        High expression of keratin 6C is associated with poor prognosis and accelerates cancer proliferation and migration by modulating epithelial–mesenchymal transition in lung adenocarcinoma

        Background Lung adenocarcinoma (LUAD) is a more frequent subtype of lung cancer and most cases are discovered in the late stages. The proliferation and metastasis of LUAD are pivotal for disease progression. Despite unremitting deeper understanding of LUAD biology, the mechanisms involved in the proliferation and metastasis of LUAD remain unclear. The objective of our article was to inquiry the expression and the function of keratin 6C (KRT6C) in LUAD cells. Methods First, the expression level and prognostic value of KRT6C in LUAD tissues were analyzed on the basis of the data acquired from TCGA database. Through qRT-PCR, the expression level of KRT6C on LUAD cell lines (A549, H1299, PC-9) and human normal lung cell line MRC-5 was tested. After that, CCK8 and colony formation assays was utilized to detect cell proliferation. In addition, to explore the influence of KRT6C on LUAD migration and invasion ability, scratch wound healing and transwell assays were utilized. Through western blotting, the protein expression levels of KRT6C, PCNA, E-cadherin, N-cadherin, Snail and Vimentin were detected. Results The outcomes revealed that KRT6C was highly expressed in LUAD tissues and cell lines. Besides, elevated level of KRT6C was related to worse prognosis in LUAD patients. Ablation of KRT6C restrained proliferation, migration and invasion of A549 cells. KRT6C deficiency augmented the expression of E-cadherin as well as reduced the expression of N-cadherin, Snail and Vimentin. Conclusion Above all, these consequences indicated that depletion of KRT6C suppressed A549 cell proliferation, migration and invasion, which might be achieved by regulating EMT. In general, KRT6C is identified as a potential therapeutic target for LUAD.

        MiR-873, as a suppressor in cervical cancer, inhibits cells proliferation, invasion and migration via negatively regulating ULBP2

        Background Cervical cancer (CC) remains a large burden in the developing countries. The tumor inhibitory role of miR873 has been verified in a variety of cancers, however, whether miR-873 has a suppressive effect on CC remains unclear. Objective The purpose of this study was to investigate the functional role of miR-873 in CC, as well as explore the underlying molecular mechanism. Methods The prognostic values of miR-873 were assessed by Kaplan–Meier methods and cox regression models using the data which were downloaded from TCGA database. The expression of miR-873 was measured by RT-qPCR. Cell counting Kit-8, clone formation, and Transwell assays were used to assess the cell viability and metastasis, appropriately. The targeting relationship between miR-873 and ULBP2 was predicted by biological software and confirmed by dual luciferase reporter assay. Rescue assays were conducted to investigate whether miR-873 affects the phenotype of CC cells via regulating ULBP2. Results We observed that miR-873 was low-expressed in CC. Up-regulation of miR-873 notably restrained the proliferation, invasion and migration of C33a cells. Meanwhile, down-regulation of miR-873 in SiHa cells presented the opposite outcomes. ULBP2 was forecasted and certified as a target of miR-873. The results of rescue assays showed that overexpression of ULBP2 could restore the proliferation and motility of CC cells that inhibited by miR-873. Conclusion MiR-873 suppressed the CC cells proliferation, invasion and migration via negatively regulating ULBP2, suggesting that miR-873 could serve as a valuable therapeutic target for CC therapy.

        Kano 모델 및 PCSI 지수를 활용한 O2O 서비스 품질에 대한 탐색적 연구:

