A novel wheat cysteine-rich receptor-like kinase gene CRK41 is involved in the regulation of seed germination under osmotic stress in Arabidopsis thaliana

        Donghua Chen,Jing Wu,Min Zhao,Xiaoyan Ma,Wei Zhang,Guangmin Xia,Mei Wang 한국식물학회 2017 Journal of Plant Biology Vol.60 No.6

The bread wheat gene TaCRK41 encodes acysteine-rich receptor kinase. It is down-regulated by variousabiotic stresses and by exposure to abscisic acid. A transientexpression experiment in Arabidopsis thaliana protoplastsinvolving a fusion between the sequence encoding greenfluorescence protein and the TaCRK41 allele from either thesalinity tolerant bread wheat cultivar Shanrong No. 3 (SR3)or its salinity sensitive progenitor cultivar Jinan 177 (JN177)showed that the TaCRK41 product is deposited in thecytoplasm. Recombinant TaCRK41 originating from bothSR3 and JN177 displayed no kinase activity in vitro. Theconstitutive expression of TaCRK41 in A. thaliana resultedin a marked reduction in the plants' sensitivity to bothexogenous abscisic acid and to salinity during germination. The level of ABA-induced transcription of ABI3, ABI5 andthe ABI5-controlled genes EM1 and EM6 was significantlyreduced in plants subjected to stress. The data support theidea that TaCRK41 is involved in the regulation of ABAdependentgermination under conditions of osmotic stress.

Astragalin reduces lipopolysaccharide-induced acute lung injury in rats via induction of heme oxygenase-1

        Donghua Zheng,Dawei Liu,Na Liu,Yukun Kuang,Qiang Tai 대한약학회 2019 Archives of Pharmacal Research Vol.42 No.8

Astragalin, a bioactive component of medicinalplants such as Rosa agrestis, has anti-inflammatory andantioxidant features. Induction of heme oxygenase (HO)-1is an effective strategy to reduce excessive generated oxidantsduring the pathogenesis of acute lung injury (ALI). The aim of the present study is to investigate that whetherthe anti-inflammatory and antioxidant features of astragalinis HO-1 dependent in lipopolysaccharide (LPS)-inducedALI. Sprague–Dawley rats were used in animal study. Intratracheal LPS was performed to induce experimentalALI model. Astragalin was administrated 1 h after LPSchallenge. Human lung epithelial cells were used in cellstudy. Samples from rats were harvested at 24 h post LPSchallenge. Astragalin treatment inhibited LPS-inducedinflammatory cells infiltration in the lung and pulmonaryedema. Astragalin treatment markedly enhanced theactivity of HO-1 compared with vehicle-treated group at24 h post LPS challenge. Levels of lipid hydroperoxide, amarker for oxidative stress, were decreased in astragalintreatedanimals compared with vehicle-treated group. However, the protective effect of astragalin on LPS-inducedALI was abolished in an inhibitor of HO-1-treatedanimals. Moreover, the astragalin-induced the upregulationof HO-1 in human lung epithelial cells was inhibited whennuclear factor erythroid-2-related factor 2 (Nrf2) wassilenced by small interfering RNA. Astragalin reducesLPS-induced ALI via activation of Nrf2/HO-1 pathway.

Procaine Attenuates Pain Behaviors of Neuropathic Pain Model Rats Possibly via Inhibiting JAK2/STAT3

        ( Donghua Li ),( Yurong Yan ),( Lingzhi Yu ),( Yong Duan ) 한국응용약물학회 2016 Biomolecules & Therapeutics(구 응용약물학회지) Vol.24 No.5

Neuropathic pain (NPP) is the main culprit among chronic pains affecting the normal life of patients. Procaine is a frequently-used local anesthesia with multiple efficacies in various diseases. However, its role in modulating NPP has not been reported yet. This study aims at uncovering the role of procaine in NPP. Rats were pretreated with procaine by intrathecal injection. Then NPP rat model was induced by sciatic nerve chronic compression injury (CCI) and behavior tests were performed to analyze the pain behaviors upon mechanical, thermal and cold stimulations. Spinal expression of Janus kinase 2 (JAK2) and signal transducer and activator of transcription 3 (STAT3) was detected by qRT-PCR and western blot. JAK2 was also overexpressed in procaine treated model rats for behavior tests. Results showed that procaine pretreatment improved the pain behaviors of model rats upon mechanical, thermal and cold stimulations, with the best effect occurring on the 15th day post model construction (p<0.05). Procaine also inhibited JAK2 and STAT3 expression in both mRNA (p<0.05) and protein levels. Overexpression of JAK2 increased STAT3 level and reversed the improvement effects of procaine in pain behaviors (p<0.01). These findings indicate that procaine is capable of attenuating NPP, suggesting procaine is a potential therapeutic strategy for treating NPP. Its role may be associated with the inhibition on JAK2/STAT3 signaling.

