        Realgar transforming solution suppresses angiogenesis and tumor growth by inhibiting VEGF receptor 2 signaling in vein endothelial cells

        Realgar (As4S4), as an arsenic sulfide mineraldrug, has a good therapeutic reputation for anticancer inTraditional Chinese Medicine, and has recently beenreported to inhibit angiogenesis in tumor growth. However,considering the poor solubility and low bioavailability ofrealgar, large dose of realgar and long period of treatmentare necessary for achieving the effective blood medicineconcentration. In present study, we resolved the crucialproblem of poor solubility of realgar by using intrinsicbiotransformation in microorganism, and investigatedunderlying mechanisms of realgar transforming solution(RTS) for antiangiogenesis. Our results demonstrated thatRTS had a strong activity to inhibit HUVECs proliferation,migration, invasion, and tube formation. Moreover, RTSinhibited VEGF/bFGF-induced phosphorylation ofVEGFR2 and the downstream protein kinases includingERK, FAK, and Src. In vivo zebrafish and chickenchorioallantoic membrane model experiments showed thatRTS remarkably blocked angiogenesis. Finally, comparedwith the control, administration of 2.50 mg/kg RTSreached more than 50% inhibition against H22 tumorallografts in KM mice, but caused few toxic effects in thehost. The antiangiogenic effect was indicated by CD31immunohistochemical staining and alginate-encapsulatedtumor cell assay. In summary, our findings suggest thatRTS inhibits angiogenesis and may be a potential drugcandidate in anticancer therapy.

        Ginsenoside Ro, an oleanolic saponin of Panax ginseng, exerts antiinflammatory effect by direct inhibiting toll like receptor 4 signaling pathway

        Background: Panax ginseng Meyer (P. ginseng), a herb distributed in Korea, China and Japan, exerts benefits on diverse inflammatory conditions. However, the underlying mechanism and active ingredients remains largely unclear. Herein, we aimed to explore the active ingredients of P. ginseng against inflammation and elucidate underlying mechanisms. Methods: Inflammation model was constructed by lipopolysaccharide (LPS) in C57BL/6 mice and RAW264.7 macrophages. Molecular docking, molecular dynamics, surface plasmon resonance imaging (SPRi) and immunofluorescence were utilized to predict active component. Results: P. ginseng significantly inhibited LPS-induced lung injury and the expression of proinflammatory factors, including TNF-a, IL-6 and IL-1b. Additionally, P. ginseng blocked fluorescence-labeled LPS (LPS488) binding to the membranes of RAW264.7 macrophages, the phosphorylation of nuclear factor-kB (NF-kB) and mitogen-activated protein kinases (MAPKs). Furthermore, molecular docking demonstrated that ginsenoside Ro (GRo) docked into the LPS binding site of toll like receptor 4 (TLR4)/myeloid differentiation factor 2 (MD2) complex. Molecular dynamic simulations showed that the MD2-GRo binding conformation was stable. SPRi demonstrated an excellent interaction between TLR4/MD2 complex and GRo (KD value of 1.16 × 10<SUP>-9</SUP> M). GRo significantly inhibited LPS488 binding to cell membranes. Further studies showed that GRo markedly suppressed LPS-triggered lung injury, the transcription and secretion levels of TNF-α, IL-6 and IL-1β. Moreover, the phosphorylation of NF-kB and MAPKs as well as the p65 subunit nuclear translocation were inhibited by GRo dose-dependently. Conclusion: Our results suggest that GRo exerts anti-inflammation actions by direct inhibition of TLR4 signaling pathway.

      • Structure Analysis of 16S rDNA Sequences from Strains of Acidithiobacillus ferrooxidans

        Four strains of Acidithiobacillus ferrooxidans with different iron oxidation capacity were isolated from different mine drainage stations. The 16S rRNA gene of these strains were cloned and sequenced. Based on our sequences analysis on the four strain and the data on the other strains deposited in Genbank, all A. ferrooxidans may be classified into three phylogenetic groups. The analysis data showed that nucleotide variables (signature sites) were detected in 21 positions, and most of them were found in the first 800bp from 5' terminal except position 970 and 1375. Interestingly, the first 13 signature sites were located in two main regions:the first region (position 175-234) located in V2 while the second region (position 390-439) were detected in constant region between V2 and V3. Furthermore, the secondary structure and minimal free energy were determined in two regions among strains of three groups. These results may be useful in characterizing the microevolutionary mechanisms of species formation and monitoring in biohydrometallurgical application.

