      • OGGI 유전자의 다형성이 방광암에 미치는 영향

        연구목적 : 손상된 유전자의 회복은 암의 발생 및 예방에 중요한 역할을 한다. 잘못된 염기의 제거 및 회복에 중요한 역할을 하는 OGGI 유전자의 다형성이 방광암에 어떠한 작용을 하는지를 환자-대조군 연구를 통해서 조사하였다. 대상 및 방법: 방광암 환자 168명과 건강인 672명을 대조군으로 하여 genomic DNA를 이용하여 SSCP (single-stranded conformational polymorphism), direct DNA sequencing 및 restriction fragment length polymorphism (RFLP) 방법을 통해 환자 및 대조군사이의 OGGI 유전자의 다형성을 조사하였다. 결과: 건강 한국인에서 OGGI 유전자의 유전형을 조사한 결과 econ 6의 Pro324Pro 부위(lb type)와 exon 7의 Ser326Cys 부위 (la type)에 유전자의 다형성이 있음을 발견하였다. Codon 324는 silent polymorphic site였으나 codon 326은 C→G로 nucleotide가 바뀌어 amino acid가 serine→cysteine으로 바뀌는 polymorphic site였다. 대조군에서 codon 326은 Ser326Ser 형이 127례 (18.9%), Ser326Cys 형이 363례 (54.0%) 및 Cys326Cys 형이 182례 (27.1%)이었으며, 168명의 방광암 환자군에서는 Ser326Ser 형이 44례 (26.2%), Ser326Cys 형이 87례 (51.8%)이고 0ys3260ys 형이 37례 (22.0%) 이었다(p=0.034) 즉 Cys326Cys 형 및 Ser326cys 형에 비하여 Ser326Ser 형에서 방광암이 생길 확율이 1.52배 (95% CI=0.439-0.969) 높았다 특히 40세 미만의 남자의 경우 Ser326Ser 형에서 방광암 발생확률이 다른 유전형에 비하여 6.1배 높았다. 결론: OGGI 유전자의 유전형은 서구인과 판이하게 달랐으며 방광암에서는 이 유전자의 변이가 빈번할 뿐아니라 codon 326의 Ser326Ser 형에서는 다른 유전자형에 비하여 방광암이 발생할 확률이 1.5배 이상 높았다. 특히 한국인 40세 미만의 남자에서는 방광암 발생확률이 6배이상 높았다. 본 연구결과 OGGI codon 326의 유전적 다형성은 방광암의 종양화 과정과 관계가 있을 것으로 여겨지며 특히 40세 미만의 한국 남자의 경우는 더욱 밀접한 관계가 있을 것으로 사료된다. Purpose: A repair of damaged DNA has been shown to be involved in the susceptibility to cancer development and prevention. Therefore, it is worth investigating genetic polymorphisms of the OGGl gene associated with the gene repair mechanism. In this study, we examined a possible association of genetic ploymorphisms in OGGl with the risk of bladder tumor. Materials and Methods: Hospital based, case-control investigation was carried out in 168 primary bladder tumor patients and 672 control subjects. We performed the SSCP, PCR-based restriction fragment length polymorphism (RFLP) and direct DNA sequencing to characterize the genetic polymorphism of OGGI in both cases and oontrols. Results: We found two polymorphic sites in OGGl. A Ser/Cys polymorphism at codon 326 (la type) in exon 7 was associated with an exchange of amino acid. Another polymorphic site at codon 324 (1b type) in econ 6 was silent. The association between codon 326 Polymorphism and the risk of the bladder tumor was examined by a age-sex adjusted analysis. We found that the distribution of OGGl Ser326Cys genotypes of controls(Ser/Ser, 18.9% ; Ser/Cys, 54.0% ; Cys/Cys, 27.1%) was significantly different from that of bladder tumor patients (36.2%, 51.8% and 22.0%, respectively) (p = 0.034, adjusted OR = 0.652, 95% Cl = 0.44 - 0.97). Especially, bladder tumor risk in Korean male under 40 years old was approximately 6 times higher than over 40 years old males. Conclusion : Our data suggested that Ser326cys polymorphism at codon 326 of OGGl male below 40 years old in Korea significantly increased the risk of tumorigenesis in the urinary bladder (p = 0.015, adjusted OR = 0.165, 95% CI = 0.04 - 0.75) Our results suggest that the OGGl Ser326Cys Polymorphism might play a role in the tumorigenesis of the bladder.

      • 식도암의 임상적 고찰

        A Clinical observation was made in 52 cases of patients with esophageal cancer at Chosun University Hospital during about 5years from january 1981 until may 1986. The following results were obtained: 1. The peak age of patients was fifth decade. The average age of patients is 55.7 years old. The sex ratio is 7.6 : 1 with a greater prevalence male. 2. Duration of the disease before admission was relatively long, about 4~6 months is 48% of all case. 3. The most common initial symptoms were swallowing difficulty, weight loss, substernal pain. 4. On the majority of patients, Heavy s.noking was manifested. 5. In blood type, A type was predominent. 6. In location of tumor, 69.2% of total cases were located in mid esophagus. 7. In histopathologic finding, 73% of total cases was squamous cell ca.