        연구목적: 본 연구는 중국 소비자들을 대상으로 O2O 서비스에서 Kano 모형을 이용해 서비스 품질을 분류하고, 고객만족계수와 불만족계수를 각각 산출하였다. 이후 순차적으로 잠재적 고객만족개선지수(PCSI Index)를 산출하였다. 그 결과 O2O 서비스 제공 시 우선적으로 고려하여 관리 및 개선해야 할 서비스 품질속성을 판단하고, 최종적으로 중국 소비자들의 고객만족을 높일 수 있는 우선적인 개선사항들을 제시하고자 한다. 논문구성/논리: 본 논문은 전체 5절로 구성되어 있다. 1절에서는 연구의 배경 및 방향에 대하여 제시한다. 이 후 2절에서는 관련 문헌에 대하여 검토한다. 3절에서는 연구모형과 연구방법을 제시하고, 4절에서는 연구방법을 통해 도출된 결과를 분석하였다. 최종적으로 5절에서는 결론을 도출하고, 연구의 시사점 및 한계점을 설명한다. 결과: 첫째로 중국 소비자들을 대상으로 하는 기업에서는 본 연구에서 확인된 당연적 품질요소나 일원적 품질요소 이외에도 이러한 매력적 품질요소에 투자를 하고 꾸준한 관리를 해야 한다. 둘째로 PCSI 지수 값이 높을수록 고객의 만족도를 증가시킬 수 있는 요인들이다. 따라서 지수 값이 높게 나온 요인들을 중점적으로 개선한다면 고객들의 만족도가 크게 증가할 것이다. 마지막으로 PCSI지수의 상위에 위치한 항목들은 개선이 가장 시급하고 고객만족이 가장 크게 증가될 수 있다. 따라서 이와 같은 항목에 투자하여 소비자들을 끌 수 있도록 해야 할 것이다. 독창성/가치: 본 연구는 중국 소비자들을 대상으로 O2O 서비스 품질을 Kano 모형에 따라 매력적 품질, 일원적 품질, 당연적 품질, 무관심 품질, 역 품질로 분류하였다. 이 후 고객만족계수를 이용하여 품질 요인들 중 고객의 만족도의 상승가능여부 또는 하락가능여부를 파악하고 잠재적 고객만족 개선지수(PCSI index)를 산출하였다. 이로써 고객만족을 높이기 위해 우선적으로 개선되어야 하는 품질 요인들이 결정되었다. 따라서 본 연구에서는 Kano 모형과 PCSI 지수를 기반으로 O2O 서비스 품질에 관한 고객만족을 높일 수 있는 우선적인 개선사항들이 효과적으로 제시되었다. Purpose: In our study, we classify the quality factors involved in the online to offline (O2O) service to Chinese consumers by using the Kano model and calculate the customer satisfaction and dissatisfaction coefficients. Next, we calculate the PCSI (Potential Customer Satisfaction Improvement) index. As a result, we determine the service quality attributes in O2O service that should be managed and improved first, and finally present some preemptive improvements that will enhance customer satisfaction among Chinese customers. Composition/Logic: This paper has five sections. In section 1, we present the objectives and overview of the study. We review related literature in section 2. We describe our research model and methodology in section 3 and analyze the results in section 4. In section 5, we draw conclusions from the study, and explain both the implications and limitations of the study. Findings: First, in the companies targeting Chinese consumers, the attractive quality factors, which are identified in this study, should be invested in and maintained at a constant level, along with the must-be and one-dimensional quality factors. Second, the higher the value of PCSI index, the more likely is the increase in the satisfaction of the customer. Thus, customer satisfaction will increase significantly if we focus on the factors that have high PCSI index values. Third, the top ranked items in the PCSI index are the quality factors that not only need the most urgent improvement but can also increase customer satisfaction the most. Therefore, consumers should be attracted by investing in these items. Originality/Value: This study categorizes O2O service quality into attractive quality, one-dimensional quality, must-be quality, indifferent quality and reverse quality by applying the Kano model to Chinese consumers. We then use the customer satisfaction coefficient to determine whether the customer s satisfaction level could increase or decrease and calculate the Potential Customer Satisfaction Improvement index (PCSI index). This has led to the identification of quality factors that should be improved first to improve customer satisfaction. Therefore, in this study, based on the Kano model and the PCSI index, the priority improvements that can improve customer satisfaction on O2O service quality are presented effectively.

