        Occurrence of biogenic amines in sufu obtained from Chinese market

        This study aims at examining the level of biogenicamines (BAs) in different kinds of sufu commonlyconsumed in China. The correlation between different BAsand physical and chemical index in sufu samples was alsoinvestigated. The results proved that different processingtechnologies altered the distribution of BAs in commercialsufu. Total BA level was significantly correlated with saltcontent and pH. Some of the sufu samples in this surveycontained higher levels of BAs, of which 26.6% of thesamples might induce histamine poisoning, 15.6% mightinduce headache in virtue of phenylethylamine, and 23.4%might cause migraine and headache in virtue of tyramine. Moreover, 6.3% of the sufu samples with total BA contentover 1000 mg/kg may be harmful to human health. Fromthe food safety perspective, some sufu should not beexcessively consumed daily and should be processed understrict sanitary conditions to decrease the BA level.

        Effect of Lactobacillus rhamnosus hsryfm 1301 on the Gut Microbiota and Lipid Metabolism in Rats Fed a High-Fat Diet

        Accumulating evidence indicates that lactic acid bacteria could improve host physiology and lipid metabolism. To investigate the effect of the gut microbiota on host lipid metabolism, a hyperlipidemic rat model was established by feeding rats a high-fat diet for 28 days, and the gut microbiota of the rats was analyzed using real-time PCR before and after administration of Lactobacillus rhamnosus hsryfm 1301 and its fermented milk for 28 days. The findings showed that the Lactobacillus spp., Bifidobacterium spp., Bacteroides spp., and Enterococcus spp. content in the hyperlipidemic rats gut was increased significantly (p < 0.05), while the Clostridium leptum and Enterobacter spp. content was decreased significantly after intervening with L. rhamnosus hrsyfm 1301 and its fermented milk for 28 days (p < 0.05). Furthermore, the lipid levels of the serum and the liver were decreased significantly (p < 0.05) and the fecal water content was increased significantly (p < 0.05) in the hyperlipidemic rats after the intervention, and hepatocyte fatty degeneration of liver tissues was also prevented. A positive correlation was observed between the Clostridium leptum content and the level of serum cholesterol, triglycerides, low-density lipoprotein, and high-density lipoprotein, and a negative correlation was observed between the Enterobacter spp. content and the Lactobacillus spp. and Bifidobacterium spp. content in the hyperlipidemic rats gut. These results suggest that the gut microbiota and lipid metabolism of hyperlipidemic rats could be improved by supplementation with L. rhamnosus hsryfm 1301 and its fermented milk.

      • Entomological Society of China: A brief introduction

        The Entomological Society of China (ESC) is the largest non-government organization in the world serving the professional and scientific needs of entomologists and people in related disciplines. 1. Founded in 1944, the ESC has more than 13800 members affiliated educational or research institutions, health agencies, private industry, and the Chinese government. Members are researchers, teachers, extension service personnel, administrators, marketing representatives, research technicians, consultants, students, and hobbyists. 2. The ESC is a not-for-profit professional society governed by a board comprised of members. ESC is affiliated with the China Association for Science and Technology and its headquarter is attached to the Institute of Zoology, Chinese Academy of Sciences, Beijing. 3. The ESC holds together members and organizes various academic activities in promoting the development of science and technology in the fields of entomology. The society endeavors to popularize scientific knowledge and plays an advisory role in solving important problems related to insects and in entomological theories. The purpose of the society is to promote entomology for the advancement of science and the benefit of society through: scientific and professional communications, outreach on science and public policy, program development, continuing education, and fostering interest in entomology. 4. ESC is vivid in international affairs and has wide relationship with many foreign societies and institutions. Now ESC has established bilateral and multilateral cooperation with the Convention of Biological Diversity, the International Society of Entomology, the Asia-Pacific Entomological Association, and etc. 5. The Supervisory Council (Governing Board) makes important decisions and carries out essential functions of the society. Its members, volunteering their time and energy, are elected by the members for every five years. 6. Activities of the ESC can be roughly grouped into following categories: meeting, programming, consulting and popularizing on entomology. The premier event of the Society is its Annual Meeting. 7. The society have 6 working committees (Science Popularization, Technical Consulting, International affairs, Organizing, Youth, Financial), and 12 special academic committees (Insect Faunistics and Systematics, Insect Physiology and Biochemistry, Ecology, Insecticide and Toxicology, Agricultural Entomology, Forest Entomology, Medical Entomology, Biological Control, Insect Resources, Urban Entomology, Acarology, and Butterflies). 8. The ESC offers journals that provide unsurpassed coverage of the broad science of entomology. The society publishes scientific journals, such as Acta Entomologica Sinica, Insects Science, Chinese Journal of Applied Entomology (former Entomological Knowledge), Acta Zootaxonomica Sinica, and Acta Parasitologica et Medica. 9. The ESC is seeking for new routines to meet the needs of developments of sciences, communities, and economy.

