      • Hypoxia-Responsive MicroRNA-101 Promotes Angiogenesis <i>via</i> Heme Oxygenase-1/Vascular Endothelial Growth Factor Axis by Targeting Cullin 3

        <P>Aims: Hypoxia induces expression of various genes and microRNAs (miRs) that regulate angiogenesis and vascular function. In this study, we investigated a new functional role of new hypoxia-responsive miR-101 in angiogenesis and its underlying mechanism for regulating heme oxygenase-1 (HO-1) and vascular endothelial growth factor (VEGF) expression. Results: We found that hypoxia induced miR-101, which binds to the 3 ' untranslated region of cullin 3 (Cul3) and stabilizes nuclear factor erythroid-derived 2-related factor 2 (Nrf2) via inhibition of the proteasomal degradation pathway. miR-101 overexpression promoted Nrf2 nuclear accumulation, which was accompanied with increases in HO-1 induction, VEGF expression, and endothelial nitric oxide synthase (eNOS)-derived nitric oxide (NO) production. The elevated NO-induced S-nitrosylation of Kelch-like ECH-associated protein 1 and subsequent induction of Nrf2-dependent HO-1 lead to further elevation of VEGF production via a positive feedback loop between the Nrf2/HO-1 and VEGF/eNOS axes. Moreover, miR-101 promoted angiogenic signals and angiogenesis both in vitro and in vivo, and these events were attenuated by inhibiting the biological activity of HO-1, VEGF, or eNOS. Moreover, these effects were also observed in aortic rings from HO-1(+/-) and eNOS(-/-) mice. Local overexpression of miR-101 improved therapeutic angiogenesis and perfusion recovery in the ischemic mouse hindlimb, whereas antagomiR-101 diminished regional blood flow. Innovation: Hypoxia-responsive miR-101 stimulates angiogenesis by activating the HO-1/VEGF/eNOS axis via Cul3 targeting. Thus, miR-101 is a novel angiomir. Conclusion: Our results provide new mechanistic insights into a functional role of miR-101 as a potential therapeutic target in angiogenesis and vascular remodeling. Antioxid.       • Hepatoprotective and Antioxidative Activities of <i>Cornus officinalis</i> against Acetaminophen-Induced Hepatotoxicity in Mice

        <P>The fruit of <I>Cornus officinalis </I>Sieb. et Zucc. is commonly prescribed in Asian countries as a tonic formula. In this study, the hepatoprotective effect of ethanolic extracts of the fruit of <I>C. officinalis</I> (ECO) was investigated in a mouse model of acetaminophen- (APAP-) induced liver injury. Pretreatment of mice with ECO (100, 250, and 500 mg/kg for 7 days) significantly prevented the APAP (200 mg/kg) induced hepatic damage as indicated by the serum marker enzymes (AST, ALT, and LDH). Parallel to these changes, ECO treatment also prevented APAP-induced oxidative stress in the mice liver by inhibiting lipid peroxidation (MDA) and restoring the levels of antioxidant enzymes (SOD, CAT, and HO-1) and glutathione. Liver injury and collagen accumulation were assessed using histological studies by hematoxylin and eosin staining. Our results indicate that ECO can prevent hepatic injuries associated with APAP-induced hepatotoxicity by preventing or alleviating oxidative stress.</P>

      • Anti-Inflammatory Effect of By-Products from<i>Haliotis discus hannai</i>in RAW 264.7 Cells

        <P>Several reports promoted the potential of shellfish due to their ability to act as antioxidant, anti-inflammatory, and antimicrobial agents. Pacific abalone,<I>Haliotis discus hannai</I>viscera is, reported to possess bioactivities such as antioxidative stress and anti-inflammatory. In this study, anti-inflammatory potential of mucus-secreting glands from shell-shucking waste of<I>H. discus hannai</I>was evaluated using RAW 264.7 mouse macrophage cell model. Results indicated that presence of<I>H. discus hannai</I>mucosubstance by-products (AM) significantly lowered the nitric oxide (NO) production along the expressional suppression of inflammatory mediators such as cytokines TNF-<I>α</I>, IL-1<I>β</I>, and IL-6 and enzymes iNOS and COX-2. Also, AM was shown to increase expression of anti-inflammatory response mediator HO-1. Presence of AM also scavenged the free radicals<I>in vitro</I>. In conclusion, by-products of<I>H. discus hannai</I>are suggested to possess notable anti-inflammatory potential which promotes the possibility of utilization as functional food ingredient.</P>

