        Probing the impact of quercetin-7-O-glucoside on influenza virus replication influence

        <P>Background: Influenza virus is still at large and seriously affects social welfare and health. Dianthus superbus is a well-known medicinal plant widely used in Mongolian and Chinese traditional medicine for anti-inflammatory purposes. Purpose: To investigate the influence of this novel herbal medicinal product over virus infection and virus-induced symptoms Method: Quercetin-7-O-glucoside was isolated by bioassay (anti-influenza)-guided fractionation. The structural elucidation was made with 1H-NMR and 13C-NMR. Influenza A/Vic/3/75 (H3N2), A/PR/8/34 (H1N1), B/Maryland/1/59 and B/Lee/40 viruses were used for the evaluation of the antiviral activity. Virus-induced reactive oxygen species and autophagy formation levels were studied. The antiviral mechanism was elucidated via time-dependent, pre-, post-incubation assay methods. The viral RNA replication inhibition of Q7G was analyzed using quantitative RT-PCR method. The blocking of polymerase basic protein subunits of influenza viral RNA polymerase by Q7G was detected by in silico molecular docking assays using AutoDock Vina program with m(7)GTP. Additionally, Q7G was tested against M-MuLV RNA polymerase. Results: Q7G was not cytotoxic (CC50 > 100 mu g/ml) in MDCK cells and it showed 3.1 mu g/ml, 6.61 mu g/ml, 8.19 mu g/ml and 5.17 mu g/ml IC50 values against influenza A/PR/8/34, A/Vic/3/75, B/Lee/40 and B/Maryland/1/59 virus strains, respectively. Treatment of Q7G highly reduced ROS and autophagy formation induced by influenza virus infection. Q7G did not reduce NA activity and did not directly interact with the virus particles. Since viral RNA synthesis was blocked by treatment of Q7G. We targeted viral RNA polymerase for further probing. Interestingly, the binding energy of Q7G on viral PB2 protein was -9.1 kcal/mol and was higher than m(7)GTP recorded as -7.5 kcal/mol. It also was observe to block M-MuLV RNA polymerase. Conclusion: Isolated compound Q7G showed strong inhibition activity against influenza A and B viruses. It also reduced virus-induced ROS and autophagy formation. Q7G does not directly bind to the virus particles and did not affect NA activity. These results indicated that Q7G inhibits viral RNA polymerase, and that it occupies the binding site of m(7)GTP on viral PB2 protein. (C) 2016 Elsevier GmbH. All rights reserved.</P>

        Genetic Variability of mtDNA Sequences in Chinese Native Chicken Breeds

        The variability of mtDNA hypervariable segment I (HVS I) sequences was investigated in a total of 48 birds belonging to 12 Chinese native chicken breeds. Sixteen haplotypes were identified from 35 polymorphic nucleotide sites which accounted for 6.4% of a sequenced 544 bp fragment. Diversity analysis of the haplotypes showed that Tibetan, Langshan and Henan cockfight chicken had only one haplotype, while ancient haplotypes existed in Taihe silky and Chahua chicken. Phylogenetic analysis of the haplotypes suggested that Chinese native chicken breeds shared 5 maternal lineages and some breeds would share the same maternal lineage, regardless of their external features and ecological types. Both divergent and phylogenetic analysis of the haplotypes indicated the close genetic relationships between the Chinese native chicken breeds and G. g. gallus and G. g. spadiceus from different areas, which implied that G. g. gallus and G. g. spadiceus were the original ancestors of the Chinese native chicken breeds.

