      • Genetic Analysis of Apoplastic Proteins in Barley Crosses

        Antifreeze proteins (AFPs) accumulate in the leaves of barley during cold acclimation, where they may inhibit ice recrystallization and produce freezing resistance of the plant. Four parental diallel crosses of the barley varieties were used to determine the heritability of AFPs and the relationship between the accumulation level of AFPs and freezing resistance. The concentration of apoplastic proteins in the cold-acclimated leaves was increased in the mean by four-fold over as compared with that of nonacclimated. The diallel cross analyses revealed that the gene of Sacheon 6 was dominant and those of Reno and Dongbori 1 were recessive. The AFPs had high narrow-sense heritabilities. The general combining ability effects of Reno and Dongbori 1 were much higher than the other parents. The bands of 32-kD for GLP, 35-& 28-kD for CLP and 25-, 22- & 16-kD for TLP were observed in the apoplastic extracts from cold-acclimated plants, but there were no clear differences between the parents and Fl hybrids. The concentrations of AFPs were significantly correlated with the degree of freezing resistance, indicating that the concentration of AFPs in the plant is the very important factor for freezing resistance.

      • 토양으로부터 分離한 Bacteriophage의 특성

        환경 미생물 분야와 산업미생물 분야 및 분자생물학 분야에서 주목을 받고 있고, 많은 연구가 된 새로운 E. coli의 bacteriophage를 토양으로 부터 분리해서, 그 물리적인 특성과 핵산을 밝혀내, phage상호간의 관련성을 검토하여 보았다. 그 결과를 요약하면 다음과 같다. 1. 토양으로 부터 분리한 phage는 숙주균에 따라 SJ-1, SJ-2, SJ-3, SJ-4, SJ-5, SJ-6, SJ-7이라 명명하였다. SJ phage들은 높은 숙주특이성을 나타내 phage SJ-2, SJ一7은 단일종 숙주균에만 감염이 되었고, SJ-1, SJ-3, SJ-4, SJ-5, SJ-6은 다중독성을 나타내었다. 2. 7종류의 SJ-phage는 본 실험조건하에서는 모두 virulent phage임이 밝혀졌다. 모든 용균반의 형태는 중앙이 투명한 둥근 모양이었으며 주변에 무리가 있는 것과 없는것 두가지 형태를 나타냈다. 용균반의 크기는 1-5mm이었으며, SJ-5phage의 용균반이 5mm로서 가장 컸고 SJ-7 phage가 가장 적었다. 숙주균은 OD_600 0.3-0.5상태에서 가장 감염되기가 좋았다. 그리고 상층배지의 한천 농도와 양은 0.5%와 3ml이 가장 좋은 조건이었다. 3. 일단 증식 실험의 결과를 보면 SJ-phage는 잠복기가 15-50분이었고 평균 phage방출수는 190-250이었다. SJ-6과 SJ-7이 잠복기가 15분으로 가장 짧았고 SJ-4가 50분으로 가장 길었다. 평균 방출수는 SJ-3과 SJ-5가 250정도로 많았으며 SJ-1과 SJ-2는 약 190정도였다. 4. SJ-phage는 PD buffer에서 안정하였고, magnesium ion 5×10^-3M의 첨가는 phage에게 큰 안정성을 주었다. SJ-phage는 pH6과 pH9사이에서 안정하였으며 그 pH는 숙주균의 최적 pH였다. SJ-phage의 온도 불활성화는 40 ℃이상에서 불활성화되기 시작하여 60 ℃이상에서는 완전히 불활성화 되었다. 자외선 조사에 의한 불활성화에서 SJ-phage는 120초 이후에 완전히 불활성화 되었다. All phages used in this experiment were isolated from soil of the surburbs of Chun-Nam, and they designated as phage SJ-1, SJ-2, SJ-3, SJ-4, SJ-5, SJ-6, SJ-7. The hosts were JM105, JM109, K802, LE392, MC1000, MV1184 and PSM CC, respectively. These phages were virulent under the experimental conditions. They produced plaques with dear and round without halo. The size of plaques was 1-5mm in diameter. SJ-phages were stable in 5×10^-3M Mg^2+. Thermostability experiments indicated that SJ-phages were stable at 37℃. Optimal pH of the phages were 6-9. Exposure of the phages to U.V. for 120 seconds resulted in complete inactivation. In one-step growth experiments, the latent period at 37 ℃ was about 15-50 min and the average burst see was 190-250. Agarose gel electrophoresis of the phage DNA showed single band of high molecular weight from SJ-1 to SJ-7 phages

      • 엔로플록사신 액제의 닭 호흡기 및 소화기 감염증에 대한 예방 및 치료효과

        We evaluated the efficacy of the liquid form of enrofloxacin against poultry respiratory and digestive infections, the major problems in the poultry industry, from diverse diagnosis viewpoints. Results obtained through this study were as follows: 1. The liquid form of enrofloxacin showed good acceptability to poultry owing to no abhorrence to the drug by chickens, which indicated its suitability for mixing in drinking water. 2. The application of the enrofloxacin formulation improved the productivity of the chicken farm to a great extent by lowering the death toll of chicken raised in the farm. 3. The use of the drug shortened the days to the market, and increased the body weight at the time of marketing. 4. Preventive and therapeutic effects of the liquid enrofloxacin were observed higher than ampicillin, a major antibiotic used in poultry farming. From above results, the liquid form of enrofloxacin would find its use for the prevention and treatment of digestive and respiratory infections, let alone the growth promotion and feed efficiency, especially in the chicken farming practice.