      • Low Temperature Sintering and Electrical Properties of Sr₂Nb₂O_(7) Ceramics by the Chemical Coprecipitation Method

        Strontium niobate, Sr_(2)Nb_(2)O_(7), was prepared by the chemical coprecipitation method at low temperatures, using Sr_(2)Cl_(2)6H_(2)O, NbCI_(5) and HO_(2)CCO_(2)H. The foil nation of Sr_(2)Nb_(2)O_(7) occurred at 800℃ via an intermediate phase of composition, close to that of Sr_(5)Nb_(4)O_(15) at 700℃. The resulting powder was observed to have finer and a more uniform partical size distribution, as compared to those obtained through conventional methods, These powder characteristics allowed the use of a much lower sintering temperature of 1400℃, without sacrificing the desired dielectric and piezoelectric properties. Grain-orientation along <0k0> direction, which is a cause for improved electrical properties, was also observed.

        GPS에 의한 도로변형량 측정 적용

        본 연구에서는 향후 폭넓게 활용될 GPS에 의하여 도로의 변형량을 측정하고, 그 결과를 검증하기 위하여 EDM으로 관측한 결과와 비교하였으며, 관측대상지역은 경상남도 일원의 국도를 산정하였다. 첫째로 GPS의 측정 Mode중 Rapid Static Mode로 측정한 결과와 EDM으로 관측한 결과를 비교하면 수평방향 변위 평균 1.9~2.3㎝, 수직방향변위 평균 3.2㎝교차가 있었다. 둘째로 GPS 관측방법중 Static Mode 는 ±(3~7)㎜, Rapid Static Mode는 ±(6~7)㎜, Stop and Go Mode는 ±(121~226)㎜ 크기의 표준편차가 있는 것으로 나타났다. 따라서 Rapid Static Mode는 도로변형량 측정에 이용할 수 있는 방법이다. 셋째로 도로시공시 변형이 우려되는 곳에 이러한 간격으로 검사점을 설치하고, GPS에 의한 초기값을 관측해 두면, 시간경과에 따른 도로의 3차원 변형량을 비교적 정확하게 관측할 수 있다. This research was to analyze the road deformation surveying by GPS(Global Positioning System), and accuracy and usefulness of GPS was founded out excellent in the road deformation surveying. To execute this study, the national road in Kyungsang Nam-Do was surveyed, and analyzed the observation data. Results form this study are as follows, First, Compared GPS data with EDM data, the average horizontal discrepancy was 1.9~2.3㎝, the average vertical discrepancy was 3.2cm. Second, In GPS surveying mode, the standard deviation computed from "Static Mode" was ±(3~7)㎜, from "Rapid Static Mode" was ±(6~7)㎜, and from "Stop and Go Mode" was ±(121~226)㎜. Therefore "Rapid Static Mode" is also available for road deformation surveying. Third, In case of road construction, monitoring point were set up at regular intervals and checked a initial value by GPS, the 3-dimensional road deformation with lapse of time could be surveying correctly.

        권역단위 농촌지역개발사업의 주민역량 측정을 위한 설문도구개발

        The purpose of this study was to develop survey tools for diagnosis of capacity levels in business promotion of rural residents when performing a rural development project of a regional unit. The cases of previous studies were analyzed to select community capacity indicators related to a rural development project. Five indicators were derived : social capital, consciousness of participation, community spirit, and leadership. Based on the five indicators, measurement items of various capacities were selected and 54 survey items were selected through evaluation of experts twice. The pilot tests were conducted and targeted at Jeonnam song ho-jung village and Gyeongnam Haegeumgang village to identify derived survey items. In addition, descriptive statistic analysis and reliability analysis were conducted. As a result, survey items were corrected by reducing 10 items of the total 54 items. This results showed that using this tool could help us understand capacity levels of rural residents.

