        Recruitment of monocytes primed to express heme oxygenase-1 ameliorates pathological lung inflammation in cystic fibrosis

        Overwhelming neutrophilic inflammation is a leading cause of lung damage in many pulmonary diseases, including cystic fibrosis (CF). The heme oxygenase-1 (HO-1)/carbon monoxide (CO) pathway mediates the resolution of inflammation and is defective in CF-affected macrophages (MΦs). Here, we provide evidence that systemic administration of PP-007, a CO releasing/O2 transfer agent, induces the expression of HO-1 in a myeloid differentiation factor 88 (MyD88) and phosphatidylinositol 3-kinase (PI3K)/ protein kinase B (AKT)-dependent manner. It also rescues the reduced HO-1 levels in CF-affected cells induced in response to lipopolysaccharides (LPS) or Pseudomonas aeruginosa (PA). Treatment of CF and muco-obstructive lung disease mouse models with a single clinically relevant dose of PP-007 leads to effective resolution of lung neutrophilia and to decreased levels of proinflammatory cytokines in response to LPS. Using HO-1 conditional knockout mice, we show that the beneficial effect of PP-007 is due to the priming of circulating monocytes trafficking to the lungs in response to infection to express high levels of HO-1. Finally, we show that PP-007 does not compromise the clearance of PA in the setting of chronic airway infection. Overall, we reveal the mechanism of action of PP-007 responsible for the immunomodulatory function observed in clinical trials for a wide range of diseases and demonstrate the potential use of PP-007 in controlling neutrophilic pulmonary inflammation by promoting the expression of HO-1 in monocytes/macrophages.

      • Cholesterol modulates ion channels via down-regulation of phosphatidylinositol 4,5-bisphosphate

Neurochem.</I> (2010) <B>112</B>, 1286–1294.</P><P>Abstract</P><P>Ubiquitously expressed Mg<SUP>2+</SUP>-inhibitory cation (MIC) channels are permeable to Ca<SUP>2+</SUP> and Mg<SUP>2+</SUP> and are essential for cell viability. When membrane cholesterol level was increased by pre-incubating cells with a water-soluble form of cholesterol, the endogenous MIC current in HEK293 cells was negatively regulated. The application of phosphatidylinositol 4,5-bisphosphate (PIP<SUB>2</SUB>) recovered MIC current from cholesterol effect. As PIP<SUB>2</SUB> is the direct modulator for MIC channels, high cholesterol content may cause down-regulation of PIP<SUB>2</SUB>. To test this possibility, we examined the effect of cholesterol on two exogenously expressed PIP<SUB>2</SUB>-sensitive K<SUP>+</SUP> channels: human <I>Ether-a-go-go</I> related gene (HERG) and KCNQ. Enrichment with cholesterol inhibited HERG currents, while inclusion of PIP<SUB>2</SUB> in the pipette solution blocked the cholesterol effect. KCNQ channel was also inhibited by cholesterol. The effects of cholesterol on these channels were blocked by pre-incubating cells with inhibitors for phospholipase C, which may indicate that cholesterol enrichment induces the depletion of PIP<SUB>2</SUB> via phospholipase C activation. Lipid analysis showed that cholesterol enrichment reduced &ggr;-<SUP>32</SUP>P incorporation into PIP<SUB>2</SUB> by approximately 35%. Our results suggest that cholesterol may modulate ion channels by changing the levels of PIP<SUB>2</SUB>. Thus, an important cross-talk exists among two plasma membrane-enriched lipids, cholesterol and PIP<SUB>2</SUB>.</P>

        Relationship between ICT supporting conditions and ICT application in Chinese urban and rural basic education

        To investigate the relationships between schools’ ICT supporting conditions and ICT application, a total of 2567 primary and secondary schools from 47 prefecture-level cities were surveyed. The questionnaire was administered to collect the ICT supporting conditions in school (ISCS) and the ICT application in school (IAIS) for each school. Among the ISCS factors, the ICT Environment and School Scale were indicated as being significant predictors of IAIS. In addition, the significant predictive relationship between the ISCS and the IAIS factors for rural schools was much stronger than for urban schools. This indicates that the rural and urban schools in China are now at different ICT development stages. Furthermore, rural schools should focus on the improvement of physical ICT infrastructure and the quality digital resources, whereas urban schools should try to improve teachers’ inadequate epistemic beliefs and design thinking ability. Nevertheless, there are other external and individual-level factors that could directly and indirectly influence schools’ ICT application in education.

