        Effects of Propranolol on the Left Ventricular Volume of Normal Subjects During CT Coronary Angiography

        Objective: The purpose of this study is to determine the effects of propranolol on the left ventricular (LV) volume during CT coronary angiography. Materials and Methods: The LV volume of 252 normal Chinese subjects (126 subjects with propranolol medication and 126 age- and gender-matched Chinese subjects without medication) was estimated using 64 slices multi-detector CT (MDCT). The heart rate difference was analyzed by the logistic linear regression model with variables that included gender, age, body height, body weight, systolic blood pressure (SBP), diastolic blood pressure (DBP) and the dosage of propranolol. The following global LV functional parameters were calculated: the real-end diastolic volume (EDV), the real-end systolic volume (ESV) and the real-ejection fraction (EF). Results: The female subjects had a greater decrease of heart rate after taking propranolol. The difference of heart rate was negatively correlated with the dosage of propranolol. The real-EDV, the real-ESV and the real-EF ranged from 48.1 to 109 mL/m2, 6.1 to 57.1 mL/m2 and 41% to 88%, respectively. There was no significant difference in the SBP and DBP between the groups without and with propranolol medication (123 ± 17 and 80 ± 10 mmHg; 120 ± 14 and 80 ± 11 mmHg, respectively). The real-EDV showed no significant difference between these two groups, but the real-ESV and real-EF showed significant differences between these two groups (69.4 ± 9.3 and 70.6 ± 8.9 mL/m2; 23.5 ± 5.7 and 25.6 ± 3.7 mL/m2, 66.5 ± 5.1% and 63.5 ± 4.6%, respectively). Conclusion: The difference of heart rate is significantly influenced by gender and the dosage of propranolol. Propranolol will also increase the ESV, which contributes to a decreased EF, while the SBP, DBP and EDV are not statistically changed.         Effect of Surface Modification on Impact Strength and Flexural Strength of Poly(lactic acid)/Silicon Carbide Nanocomposites

        Silicon carbide (SiC) nanoparticles were surface modified using a silane coupling agent, and their properties were characterized using Fourier transform infrared, thermogravimetric analysis, and scanning electron microscope. Biodegradable poly(lactic acid) (PLA) composites were manufactured from PLA and SiC nanoparticles using a solution-blending method. The effect of SiC surface modification on the impact strength, flexural strength, and morphology of the PLA/SiC nanocomposites was studied. The impact strength of the PLA/surface-modified SiC (S-SiC) nanocomposites improved remarkably with increasing S-SiC content and scanning electron micrographs revealed that the PLA/S-SiC nanocomposites possessed a more ductile fracture surface than neat PLA or the PLA/SiC nanocomposites.

        Dynamic behavior analysis of push-the-bit rotary steerable bottom hole assembly

        Rotary steerable drilling technology is currently an advanced technology in the field of drilling engineering. A good understanding of dynamic behavior of rotary steerable system will benefit the drilling progress. Aimed at push-the-bit type of rotary steerable tool, finite element method was employed to model the rotary steerable bottom hole assembly (RSBHA) and establish the motion equation. Contact interaction between drill string and wellbore wall was analyzed and taken as the constraint condition in solving RSBHA motion equation. Force applied by steering pads was as the external force acting at the pads node. Then RSBHA motion state and bit lateral force were obtained by using Newmark method to solve the motion equation. A case was given and effects of influencing factors were analyzed. The results show that stick-slip phenomenon occurs under 60 r/min rotary speed condition and disappears when rotary speed rises to 90 r/min. Fully developed backward whirl comes into being at the near-bit stabilizer when rotary speed increases to 120 r/min. High weight on bit intensifies bit torsional vibration while larger pads’ steering force decreases the bit torsional vibration. Upper stabilizer plays a part in weakening RSBHA backward whirl under high rotary speed condition but reduces the inclination force. Time average of bit lateral force is mainly determined by pads’ steering force. Based on the results, corresponding suggestions were proposed to make the RSBHA be better used in the field applications.

        Prognostic Impact of Progesterone Receptor Status in Chinese Estrogen Receptor Positive Invasive Breast Cancer Patients

