멜라닌 색소는 포유동물의 피부, 머리카락, 눈, 신경계에 풍부하게 존재한다. 멜라닌은 다양한 환경적 스트레스로부터 피부를 보호하며, 생리학적 산화-환원 완충 작용을 통해 항상성을 유지한다. 그러나, 과도한 멜라닌 축적은 간반, 주근깨, 노인성 흑자, 염증성 색소침착을 일으킬 수 있다. 티로시나아제는 멜라닌의 생합성 경로 조절에 아주 중요한 역할을 하는 것으로 알려져 있다. 티로시나아제의 활성을 저해하는 다양한 미백제가 개발되었지만 알러지, DNA손상, 세포독성, 돌연변이 유발 등을 야기하는 부작용으로 인해 임상적 적용이 제한되었다. 본 논문에서 여러 4-크로마논 유도체를 합성하여 티로시나아제 억제 활성을 조사하였다. 이들 화합물 중 MHY1294는 IC50가 5.1±0.86 μM으로 양성 대조군인 코직산(14.3±1.43 μM) 보다 나은 티로시나아제 효소 억제 활성을 나타냈다. 또한 MHY1294는 티로시나아제의 촉매 부위에서 경쟁적인 억제 작용을 보였으며 코직산보다 더 큰 기질 결합 친화성을 가지는 것으로 확인되었다. 뿐만 아니라, MHY1294는 B16F10 흑색종 세포에서 멜라닌 세포 자극 호르몬(α-MSH)에 의해 유도되는 멜라닌 합성과 세포 내 티로시나아제 활성을 유의적으로 억제하였다. 결론적으로 본 연구는 MHY1294가 과도한 멜라닌 축적에 대한 약물 제제 및 미백제로서의 개발 가능성이 있음을 시사한다. Melanin pigments are abundantly distributed in mammalian skin, hair, eyes, and nervous system. Under normal physiological conditions, melanin protects the skin against various environmental stresses and acts as a physiological redox buffer to maintain homeostasis. However, abnormal melanin accumulation results in various hyperpigmentation conditions, such as chloasma, freckles, senile lentigo, and inflammatory pigmentation. Tyrosinase, a copper-containing enzyme, plays an important role in the regulation of the melanin pigment biosynthetic pathway. Although several whitening agents based on tyrosinase inhibition have been developed, their side effects, such as allergies, DNA damage, mutagenesis, and cytotoxicity of melanocytes, limit their applications. In this study, we synthesized 4-chromanone derivatives (MHY compounds) and investigated their ability to inhibit tyrosinase activity. Of these compounds, (E)-3-(4-hydroxybenzylidene)chroman-4-one (MHY1294) more potently inhibited the enzymatic activity of tyrosinase (IC50 = 5.1±0.86 μM) than kojic acid (14.3±1.43 μM), a representative tyrosinase inhibitor. In addition, MHY1294 showed competitive inhibitory action at the catalytic site of tyrosinase and had greater binding affinity at this site than kojic acid. Furthermore, MHY1294 effectively inhibited α-melanocyte stimulating hormone (α-MSH)-induced melanin synthesis and intracellular tyrosinase activity in B16F10 melanoma cells. The results of the present study indicate that MHY1294 may be considered as a candidate pharmacological agent and cosmetic whitening ingredient.
발달장애 학생을 위한 교과교육 관련 단일대상 연구 분석

