        Genotype­specific methylation of HPV in cervical intraepithelial neoplasia

        Objective: Hypermethylation of human papillomavirus (HPV) and host genes has beenreported in cervical cancer. However, the degree of methylation of different HPV typesrelative to the severity of the cervical lesions remains controversial. Studies of the degree ofmethylation associated with the host gene and the HPV genome to the severity of cervicallesions are rare. We examined the association of methylation status between host genes andlate gene 1 (L1) regions of HPV16, 18, 52, and 58 in cervical brushings. Methods: Cervical brushings from 147 HPV-infected patients were obtained. The samplescomprised normal (n=28), cervical intraepithelial neoplasia (CIN) 1 (n=45), CIN2 (n=13), andCIN3/carcinoma in situ (n=61). The methylation status of HPV and host genes was measuredusing bisulfite pyrosequencing and quantitative methylation-specific polymerase chainreaction (PCR). Results: The degree of methylation of L1 in HPV16, 18, and 52 was associated with theseverity of the cervical lesion. In HPV52, C-phosphate-G (CpG) sites 6368m, 6405m, and6443m showed significantly higher methylation in lesions ≥CIN3 (p=0.005, 0.003, and0.026, respectively). Methylation of most HPV types except HPV52 (r<−0.1) was positivelycorrelated with the degree of methylation of host genes including PAX1 and SOX1 (0.4≤r≤0.7). Combining HPV methylation with PAX1 methylation improved the clustering for ≥CIN2. Conclusion: Our study showed that the degree of L1 methylation of HPV16, 18, and 52but not 58 is associated with the severity of cervical lesions. The association betweenHPV methylation and host gene methylation suggests different responses of host cellularepigenetic machinery to different HPV genotypes.

        Large displacement analysis of inelastic frame structures by convected material frame approach

        This paper presents the convected material frame approach to study the nonlinear behavior of inelastic frame structures. The convected material frame approach is a modification of the co-rotational approximation by incorporating an adaptive convected material frame in the basic definition of the displacement vector and strain tensor. In the formulation, each discrete element is associated with a local coordinate system that rotates and translates with the element. For each load increment, the corresponding strain-displacement and nodal force-stress relationships are defined in the updated local coordinates, and based on the updated element geometry. The rigid body motion and deformation displacements are decoupled for each increment. This modified approach incorporates the geometrical nonlinearities through the continuous updating of the material frame geometry. A generalized nonlinear function is used to derive the inelastic constitutive relation and the kinematic hardening is considered. The equation of motion is integrated by an explicit procedure and it involves only vector assemblage and vector storage in the analysis by assuming a lumped mass matrix of diagonal form. Several numerical examples are demonstrated in close agreement with the solutions obtained by the ANSYS code. Numerical studies show that the proposed approach is capable of investigating large deflection of inelastic planar structures and providing an excellent numerical performance.

        Vimentin Is Involved in Peptidylarginine Deiminase 2-Induced Apoptosis of Activated Jurkat Cells

        Peptidylarginine deiminase type 2 (PADI2) deiminates (or citrullinates) arginine residues in protein to citrulline residues in a Ca2+-dependent manner, and is found in lymphocytes and macrophages. Vimentin is an intermediate filament protein and a well-known substrate of PADI2. Citrullinated vimentin is found in ionomycin-induced macrophage apoptosis. Citrullinated vimentin is the target of anti-Sa antibodies, which are specific to rheumatoid arthritis, and play a critical role in the pathogenesis of the dis-ease. To investigate the role of PADI2 in apoptosis, we generated a Jurkat cell line that overexpressed the PADI2 transgene from a tetracycline-inducible promoter, and used a combination of 12-O-tetradecanoylphorbol-13-acetate and ionomycin to activate Jurkat cells. We found that PADI2 overexpression reduced the cell viability of activated Jurkat cells in a dose- and time-dependent manner. The PADI2-overexpressed and -activated Jurkat cells presented typical manifestations of apoptosis, and exhibited greater levels of citrullinated proteins, including citrullinated vimentin. Vimentin overexpression rescued a portion of the cells from apoptosis. In conclusion, PADI2 overexpression induces apoptosis in activated Jurkat cells. Vimentin is involved in PADI2-induced apoptosis. Moreover, PADI2-overexpressed Jurkat cells secreted greater levels of vimentin after activation, and expressed more vimentin on their cell surfaces when undergoing apoptosis. Through artificially highlighting PADI2 and vimentin, we demonstrated that PADI2 and vimentin participate in the apoptotic mechanisms of activated T lymphocytes. The secretion and surface expression of vimentin are possible ways of autoantigen presentation to the immune system.