        Purpose: Estrogen receptor (ER) and progesterone receptor (PR) have been used as indicators of endocrine system status since the mid-1970s in the clinical management of breast cancer. The predictive role of ER in endocrine therapy is undisputed, but the prognostic value of PR is still debated. The aim of this study was to investigate the clinical characteristics and prognosis of ER positive breast cancer with different PR expression levels. Methods: A population cohort of 3,030 primary invasive ER positive breast cancer patients from a single cancer center underwent surgery and received adjuvant endocrine therapy from 2004 to 2010. The clinical and biological features of these patients with high PR-expressing tumors were compared with those of patients with low PR-expressing tumors. The follow-up data for disease-free survival (DFS), overall survival (OS), and breast cancer specific survival (BCSS) was obtained from 2,778 patients. Cox regression analysis was used to correlate biomarkers and tumor characteristics with DFS, OS, and BCSS. Results: Tumors with low PR expression had more invasive pathological features and biological indexes than those with high PR expression. Low PR expression was an independent poor prognostic factor for DFS (p=0.014; hazard ratio [HR], 0.781; 95% confidence interval [CI], 0.641–0.950), OS (p=0.002; HR, 0.699; 95% CI, 0.560–0.873), and BCSS (p=0.005; HR, 0.714; 95% CI, 0.566–0.902). Furthermore, in low PR expressing tumors, patients who received chemotherapy had better DFS (p=0.002; HR, 0.449; 95% CI, 0.268–0.751), OS (p<0.001; HR, 0.341; 95% CI, 0.192–0.606), and BCSS (p<0.001; HR, 0.292; 95% CI, 0.156–0.549) than patients who did not received chemotherapy. Conclusion: Patients with ER positive invasive breast cancer with low PR expressing tumors have a worse prognosis than those with high PR expressing tumors, and these patients can benefit from chemotherapy.

      • Autophagy Inhibition Promotes Gambogic Acid-induced Suppression of Growth and Apoptosis in Glioblastoma Cells

        Objective: To investigate the effects of gambogic acid (GA) on the growth of human malignant glioma cells. Methods: U251MG and U87MG human glioma cell lines were treated with GA and growth and proliferation were investigated by MTT and colony formation assays. Cell apoptosis was analyzed by annexin V FITC/PI flow cytometry, mitochondrial membrane potential assays and DAPI nuclear staining. Monodansylcadaverine (MDC) staining and GFP-LC3 localisation were used to detect autophagy. Western blotting was used to investigate the molecular changes that occurred in the course of GA treatment. Results: GA treatment significantly suppressed cell proliferation and colony formation, induced apoptosis in U251 and U87MG glioblastoma cells in a time- and dose-dependent manner. GA treatment also lead to the accumulation of monodansylcadaverine (MDC) in autophagic vacuoles, upregulated expressions of Atg5, Beclin 1 and LC3-II, and the increase of punctate fluorescent signals in glioblastoma cells pre-transfected with GFP-tagged LC3 plasmid. After the combination treatment of autophagy inhitors and GA, GA mediated growth inhibition and apoptotic cell death was further potentiated. Conclusion: Our results suggested that autophagic responses play roles as a self-protective mechanism in GA-treated glioblastoma cells, and autophagy inhibition could be a novel adjunctive strategy for enhancing chemotherapeutic effect of GA as an anti-malignant glioma agent.

        Glechomanamides A-C, Germacrane Sesquiterpenoids with an Unusual Δ<sup>8</sup>-7,12-Lactam Moiety from <i>Salvia scapiformis</i> and Their Antiangiogenic Activity

        <P>Three new germacrane sesquiterpenoid-type alkaloids with an unusual Δ<SUP>8</SUP>-7,12-lactam moiety, glechomanamides A-C (<B>1</B>-<B>3</B>), and two pairs of 7,12-hemiketal sesquiterpenoid epimers (<B>4a</B>/<B>b</B>, <B>5a</B>/<B>b</B>) were isolated from <I>Salvia scapiformis</I>. Their structures were elucidated by spectroscopic methods including HRESIMS, IR, UV, and 1D and 2D NMR and also confirmed by single-crystal X-ray diffraction analysis. The chemical transformation of compounds <B>1</B>-<B>5</B> in a solution environment was analyzed by 2D NMR spectroscopy. The aza acetallactams (<B>1</B>-<B>3</B>) were stable in organic solvent, while single crystals of the hemiacetal esters (<B>4a</B>/<B>b</B>, <B>5a</B>/<B>b</B>) underwent a tautomeric equilibrium after being dissolved. Single crystals of <B>4a</B>, <B>4b</B>, and <B>5a</B> were obtained for the first time as their naturally occurring forms. Glechomanamide B (<B>2</B>) exhibited antiangiogenic activity by suppression of vascular endothelial growth factor (VEGF)-induced tube formation through modulation of VEGF receptor 2 (VEGFR2)-mediated signaling pathways in human umbilical vascular endothelial cells (HUVECs). In addition, compound <B>2</B> also showed the significant suppression of mRNA expression associated with glycolysis and angiogenesis biomarkers in high glucose (30 mM)-induced HUVECs. These findings suggest that compound <B>2</B> might be a potential lead compound candidate for the management of diabetic retinopathy.</P> [FIG OMISSION]</BR>