        Continuous Cooling Transformation Behavior and Impact Toughness in Heat-Affected Zone of Nb-Containing Fire-Resistant Steel

        Simulated heat-affected zone continuous cooling transformation diagram was developed for advanced fireresistantsteel. Over a wide range of cooling rates, corresponding to t8/5 from 6 s to 150 s, granular bainite was thedominant transformation constituent, while the morphology of less dominant martensite-austenite (M-A) constituentchanged from film-like to block-type constituent; but the hardness remained similar to the average value of 190-205 HV (0.2). The start and finish transformation temperature was high at 700 °C and 500 °C, and is different fromthe conventional high strength low alloy steels. It is believed that the high-content (0.09 wt%) of Nb may promotebainite transformation at relatively high temperatures. Martenistic matrix was not observed at high cooling rateand the film-like M-A constituent and blocky M-A constituent with thin film of retained austenite and lath martensitewere observed on slow cooling. Excellent impact toughness was obtained in the heat-affected zone with15-75 kJ/cm welding heat input.

        중국의 많은 역사지구는 수천년에 걸쳐 형성되었다. 역사 가구(街區)의 보존과 개발 간의 관계를 처리하기 위해서는 역사적인 ‘진실성’과 변화 과정에 대해 충분하고 완전한 이해가 필요하며 이를 바탕으로 적절한 경관관리계획을 수립할 수 있다. 복주 ‘삼방칠항(三坊七巷)’지구의 공간패턴은 당나라 후기 도시계획에 방리제를 도입하면서 시작되었으며 송·명 왕조시대를 거치면서 청나라 중기에 전성기를 이루었다. 이때부터 중정 형식의 전통주택과 ‘시란조(柴欄?)’목조 주택과 같은 독특한 주택이 나타날 뿐만 아니라 위대한 문화적 영향과 교육 제도를 완비하여 인재를 많이 배출하고 상업도 활발하게 이루어졌다. 그러나 1955년에서 21세기 초반까지 ‘삼방칠항’은 더 이상 발전하지 않았고 빈민가로 퇴락되었다. 2005년부터는 ‘삼방칠항’의 관리 전략을 ‘문화관광거리’ 모델에서 포괄적인 ‘커뮤니티 박물관’ 모델로 변경하면서 획기적인 변화를 모색하였지만 일부 문제는 여전히 존재하여 개선 방안이 필요하다. The formation of many historic districts in China could be traced back to millennia ago. To deal with the relationship between conservation and development of historical blocks, it is necessary to gain a full understanding of the “authenticity” and the change process of historic districts. On this basis, making appropriate landscape management decisions could be possible. The spatial pattern of Fuzhou “Three Lanes and Seven Alleys” district was established in the late Tang Dynasty by introducing Li-Fang system in city planning; after the accumulation in Song, Yuan and Ming Dynasties, this district reached the peak of development in the middle of Qing Dynasty. It was not only manifested in the emergence of characteristic dwellings, such as courtyard-style folk houses and the “Chai-lan-cuo” wooden house; but also in the great cultural influence, the formation of perfect education system, the talent people coming forth in large numbers and the prosperous commercial atmosphere. From 1955 to early twenty-first Century, “three lanes and Seven Alleys” were no longer brilliant, even once reduced to “shanty towns”. Since 2005, the management strategy of “San-fang Qi-xiang” have changed from one-side “Tourism Culture Street” model to a more comprehensive “community museum” model, but some problems still exist and need revamp.