        Effects of light and heavy metals on Cordyceps militaris fruit body growth in rice grain-based cultivation

        The objective of this study was to examine the light and heavy metals on the fruit body growth of Cordyceps militaris in the rice-based cultivation. Since heavy metals are commonly detected in the paddy field, we investigated the effect of lead, cadmium and mercury on the rice grain-based cultivation of C. militaris. Cordycepin and cordycepic acid were determined by HPLC method. The result showed that the best fruit body growth and bioactive complements was obtained in rice I under 12 h light/dark cycle conditions. The effects of heavy metals (Pb, Hg, and Cd) to the fruit body were remarkable-the inhibition carried a dose-dependent behavior.

Based on the study sample, the new findings help comprehend the overall interconnected relationship which includes the direct and indirect (mediating) effects of information technology service capability and human response capability as well as their interaction (moderation) on customer relationship management advantage. The new insights interprets the two capabilities (human and information technology) are vital to business because they are the foundation set of service resources significantly to enhance customer relationship management advantage.         Evaluating Extreme Rainfall Changes over Taiwan Using a Standardized Index

        <P>The annual daily maximum precipitation (rx1day) is widely used to represent extreme events and is an important parameter in climate change studies. However, the climate variability in rx1day is sensitive to outliers and has difficulty representing the characteristics of large areas. We propose to use the probability index (PI), based on the cumulative density function (CDF) of a generalized extreme value (GEV) distribution to fit and standardize the rx1day to represent extreme event records in this study. A good correlation between the area-averaged PIs of the observed stations and those of the gridded dataset can be found over Taiwan. From the past PI records, there is no distinct trend in western Taiwan before the end of the 20th century, but a climate regime change happened during 2002 - 2003. The dual change effects from both the variance and linear trend of extreme events are identified over the northeastern and southern parts of Taiwan, along with the island's central and southern regions, showing different abrupt changing trends and intensity. The PI can also be calculated using climate projection data to represent the characteristics of future extreme changes. The climate variability of PIs on the present (ALL) and future (RCP4.5 and RCP8.5) scenarios were evaluated using the 16 Couple Model Intercomparison Projects Phase-5 models (CMIP5). The simulated present fluctuations in PIs are smaller than those of actual observations. In the 21st century, the RCP8.5 scenario shows that the PI significantly increases by 10% during the first half of the century, and 14% by the end of the century.</P>

        Overexpression of Ornithine Decarboxylase Suppresses Thapsigargin-Induced Apoptosis

        Ornithine decarboxylase (ODC), the key enzyme of poly-amine biosynthesis, has paradoxical roles in apoptosis. Our published papers show overexpression of ODC pre-vents the apoptosis induced by many cytotoxic drugs. Thapsigargin (TG) is an inhibitor of the sarcoplasmic/en-doplasmic reticulum (ER) Ca2+ ATPase (SERCA) pumps and causes ER stress-induced apoptosis. We used ODC overexpressing cell lines to examine whether overexpres-sion of ODC inhibits TG-induced apoptosis. Our results indicated overexpression of ODC attenuated TG-induced apoptosis. Overexpression of ODC blocked procaspse-4 cleavage and phosphorylation of protein kinase-like ER-resident kinase (PERK), triggered by TG. It also attenuated the increase in CAAT/enhancer binding protein homolo-gous protein (CHOP). Cells with overexpressed ODC had greater Bcl-2 expression. Overexpression of ODC pre-served the expression of Bcl-2, inhibited the increase in Bak and stabilized mitochondrial membrane potential without the influences of TG. Cytochrome c release and downstream caspase activation were blocked. That is, overexpression of ODC inhibits the mitochondria-medi-ated apoptotic pathway, induced by TG. Finally, overex-pression of ODC maintains the protein and mRNA expres-sion of SERCA. In conclusion, overexpression of ODC suppresses TG-induced apoptosis by blocking caspase-4 activation and PERK phosphorylation, attenuating CHOP expression and inhibiting the mitochondria-mediated apoptotic pathway.
