        Commercial strain typing of Lentinula edodes with inter-retrotransposon amplified polymorphism-PCR method

        [Aims] Lentinula edodes (Berk.) Pegler has many commercial strains both morphologically and physiologically similar to each other. At present, detection of polymorphism in rDNA-IGS region (Babasaki, 2006) and/or RAPD marker (Zhang and Molina, 1995) is generally used for strain typing of L. edodes. However, it is rather time-and-cost consuming. Inter-retrotransposon amplified polymorphism (IRAP)-PCR method mainly used for horticultural crops takes less time and lower in cost in strain typing (Kalendar et al, 1999). In this study, we designed IRAP primers for L. edodes and verified their strain typing efficiency. [Method] Thirty three strains were provided for this study. Either fungal cultures on PDA or fungal tissues of fruit bodies were cut into approximately 4 x 4 x 4 mm. Total DNA of each samples were extracted by DNeasy Plant Mini Kit (QIAGEN). For PCR, IRAP primer set and Pfu-X polymerase (greiner) were performed. Based on LTR (Long Terminal Repeat) sequence in L. edodes, we designed one set of primers amplifying the regions between retrotransposons. Ampricons were electrophored for 50 min at 100 V on 1.7 % agarose gel with GelRed (Biotium) and evaluated under UV irradiation. [Results] The products obtained by IRAP-PCR were determined using mini-gel electrophoresis system. The band patterns of IRAP-PCR products differ among strains except the ones having the same parental cultivar. The detected bands were bright and clear without smearing. The IRAP-PCR products of fungal cultures on PDA and correlating fungal tissues of fruit bodies showed the same band pattern, suggesting that the procedure is highly reproducible. Thus, it is considered that IRAP-PCR with short ranged (ca. 1 kb) electrophoresis is a time-efficient and practical strain typing method of L. edodes.

      • Speaker Diarization Using Multi-Modal i-vectors

        We propose multi-modal i-vectors, which extend the audio i-vector framework for speaker verification to a multi-modal speaker diarization in movies. In addition to the audio i-vector, which represents a speech utterance in an audio stream by a low-dimensional vector, we extract a visual i-vector from faces in a video segment. The audio and visual i-vectors are concatenated as a multi-modal i-vector clustered in an unsupervised way. We evaluate our method on the Hannah movie dataset. Our experiments show that diarization error rate is improved from 68.3% to 65.5% compared with audio stream only.

      • MURASAKI: Web-based Word Sense Description System

        In this paper, we propose a method to descriptive a Japanese word sense with associative knowledge. The associative knowledge, which are called as descriptors, are extracted from world wide web(www) by focusing on collocation with comparative structure. For extracting descriptors, the specific comparative expressions with context which was generated with a pair of a query word and each related descriptor are retrieved from www. Some experiments have been conducted and those experimental results showed that the performace of our method overcome the baseline and we regard our method as a promising approach.

        8세기 중반 나일관계(羅日關係)의 추이(推移)와 김태렴(金泰廉) 사절단(使節團)의 도일(渡日)