        Effects of epigallocatechin gallate on CoCl_(2)-induced apoptosis in PC12 cells

        Neuronal apoptotic events, consequently resulting in neuronal cell death, are occurred in hypoxic/ischemic condition. This cell death has been shown to be accompanied with the production of reactive oxygen species (ROS), which can attack cellular components such as nucleic acids, proteins and phospholipid. However, the underlying mechanisms of apoptosis induced in hypoxic/ischemic condition and its treatment methods are unsettled. Cobalt chloride (CoCl_(2)) has been known to mimic hypoxic condition including the production of ROS. Epigallocatechin gallate (EGCG). a green tea polyphenol, has diverse pharmacologial activities in cell growth and death. This study was aimed to investigate the apoptotic mechanism by CoCl_(2) and effects of EGCG on CoCl_(2)-induced apoptosis in PC12 cells. Administration of CoCl_(2) decreased cell survival in dose- and time-dependent manners and induced genomic DNA fragmentation. Treatment with 100 µM EGCG for 30 min before PC12 cells were exposed to 150 µM CoCl_(2), being resulted in the cell viability and DNA fragmentation being rescued. CoCl_(2) caused morphologic changes such as cell swelling and condensed nuclei, whereas EGCG attenuated morphologic changes by CoCl_(2). EGCG suppressed the apoptotic peak and a loss of Δψ_(m) induced by CoCl_(2). CoCl_(2) decreased Bcl-2 expression but Bax expression was not changed in CoCl_(2)-treated cells. EGCG attenuated the Bcl-2 underexpression by CoCl_(2). CoCl_(2) augumented the cytochrome c release from mitochondria into cytoplasm and increased caspase-8, -9 and caspase-3 activity, a marker of the apoptotic executing stage. EGCG ameliorated the incruement in caspase-8, -9 and -3 activity, and cytochrome c release by CoCl_(2). NAC (N-acetyl-cysteine), a scavenger of ROS, attenuated CoCl_(2)-induced apoptosis in consistent with those of EGCG. These results suggest, that CoCl_(2) induces apoptotic cell death through both mitochondria- and death receptor-dependent pathway and EGCG has neuroprotective effects against CoCl_(2)-induced apoptosis in PC12 cells. 신경세포자멸사는 저산소 및 허혈환경에서 일어나며 이러한 세포죽은 reactive oxidant species (ROS) 생성을 동반함이 알려져있다. 그러나, 저산소 및 허혈환경에서 일어나는 세포자멸사의 기전 및 그 치료방법은 아직 정립되어 있지 않다. CoCl_(2)는 ROS를 생성하는 등 저산소환경과 유사한 조건을 초래하는 것으로 알려져 있다. Epigallocatechin gallate (EGCG)는 녹차의 polyphenol 성분으로서 세포성장과 죽음에 다양한 약리학적 효과를 나타냄이 알려져 있다. 본 연구는 PC12 세포에서 CoCl_(2)에 의한 세포자멸사기전을 밝히고 이에 미치는 EGCG의 효과를 조사하는데 목적이 있다. Cell viability는 MTT 측정으로 조사되었고, DNA fragmentation은 DNA laddering으로 조사되었다. Bcl-2와 Bax발현 정도는 RT-PCR로, caspase-3와 -9의 활성은 spectrophotometer, caspase-8의 활성은 flow cytometry에 의해 측정되었다. 미토콘드리아에서 세포질로 분비된 cytochrome c는 western blot으로, 분해된 DNA 양과 미토콘드리아 세포막전위 (Δψ_(m))는 FACScan으로 조사되었다. CoCl_(2)투여로 PC12 세포수는 용량 및 시간 의존형태로 감소하였고, genomic DNA fragmentation이 발생하였다. CoCl_(2)투여로 야기된 cell viability의 감소와 DNA fragmentation은 EGCG 전처치에 의해 억제되었다. CoCl_(2)은 세포용적팽창과 condensed nuclei 같은 형태적 변화를 일으켰으며, apoptotic peak, Δψ_(m)감소 및 cytochrome c 유리를 야기하였다. EGCG는 CoCl_(2)에 의한 세포형태변화, apoptotic peak, Δψ_(m)소실 및 cytochrome c 유리를 억제시켰다. CoCl_(2)는 Bcl-2 발현을 감소시켰지만, Bax 발현에는 영향을 미치지 않았다. EGCG는 CoCl_(2)에 의해 야기된 Bcl-2 발현 감소를 억제시켰다. CoCl_(2)는 caspase-3, -8, 그리고 -9의 활성을 증가시켰으며, EGCG는 CoCl_(2)에 의한 세포자멸사를 억제시켰다. 본 실험결과는 PC12 세포에서 CoCl_(2)가 미토콘드리아 의존 및 death receptor의존 기전으로 세포자멸사를 일으키며, EGCG는 세포자멸사기전을 억세지킴으로 신경보호기능을 가짐을 시사하였다.