      • 저수온기 돔류의 녹간증 혈청성분

        본 연구는 2001년 3월에서 5월까지 어류 해상가두리 양식장에서 사육하고 있던 찰돔, Pagrus major, 감성돔, Acanthopagrus schlegeli 및 돌돔, Oplegnathus fasciatuas과 육상에서 사육한 강담돔, Oplegnathus punctatus의 혈청성분을 분석하여 월동시기 이후에 돔류의 폐사원인으로 판단되는 녹간증을 나타내는 병어의 혈청생화학성분을 분석하여 생화학적특성을 조사하고자 하였다. 조사한 어류는 양식자의 그물 가장자리에서 균형을 잃고 옆으로 눕거나 뒤집어져 유영하는 개체들을 수집하여 혈액을 체취, 혈청을 분리한 후 생화학분석기(DRI-CAM 3000)으로 Albumin 의 9가지의 혈청성분을 분석한 결과, GOT, GPT값이 정상어에 비해 매우 높아 돔류의 정상적인 간기능에 장애가 생긴 것으로 판단되었다. 또한 Albumin, Ca, Glucose 값에는 뚜렷한 영향을 주지 않았으나 NH_3는 기준값에 비해 다소 높았다. TBIL, TP, UA는 정상어에 비해 매우 낮은 수치였다. 따라서 이러한 돔류에는 겨울철 저수온으로 사료를 정상적으로 먹지 못하여 어체에 영향결핍을 초래하고 있어 저수온 스트레스 등의 생태습성 부적합에 의하여 논간현상이 발생하는 것으로 판단되었다 A blood biochemical analysis was carried out for the serum of wintering sea bream (red sea bream Pagrus major, black sea bream Acanthopagrus schlegeli, rock bream Oplegenathus fasciantuas) with green liver syndrome. GOT and GPT were significantaly elavated in the diseased sea bream. The value of NH_3 was slightly higher but those of TBIL. TP and UA were lower than the control. But there were no differences in albumin. Ca and glucose between the diseased and the control. These results suggent that a green liver syndrome in the sea bream might be related with nutritional defects and the stress of low water temperature during the winter.

        This study is prepared to introduce design quality improvement measures by the trend and cause analysis of design changes which have been made during operations of construction project for power plant. In this study, two types of analysis tool have mainly taken for the use of the improvement measures. One tool is the trend analysis to identify which action should be taken to cut down design changes. This is also useful to monitor whether project participant's endeavour for the action taken is effective or not, to the project performance and design quality improvement. The other is the cause analysis, by using cause and effect diagram, which is useful to identify main causes of the design changes and to establish the measures to reduce recurrence of the design changes.

      • Finite Element Analysis of the Rolling Contact Fatigue Life of Railcar Wheels

        <P>Railcar wheels and axles belong to the most critical components in railway vehicles. The service conditions of railway vehicles have been more severe in recent years due to speed-up. Therefore, a more precise evaluation of railcar wheel life and safety has been requested. Wheel/rail contact fatigue and thermal cracks due to braking are two major mechanisms of the railcar wheel failure. One of the main sources influencing on the contact zone failure is residual stress. The residual stress in wheels formed during heat treatment in manufacturing changes in the process of braking. Thus the fatigue life of railcar wheels should be estimated by considering both thermal stress and rolling contact. Also, the effect of residual stress variation due to manufacturing process and braking process should be included in simulating contact fatigue behavior. In this paper, an evaluation procedure for the contact fatigue life of railcar wheels considering the effects of residual stresses due to heat treatment, braking and repeated contact load is proposed. And the cyclic stressstrain history for fatigue analysis is simulated by finite element analysis for the moving contact load.</P>

        Fatigue Life Evaluation of Welded Joints by a Strain-life Approach Using Hardness and Tensile Strength

        To evaluate the fatigue lifetime of structures, it is necessary to identify the values of parameters through tests. From the viewpoint of time and cost it is difficult for engineers to get the necessary data through tests. In this study, we surveyed literature and proposed a procedure to identify the fatigue parameters expressed with the Brinell hardness and elastic modulus. After obtaining stress concentration factors by finite element analysis, we calculated fatigue notch factors using Peterson's formula. Taking into account the welding residual stress, which was also obtained by finite element analysis, we evaluated the fatigue lifetime of four kinds of welded joints using the proposed approach. The estimated results are in a good agreement with the experimental results.

        Thermal cracks are among the key factors that control the quality of a brake disk. Thermal cracks may shorten the lifetime of the disc and increase brake noise. Therefore, high heat-resistant brake disk materials are needed. In this study, three kinds of disk material were tested. They are composed of C, Si, Mn, P, S, Cu, Cr, Mo, and Ni. For the three materials, tensile tests, hardness measurement, metallurgical structure analysis, image analyzer analysis, etc were carried out. And friction tests were performed by a small scale dynamometer.

        To evaluate the fatigue lifetime of structures, it is necessary to identify the value of parameters through tests or literature. it is difficult for field engineers to get the necessary data through tests from the viewpoint of time and cost. In this study, we surveyed literature and proposed a procedure to identify the fatigue parameters for fatigue life evaluation by local strain approach.