      • Overexpression of Hiwi Promotes Growth of Human Breast Cancer Cells

        The Piwi subfamily comprises two argonaute (Ago) family proteins, which are defined by the presence of PAZ and Piwi domains, with well known roles in RNA silencing. Hiwi, a human Piwi subfamily member, has been shown to play essential roles in stem cell self-renewal and gametogenesis. Recently, accumulating reports have indicated that abnormal hiwi expression is associated with poorer prognosis of multiple types of human cancers, including examples in the breast. However, little is known about details of the oncogenic role of hiwi in breast cancers. In present study, we confirmed overexpression of hiwi in breast cancer specimens and breast cancer cell lines at both mRNA and protein levels. Thus both RT-qPCR and Western blot data revealed significantly higher hiwi in intratumor than peritumor specimens, overexpression being associated with tumor size, lymph node metastasis and histological grade. Hiwi overexpression was also identified in breast cancer cell lines, MDA-MB-231 and MCF-7, and gain-of-function and loss-of-function strategies were adopted to identify the role of hiwi in the MCF-7 cell growth. Results demonstrated that hiwi expression in MCF-7 cells was significantly up- or down-regulated by the two strategies. We next evaluated the influence of hiwi overexpression or knockdown on the growth of breast cancer cells. Both cell count and colony formation assays confirmed promoting roles of hiwi in MCF-7 cells, which could be inhibited by hiwi specific blockage by siRNAs. In summary, the present study confirmed overexpression of hiwi in breast cancer specimens and breast cancer cell lines, and provided e vidence of promotion by hiwi of cell growth. The results imply an oncogenic role of hiwi in breast cancers.

      • Bufalin Induces Mitochondrial Pathway-Mediated Apoptosis in Lung Adenocarcinoma Cells

        Background: To evaluate the effects of bufalin in A549 human lung adenocarcinoma epithelial cells in vitro and assess the underlying mechanisms. Materials and Methods: Human A549 non-small cell lung cancer (NSCLC) cells were treated with various concentrations of bufalin. Cell proliferation was measured by CCK-8 assay, apoptotic cell percentage was calculated by flow cytometry and morphological change was observed by inverted phase contrast microscopy/transmission electron microscopy. In addition, the membrane potential of mitochondria was detected by JC-1 fluorescence microscopy assay, and the related protein expression of cytochrome C and caspase-3 was analyzed by Western blotting. Results: Bufalin could inhibit the proliferation of A549 cells via induction of apoptosis, with the evidence of characteristic morphological changes in the nucleus and mitochondria. Furthermore, bufalin decreased the mitochondrial membrane potential with up-regulation of cytochrome C in the cytosol, and activation of caspase-3. Conclusions: Bufalin inhibits the proliferation of A549 cells and triggers mitochondria-dependent apoptosis, pointing to therapeutic application for NSCLC.

      • The Strategic Impact of the Summer Palace on China′s ″Silicon Valley″

        The Haidian Garden of Zhongguancun Science and Technology Zone is an area with high intellectual intensity. It is therefore known as China's Silicon Valley. The Summer Palace, a World Heritage Site, sits in the northwest part of it. 250 years ago, the construction of imperial gardens in this area, including the Summer Palace, gave birth to the appearance and prosperity of the town of Haidian. This will also provide various opportunities for the growth of the science and technology. Today the green space, the cultural and ecological environment of the Summer Palace, and its 3-kilometer buffer zone are becoming one important strategic factor for the Haidian Garden to attract talents, improve international competitiveness and realize sustainable development. How to taking advantage of the favorable resources, to achieve balance between protection of the environment around the Summer Palace and urban development, will be of great importance in the future plans of the science and technology.