        Inhibitory Effect of Black Radish (Raphanus sativus L. var. niger) Extracts on Lipopolysaccharide-Induced Inflammatory Response in the Mouse Monocyte/Macrophage-Like Cell Line RAW 264.7

        Black radish (Raphanus sativus L. var. niger), which is cultivated worldwide, is used in traditional medicine as it aids liver function, gastric secretion, gallbladder function, and gallstone mitigation. In this study, we examined the anti-inflammatory effects of black radish extract (BRE) on the lipopolysaccharide (LPS)- and interleukin (IL)-6-mediated inflammatory responses in the RAW 264.7 cell lines. Our findings show that BRE significantly ameliorated LPS-induced nitric oxide (NO) release and production of pro-inflammatory cytokines, such as IL-1β, IL-6, tumor necrosis factor (TNF)-α,and prostaglandin E2. The levels of cyclooxygenase (COX)-2 and inducible NO synthase (iNOS) in LPS-stimulated RAW 264.7 cells were found to be suppressed by BRE. Further, BRE significantly suppressed the LPS-induced expression of mRNAs encoding COX-2, iNOS, IL-1β, IL-6, and TNF-α in a concentration-dependent manner. BRE treatment significantly inhibited Janus kinase 2 (JAK2) and signal transducer and activator of transcription 3 (STAT3) phosphorylation in IL-6- and LPS-treated RAW 264.7 cells. In addition, BRE decreased the levels of phosphorylated extracellular signal-regulated protein kinases and c-Jun N-terminal kinase under the same conditions. Moreover, BRE induced high nuclear factor erythroid 2-related factor 2 (NRF2) levels and its target gene heme oxygenase 1 (HO-1) in the absence of LPS. These data demonstrate that BRE may be beneficial for treating inflammation through selective immunomodulatory effects, which may be mediated by inhibition of the STAT3/JAK2 and activation of the NRF2/HO-1 signal transduction pathways.

      • 林土의 有機物의 分解速度와 無機養料의 垂直分布에 關한 考察

        Kim Choon Min, Chang Nam Kee and Chung Whan Ho: Decomposition rate of plant residue and the vertical distibution of mineral nutrients in the woodland soil. The relationship between decomposition rate of litter and the amount of mineral nutrients dispersed to the soil horizons were studied on the oak and pine forest stand. The results are as follows: 1. Decomposition rate of litter was highest in the F horizon of the oak stand, however in the pine stand it was highest in H horizon. 2. The correlation coefficients between decomposition rate and humus, organic carbon, moisture content, calcium, phosphorus, and nitrogen in the pine stand are; 0.90, 0.98, 0.76, 0.98, and 0.88, in oak stand; 0.97, 0.93, 0.92, 0.97, 0.74 and 0.73. 3. Factors which affect the decomposition rate of litter are humus, organic carbon, moisture content, calcium, phosphorus and nitrogen. 4. Among the variables, PH, moisture content and phosphorus under the oak and pine forest showed significant differences in each horizon. 5. C/N ratio of pine stands is higher than that of oak stands studied. 6. The vertical distribution of mineral nutrients is highly related with the amount of humus accumulated on top soil.