본 연구의 목적은 발달장애 학생을 위한 교과교육 관련 단일대상 연구 논문을 분석하는 것이었다. 본 연구의 분석대상은 2010년부터 2019년까지 국내에서 발표된 발달장애 학생을 위한 교과교육 관련 단일대상 연구 28편이었다. 분석대상 논문의 일반적 특성, 중재변인 및 중재결과를 분석하고, Horner 외(2005)와 CEC(2014)의 증거기반 실제의 질적지표에 근거하여 질적분석을 하였다. 연구 결과는 다음과 같다. 첫째, 일반적 특성에 따르면 초등학생대상 연구가 32.2%이고, 중고등학생 대상 연구가 67.8%였으며, 연구대상은 지적장애 학생이 자폐장애 학생보다 많았다. 실험설계는 중다(간헐)기초선 연구가 70%로 가장 많았다. 둘째, 중재 변인의 분석 결과에 따르면 국어와 수학 관련 논문이 가장 많았으며, 중재프로그램을 구성하기 위해 교육과정을 분석한 논문은 63.3%이었고, 관찰자간 신뢰도, 중재충실도, 사회적 타당도에 대해 보고한 논문이 각각 60.7%, 50.0%, 75%이었다. 셋째, 중재결과의 분석에 따르면, 중재효과, 유지 및 일반화효과가 있다고 보고하였으며, 시각적 분석 방법으로 모든 분석대상 논문이 평균선을 이용하여 분석을 하였고, 비중복비율(중복비율 포함)을 이용하여 분석한 논문은 32.1%였다. 넷째, 질적분석 결과, 모든 질적지표를 충족한 논문은 없었으나 중재자 자격여부 정보와 사회적 타당도 영역을 제외한 모든 질적지표에 대해 논문의 60% 이상 충족하였다. 본 연구 결과를 바탕으로 발달장애 학생을 위한 교과교육 관련 단일대상 연구에 대한 논의와 시사점을 제시하였다. The purpose of this study was to analyze the single-subject design studies related to subject matter education for students with developmental disabilities based on the quality indicators of evidence-based practices. Twenty eight single-subject studies published from 2010 to 2019 were analyzed using the quality indicators of evidence-based practices developed by Horner and colleagues(2005) and CEC(2014). The research findings were as follows. First, the most widely selected study subjects were students with intellectual disabilities. Multiple probe designs were the most widely used. Second, the most widely selected subjects were Korean and math. Third, in all 28 studies interventions were effective for improving dependent variables. Generalization effects were tested in only eight of 28 studies, while maintenance effects were tested in 27 of 28 studies. The following quality indicators were not entirely fulfilled: intervention settings, background informations, qualifications of experimenters, operant definitions of dependent variables, fidelity of intervention, baselines, and internal validity. Educational implications and suggestions for future studies were discussed.
A Case of Ischiofemoral Impingement Syndrome as a Differential Diagnosis of Ankylosing Spondylitis

Ischiofemoral impingement (IFI) syndrome is an uncommon cause of gluteal and hip pain. We report on a case of a 20-year-old man who presented with chronic gluteal and hip pain with low back pain without a history of trauma or surgery. He was misdiagnosed with ankylosing spondylitis (AS) at another clinic. The patient was finally diagnosed with IFI syndrome according to pelvic magnetic resonance imaging findings at our hospital. After two weeks of medical and physical treatment, his pain showed gradual improvement. Because IFI syndrome is rarely reported in male patients, it might be misdiagnosed as AS. Therefore, IFI syndrome should be considered as a differential diagnosis of AS, particularly in young male patients with atypical pain characteristics. (J Rheum Dis 2016;23:179-182)
Detection of Waterborne Pathogens by PCR-reverse Blot Hybridization

The present study was set to develop comprehensive system for assessing the safety of drinking water using PCR-reverse blot hybridization assay (REBA). The REBA developed in this study can detect waterborne pathogens such as Shigella spp., Salmonella spp., Citrobacter spp., Enterobacter spp., Klebsiella spp., Yersinia spp., Mycobacterium spp., Listeria spp. at the genus level, and Escherichia coli, Citrobacter freundii, Klebsiella pneumoniae, Pseudomonas aeruginosa, Yersinia enterocolitica, Y. pseudotuberculosis, Mycobacterium avium complex, M. marinum, Enterococcus faecalis, and Staphylococcus aureus at the species level, and E. coli O157:H7 at the strain level.

<P>Drug resistance in Mycobacterium leprae is a significant problem in countries where leprosy is endemic. A sensitive, specific, and high-throughput reverse blot hybridization assay (REBA) for the detection of genotypic resistance to rifampicin (RIF) was designed and evaluated. It has been shown that resistance to RIF in M. leprae involves mutations in the rpoB gene encoding the -subunit of the RNA polymerase. The PCR-REBA simultaneously detects both 6 wild-type regions and 5 different mutations (507AGC, 513GTG, 516TAT, 531ATG, and 531TTC) including the most prevalent mutations at positions 507 and 531. Thirty-one clinical isolates provided by Korea Institute of Hansen-s Disease were analyzed by PCR-REBA with RIF resistance of rpoB gene. As a result, missense mutations at codons 507 AGC and 531ATG with 2-nucleotide substitutions were found in one sample, and a missense mutation at codon 516 TAT and ??WT6 (deletion of 530-534) was found in another sample. These cases were confirmed by DNA sequence analysis. This rapid, simple, and highly sensitive assay provides a practical alternative to sequencing for genotypic evaluation of RIF resistance in M. leprae.</P>
