were validated by Real-time PCR in an independent rectal cancer samples. The results showed that the most frequent gains in stage II rectal cancer were at 1q21.2-q23.1, 3p21.31, 11q12.2-q23.3, 12q24.11-q24.31, 12q13.11-q14.1 and losses in 18q11.2-q23, 17q21.33-q22, 13q31.1-q31.3, 21q21.1-q21.3, 8p23.3-p23.1 and 4q22.1-q23. Twenty-two amplifications and five homozygous deletions were also identified. We further found that S100A1 (1q21.3-q23.1), MCM7 (7q22.1) and JUND (19p13.11) were amplified and overexpressed in stage II rectal cancer. Interestingly, the genomic aberrations affected 14 signaling pathways including VEGF signaling pathway and fatty acid metabolism. Most importantly, loss of 13q31.1-q34 and gain of 1q44 were associated with distant metastasis. Our results indicated that these metastasis associated genomic changes may be useful to reveal the pathogenesis of rectal cancer metastasis and identify candidate biomarkers.
Ozone Synthesis and Regeneration of DPF using Dielectric Barrier Discharges

Non-thermal plasma technology has many applications in various areas. One of the applications is regenerating diesel particulate filter (DPF). DPF is a widely applied device to control the particulate emission of diesel engines. However, clogging of the DPF occurs after hours of operation resulting in reduced engine efficiency. In order to solve this problem carbon soot collected inside the DPF should be cleaned out periodically. Conventional high-temperature removal processes easily leads to the breakage of DPF. Herein, low-temperature plasma formed in a dielectric-barrier discharge(DBD) reactor was used to form active oxidants such as ozone and monatomic oxygen. Two types of plasma reactors: surface DBD and volume DBD were studied. Experimentally, the effects of discharge power and frequency on the performance of DBD reactors were studied. It has found that design of surface DBD used in this study is much more efficient than volume DBD. Regeneration of the DPF was carried out with oxygen flow rate 3.5 L/min; with applied discharge power 19 W using volume DBD. It could remove 2.1g of carbon soot after a continuous regeneration for 2.5 hrs.

Objective Coronavirus disease-2019 (COVID-19) pandemic-related psychological symptoms can lead to smartphone addiction (SPA) risk and other behavioral disorders, thus impacting individuals’ mental health and well-being. The present study aims to propose a mediation model to investigate the association between emotional intelligence (EI) and SPA, and the mediating role of future anxiety (FA) during the post-COVID-19 era. Methods An online questionnaire including the Emotional Intelligence Scale, the Dark Future Scale, and the Smartphone Addiction Scale among university students from China, was conducted between September 14 and November 22, 2022. Finally, 1,154 valid questionnaires were collected. The reliability and confirmatory factor analysis results showed that all three scales had good reliability and validity. Results Structural Equation Model demonstrated that EI significantly and negatively influenced SPA (β=0.211, p<0.001), university students’ FA significantly and positively effected SPA (β=0.315, p<0.001), EI significantly predicted SPA in university students, and FA partially mediated the association between EI and SPA. The mediation effect of FA was 0.110, which accounted for 34.27% of the total effect. Bootstrap results furthermore tested the rigor of the mediating effect. Conclusion These findings broaden our understanding regarding the relationship between EI and SPA and the mediating role of FA, providing new sights for educators on how to reduce the risk of SPA when confronting the ongoing and possible future pandemics.

Intestinal enterochromaffin (EC) cell hyperplasia and increased 5-hydroxytryptamine (5-HT) availability play key roles in the pathogenesis of abdominal hypersensitivity of irritable bowel syndrome (IBS). This study aims to study the effect of quercetin on visceral pain and 5-HT availability in postinflammatory IBS (PI-IBS) rats. PI-IBS model rats were administered quercetin by gavage at doses of 5, 10, and 20 mg/kg for 14 days. Compared with normal rats, the visceral pain threshold of PI-IBS rats was markedly decreased and the abdominal motor response to colon distension was markedly increased. The EC cell count and 5-HT level, as well as tryptophan hydroxylase (TPH) protein, were all significantly elevated in PI-IBS rats, while the 5-HT reuptake transporter (serotonin transporter) was reduced. Genes that are responsible for enteroendocrine cell differentiation, that is, Ngn3 and pdx1, were significantly increased in the PI-IBS group. Quercetin treatment markedly elevated the pain threshold pressure and decreased the visceral motor response of PI-IBS animals; and EC cell density and 5-HT level, as well as TPH expression, in the PI-IBS group were all reduced by quercetin. Quercetin treatment also significantly reduced colonic expression of Ngn3 and pdx1 of PI-IBS. Findings from the present study indicated that the analgesic effect of quercetin on PI-IBS may result from reduction of 5-HT availability in the colon, and the regulatory role of quercetin in endocrine progenitors may contribute to reduced EC cells.