      • 엔로플록사신 액제의 닭유래 병원세균에 대한 시험관내 항균효과 시험

        In this study the antimicrobial spectrum and sensitivity of a liquid formulation of enrofloxacin against the major six pathogenic bacteria isolated from the diseased poultry in Korea were evaluated in comparison with ciprfloxacin (CFX), gentamicin (GM) and penicillin (PC). A liquid form of enrofloxacin, a test material and ciprofloxacin showed at least 83% antibacterial activity against all of six test pathogenic microorganisms at the fixed concentration of 10 ㎍/㎖. Gentamicin and penicillin revealed much lower antibacterial activity at the same condition. We evaluated the liquid form of enrofloxacin as having widespread antibacterial spectrum against poultry infections. The liquid form of enrofloxacin showed potent antibacterial activity, with the MIC range being 0.12∼2 ㎍/㎖ against Gram positives, 0.06∼2 ㎍/㎖ against Gram negatives and 0.5∼2 ㎍/㎖ against Mycoplasma spp. These values were equivalent to or somewhat higher than those of ciprofloxacin, but much lower than those of gentamicn and penicillin. This new liquid form of enrofloxacin would find its way in application of mixing with drinking water for the prevention and treatment of diverse infections in chickens.

        사인별 평균수명 손실량 변화에 관한 연구

        The major purpose of this study is to analyse trends of life expectancy which were lost by cause of death. Data used in Cause-Elimination Model are 'Cause of Death Statistics from 1983 to 1997 published by National Statistical Office of Korea. The major findings may be summarized as follows: (1) The theoretical limit to life expectancy by Cause-Elimination Model is 78.65 in 1983 and 89.72 in 1997 for male, and 85.23 in 1983 and 89.89 in 1997 for female. (2) The effect on life expectancy at birth of eliminating Malignant Neoplasms is 5.06 years for male and 2.63 years for female in 1997. Eliminating the External Causes of Death increase life expectancy at birth by 3.52 years for male and 1.38 years for female in 1997.

      • 냉장고용 폴리우레탄 발포체의 재활용에 관한 연구

        The recycled polyols by the depolymerization of the waste PU foam for refrigerator insulation obtained. The depolymerization was experimented with glycols such as EG and PG. The rigid PU foams were manufactured by Polyols of both virgin and recycled polyols at various mixing ratios and their properties were characterized. Faster reactivity in the synthesis of PU foam was shown with increasing contents of the recycled polyol in the mixed Polyols. The thermal insulation of PU foams by mixed polyols of virgin/recycled polyols was increased but the compession strength was slightly decreased.

