Synthesis and Anti-inflammatory Activity of Certain Piperazinylthienylpyridazine Derivatives

In this study, a novel series of 2-(4-substituted piperazin-l-ylmethyl)-6-(thien-2-yl)-2H-pyridazin- 3-ones (3a-f), 2-(4-substituted piperazin-l-yl carbonylmethyl)-6-(thien-2-yl)-2H-pyridazin-3-ones (4a-c) and 2-[2-(4-substituted piperazin-l-ylcarbonylethyl)]-6-(thien-2-yl)-2H-pyridazin-3-ones (5a,b) were prepared from 6-(thien-2-yl)-2H- pyridazin-3-one (1). In addition, 3-(4-substituted piperazin-l-ylcarbonyl methyl thio)-6-(thien-2-yl) pyridazines (6a-c) and 3-[2-(4-substitutedpiperazin- l-ylcarbonyl ethylthio]-6-(thien-2-yl) pyridazines (7a,b) were synthesized. Furthermore, 5-(4-substituted piperazin-l-ylmethyl)-6-(thien-2-yl)-2H-pyridazin-3-ones (12a,b) were prepared. The structures of the new compounds were confirmed by elemental analysis as well as by 1H-NMR, IR and MS data. Some of the newly prepared compounds were subjected to evaluation for their anti-inflammatory activity against carrageenan-induced paw edema at a dose of 10 mg/kg using indomethacin as the reference standard.

Objectives: Previous researches have investigated depression in postmenopausal women (PMW) with osteoporosis and fractures, but little is known regarding Moroccan PMW without fractures. We investigated depression prevalence and severity in Moroccan PMW with osteoporosis without fractures and its relationship with quality of life (QoL) and physical and psychological state. Methods: This cross-sectional study enrolled 100 PMW with osteoporosis without fractures. Depressive symptoms, QoL, self-esteem, and fatigue were evaluated using the Patient Health Questionnaire-9 (PHQ-9), Arabic version of ECOS-16 questionnaire, Rosenberg self-esteem scale, and Arabic version of the Multidimensional Assessment of Fatigue scale, respectively. A questionnaire including sociodemographic factors, bone density features, pain intensity, and sleep disturbance was completed. Results: Overall, 58% patients suffered from depression and 55% from pain (63.8% depressed women vs. 42.9% nondepressed patients; P = 0.03). Bone mineral density, lumbar spine T-score, ECOS-16, and self-esteem in depressed and nondepressed women were 0.791 (0.738–0.840) vs. 0.835 (0.790–0.866); –3.25 (–3.8 to –2.875) vs. –2.9 (–3.425 to –2.700), P = 0.02; 2.338 ± 0.605 vs. 1.638 ± 0.455; and 13.517 ± 5.487 vs. 18.404 ± 5.771, P < 0.0001, respectively. Depression severity correlated with pain, QoL, self-esteem, and fatigue (r = 0.367, r = –0.390, r = –0.390, and r = 0.369, respectively; P < 0.0001) as well as lumbar spine bone mineral density and T-score (r = –0.258 and r = –0.255, respectively; P = 0.01). Multiple linear regression analysis revealed impaired QoL (β = 0.526; P < 0.0001), fatigue (β = 0.177; P = 0.02), and lower self-esteem (β = –2.170; P = 0.005) as the strongest risk factors of depression. Conclusions: Our study shows that even without fractures, Moroccan PMW with osteoporosis suffered from depression, pain, impaired QoL, and lower self-esteem.

Epidemiological data and animal models indicate that Helicobacter pylori and dietary NaCl have a synergistic ill effect on gastric maladies. Here we show that the Ferric Uptake Regulator (Fur), which is a crucial regulatory factor required for H. pylori colonization, is essential for growth in the presence of high NaCl concentrations. Moreover, we demonstrate that the transcriptional response induced by sodium chloride stress exhibits similarities to that seen under iron depletion.
FTIR study of gamma and electron irradiated high-density polyethylene for high dose measurements

A reliable and well-characterized dosimetry system which is traceable to the international measurementsystem, is the key element to quality assurance in radiation processing with cobalt-60 gamma rays, Xrays,and electron beam. This is specifically the case for health-regulated processes, such as the radiationsterilization of single use medical devices and food irradiation for preservation and disinfestation. Polyethylene is considered to possess a lot of interesting dosimetric characteristics. In this work, adetailed study has been performed to determine the dosimetric characteristics of a commercialized highdensitypolyethylene (HDPE) film using Fourier transformed infrared spectrometry (FTIR). Correlationshave been established between the absorbed dose and radiation induced infrared absorption in polyethylenehaving a maximum at 965 cm 1 (transvinylene band) and 1716 cm 1 (ketone-carbonyl band). We have found that polyethylene dose-response is linear with dose for both bands up to1000 kGy. Fortransvinylene band, the dose-response is more sensitive if irradiations are made in helium. While, forketone-carbonyl band, the dose-response is more sensitive when irradiations are carried out in air. Thedose-rate effect has been found to be negligible when polyethylene samples are irradiated with electronbeam high dose rates. The irradiated polyethylene is relatively stable for several weeks after irradiation.

In this study, six in situ gelling formulations based on Gelrite were prepared and evaluated for the retained ophthalmic delivery of Moxifloxacin (Mox). The effectiveness of the best developed formula G5 was compared with photodynamic therapy (PDT), the recent expanding approach for the treatment of ophthalmologic disorders after the assessment of optimum photodynamic inactivation parameters that permit efficient pathogens eradication. It was found that, Staphylococcus aureus (S. aureus) (Gram-positive) was more susceptible to effective lethal photosensitization that reaches 93.5% reduction in viable count than Escherichia coli (E. coli) (Gramnegative) of 76.1% using 3 mg/mL Hematoporphyrin (HP), illuminated by 630 nm Light Emitting Diode (LED) at 9 J/cm^2 and incubated for 15 min. Following topical instillation of G5 to rabbits corneas, higher amount of Mox was retained in the aqueous humor up to 24 h with significant 6-fold increase in the C_max and AUC_(0-∞) compared to vigamox^® commercial eye drops. After post corneal infection with S. aureus, both approaches were effectively treating the infection without causing ocular irritation or collateral damage to corneal tissue where G5 showed remarkable improvement after four days compared to seven days of PDT treatment.