Two isocoumarin derivatives, stoloniferol A (1) and B (2), a known $5{\alpha}$, $8{\alpha}-epidioxy-23-methyl-(22E, 24R)-ergosta-6$, $22-dien-3{\beta}-ol$ (3), and a known dihydrocitrinone (4) were isolated from the ethyl acetate extract of the sea squirt-derived fungus, Penicillium stoloniferum QY2-10, and a halophilic fungus, Penicillium notatum B-52, respectively. Their structures were elucidated by spectroscopic methods and optical rotation. The stereochemistry of 2 was determined on the basis of different NOE experiments and chemical transformation. Compound 3 showed cytotoxicity against P388 cells, with an $IC_{50}$ value of 4.07 ${\mu}M$.

Aims: Late recurrence (> 2 years) after liver resection of hepatocellular carcinoma (HCC) is usually considered as multi-centric tumors or de novo cancer formation. We aimed to investigate risk factors, patterns and outcomes of late recurrence after HCC resection. Methods: From a multicenter database from 2001 to 2015, 734 patients who were alive and recurrence-free at 2 years after curative resection of initial HCC were enrolled into this retrospective study. Univariate and multivariate Cox-regression analysis were used to identify independent risk factors of late recurrence. Patterns, treatments and outcomes of late recurrence were investigated and analyzed. Results: During a median follow-up of 78.0 months after surgery, 303 patients (41.3%) developed late recurrence. Multivariate analysis revealed that cirrhosis, macroscopic vascular invasion, satellites, and tumor size > 5cm were independent risk factors of late recurrence. Among them, 273 (90.1%) were sole intrahepatic recurrence, 30 (9.9%) were concurrent intrahepatic and extrahepatic recurrence, and none of them was sole extrahepatic recurrence; 165 (54.4%) patients received curative treatments for recurrent HCC, including re-resection, transplantation and local ablation. Multivariate analysis showed regular postoperative surveillance and receiving curative treatments were two independent protective factors of prolonging survival for those patients with late recurrence. Conclusions: Late recurrence is correlated with cirrhosis and certain tumor-related characteristics of initial HCC. The patterns of late recurrence suggest that postoperative surveillance after 2 years of surgery could be adjusted and more targeted. Regular postoperative surveillance improves the probability to receive curative treatments again, yielding to better outcomes for patients with late recurrence.
The Effect of Mn/Al Substitution on the Structural Stability and Magnetic Properties of Mn₃AlC

The structural stability and magnetic properties of Mn3+xAl1-xC antiperovskite with varied Mn/Al substitution were studied systematically. Single phase Mn3+xAl1-xC alloys with antiperovskite structure were obtained in samples with x = −1/4, 0, 1/4, 1/2. An additional Mn23C₆ phase was precipitated from Mn3+xAl1-xC antiperovskite for x = 3/4 while Mn23C₆ phase was formed as major phase for x = 1. The mutual substitution of Mn and Al atoms has substantial effect on the Curie temperature and the saturation magnetization of the Mn3+xAl1-xC alloys. In comparison with the as-cast alloys, the as-annealed Mn3+xAl1-xC alloys exhibit reduced Mn/Al substitutions after high temperature homogenization, which enhances the ordering of Mn and Al atoms in the lattices. The Curie temperature of the homogenized Mn3+xAl1-xC increases with increasing Mn substitution to Al. The Mn₃AlC alloy shows the highest saturation magnetization among all samples with varied Mn/Al ratios. Most samples show zero coercivity and zero remanent magnetization. The maximum value of the magnetic entropy changes of Mn2.75Al1.25C at 285 K is 2.26 J/㎏ K in fields up to 3 T.
Causal Relations between Exposome and Stroke: A Mendelian Randomization Study

Background and Purpose To explore the causal relationships of elements of the exposome with ischemic stroke and its subtypes at the omics level and to provide evidence for stroke prevention. Methods We conducted a Mendelian randomization study between exposure and any ischemic stroke (AIS) and its subtypes (large-artery atherosclerotic disease [LAD], cardioembolic stroke [CE], and small vessel disease [SVD]). The exposure dataset was the UK Biobank involving 361,194 subjects, and the outcome dataset was the MEGASTROKE consortium including 52,000 participants. Results We found that higher blood pressure (BP) (systolic BP: odds ratio [OR], 1.02; 95% confidence interval [CI], 1.01 to 1.04; diastolic BP: OR, 1.03; 95% CI, 1.01 to 1.05; pulse pressure: OR, 1.03; 95% CI, 1.00 to 1.06), atrial fibrillation (OR, 1.18; 95% CI, 1.13 to 1.25), and diabetes (OR, 1.13; 95% CI, 1.07 to 1.18) were significantly associated with ischemic stroke. Importantly, higher education (OR, 0.69; 95% CI, 0.60 to 0.79) decreased the risk of ischemic stroke. Higher systolic BP (OR, 1.06; 95% CI, 1.02 to 1.10), pulse pressure (OR, 1.08; 95% CI, 1.02 to 1.14), diabetes (OR, 1.28; 95% CI, 1.13 to 1.45), and coronary artery disease (OR, 1.58; 95% CI, 1.25 to 2.00) could cause LAD. Atrial fibrillation could cause CE (OR, 1.90; 95% CI, 1.71 to 2.11). For SVD, higher systolic BP (OR, 1.04; 95% CI, 1.00 to 1.07), diastolic BP (OR, 1.06; 95% CI, 1.01 to 1.12), and diabetes (OR, 1.22; 95% CI, 1.10 to 1.36) were causal factors. Conclusions The study revealed elements of the exposome causally linked to ischemic stroke and its subtypes, including conventional causal risk factors and novel protective factors such as higher education.
중국 수출시장 진출을 위한 한방화장품 포장디자인 연구 - 경북 한방화장품을 중심으로 -