      • Workpiece surface defects recognition based on improved lightweight YOLOv4

        Donghua Hu,Defu Chen,Kun Yan,Yu Cao 제어로봇시스템학회 2022 제어로봇시스템학회 국제학술대회 논문집 Vol.2022 No.11

A novel metal surface defect recognition scheme based on improved lightweight YOLOv4 (You Only Look Once version 4) is proposed for workpiece defects recognition in mass production. First, the image preprocessing method accurately cuts the region of interests. Second, use the lightweight network model MobileNetV2 (Mobile Networks Version2) and depthwise separable convolution replace the original backbone feature extraction network and 3 X 3 Standard convolution of YOLOv4 respectively. Therefore the model size is greatly reduced. The detection accuracy of this method for workpiece defects reaches 90.63%, and the detection speed is 33 frames per second. Compared to the original YOLOv4 model, the model size is reduced by 82.1%, the recognition accuracy is increased by 1.8%, and the processing speed increases by 150%.

      • Precise Inductor Current Ripple Distribution of Three-Level Active Neutral-Point-Clamped Inverter

        Donghua Pan,Mengxing Chen,Xiongfei Wang,Huai Wang,Frede Blaabjerg,Wei Wang 전력전자학회 2019 ICPE(ISPE)논문집 Vol.2019 No.5

In power electronic converters, the inductor current ripple affects the system power loss and should be minimized. This paper studies in detail the output filter inductor current ripple distribution of a three-level active neutral-point-clamped (3L-ANPC) inverter. The peak-topeak current ripple per line cycle is precisely derived according to the modulation index. Then, the maximum current ripple is identified, and the concept of average current ripple is proposed to quantify the overall ripple level. An optimal operating point with minimum ripple level is found, and it is chosen as the nominal operation condition of the 3L-ANPC inverter. Simulations are performed in a case study to confirm the theoretical expectations.

Axial Movements and Length Changes of the Human Lower Esophageal Sphincter During Respiration and Distension-induced Secondary Peristalsis Using Functional Luminal Imaging Probe

( Donghua Liao ),( Christian Lottrup ),( Lotte Fynne ),( Barry P Mcmahon ),( Klaus Krogh ),( Asbjørn M Drewes ),( Jingbo Zhao ),( Hans Gregersen ) 대한소화기기능성질환·운동학회(구 대한소화관운동학회) 2018 Journal of Neurogastroenterology and Motility (JNM Vol.24 No.2

Background/Aims Efficient transport through the esophago-gastric junction (EGJ) requires synchronized circular and longitudinal muscle contraction of the esophagus including relaxation of the lower esophageal sphincter (LES). However, there is a scarcity of technology for measuring esophagus movements in the longitudinal (axial) direction. The aim of this study is to develop new analytical tools for dynamic evaluation of the length change and axial movement of the human LES based on the functional luminal imaging probe (FLIP) technology and to present normal signatures for the selected parameters. Methods Six healthy volunteers without hiatal hernia were included. Data were analyzed from stepwise LES distensions at 20, 30, and 40 mL bag volumes. The bag pressure and the diameter change were used for motion analysis in the LES. The cyclic bag pressure frequency was used to distinguish dynamic changes of the LES induced by respiration and secondary peristalsis. Results Cyclic fluctuations of the LES were evoked by respiration and isovolumetric distension, with phasic changes of bag pressure, diameter, length, and axial movement of the LES narrow zone. Compared to the respiration-induced LES fluctuations, peristaltic contractions increased the contraction pressure amplitude (P < 0.001), shortening (P < 0.001), axial movement (P < 0.001), and diameter change (P < 0.01) of the narrow zone. The length of the narrow zone shortened as function of the pressure increase. Conclusions FLIP can be used for evaluation of dynamic length changes and axial movement of the human LES. The method may shed light on abnormal longitudinal muscle activity in esophageal disorders. (J Neurogastroenterol Motil 2018;24:255-267)
