Extracellular vesicles (EVs) play an essential role in the communication between cells and the tumor microenvironment. However, the effect of tumor-derived EVs on the growth and metastasis of lung adenocarcinoma (LUAD) remains to be explored. This study aimed to elucidate the role of miR-153-3p-EVs in the invasion and migration capabilities of LUAD cells and explore its mechanism through in vivo and in vitro experiments. We found that miR-153-3p was specifically and highly expressed in LUAD and its secreted EVs. Furthermore, the expression of BANCR was negatively regulated by miR-153-3p and identified as a target gene of miR-153-3p using luciferase reporter assays. Through further investigation, we found that the downregulation of BANCR activates the PI3K/AKT pathway and accelerates the process of epithelial-mesenchymal transition (EMT), which ultimately leads to the aggravation of LUAD. The orthotopic xenograft mouse model was established to illustrate the effect of miR-153-3p-EVs on LUAD. Animal studies showed that miR-153-3p-EVs accelerated tumor growth in mice. Besides, we found that miR-153-3p-EVs could damage the respiratory ability of mice and produce a mass of inflammatory cells around the lung tissue of mice. Nevertheless, antagomir-153-3p treatment could inhibit the deterioration of respiratory function and inhibit the growth of lung tumors in mice. In conclusion, our study reveals the potential molecular mechanism of miR-153-3p-EVs in the development of LUAD and provides a potential strategy for the treatment of LUAD.

Streptomyces sp. 139 generates a novel exopolysaccharide (EPS) designated as Ebosin, which exerts an antagonistic effect on IL-1R in vitro and anti-rheumatic arthritis activity in vivo. A ste gene cluster for Ebosin biosynthesis consisting of 27 ORFs was previously identified in our laboratory. In this paper, ste16 was expressed in Escherichia coli BL21 and the recombinant protein was purified, which has the ability to catalyze the transfer of the methyl group from S-adenosylmethionine (AdoMet) to dTDP-4-keto-6-deoxy-D-glucos, which was thus identified as a methyltransferase. In order to determine the function of ste16 in Ebosin biosynthesis, the gene was disrupted with a double crossover via homologous recombination. The monosaccharide composition of EPS-m generated by the mutant strain Streptomyces sp. 139 (ste16-) was found to differ from that of Ebosin. The IL-1R antagonist activity of EPS-m was markedly lower than that of Ebosin. These experimental results have shown that the ste16 gene codes for a methyltransferase which is involved in Ebosin biosynthesis.

Members of the genus Bacillus are known to play an important role in promoting plant growth and protecting plants against phytopathogenic microorganisms. In this study, 21 isolates of Bacillus spp. were obtained from the root micro-ecosystem of Suaeda glauca. Analysis of the 16S rRNA genes indicated that the isolates belong to the species Bacillus amyloliquefaciens, Bacillus velezensis, Bacillus subtilis, Bacillus pumilus, Bacillus aryabhattai and Brevibacterium frigoritolerans. One of the interesting findings of this study is that the four strains B1, B5, B16 and B21 are dominant in rhizosphere soil. Based on gyrA, gyrB, and rpoB gene analyses, B1, B5, and B21 were identified as B. amyloliquefaciens and B16 was identified as B. velezensis. Estimation of antifungal activity showed that the isolate B1 had a significant inhibitory effect on Fusarium verticillioides, B5 and B16 on Colletotrichum capsici (syd.) Butl, and B21 on Rhizoctonia cerealis van der Hoeven. The four strains grew well in medium with 1-10% NaCl, a pH value of 5-8, and promoted the growth of Arabidopsis thaliana. Our results indicate that these strains may be promising agents for the biocontrol and promotion of plant growth and further study of the relevant bacteria will provide a useful reference for the development of microbial resources.
Improvement of Tracking Performance Using Prediction-Based Algorithms for a Maneuvering Target

This paper presents a study involving prediction of a complicated maneuvering target, with the aim of improving the tracking performance of a fire control system (FCS). In this study, we predict the position of a complicated maneuvering target 5 s in advance using the information up to the current time. Because of the large error caused by the complicated maneuvers and the long prediction time in-terval, the mechanical system of the fire control system will take a heavy load. In order to cope with this problem, several approaches to decreasing the prediction error have been proposed including the prediction algorithms based on the multiple model(MM) filter, interacting multiple model (IMM) filter, and variable dimension with input estimation (VDIE) filter. Finally, comparative simulation results are presented to verify the performance of the filters.