연구배경 경북 한방화장품은 대기업의 꾸준한 화장품 성장 시장과는 달리 제품의 질적 수준이 우수함에도 불구하고 수준 낮은 포장디자인과 마케팅 부족으로 저가화장품 이미지를 벗어나지 못하고 있어 국내 소비자 뿐 만 아니라 해외 소비자 시장 개척이 힘들어 많은 경북 한방화장품 중소기업들이 어려움을 겪고 있다. 따라서 경북 한방화장품이 중국을 중심으로 해외시장으로 진출하기 위해 브랜드별 시각적 조형요소 분석을 통해 브랜드이미지 구축을 위한 방향을 설정하고 중국 소비자 욕구에 맞는 포장디자인 개발 방향을 제시하고자 한다. 연구방법 연구범위는 국내 생산 한방화장품 중 경북에서 생산되는 한방화장품 8개 브랜드로 제한하였으며, 연구방법으로는 국내외 문헌을 통하여 이론적 근거를 정립하였고 한방화장품 업계의 시장조사 자료 및 한방화장품 전문자료, 경북 한방화장품 포장디자인 사례 등을 비교 분석하여 정리 하였다. 또한 심층적인 연구를 위해 국내 거주 중국인 100명을 대상으로 경북 한방화장품 포장디자인 이미지에 대한 설문조사를 통해 그에 따른 결과 및 시사점을 제시하였다. 연구결과 본 연구의 결과 경북 한방화장품이 중국 시장 진출을 위해서는 제품이 갖는 기능성은 물론이고 보다 해외 소비자들의 눈높이에 맞는 브랜드 포장디자인 개발이 시급한 것으로 나타났다. 이러한 현상은 경북 한방화장품 생산업체들이 브랜드 포장디자인에 대한 인식 부족하며 전문 디자이너 부족에서 나오는 현상으로 보다 체계적인 디자인 개발이 요구되고 있다. 결론 경북 한방 화장품이 중국 수출 시장으로 진출하기 위해서는 보다 철저한 중국시장의 문화를 이해하는 연구가 필요할 것이다. 특히 중국은 전 세계 화장품 시장의 절반을 차지하는 만큼 해외 시장 개척을 위해서는 피할 수 없는 시장으로 식지 않는 한류를 배경으로 보다 체계적이고 전문 디자이너들에 의한 디자인 개발이 필요하다. Background In terms of oriental medicine cosmetics in the Gyeongbuk region, many SMEs are experiencing difficulty in sales due to low domestic demand and the global economic recession. Therefore, in order for Gyeongbuk oriental medicine to enter overseas markets especially targeting China and Southeast Asia, this study aims to set directions for building brand images by analyzing elements of the visual designs of various brands and to provide direction regarding packaging design that will meet the needs of overseas consumers. Methods The scope of this study was limited to eight oriental medicine cosmetics brands produced in the Gyeongbuk area among all products manufactured in Korea. The theoretical basis of this research was established from domestic and foreign literature, and market research data from the oriental cosmetics industry and professional material on oriental medicine cosmetics, and examples of packaging design of oriental medicine in the Gyeongbuk area were compared, analyzed, and organized. Also, for further research, surveys of 100 Chinese residents living in Korea were conducted about the packaging design images of Gyeongbuk oriental medicine cosmetics. The results and implications are provided in this study. Result The study found that in order for Gyeongbuk oriental medicine cosmetics to enter overseas markets, not only is the function of the cosmetics important but the development of brand packaging design that satisfies the eyes of foreign consumers is urgent. The urgency rises from a lack of awareness of Gyeongbuk oriental medicine cosmetics manufacturers on brand packaging design and a lack of expert designers, and a systematic development of design is in need. Conclusion For Gyeongbuk oriental medicine cosmetics to enter the overseas export market, studies on understanding the culture of the overseas market more thoroughly should be conducted. As the market in China accounts for as much as half of the global cosmetics market, it is a market that cannot be neglected. With the aid of the still popular Korean wave, a systematic design created by professional designers needs to be developed.