This study investigated for the first time the antimicrobial effect of methanolic extract of Grewia mollis Juss (mGM) against Staphylococcus aureus KCCM 11256 (Gram positive) and Escherichia coli KCCM 11234(Gram negative). The minimum inhibitory concentration (MIC) values for each bacterial strain were recorded in a liquid medium. In addition, ultrafine poly(D,L-lactide-co-glycolide) (PLGA) fiber mats containing a methanolic crude extract (7.5 wt%) of Grewia mollis (mGM) were fabricated from the neat PLGA solution by electrospinning. The PLGA ultrafine fiber mat containing mGM were characterized by SEM, FE-SEM, Bio-TEM, TGA, and EDX analyses. Our findings indicate that the incorporation of mGM in the polymer media affected both the morphology and size of the mGM-loaded PLGA ultrafine fibers (UFs). SEM-EDX results confirmed well oriented ultrafine fibers with good incorporation of mGM. Results of the structure characterization indicated that the incorporated mGM interacted strongly with the PLGA matrix and the mechanical properties were effectively improved. Moreover,mGM loaded PLGA ultrafine fiber mats were able to inhibit the growth of the bacterial strains, which indicate that mGM blended PLGA fibers could be used as a potential effective antimicrobial agent for many biomedical applications including wound dressing.
Perioperative effects of various anesthetic adjuvants with TIVA guided by bispectral index

Background: This prospective, randomized, double blinded, controlled study was designed to compare effects of intravenous co-administration of clonidine, magnesium, or ketamine on anesthetic consumption, intraoperative hemodynamics, postoperative analgesia and recovery indices during Bispectral Index (BIS) guided total intravenous anesthesia (TIVA). Methods: After ethical committee approval and written informed consent, 120 adult patients ASA I and II scheduled for open cholecystectomy were randomly assigned to one of 4 equal groups. Group CL received clonidine 3 μg/kg and maintained by 2 μg/kg/h. Group MG received magnesium sulphate 50 mg/kg and maintained by 8 mg/kg/h. Group KET received racemic ketamine 0.4 mg/kg and maintained by 0.2 mg/kg/h. Control group (CT) received the same volume of isotonic saline. Anesthesia was induced and maintained by fentanyl, propofol and rocuronium. Propofol infusion was adjusted to keep the BIS value between 45-55. Intraoperative hemodynamics, induction time, anesthetic consumption, recovery indices, and PACU discharge were recorded. Results: Induction time, propofol requirements for induction and maintenance of anesthesia, intraoperative fentanyl and hemodynamic values were significantly lower with Groups CL and MG compared to Groups KET and CT (P < 0.05). Patients in Group MG showed significantly lower muscle relaxant consumption, delayed recovery and PACU discharge than other groups (P < 0.05). First, analgesic requirement was significantly longer and total postoperative analgesic consumption was significantly lower in the adjuvant groups versus Group CT (P < 0.05). Conclusions: Clonidine, magnesium, and ketamine can be useful adjuvant agents to BIS- guided TIVA. Pharmacokinetic studies of such drug combinations were recommended to investigate their interaction. Axial.Flexural Interaction in FRP-Wrapped RC Columns

The study reported herein aims at investigating the behavior of medium-scale circular reinforced concrete columns wrapped with fiber reinforced polymer (FRP) sheets under concentric and eccentric axial loads. The experimental program was devised to assess the effects of loading conditions, absence/presence of an FRP jacket as well as the FRP wrapping system. To achieve the study objectives, four column groups were tested under axial compression at 0, 25, 50 and 65 mm loading eccentricities corresponding to eccentricity-to-diameter ratios of 0, 0.13, 0.26 and 0.34, respectively. Specimens in a fifth group were tested in pure bending simulating axial compression at infinite loading eccentricities. Three column subcategories were tested under each of the 5 loading eccentricities: unwrapped; wrapped with one ply of hoop FRP sheets; and wrapped with two FRP plies with fibers oriented at 0 and 90° to the longitudinal column axis thereby providing externally-bonded longitudinal reinforcement and hoop confinement, respectively. Tests confirmed that FRP confinement enhances the axial–flexural column resistance even at large eccentricities that exceed the balanced state of unconfined columns. Although axial column resistance decreased with increasing bending moments, relative enhancements (25–35%) in axial resistance provided by FRP confinement were found to be more significant under eccentric loading than in pure compression. Compared to hoop FRP-confined columns, using additional longitudinal sheets resulted in minor (7–9%) but stable enhancements in axial resistance that were unaffected by the increase in loading eccentricity. The FRP hoop wraps had a minor effect on the flexural resistance of specimens tested in pure bending but managed to double their resistance when combined with the externallybonded longitudinal FRP sheets. Finally, three stress–strain models of FRP-confined concrete were used in conventional section analysis to assess the axial–flexural interaction in the FRP-jacketed columns. Strength predictions made using the stress–strain model proposed in ACI 440.2R-17 design guidelines did not agree with the test results of the eccentrically-loaded columns and underestimated the moment resistance at a given axial force even when considering higher confinement ratios than those permitted by the guidelines. Strength predictions made using eccentricitydependent stress–strain models showed better results especially when accounting for the increase in ultimate axial strains under eccentric loading.