      • How Student Classroom Engagement Affects Students’ Study Results in Mathematics Classroom

        To improve students’ classroom engagement is not only the demand of curriculum revolution, but also the reflection of learning democracy. Students' responses and thinking are the main manifestations of students' participation in classroom learning. To reduce the amount of questions and increase the opportunities and time for students to think, this study, by employing SPSS, makes attempts to analyze the data by using multivariate GLM analysis to explore the effects of students' responses and thinking on learning results. The results indicated the students learning effect will be promoted through reducing the quantity and increasing the quality of question and adding the thinking opportunities.

        The purified extract of steamed Panax ginseng protects cardiomyocyte from ischemic injury via caveolin-1 phosphorylation-mediating calcium influx

        Background: Caveolin-1, the scaffolding protein of cholesterol-rich invaginations, plays an important rolein store-operated Ca2þ influx and its phosphorylation at Tyr14 (p-caveolin-1) is vital to mobilize protectionagainst myocardial ischemia (MI) injury. SOCE, comprising STIM1, ORAI1 and TRPC1, contributesto intracellular Ca2þ ([Ca2þ]i) accumulation in cardiomyocytes. The purified extract of steamed Panaxginseng (EPG) attenuated [Ca2þ]i overload against MI injury. Thus, the aim of this study was to investigatethe possibility of EPG affecting p-caveolin-1 to further mediate SOCE/[Ca2þ]i against MI injury in neonatalrat cardiomyocytes and a rat model. Methods: PP2, an inhibitor of p-caveolin-1, was used. Cell viability, [Ca2þ]i concentration were analyzedin cardiomyocytes. In rats, myocardial infarct size, pathological damages, apoptosis and cardiac fibrosiswere evaluated, p-caveolin-1 and STIM1 were detected by immunofluorescence, and the levels ofcaveolin-1, STIM1, ORAI1 and TRPC1 were determined by RT-PCR and Western blot. And, release of LDH,cTnI and BNP was measured. Results: EPG, ginsenosides accounting for 57.96%, suppressed release of LDH, cTnI and BNP, and protectedcardiomyocytes by inhibiting Ca2þ influx. And, EPG significantly relieved myocardial infarct size, cardiacapoptosis, fibrosis, and ultrastructure abnormality. Moreover, EPG negatively regulated SOCE viaincreasing p-caveolin-1 protein, decreasing ORAI1 mRNA and protein levels of ORAI1, TRPC1 and STIM1. More importantly, inhibition of the p-caveolin-1 significantly suppressed all of the above cardioprotectionof EPG. Conclusions: Caveolin-1 phosphorylation is involved in the protective effects of EPG against MI injury viaincreasing p-caveolin-1 to negatively regulate SOCE/[Ca2þ]i.

        생산력 발전 불균형은 중국 경제발전의 가장 기본적인 특징이며, 중국경제발전 상황에 적합한 경제이론을 선택하고 적용하여 중국 지역경제발전을 분석하고 연역하는 것은 중요한 이론적 가치와 현실적 의미를 갖는다. 본 논문은 생산력 불균형 구조라는 각도에서 출발하여 중국 지역경제발전 격차가 확대되는 변화과정 및 이 격차가 경제발전에 어떤 영향을 미치는지 분석하였다. 분석에 근거하여 정부의 거시조절이 지역격차를 조절하는데 필수불가결하다고 본다.

      • Celebrity Endorsement Problem on Social Media : Formulation, Analysis and Recommendation Algorithm

        In this paper, a new matching recommendation algorithm is proposed to help enterprises find one or more proper celebrities from social media for their product endorsement. The fans group of a celebrity, his impaction value in the social media, as well as the matching degree between the celebrity and the product are selected to measure a celebrity. The attribute similarities between target customers of the product and fans of the celebrity are calculated via the Pearson similarity formula. Then, considering the impaction value of the celebrity and the matching degree of the celebrity and the product which can be accessed on the website or usually available from the enterprise, an evaluation index is proposed. We use some data from Sina Micro-blog, which is the most popular social media platform in China, to show the effectiveness of our proposed matching recommendation algorithm. Moreover, the analysis shows that a particular celebrity may suitable to endorse different product on-line and off-line.