        고온기 팽이버섯 병재배 배지제조시 저온수 이용 효과

        This study was conducted to verify the cause of suppression symptom in mycelial growth during summer and to be able to establish a countermeasure. Cultivation of Flammulina velutipes was experimented with varying elapsed time of 0, 3, 6, 9 hours after mixing the sawdust media and two kinds of water temperature (normal water, 24℃ and cold water, 6℃) for mixing sawdust media. There were trends of increased media temperature from 24℃ to 31℃ and decreased pH from 6.5 to 5.2~5.6 with varying elapsed time from mixing the media to sterilization. Bacterial density also increased with bacterial density in Medium 24℃ being 1.9~4.1 times higher than that in Medium 6. Growth of F. velutipes was delayed with dual culture of bacteria isolated from sawdust media. Total nitrogen content of sawdust media was lowered by elapsed time from mixing the media to sterilization. The use of normal water(24℃) delayed mushroom growth by 1~2 days compared with that of cold water(6℃). Furthermore, mycelial growth of F. velutipes in the bottle cultivation ceased 9 hours after mixing the media. Primordia formation of F. velutipes was delayed by 1~3 days by elapsed time after mixing sawdust media, while fruit-body yield decreased by 7~12% 6 hours after mixing the media. Fruit-body yield was more stabilized with the use of cold water(6℃) than with that of normal water(24℃). Results showed that it is effective to use cold water as low as 6℃ in mixing media for F. velutipes cultivation, especially during summer when environmental temperature is high, high pressure sterilization after bottling work can prevent bacterial propagation in the media and can stabilize media ingredient. 여름철 팽이버섯 병재배시 톱밥배지에서 발생하는 균사 생장 억제증상의 원인을 구명하고 그 경감대책을 확립하기 위하여 톱밥배지 배합시 물온도를 24℃(상온수)와 6℃(저온수)로 하고, 배지제조후 살균전 경과시간을 0, 3, 6, 9시간으로 달리하여 팽이버섯의 재배시험을 수행하였다. 배지제조 후 살균전 시간이 경과함에 따라 배지온도가 상승하고, pH는 6.5에서 5.2~5.6으로 낮아지는 경향이었다. 이때 톱밥배지 내에서 세균밀도가 증가하였으며, 물온도 24℃에서는 6℃보다 6시간 이후의 세균밀도가 1.9~4.1배 높았다. 톱밥배지에서 분리한 세균과 대치배양시 팽이버섯균의 생장이 억제되었다. 또한 배지제조후 살균전 시간이 경과함에 따라 배지내의 전질소 함량이 감소하였으며 유리 당의 함량도 종류에 따라서 증가 또는 감소하였다. 배지배합시 24℃물 처리구는 6℃물 처리구에 비하여 팽이버섯 병재배사에서 균배양기간 및 버섯발생기간이 각각 1~2일 지연되는 경향이었으며, 자실체 수량도 저온수 처리구에서 안정적이었다. 그리고 9시간 경과한 배지에서는 균사생장이 정지되었다. 배지제조후 경과시간에 따라 팽이버섯 발생기간은 1~3일이 지연이 되었으며, 자실체 수량은 6시간이 경과된 배지에서 7~12% 감소하는 경향이었다. 본 시험은 여름철 고온기의 팽이버섯 재배시 배지 제조에 6℃이하의 저온수를 사용하여 배지내 미생물 증식을 억제시키고, 입병작업을 마치는 즉시 고압살균 함으로써 배지의 성분을 안정화시켜 버섯의 안정생산에 기여할 것으로 기대된다.

      • 팬티스타킹의 치수규격 제시를 위한 기초 연구

        This study was initiated to investigate the need of sizing system for the production of domestic panty-hose. The study was composed of two parts. At the first part, sizing systems of imprted panty-hoses and domestic one were compared. At the second part, 350 female subjects age 16 to 48 were surveyed to measure their need for the quality of panty-hose. The results were as follows: 1. The imported panty-hose have been produced with various sizes and functions to improve quality of it. Most imported panty-hose were produced in 3 to 6 different sizes and with various fiber contents. 2. The subjects believed that the quality and fit of the imported panty-hose are superior to the domestic ones. 3. The two most important aspects of panty-hose that subjects considered when purchasing panty-hose were fit(37.0%) and color(35.4%). 4. The subjects' satisfaction of fit was differ by their age and body types. The younger subjects(10's and 20's) wanted the longer leg length and the shorter crotch length of panty-hose. The older subjects(40's) felt that the leg length is too long and the hips area was too tight. 5. The shorter subjects(shorter than 156cm) felt that leg length and crotch length are too long. The heavier subjects(heavier than 59kg) felt that hips and thigh were too tight. 6. The younger subjects(10's and 20's)wanted more diversity on color of panty-hose. The 20's also wanted better quality on color fastness after washing it and sweat control at feet while wearing it.

        수액치한법을 이용한 소경재의 산지처리(Ⅰ) : 증산법과 원구법을 이용한 처리 가능성 Feasibility of Treatment Using Transpiration Method and Butt-end Method

        본 연구는 수액치환법을 이용한 침엽수 간벌소경재나 미(저)이용 활용수재의 산지 보존처리 기술을 개발하기 위하여 수행되었다. 본 논문에서는 증산법과 원구법을 이용한 3종 침엽수와 3종 활엽수의 방부제, 난연제, 치수안정제 처리 가능성을 조사하여 보고한다. 수종과 처리약제의 조합에 관계없이 원구법이 증산법에 비하여 약제의 목재내 침투가 양호할 뿐만 아니라 약제의 침투가 균일한 관계로 수분 이동경로를 이용한 처리재 생산은 원구법의 사용이 바람직하였다. 원구법 처리는 수관부를 절단한 후 실시하기 때문에 증산법에 비하여 처리가 용이할 뿐만 아니라 벌채 및 조재 후 산지의 일정 장소로 운재 후 처리를 실시하기 때문에 증산법과 비교할 때 집약적 처리로 단위시간당 처리재 생산량이 높고 산지 환경오염 문제도 예방할 수 있는 장점이 있다. This research was carried out to develop field treatment techniques of thinned small diameter softwood logs and less utilized hardwood logs using sap displacement method. In this paper, we report the feasibility of using transpiration method and butt-end method for the treatment of three softwood species and three hardwood species with preservatives, fire-retardant chemicals, and dimensional stabilizer. Butt-end method was effective as a field treatment technique compared to transpiration method when considered the treatability, easiness of treatment, productivity of treated wood, and environmental aspects related to chemical treatment, regardless of the combination of wood species and chemicals.