        8세기 중반 신라와 일본의 관계는 정치적 긴장이 표면화된 상태에 있었다. 735년 2월, 신라 조정은 일본에 사신을 파견하여 자국이 “王城國”임을 표방하자 일본 조정은 사신을 돌려보냈다. “왕성국”은 신라가 당의 제후국임을 표방한 상징적 표현으로서, 천황을 중심으로 한 독자적 세계관을 가지는 일본에 대해 동아시아 국제 질서 속에서 자국이 우위에 있음을 확고히 하는데 작용하였다. 이후 양국 사이에 외교 방식을 둘러싼 갈등이 발생하는 등 정치적 긴장이 표면화되었다. 이와 같은 상황에서 752년에 약 9년 만에 양국 간의 외교가 진행되었다. 이때 신라는 왕자 김태렴을 필두로 한 700여 인이라는 사상 최대의 규모를 갖춘 사절단을 일본에 파견하였다. 김태렴 사절단은 신라 사신으로서 약 17년 만에 일본의 京에 들어가 교역 활동을 비롯한 외교를 진행하였다. 김태렴 사절단에 관한 기록을 보면 대체로 그들이 일본에서 朝貢國으로서의 禮를 갖추어 외교를 진행하였다고 서술되어 있다. 기존 연구에서는 김태렴 사절단이 파견 목적을 이루기 위해 그와 같은 외교 방식을 취하였다고 이해해 왔다. 그러나 이와 같은 이해는 일본 율령법의 이념에 의해 윤색된 기록을 근거로 도출된 것이다. 일본 측 사료에는 김태렴 사절단이 상위 국가로서의 예를 갖추어 일본과의 외교를 진행하였다는 기록이 확인되나, 기존 연구에서는 이를 간과해 왔다. 일본 조정은 김태렴 사절단에 대해 외교 방식에 관한 요구 사항을 제시하였다. 이는 752년 4월에 거행된 東大寺 大佛開眼會를 통해 형성된 새로운 세계관에 입각한 것으로서, 신라에 대해 조공국으로서의 예를 갖추도록 요구한 것이다. 그러나 신라는 이를 받아들이지 않고, 종래의 대일 외교 방침을 유지하였다. 김태렴 사절단의 도일은 일본 조정의 요청에 따른 것이었다. 관련 사료에 따르면 신라 조정은 김태렴 사절단을 파견하여 요청된 물품을 전달하는 동시에 경제적 이익을 추구하기 위한 교역 활동을 전개하였음을 알 수 있다. 이는 신라 조정의 정치적 의도에 따른 것으로서, “왕성국” 표방 이래 일본과의 사이에 표면화된 정치적 긴장 속에서 다시금 자국의 문화적․권위적 우위성을 과시하며, 나아가 대내적으로도 왕권의 위상을 높이기 위한 정책이었다. In the middle of the 8th century, political tensions had surfaced in Silla-Japan relations. In February 735, the court of Silla claimed that his country was a "Wand Seong Guk" through his messenger, and the Japanese court sent his messenger home. “Wand Seong Guk” is a symbolic expression that advocates that Silla is a Tang lord, and its own country is superior to Japan, which has its own world view centered on the Emperor, in the international order of East Asia Worked to ensure that Later, political tensions surfaced, with a conflict between the two countries over diplomatic systems. Under these circumstances, diplomacy between the two countries took place in 752 for the first time in about nine years. At this time, Silla dispatched the largest mission ever in Japan, with about 700 people led by prince Kim, Tae-ryeom. The Kim, Tae-ryeom Mission entered the capital of Japan for the first time in about 17 years as an ambassador to Silla and conducted extensive diplomacy, including extensive trade activities. According to the records of the Kim, Tae-ryeom Mission, they stated that they had advanced diplomacy in Japan as the Empire. It has been understood in previous research that Kim, Tae-ryeom's diplomatic approach was used to achieve the mission purpose. However, the record of the Kim, Tae-ryeom Mission is not likely to convey the actual situation, because it is based on the principles of the Japanese Ritsuryo Law, centered on the Emperor. In fact, it has been confirmed in the Japanese archives that the Kim, Tae-ryeom Mission adopted a diplomatic system as a high-ranking nation and advanced diplomacy with Japan. The Imperial Court of Japan has presented the mission to Kim, Tae-ryeom with diplomatic requirements. This was based on a new world view formed through the opening of the Great Buddha Eye of Todaiji Temple in April 752, and demanded that Silla establish a gratitude as a tributary country. However, Silla did not accept this, and maintained its foreign policy with Japan, which has been adopted since the "Wand Seong Guk Case". The arrival of the Kim, Tae-ryeom Mission to Japan was at the request of the Japanese court. The Shilla Imperial Court dispatched the Kim, Tae-ryeom Mission to send the requested goods and at the same time conduct trade activities to pursue economic benefits. The dispatch of a large delegation, led by the Prince, was due to the political intentions of the Silla Court. It was to show off and to increase the status of the kingdom inward.

        X-band Thermionic Cathode RF Gun and Multi-Beam Compton Scattering Monochromatic Tunable X-ray Source

        A Compton scattering X-ray source consisting of an X-band (11.424 GHz) electron linear accelerator (linac) and Q-switched Nd:YAG laser is currently under construction. Monochromatic hard X-rays are required for a variety of medical and biological applications. The new hard X-ray source produces monochromatic X-rays via collision between a 35-MeV multi-bunch (104 bunches in a 1 μs RF pulse) electron beam and 1.4 J/10 ns (532 nm) Nd:YAG laser beam. The linac uses an X-band 3.5-cell thermionic cathode RF gun and an alpha magnet as an injector. The thermionic cathode RF gun is the first of its kind and can generate a high current (2 μA) and a multi-bunch 2-MeV electron beam. To increase the efficiency of the X-ray yield, a laser pulse circulation system is adopted, that can increase the X-ray intensity by up to 50 times. This scheme can produce monochromatic tunable X-rays (10 40 keV) with intensities of 108 109 photons/sec. In addition, the X-ray energy can be changed rapidly over 40 ms by two different wavelength lasers (YAG fundamental (1064 nm), 2nd harmonic (532 nm)) and an optical switch. This quick energy change is very important for living specimens and is very difficult to achieve in light sources such as a large SR (Synchrotron Radiation) source. The system can be used for dual-energy X-ray CT and subtraction X-ray CT to determine the 3D distribution of the atomic number density and electron density, and specified atomic distribution, respectively. In this paper, we describe the details of the system, report on experiments on the X-band thermionic cathode RF gun, and discuss applications of monochromatic X-rays.?

        Molecular genetic analysis of the genes for salinomycin biosynthesis in Streptomyces albus

        Streptomyces albus wild type produced salinomycin and the strain 14∼33 chromosomal DNA was used for the construction of genomic library using pKC 505 cosmid vector. The genomic library was screened using act 1 (gene for actinorhodin synthase-condensing enzyme) probe and p22∼81 was isolated. TLC, HPLC, and MS analyses revealed that S. lividans transformed with the p22∼81 produced salinomycin. The p22∼81 contained about 24 kb insert and a region of 4.4 kb fragment of the insert hybridized with actI and ACP (acyl-carrier protein) probes was sequenced. Five ORFs found in the same direction were homologus to tcmI, tcmK, tcmL, tcmM and actIII, respectively (tcm; tetracenomycin, act; actinorhodin), tcmI is a cyclase in tetracenomycin synthase gene clusters and tcmK & tcmL are gene for heterodimeric beta-acyl synthase. tcmM codes ACP and actIII is a gene for ketoreductase.
