The ginsenoside-hydrolyzing β-glucosidase gene (bgy2) was cloned from Lactobacillus brevis. We expressed this gene in Escherichia coli BL21(DE3), isolated the resulting protein, and then utilized the enzyme for the biotransformation of ginsenosides. The bgy2 gene contains 2,223 bp, and encodes a protein of 741 amino acids that is a member of glycosyl hydrolase family 3. β-Glucosidase (Bgy2) cleaved the outer glucose moieties of ginsenosides at the C-20 position, and the inner glucose at the C-3 position. Under optimal conditions (pH 7.0, 30˚C), we used 0.1 mg/ml Bgy2 in 20 mM sodium phosphate buffer (PBS) for enzymatic studies. In these conditions, 1.0 mg/ml ginsenoside Rb1 and ginsenoside F2 were converted into 0.59 mg/ml ginsenoside Rd and 0.72mg/ml compound K, with molar conversion productivities of 69% and 91%, respectively. In pharmaceutical and commercial industries, this recombinant Bgy2 would be suitable for producting ginsenoside Rd and compound K.

In this paper, we consider the stabilization of the viscoelastic wave equation with variable coefficients in a bounded domain with smooth boundary, subject to linear dissipative internal feedback with a delay. Our stabilization result is mainly based on the use of the Riemannian geometry methods and Lyapunov functional techniques.

This paper deals with a degenerate parabolic system with coupled nonlinear localized sources subject to weighted nonlocal Dirichlet boundary conditions. We obtain the conditions for global and blow-up solutions. It is interesting to observe that the weight functions for the nonlocal Dirichlet boundary conditions play substantial roles in determining not only whether the solutions are global or blow-up, but also whether the blowing up occurs for any positive initial data or just for large ones. Moreover, we establish the precise blow-up rate.

Transferring the recombinant S. cerevisiae T73-63 from 30 oC to 4 oC resulted in 41.4% increment of β-carotene concentration (3.96mg/g dry cell weight) relative to that of 30 oC, which was accompanied with the accumulation of fatty acid and ergosterol. The comparisons of the transcriptional levels of mevalonate pathway genes indicated that the expressions of HMG1, ERG9, ERG19, ERG20 and IDI1 at 4 oC were all higher than those of 30 oC, respectively. This suggested that increased transcriptions of mevalonate pathway genes contribute to the improvement of β-carotene production at low temperature. We also found that supplementation of 30mg/L triclosan, an inhibitor of fatty acid synthesis, led to further 28.3% enhancement of β-carotene concentration (4.94mg/g DCW), which was 18.8% higher than that of 30 oC with the same concentration of triclosan. The higher expressional levels of HMG, ERG19 and ERG20 and the simultaneous increment of ergosterol content (17.8%) suggested that more carbon source was transferred from fatty acid synthesis to mevalonate pathway under the circumstance of appropriately blocking fatty acid synthesis at low temperature (4 oC), which resulted in a higher increment of β-carotene production compared to that of 30 oC. The results of this study collectively suggest that the combination of reducing temperature and adding fatty acid synthesis inhibitors is a potential approach to improve the production of desirable isoprenoid compounds such as carotenoids.

As the most common type of endocrine malignancy, papillary thyroid cancer (PTC) accounts for 85-90% of all thyroid cancers. In this study, we presented the hypothesis that SDC4 gene silencing could effectively attenuate epithelial mesenchymal transition (EMT), and promote cell apoptosis via the Wnt/β-catenin signaling pathway in human PTC cells. Bioinformatics methods were employed to screen the determined differential expression levels of SDC4 in PTC and adjacent normal samples. PTC tissues and adjacent normal tissues were prepared and their respective levels of SDC4 protein positive expression, in addition to the mRNA and protein levels of SDC4, Wnt/β-catenin signaling pathway, EMT and apoptosis related genes were all detected accordingly. Flow cytometry was applied in order to detect cell cycle entry and apoptosis. Finally, analyses of PTC migration and invasion abilities were assessed by using a Transwell assay and scratch test. In PTC tissues, activated Wnt/β-catenin signaling pathway, increased EMT and repressed cell apoptosis were determined. Moreover, the PTC K1 and TPC-1 cell lines exhibiting the highest SDC4 expression were selected for further experiments. In vitro experiments revealed that SDC4 gene silencing could suppress cell migration, invasion and EMT, while acting to promote the apoptosis of PTC cells by inhibiting the activation of the Wnt/β-catenin signaling pathway. Besides, si-β-catenin was observed to inhibit the promotion of PTC cell migration and invasion caused by SDC4 overexpression. Our study revealed that SDC4 gene silencing represses EMT, and enhances cell apoptosis by suppressing the activation of the Wnt/β-catenin signaling pathway in human PTC.

The main purpose of this study is to analyse changes of housing lifestyle in Shanghai with the flow of historial events such as the open door policy following the Opium War. This study aims to find out the potential residential development in the environment coexisting the East and the West. And it discussed the phases of the times in Shanghai in 1949, the year established the People's Republic of China. At last, it mentioned the effects and the development tendency from residential and cultural complex in Shanghai since 1949.
