This paper proposes a simulation-based prediction model to predict the restraining and normal force of draw-beads for the sheet metal forming process. A reliable prediction model is constructed for the equivalent draw-bead by a modified DOE (Design of Experiment) method, which consists of the Box-Behnken design and a simplified full factorial design. To construct prediction models of draw-bead forces, draw-bead forces are first calculated by finite element analysis and confirmed by experiments followed by an approximation with second order regression equations in various design cases. To increase the accuracy of prediction models,normalization of draw-bead forces is conducted based on the effectiveness ratio of design variables in a regres-sion analysis. The normalized draw-bead forces are then approximated by second order regression equations again. The accuracy of the prediction models constructed is verified by comparing the prediction results with the simulation results in the entire design space.

Backgrounds: Bee venom has been used as an alternative medicine for immune-related diseases such as multiple sclerosis (MS). Previously, we showed that Bee venom exerts a therapeutic effect in a mouse model of MS through regulatory T cells (Tregs) and Phospholipase A2 from bee venom (bvPLA2) induces a differentiation of Tregs. Methods: To induce EAE, C57BL/6 mice were injected MOG35-55 peptide in CFA and pertussis toxin. To ascertain whether Tregs were involved in the neural protective effect of bvPLA2, the mice received a PC61 anti- CD25 mAb to deplete Tregs or normal anti-rat IgG1 for an isotype control. To verify the necessity of enzymatic activity experimentally, we synthesized a recombinant bvPLA2 and mutant bvPLA2, which has no catalytic ability. Results: The limb paralysis caused by EAE was significantly attenuated in the bvPLA2-treated mice compared to the PBS-treated mice. The beneficial effects of bvPLA2 disappeared when Tregs were depleted. Synthetic bvPLA2 showed therapeutic effects such as those of the natural bvPLA2; however, the mutant which has no catalytic ability did not. Conclusion: Our findings suggest that bvPLA2 contributes to the control of MS and that its catalytic ability is needed for its therapeutic action.

Previously, we found that Foxp3-expressing CD4+ regulatory T (Treg) cells attenuate cisplatin-induced acute kidney injury in mice and that bee venom and its constituent phospholipase A2 (PLA2) are capable of modulating Treg cells. Here we tested whether PLA2 could inhibit cisplatin-induced acute kidney injury. As a result of treatment with PLA2, the population of Treg cells was significantly increased, both in vivo and in vitro. PLA2-injected mice showed reduced levels of serum creatinine, blood urea nitrogen, renal tissue damage, and pro-inflammatory cytokine production upon cisplatin administration. These renoprotective effects were abolished by depletion of Treg cells. Furthermore, PLA2 bound to CD206 mannose receptors on dendritic cells, essential for the PLA2-mediated protective effects on renal dysfunction. Interestingly, PLA2 treatment increased the secretion of IL-10 in the kidney from normal mice. Foxp3+IL-10+ cells and CD1 1c+IL-10+ cells were increased by PLA2 treatment. The anticancer effects of repeated administrations of a low dose of cisplatin were not affected by PLA2 treatment in a tumor-bearing model. Thus, PLA2 may prevent inflammatory responses in cisplatin-induced acute kidney injury by modulating Treg cells and IL-10 through the CD206 mannose receptor.

This paper proposes prediction models for the quality of a double-layer steel cord. Main process parameters were first selected from experiments according to the usage of straightening devices in the stranding machine. The stranding experiments for selected process parameters were conducted based on the experimental table established by the Taguchi method. Prediction models were constructed from the regression analysis with the measured assessment items of a doublelayer steel cord. The prediction models constructed can be utilized as a design guideline effectively for the stranding process design of a double-layer steel cord.