        Traditional Chinese medicine for COVID-19 pandemic and emerging challenges: An online cross-sectional survey in China

        Background We aimed to investigate use of infection control behaviours, preventative and therapeutic interventions, and outcomes among respondents to an online survey during the COVID-19 pandemic in China. Methods The survey was designed by an international team, translated and adapted to simplified Chinese, including 132 kinds of traditional Chinese medicine (TCM) preparation recommended by guidelines. It was distributed and collected from February to May 2021, with data analysed by WPS spreadsheet and wjx.cn. Descriptive statistics were used to describe demographics and clinical characteristics, diagnosis, treatments, preventative behaviours and interventions, and their associated outcomes. Results The survey was accessed 503 times with 341 (67.8%) completions covering 23 provinces and four municipalities in China. Most (282/341, 82.7%) respondents reported no symptoms during the pandemic and the majority (290/341, 85.0%) reported having a SARS-CoV-2 PCR test at some point. Forty-five (13.2%) reported having a respiratory infection, among which 19 (42.2%) took one or more categories of modern medicine, e.g. painkillers, antibiotics; 16 (35.6%) used TCM interventions(s); while seven respondents combined TCM with modern medicine. All respondents reported using at least one behavioural or medical approach to prevention, with 22.3% taking TCM and 5.3% taking modern medicines. No respondents reported having a critical condition related to COVID-19. Conclusion We found evidence of widespread use of infection control behaviours, modern medicines and TCM for treatment and prevention of COVID-19 and other respiratory symptoms. Larger scale studies are warranted, including a more representative sample exploring TCM preparations recommended in clinical guidelines.       • Hepatic Re-resection Versus Transarterial Chemoembolization for the Treatment of Recurrent Hepatocellular Carcinoma after Initial Resection: a Systematic Review and Meta-analysis

        Background: A systematic review and meta-analysis were performed to compare the post-recurrence survival with hepatic re-resection versus transarterial chemoembolization (TACE) for recurrent hepatocellular carcinoma (HCC) after initial resection. Materials and Methods: All relevant papers were searched via PubMed, EMBASE, and Cochrane Library databases. Hazard ratios (HRs) with 95% confidence intervals (CIs) were pooled using a random-effects model. Subgroup analysis was performed according to country. Sensitivity analysis was performed in studies which clearly reported the recurrent regions, in moderate/high-quality studies, in studies published in full-text form, and in studies published after 2005. Results: In total, twelve papers were included in our study. Five and seven of them were of moderate- and poor-quality, respectively. The overall meta-analysis demonstrated a statistically significantly higher post-recurrence survival in the hepatic re-resection group than in those undergoing TACE (HR=0.64, 95%CI=0.52-0.79, P<0.0001). Heterogeneity was statistically significant and statistical significance remained in the subgroup analysis. Sensitivity analyses were also consistent with the overall analysis. Conclusions: Hepatic re-resection might provide a better post-recurrence survival than TACE for recurrent HCC after initial resection. However, considering the low quality of published studies and the potential bias of treatment selection, further randomized trials should be warranted to confirm these findings.

        Comparison of lifetime-based methods for 2D phosphor thermometry in high-temperature environment

        <P>This paper discusses the currently available techniques for 2D phosphor thermometry, and compares the performance of two lifetime-based methods: high-speed imaging and the dual-gate. High-speed imaging resolves luminescent decay with a fast frame rate, and has become a popular method for phosphor thermometry in recent years. But it has disadvantages such as high equipment cost and long data processing time, and it would fail at sufficiently high temperature due to a low signal-to-noise ratio and short lifetime. The dual-gate method only requires two images on the decay curve and therefore greatly reduces cost in hardware and processing time. A dual-gate method for phosphor thermometry has been developed and compared with the high-speed imaging method through both calibration and a jet impingement experiment. Measurement uncertainty has been evaluated for a temperature range of 473–833 K. The effects of several key factors on uncertainty have been discussed, including the luminescent signal level, the decay lifetime and temperature sensitivity. The results show that both methods are valid for 2D temperature sensing within the given range. The high-speed imaging method shows less uncertainty at low temperatures where the signal level and the lifetime are both sufficient, but its performance is degraded at higher temperatures due to a rapidly reduced signal and lifetime. For <I>T</I>  >  750 K, the dual-gate method outperforms the high-speed imaging method thanks to its superiority in signal-to-noise ratio and temperature sensitivity. The dual-gate method has great potential for applications in high-temperature environments where the high-speed imaging method is not applicable.</P>

        Failure simulation of ice beam using a fully Lagrangian particle method

        A realistic numerical simulation technology using a Lagrangian Fluid-Structure Interaction (FSI) model was combined with a fracture algorithm to predict the fluid-ice-structure interaction. The failure of ice was modeled as the tensile fracture of elastic material by applying a novel FSI model based on the Moving Particle Semi-implicit (MPS) method. To verify the developed fracture algorithm, a series of numerical simulations for 3-point bending tests with an ice beam were performed and compared with the experiments carried out in an ice room. For application of the developed FSI model, a dropping water droplet hitting a cantilever ice beam was simulated with and without the fracture algorithm. The simulation showed that the effects of fracture which can occur in the process of a FSI simulation can be studied.