      • Hypoxia Induced Multidrug Resistance of Laryngeal Cancer Cells via Hypoxia-inducible Factor-1α

        Objectives: To investigate whether hypoxia has an effect on regulation of multidrug resistance (MDR) to chemotherapeutic drugs in laryngeal carcinoma cells and explore the role of hypoxia-inducible factor-$1{\alpha}$ (HIF-$1{\alpha}$). Methods: Laryngeal cancer cells were cultured under normoxic and hypoxic conditions. The sensitivity of the cells to multiple drugs and levels of apoptosis induced by paclitaxel were determined by MTT assay and annexin-V/propidium iodide staining analysis, respectively. HIF-$1{\alpha}$ expression was blocked by RNA interference. The expression of HIF-$1{\alpha}$ gene was detected by real-time quantitative RT-PCR and Western blotting. The value of fluorescence intensity of intracellular adriamycin accumulation and retention in cells was evaluated by flow cytometry. Results: The sensitivity to multiple chemotherapy agents and induction of apoptosis by paclitaxel could be reduced by hypoxia (P<0.05). A the same time, the adriamycin releasing index of cells was increased (P<0.05). However, resistance acquisition subject to hypoxia in vitro was suppressed by down-regulating HIF-$1{\alpha}$ expression. Conclusion: HIF-$1{\alpha}$ could be considered as a key regulator for mediating hypoxia-induced MDR in laryngeal cancer cells via inhibition of drug-induced apoptosis and decrease in intracellular drug accumulation.

        Polymer Nanostructured Components Machined Directly by the Atomic Force Microscopy Scratching Method

        This study aims to machine complex three dimensional nano-structures on polymer surfaces directly using the Atomic Force Microscopy (AFM)-based nano-mechanical processing method. Processing parameters including the normal load, the feed rate, the machining speed and the scanning direction are optimized by the Taguchi method. Their effects on the machined depth and the surface roughness of the machined structures are analyzed. Meanwhile, the AFM PZT tube is controlled to achieve the tip’s three-dimensional motions in space. Using the optimized processing parameters, three-dimensional nanostructures on the surface of PMMA are directly fabricated. It is a novel low-cost manufacturing method for machining complex three-dimensional nano-structures or nano parts on polymer surfaces directly.

        Tucumã (Astrocaryum aculeatum) Prevents Oxidative and DNA Damage to Retinal Pigment Epithelium Cells

        Eye diseases have a negative impact on the eyesight quality of the world population. The age-related macular degeneration (AMD) draws special attention since it is a chronic disorder characterized by oxidative and inflammatory damage to the retinal epithelial pigment, which triggers progressive vision loss. In the Brazilian Amazon, Astrocaryum aculeatum is an Amazonian fruit (Tucumã) used by riverside communities in traditional medicine to treat a number of ailments. These communities have recently shown to have increased longevity and reduced prevalence of age-related morbidity. Thus, the aim of this research was to chemically characterize and analyze the in vitro antioxidant effect and molecular damage prevention of the Tucumã ethanolic extract in retinal pigment epithelium (RPE) cells in a model for AMD. The extract was chemically characterized by ultra-high-performance liquid chromatography (HPLC) coupled with diode-array detection and mass spectrophotometry (HPLC-DAD-MS). In vitro protocols were performed, and the cytopreventive effect of Tucumã on RPE cells exposed to high concentrations of superoxide anion, an oxidant and genotoxic molecule, as well as the effect of Tucumã extract on oxidative and molecular makers were assessed. Biochemical and flow cytometry analyses were conducted in these protocols. The extract presents high concentrations of caffeic acid, gallic acid, catechin, luteolin, quercetin, and rutin. Treatment did not show cytotoxic effects in cells treated only with extract at 50 μg/mL. In fact, it improved cell viability and was able to prevent necrosis and apoptosis, and oxidative and molecular damage was significantly reduced. In summary, Tucumã is an important Amazon fruit, which seems to contribute significantly to improve human health conditions, as our findings suggest that its extract has a relevant chemical matrix rich in antioxidant molecules, and its consumption could improve eye health and contribute to prevention against oxidative stress through cytoprevention, reactive oxygen species reduction, and maintenance of DNA integrity in retinal pigment epithelium (RPE) cells.