        New Evidence of Alleles (V199I and G52S) at the PRKAG3 (RN) Locus Affecting Pork Meat Quality

        The porcine PRKAG3 (RN) gene encodes the regulatory gamma subunit of adenosine monophosphate-activated protein kinase (AMPK), which is a good candidate gene affecting meat quality. In this study, the effects of two missense mutations A595G (Ile199Val) and G154A (Gly52Ser) in porcine PRKAG3 gene on meat quality traits were studied in M. Longissimus dorsi (LD), M. Semispinalis capitis (SC) and M. Biceps femoris (BF) from different populations of 326 pigs. The PRKAG3 alleles 199I, 199IV, 52S and 52G were identified with PCR-RFLPs and all genotypes - 199I/199I, 199I/199V, 199V/199V, 52S/52S, 52S/52G and 52G/52G - were found. The frequency of V allele was larger than that of I allele in all populations. I allele frequency was zero in Chinese Meishan pigs (population D) especially. G allele frequency was larger than that of S allele in all populations except Large White (population A). Both variations at the PRKAG3 locus significantly affected these meat quality traits. The pork meat quality has not previously been established in Meishan or crosses thereof. The results suggested that generally pH of LD, SC and BF was higher in Meishan pigs than that in other populations. Moreover, Meishan pigs showed higher water-holding capacity and intramuscular fat (IMF), lower water content and water loss percentage compared to other populations in terms of the two variations. The results present here supply new evidence that alleles V199I and G52S at the PRKAG3 locus affect pork meat quality and provide useful information on pork production.

        Comparison of synthesis methods for BiOI/g-C3N4 heterojunction photocatalysts and testing their visible light activity in sugar processing wastewater treatment

        The study focused on synthesis of BiOI/g-C3N4 heterojunction photocatalysts by various methods toinvestigate their physicochemical properties as well as their performances on photocatalytic treatmentof sugar processing wastewater. The heterojunction catalysts possessed enhanced catalytic activity comparedto the pristine BiOI and g-C3N4. In-situ growth of BiOI on g-C3N4 was determined as the mostpromising method when the pH of the synthesis medium was adjusted to 10. This method provided amore homogeneous distribution and smaller size for the BiOI clusters that formed a unique flower shapewhich enhanced the catalytic activity remarkably. This depicts that synthesis conditions are especiallyimportant to obtain desired physicochemical properties for a photocatalyst since it has a key role inthe crystal structure of BiOI. The optimum nominal weight ratio was determined as 75:25 for BiOI andg-C3N4, respectively by considering the superior morphology and the highest photocatalytic activityfor 45.30% and 23.40% total saccharide (TSC) and total organic carbon (TOC) removal, respectively. Thephotocatalytic mechanism depicted that charge separation in the heterojunction structure was accomplishedvia the new S-scheme and the oxidation of saccharides and intermediates followed differentpathways by reacting with different radicals.

        Effects of Intra-duodenal Infusion of Limiting Amino Acids on Plasma Insulin-like Growth Factor I, Growth Hormone, Insulin and Liver Insulin-like Growth Factor I mRNA in Growing Goat Fed a Maize Stover-based Diet

        The effects of intra-duodenal infusion of methionine (Met), lysine (Lys) and leucine (Leu) on dry matter intake (DMI), the concentrations of insulin-like growth factor I (IGF-I), growth hormone (GH) and insulin in plasma, and liver IGF-I mRNA level were investigated in two experiments for Liuyang Black growing wether goats. In Experiment 1, three goats ($10.0{\pm}0.1$ kg) were fitted with ruminal, proximal duodenal and terminal ileal fistulaes to determine the infusion amounts of Met, Lys and Leu at the duodenum according to essential amino acid flows into the duodenum and their apparent digestibility. The infusion amounts were 0.77 g/d, 0.91 g/d and 0.58 g/d respectively. In Experiment 2, 4 groups of goats (($10.0{\pm}0.2$ kg) for each group, were cannulated at the duodenum, and were infused with a mixture of Met, Lys and Leu (Control), or mixtures with 21% Met, Lys or Leu replaced with glutamate respectively on a nitrogenous basis. The replacement of 21% Met, Lys or Leu with glutamate did not affect intakes of maize stover, concentrate or both (p>0.05) when compared with the control. The replacement of 21% Met or Lys significantly (p<0.05) reduced plasma GH, insulin and IGF-I concentrations and liver IGF-I mRNA level. The replacement of 21% Leu with glutamate reduced (p<0.05) plasma IGF-I concentration only, but not plasma insulin and GH, as well as liver IGF-I mRNA level (p>0.05). The close relationships between supplying Met and Lys in the lumen of the duodenum and plasma IGF-I, GH and insulin concentrations, as well as liver IGF-I mRNA level in this study indicate that the effects of the limiting amino acids on nutrition of animals are likely intermediated via their effects on these hormones, and these hormone profiles could be used as intermediate markers for the limiting order of amino acids.

