        DnaJC18, a Novel Type III DnaJ Family Protein, is Expressed Specifically in Rat Male Germ Cells

        Mammalian spermatogenesis occurs in a precise and coordinated manner in the seminiferous tubules. One of the attempts to understand the detailed biological process during mammalian spermatogenesis at the molecular level has been to identify the testis specific genes followed by study of the testicular expression pattern of the genes. From the subtracted cDNA library of rat testis prepared using representational difference analysis (RDA) method, a complimentary DNA clone encoding type III member of a DnaJ family protein, DnaJC18, was cloned (GenBank Accession No. DQ158861). The full-length DnaJC18 cDNA has the longest open reading frame of 357 amino acids. Tissue and developmental Northern blot analysis revealed that the DnaJC18 gene was expressed specifically in testis and began to express from postnatal week 4 testis, respectively. In situ hybridization studies showed that DnaJC18 mRNA was expressed only during the maturation stages of late pachytene, round and elongated spermatids of adult rat testis. Western blot analysis with DnaJC18 antibody revealed that 41.2 kDa DnaJC18 protein was detected only in adult testis. Immunohistochemistry study further confirmed that DnaJC18 protein, was expressed in developing germ cells and the result was in concert with the in situ hybridization result. Confocal microscopy with GFP tagged DnaJC18 protein revealed that it was localized in the cytoplasm of cells. Taken together, these results suggested that testis specific DnaJC18, a member of the type III DnaJ protein family, might play a role during germ cell maturation in adult rat testis.

        Magnitude Modulation for VSAT's Low Back-Off Transmission

        This paper addresses the problem of controlling the envelope's power peak of single carrier modulated signals, band limited by root-raised cosine (RRC) pulse shaping filters, in order to reduce power amplifier back-off for very small aperture terminals ground stations. Magnitude modulation (MM) is presented as a very efficient solution to the peak-to-average power ratio problem. This paper gives a detailed description of the MM concept and its recent evolutions. It starts by extending the look-up-table (LUT) based approach of the MM concept to M-ary constellations with M ${\leq}$ 16. The constellation and RRC symmetries are explored, allowing considerable reduction on LUT computation complexity and storage requirements. An effective multistage polyphase (MPMM) approach for the MM concept is then proposed. As opposed to traditional LUT-MM solutions, MM coefficients are computed in real-time by a low complexity multirate filter system. The back-off from high-power amplifier saturation is almost eliminated (reduction is greater than 95%) with just a 2-stage MPMM system even for very demanding roll-off cases (e.g., ${\alpha}$ = 0,1). Also, the MPMM is independent of modulation in use, allowing its easy application to constellations with M > 16.

        XCM Biologic Tissue Matrix xenograft and autologous micromucosa graft for vaginal reconstruction in Mayer-Rokitansky-Küster-Hauser syndrome

        Mayer-Rokitansky-Küster-Hauser (MRKH) syndrome can be treated through numerous surgical and nonsurgical methods. We present a surgical technique in which a neovagina was reconstructed and shaped by a vaginal expander with acellular porcine dermal matrix (XCM Biologic Tissue Matrix) and mucosal interposition using microfragments harvested from the hymen. In our case, we found this procedure to be safe and effective, resulting in satisfactory sexual function and good cosmetic results, without donor site morbidity. To our best knowledge, this tissue-engineered biomaterial has never been used for vaginal reconstruction before.

        The impact of digital influencers’ characteristics on purchase intention of fashion products

        Research on digital influencers’ role in marketing strategies under ongoing development. This study explores the role of digital influencer marketing on consumer purchase intention in fashion products. The goal is to investigate the direct and indirect relationships of trustworthiness, expertise, content quality, similarity, para-social interaction, and attitude towards sponsored posts on purchase intention. A mixed-method approach was conducted combining interviews with five digital influencers and a consumer-based survey with a sample of 206 fashion consumers, among them 96.1% were women and 51.5% were between 18 and 31 years old. Results indicate that blog content quality and para-social interaction positively influence purchase intention in fashion brands. Additionally, blog content quality plays a mediating role between trustworthiness and purchase intention. Moreover, consumers’ attitude towards sponsored posts influences directly and positively purchase intention and mediates the relationship between trustworthiness and purchase intention. Consumers’ attitude towards sponsored posts was also found to mediate the relationship between expertise and purchase intention. This study highlights the digital influencers’ marketing characteristics, which affect the fashion consumer purchase intention, enriching the research in this area and providing recommendations for further investigation. The results also highlight the importance of aligning communication strategies of fashion brands with influencers’ lifestyle.