Exposure to cadmium (Cd) induces apoptosis in osteoblasts (OBs); however, little information is available regarding the specific mechanismsof Cd-induced primary rat OB apoptosis. In this study, Cd reduced cell viability, damaged cell membranes and induced apoptosis in OBs. We observed decreased mitochondrial transmembrane potentials, ultrastructure collapse, enhanced caspase-3 activity, and increasedconcentrations of cleaved PARP, cleaved caspase-9 and cleaved caspase-3 following Cd treatment. Cd also increased the phosphorylationof p38-mitogen-activated protein kinase (MAPK), extracellular signal-regulated kinases (ERK)1/2 and c-jun N-terminal kinase (JNK) in OBs. Pretreatment with the caspase inhibitor, N-benzyloxycarbonyl-Val-Ala-Asp-fluoromethylketone, ERK1/2 inhibitor (U0126), p38 inhibitor(SB203580) and JNK inhibitor (SP600125) abrogated Cd-induced cell apoptosis. Furthermore, Cd-treated OBs exhibited signs of oxidativestress protection, including increased antioxidant enzymes superoxide dismutase and glutathione reductase levels and decreased formationof reactive oxygen species. Taken together, the results of our study clarified that Cd has direct cytotoxic effects on OBs, which are mediatedby caspase- and MAPK pathways in Cd-induced apoptosis of OBs.

Aims: Hepatocellular carcinoma (HCC) is a leading cause of cancer-related deaths worldwide. The metabolic biochemical pathway disturbances associated with HCC progression and recurrence remain unsatisfactorily characterized. Dysregulated metabolism is a hallmark of cancer including HCC, manifested through alterations in metabolites. The emerging field of metabolomics has increased the hope of discovering metabolites and biochemical pathways as the novel biomarkers or therapeutic targets of HCC and tracking tumor recurrence. Methods: Metabolomic profiling was conducted with the Metabolon platform for one hundred and twenty human recurrent/non-recurrent HCC tumor/normal paired tissues. Standard statistical analyses are performed in the Partek Genomics Suite on log transformed data. Principal component analysis (PCA) was conducted using all and dysregulated metabolites. Random Forest classification was applied to compare tumor vs. adjacent margin tissues and recurrent vs. non-recurrent tumors using named metabolites. Results: We found that HCC recurrence is characterized by increasing glycolysis and glutaminolysis. Tumor recurrence was significantly associated with increasing of glucose, pyruvate, α-ketoglutarate and NAD+/NADH metabolites to increase glycolysis and glutaminolysis. Higher acylcarnitine levels in non-recurrent HCC indicate elevated fatty acid β-oxidation. Besides, we also found that Coenzyme A levels were higher and plasmalogen lipids were uniformly lower in recurrent HCC. Conclusions: Our study generates a comprehensive metabolomics dataset on HCC and highlight the massive reorganization of cellular metabolism as tumors progress and acquire recurrence feature.

This paper presents an improved method for predicting the bearing capacity of single jacked pile based on piezocone penetration test (CPTu) data which contain tip resistance (qt), side friction ( fs) and excess dynamic pore water pressure (u2). Firstly, the average value of qt was determined to calculate the pile unit base resistance according to the influence zone of the logarithmic spiral concerning the effect of soil types. Secondly, fs and u2 were used to calculate the pile unit shaft resistance considering the effect of friction fatigue. The pile jacking force was obtained in terms of unit base resistance and unit shaft resistance. The ultimate bearing capacity was finally determined using pile jacking force multiplied by a force ratio denoted as Rf. The ultimate bearing capacities calculated for two case projects show a good agreement with the measured results, which indicates that the proposed method is feasible and effective. Reliability assessment was carried out on the proposed method and other five estimation methods with respect to the reliability index obtained by advanced first-order second-moment (AFOSM) method. Comparison result showed that the method proposed in this study has a higher reliability index over other five existing methods. Therefore, the proposed method can be used as a reliable alternative to predict the bearing capacity in the engineering practice of jacked pile foundation.

Methylenetetrahydrofolate reductase (MTHFR) plays a central role in folate metabolism. This study with 381 esophageal cancer patients and 432 healthy controls was conducted to examine the association of MTHFR C677T and A1298C polymorphisms with susceptibility to esophageal cancer (EC) in a Chinese population. Compared with the CC genotype of MTHFR C677T, subjects carrying homozygote TT and variant genotypes (CT+TT) demonstrated reduced risk of EC with adjusted ORs (95% CI) of 0.44 (0.28-0.71) and 0.57 (0.37-0.88), respectively. However, no association was found between the MTHFR A1298C polymorphism and the risk of EC. Comparing to haplotype CA, haplotypes TA and TC could reduce the susceptibility to EC with adjusted ORs (95% CI) of 0.61(0.47-0.79) and 0.06 (0.01-0.43), respectively. In conclusion, the present study suggested that the MTHFR C677T polymorphism can markedly influence the risk of EC in Chinese.