        Role of miR-511 in the Regulation of OATP1B1 Expression by Free Fatty Acid

        MicroRNAs (miRNAs) are a family of non-coding RNA that are able to adjust the expression of many proteins, including ATP-binding cassette transporter and organic cation transporter. We sought to evaluate the effect of miR-511 on the regulation of OATP1B1 expression by free fatty acids. When using free fatty acids to stimulate Chang liver cells, we found that the expression of miR-511 increased significantly while the expression of OATP1B1 decreased. We also proved that SLCO1B1 is the target gene of miR-511 with a bioinformatics analysis and using the dual luciferase reporter assay. Furthermore, the expressions of SLCO1B1 and OATP1B1 decreased if transfecting Chang liver cells with miR-511, but did not increase when transfecting the inhibitors of miR-511 into steatosis cells. Our study indicates that miR-511 may play an important role in the regulation of OATP1B1 expression by free fatty acids.

      • Research on Peak-detection Algorithm for High-precision Demodulation System of Fiber Bragg Grating

        In order to improve the detection accuracy of wavelength, the filtering and curve fitting technologies were applied in the FBG wavelength demodulation system based on tunable F-P filter. These methods could realize the accurate peak-location of output signals of the photo detector. According to the characteristics of noise, the FIR low-pass filter was designed to filter the obtained light power signals so as to provide the input signals with high SNR for the peak-detection algorithms. By analyzing and comparing several typical peak-searching algorithms, the algorithm of Gauss formula nonlinear curve fitting (L-M) was chosen to fit the digitized light power signals. The experimental results show that L-M fitting algorithm reduces the mean square error by 7.5% compared with the Gauss fitting algorithm. For the Gauss signal in the wavelength demodulation system designed in the paper, the L-M algorithm has lower mean square error than other peak-searching algorithms. This algorithm is suitable for FBG wavelength demodulation system based on tunable F-P filter. It can efficiently raise the accuracy of wavelength demodulation system.

      • Association of Reduced Immunohistochemical Expression of E-cadherin with a Poor Ovarian Cancer Prognosis - Results of a Meta-analysis

        Purpose: E-cadherin is a transmemberane protein which is responsible for adhesion of endothelial cells. The aim of our study was to assess existing evidence of associations between reduced expression of E-cadherin and prognosis of ovarian cancer with a discussion of potential approaches to exploiting any prognostic value for improved clinical management. Methods: We conducted a meta-analysis of 9 studies (n=915 patients) focusing on the correlation of reduced expression of E-cadherin with overall survival. Data were synthesized with random or fixed effect hazard ratios. Results: The studies were categorized by author/year, number of patients, FIGO stage, histology, cutoff value for E-cadherin positivity, and methods of hazard rations (HR) estimation, HR and its 95% confidence interval (CI). Combined hazard ratios suggested that reduced expression of E-cadherin positivity was associated with poor overall survival (OS), HR= 2.10, 95% CI:1.13-3.06. Conclusion: The overall survival of the E-cadherin negative group with ovarian cancer was significant poorer than the E-cadherin positive group. Upregulation of E-cadherin is an attractive therapeutic approach that could exert significant effects on clinical outcome of ovarian cancer.

        Simulation Analysis and Experimental Verification for the Adhesive Strength Properties of Aluminum Foam Specimen for the Slip Mode

        In this study, the thickness was set as the variable and TDCB specimen was designed in order to investigate the adhesion strength characteristics of the slip mode made from aluminum foam. The dimensions of the models are the length of 200 mm and various thicknesses of 15 mm, 30 mm, 45 mm, and 75 mm. According to analysis results of the model with 15 mm thickness, when the forced displacement became about 5.3 mm, the maximum load appeared to be 188 N and a maximum equivalent stress of about 2.99 MPa occurred, respectively. According to the analysis results of specimens, when the propagation of the forced displacement is about 5.3 mm, a maximum reaction force of about 940 N appears whereas when the propagation of the forced displacement is about 22 mms, the reaction force almost disappears. When the trend of experiments and analysis results are applied to the four models used in this study, the adhesion strength of TDCB adhesive structure made from the aluminum foam of varying thickness can be evaluated. Therefore, it is expected that the structural safety can be anticipated by only an analysis without experiments in the actual adhesive structure.