        Dietary Requirement of True Digestible Phosphorus and Total Calcium for Growing Pigs

        Sixty healthy growing pigs ($Duroc{\times}Landrace{\times}Yorkshire$ with an average BW of 21.4 kg) were used to determine the true digestible phosphorus (TDP) requirement of growing pigs on the basis of growth performance and serum biochemical indices. Pigs were assigned randomly to one of five dietary treatments (12 pigs/diet), representing five levels of TDP (0.16%, 0.20%, 0.23%, 0.26% and 0.39%). There were three replications per treatment, with four pigs (2 barrows and 2 gilts) in each replication (2 pigs/pen) A randomized-block design was used, with pen as the experimental unit. Experimental diets were formulated to provide the 5 TDP levels with a total calcium (Ca) to TDP ratio of 2:1, and offered to pigs at 5% BW for 28 d. The total Ca contents of the five diets were 0.33, 0.38, 0.45, 0.51 and 0.79%, respectively. During the 28-d experimental period, the ADG of pigs was affected by dietary TDP levels as described by Equation 1: y = $-809,532x^4+788,079x^3-276,250x^2+42,114x-1$,759; ($R^2$ = 0.99; p<0.01; y = ADG, g/d; x = dietary TDP, %). The feed:gain ratio for pigs was affected by dietary TDP levels as described by Equation 2: y = $3,651.1x^4-3,480.4x^3+1,183.8x^2-172.5x+10.9$ ($R^2$ = 0.99; p<0.01; y = feed:gain ratio; x = dietary TDP, %). Total P concentrations in serum were affected by dietary TDP levels as described by Equation 3: y = $-3,311.7x^4+3,342.7x^3-1,224.6x^2+195.6x-8.7$ ($R^2$ = 0.99; p<0.01; y = total serum P concentration and x = dietary TDP, %). The highest ADG (782 g/d), the lowest feed:gain ratio (1.07), and the highest total serum P concentration (3.1 mmol/L) were obtained when dietary TDP level was 0.34%. Collectively, these results indicate that the optimal TDP requirement of growing pigs is 0.34% of the diet (e.g., 5.1 g/day for a 30-kg pig that consumed 1.5 kg feed daily) at a total Ca to TDP ratio of 2:1.

        Evaluation of Ni Film Interfacial Energy Release Rate on Titanium and Stainless Steel Substrates

        An expression including the effect of residual stress on the interfacial energy release rate is proposed for peeling experiments according to the energy-balance argument. The influence of residual stress on the external work is also contained in the expression. Two numerical methods are employed to evaluate the values of the work expenditure G_db, which is the actual energy dissipated during bending of the peel arm near the peel front. The peeling method is employed to test the interfacial energy release rates, G, for Ni films on Titanium and stainless steel substrates. The results indicate that the value of G for Ni films on stainless steel substrate is about 5.47-6.03 N/m, while 5.23-6.71 N/m for Ni films on titanium substrate; the interfacial energy release rates, G, do not depend on the residual stress in film, film thickness nor peel angle. The effect of residual stress in film on peel strength P/h is also discussed.

        MINERAL NUTRITION OF GRAZING SHEEP IN NORTHERN CHINA I. MACRO-MINERALS IN PASTURE, FEED SUPPLEMENTS AND SHEEP

        This study determined the macro-mineral levels in plants and sheep, at different times during the year, at three farms in northern China. Samples of plants, animal tissues and faeces were collected at 5 to 8 times during the year from each site. They were analysed for calcium, sodium, phosphorus, magnesium and potassium. Sodium concentrations in plants were below those recommended for optimum animal production at all sites for all or part of the year (0.01-1.66 g/kg DM). Low concentrations of sodium in faeces were measured and signs of sodium deficiency (soil ingestion) were observed on one farm. There were seasonal trends in other mineral levels in plants and animals. Plants were lowest in potassium (2.3-13.4 g/kg DM), magnesium (1.28-4.82 g/kg DM) and phosphorus (0.24-1.62 g/kg DM) in winter and spring. However, high levels of these elements were supplied in the feed supplements used at this time of the year. During the periods of rapid pasture growth, in summer and autumn, supplements of feed and salt are often not provide even though pasture concentrations of phosphorus and sodium are low. It may be at these times that sheep will be most susceptible to deficiencies of these elements.