        Shock temporality: international students coping with disrupted lives and suspended futures

        During the COVID-19 pandemic, people around the word experienced periods of local, state and international immobility due to lockdowns, border closers and travel restrictions. For transient migrants such as international students, these kinds of immobility have resulted in disrupted lives with professional and personal futures suspended as careers and relationships become stuck in limbo. Moreover, such sudden and extended periods of immobility have not been sufficiently covered in temporality literature due to the novelty of the pandemic crisis in the international education, migration and mobility studies spaces. By conducting a pilot project investigating current and recently graduated higher degree by research (HDR) international students (PhD, Masters and Honours) from public institutions in the Australian city of Melbourne, this paper, thus, introduces the concept of ‘shock temporality’ caused by the global COVID-19 pandemic. Shock temporality takes place when the expected and finite temporary/transitory experience becomes forcefully broken and appears to be ongoing outside of the individual’s control. While shock temporality has left HDR international students’ professional and personal agendas and aspirations in suspension, students interviewed use the time to plan and prepare for truncated futures. The findings of this paper, thus, become relevant in assisting higher education student support services in creating potential approaches and strategies for a post-pandemic future.

        Intraoperative discomfort associated with the use of a rotary or reciprocating system: a prospective randomized clinical trial

        Objectives: The aim of this randomized, controlled, prospective clinical study was to evaluate patients' intraoperative discomfort during root canal preparations in which either multi-file rotary (Mtwo) or single-file reciprocating (Reciproc) systems were used. Materials and Methods: Fifty-five adult patients, aged between 25 and 69 years old, with irreversible pulpitis or pulp necrosis participated in this study. Either the mesiobuccal or the distobuccal canals for maxillary molars and either the mesiobuccal or the mesiolingual canals for mandibular molars were randomly chosen to be instrumented with Mtwo multi-file rotary or Reciproc single-file reciprocating systems. Immediately after each canal instrumentation under anesthesia, patient discomfort was assessed using a 1 - 10 visual analog scale (VAS), ranging from 'least possible discomfort' (1) to 'greatest possible discomfort' (10). The Wilcoxon signed-rank test was used to determine significant differences at p < 0.05. Results: Little intraoperative discomfort was found in all cases. No statistically significant differences in intraoperative discomfort between the 2 systems were found (p = 0.660). Conclusions: Root canal preparation with multi-file rotary or single-file reciprocating systems had similar and minimal effects on patients' intraoperative discomfort.

        Sensory profile and technological characterization of boneless dry-cured ham with lactulose added as a prebiotic

        Objective: This study investigates the technological and sensory profile of boneless dry-cured ham with different contents of lactulose added as a prebiotic ingredient. Methods: In addition to the control samples (without the addition of lactulose), three treatments were formulated to contain 2%, 4%, or 6% lactulose. Technological (lactulose content, instrumental color and texture profile analysis) and sensory (acceptance and check-all-thatapplies tests) analyses were performed on the final product. Results: The lactulose content in the finished product (1.86%±0.23%, 3.16%±0.18%, and 2.51%±1.35%) was lower than the lactulose originally added (2%, 4%, and 6%, respectively). The addition of 4% and 6% lactulose made (p<0.05) the products darker (lower L<sup>⋆</sup>) and redder (lower h) with higher hardness and chewiness values, when compared to control samples. The additions of 2% and 4% lactulose reduce the appearance acceptability of the products, but overall the treatments were well accepted. Conclusion: The use of up to 4% lactulose as a prebiotic in the production of boneless dry-cured hams provides an alternative to improving its nutritional value with little alteration in the technological characteristics and still meeting the sensory characteristics desired by consumers.

        Transcutaneous Electrical Nerve Stimulation of PC5 and PC6 Acupoints Increases Sympathovagal Balance but Not Oxidative Stress in Healthy Subjects: A Randomized Clinical Trial

        Background: Transcutaneous electrical acupoint stimulation (TEAS) improves autonomic balance and reduces oxidative stress in subjects with chronic diseases, that decreases the risk of low-grade chronic inflammatory diseases, including cardiovascular diseases. However, these beneficial effects have never been demonstrated in healthy subjects. Objectives: To evaluate the acute effects of TEAS on autonomic balance and oxidative stress of healthy subjects. Methods: A randomized clinical trial was conducted with male healthy subjects (18-30 years old), randomly allocated to control (no intervention; n = 14), placebo (placebo intervention; n = 14) and TEAS group (at PC5 and PC6 acupoints; n = 13). The protocol consisted of accommodation (20 min), intervention (40 min), and recovery (30 min) periods. The acute effects of TEAS on hemodynamics were studied through measurements of heart rate, blood pressure and double product; on the autonomic nervous system by assessing heart rate variability; and on oxidative stress by quantifying reactive oxygen species in saliva samples, collected at the end of each period. Results: TEAS increased heart rate and double-product compared to control and placebo groups (p < 0.01). Moreover, TEAS increased sympathetic and reduced parasympathetic tonus, increasing the sympathovagal balance compared to the control and placebo groups. However, TEAS exerted no effect on oxidative stress in saliva samples. Conclusion: In healthy subjects, TEAS at PC5 and PC6 acupoints acutely improved autonomic balance, increasing sympathetic and reducing parasympathetic tonus, reflecting little improvement on hemodynamic responses. Whether it could be used as a cardioprotective strategy remains uncertain since it exerted no effect on oxidative stress.