        True Digestibility of Phosphorus in Different Resources of Feed Ingredients in Growing Pigs

        To determine the true digestible phosphorus (TDP) requirement of growing pigs, two experiments were designed with the experimental diets containing five true digestible P levels (0.16%, 0.20%, 0.23%, 0.26% and 0.39%) and the ratio of total calcium to true digestible P (TDP) kept at 2:1. In Experiment 1, five barrows (Duroc${\times}$Landrace${\times}$Yorkshire) with an average initial body weight of 27.9 kg were used in a $5{\times}5$ Latin-square design to evaluate the effect of different dietary P levels on the digestibility and output of P and nitrogen. In Experiment 2, sixty healthy growing pigs (Duroc${\times}$Landrace${\times}$Yorkshire) with an average body weight (BW) of 21.4 kg were assigned randomly to one of the five dietary treatments (12 pigs/diet), and were used to determine the true digestible phosphorus (TDP) requirement of growing pigs on the basis of growth performance and serum biochemical indices. The results indicated that the true digestibility of P increased (p<0.05) linearly with increasing dietary TDP level below 0.26%. The true P digestibility was highest (56.6%) when dietary TDP was 0.34%. Expressed as g/kg dry matter intake (DMI), fecal P output increased (p<0.05) linearly with increasing P input. On the basis of g/kg fecal dry matter (DM), fecal P output was lowest for Diet 4 and highest (p<0.05) for Diet 5. The apparent digestibility of crude protein (CP) did not differ (p>0.05) among the five diets, with the average nitrogen output of 12.14 g/d and nitrogen retention of 66% to 74% (p>0.05), which suggested that there was no interaction between dietary P and CP protein levels. During the 28-d experimental period of Experiment 2, the average daily gain (ADG) of pigs was affected by dietary TDP levels as described by Eq. (1): $y=-809,532x^4+788,079x^3-276,250x^2+42,114x-1,759$; ($R^2=0.99$; p<0.01; y = ADG, g/d; x = dietary TDP, %), F/G for pigs by Eq.         Investigation of PCR-RFLPs within Major Histocompatibility Complex B-G Genes Using Two Restriction Enzymes in Eight Breeds of Chinese Indigenous Chickens

        New polymorphism of major histocompatibility complex B-G genes was investigated by amplification and digestion of a 401bp fragment including intron 1 and exon 2 using polymerase chain reaction-restriction fragment length polymorphism (PCRRFLP) technique with two restriction enzymes of Msp I and Tas I in eight breeds of Chinese indigenous chickens and one exotic breed. In the fragment region of the gene, three novel single nucleotide polymorphisms (SNPs) were detected at the two restriction sites. We found the transition of two nucleotides of A294G and T295C occurred at Tas I restriction site, and consequently led to a nonsynonymous substitution of asparagine into serine at position 54 within the deduced amino acid sequence of immunoglobulin variableregion- like domain encoded by the exon 2 of B-G gene. It was observed at rare frequency that a single mutation of A294G occurring at the site, also caused an identical substitution of amino acid, asparagine 54-to-serine, to that we described previously. And the transversion of G319C at Msp I site led to a non-synonymous substitution, glutamine 62-to-histidine. The new alleles and allele frequencies identified by the PCR-RFLP method with the two enzymes were characterized, of which the allele A and B frequencies at Msp I and Tas I loci were given disequilibrium distribution either in the eight Chinese local breeds or in the exotic breed. By comparison, allele A at Msp I locus tended to be dominant, while, the allele B at Tas I locus tended to be dominant in all of the breeds analyzed. In Tibetan chickens, the preliminary association analysis revealed that no significant difference was observed between the different genotypes identified at the Msp I and Tas I loci and the laying performance traits, respectively.
