        Vegetation change and emerging research feedback for Korean National Long Term Ecological Research (KNLTER)

        Various responses of forest ecosystems to climate change underscore the need to improve our understanding of the environmentally-driven changes in forests, most effectively by long-term monitoring protocols. We have explored vegetation dynamics based on changes in community structure, species composition, diversity and demographics in four Korean National Long Term Ecological Research (KNLTER) sites--Mt. Nam, Mt. Jeombong, Mt. Worak, and Mt. Jiri--between 2004 and 2009. Most of the sites and forests studied exhibited increments in total basal area, but this was not observed in Quercus mongolica forests in Mt. Nam and Mt. Worak. Stem density exhibited various changes. Altitude gradient was the representative factor in differences in species composition. Two patterns of compositional change--convergence and divergence--were detected. The vegetation of Mt. Nam and Q. mongolica community of Mt. Work showed relatively larger changes in composition. However, in the other sites, few changes were observed. Changes of species richness were not notable except for Mt. Nam, where three species were added in the pine forest, whereas one species disappeared in the oak forest. In the oak forests, mortality rate was as follows (in descending order): Mt. Nam (25.5%), Mt. Jeombong (24.3%), Mt. Worak (16.4%) and Mt. Jiri (0.8%). In the pine forest, the recruitment rate was as follows (in descending order): Mt. Nam (63.7%), Mt. Worak (12.9%), Mt. Jeombong (7.6%) and Mt. Jiri (7.3%). The mortality rate and change rate of basal area were strongly negatively correlated (r = -0.9, P = 0.002), and the recruitment rate and change rate of density were positively correlated (r = 0.77, P = 0.026). In the KNLTER sites, larger vegetation changes were attributed to anthropogenic activities such as salvage logging. Suppression or competition for resources would also affect these changes. Research suggestions such as monitoring to clarify the causes of species mortality were discussed.

      • Tilt test에 依한 循環器機能變化에 關한 硏究

        The Cardiovascular functions are regulated by autonomic nervous system and many physical and hormonal or chemical tests for evaluation of this function have been used. Auther considered that the head-up tilt test may be simple and easy appropriate method to evaluate the function of baroreflex control of cardiovascular functions. Since there is no report on normal and diseases by this method in Korea, auther studied the changes of cardiovascular function by 70 degree Head-up Tilt test using tilting table in 41 cases of normal adult(age 20-39 year group, 25 cases, age 60-75 year group, 16 cases), 16 cases of untreated mild essential hypertension, 9 cases of treated hypertension with pindolol and clonidine and 4 cases of diabetes mellitus with peripheral neuropathy. The cardiovascular functions studied were heart rate, blood pressure and systolic time interval (PEP, ET, PEP/ET, QS_(2)) at 1,5,12 minutes after the head-up tilt. The results were as follows; 1. In normal adult, after 70 degree head-up tilt, the cardiovascular functions were changed since 1 minute and the heart rate increased (average 13/min. after 5 minutes), systolic blood pressure decreased slightly (average 2 ㎜Hg after 5 minutes), diastolic blood pressure increased (average 6 ㎜Hg after 5 minutes), pre-ejection period(PEP) prolonged (average 18 msc increase after 5 minutes), PEP/ET increased (average 0.148 after 5 minutes), ET was shortend (average 52 msc decrease after 5 minutes) and QS_(2) was shortend (average 32 msc decrease after 5 minutes). The systolic blood pressure change was least noticeable up to 12 minutes after the tilt and the heart rate corrected systolic time intervals showed also similar changes. 2. Comparing the change of above cardiovascular functions in young adult and old adult, the changes of the functions were less marked in old adult than in young adult and the systolic blood pressure was rather slightly increased at 1 and 5 minute in old adult and decreased at 12 minute after tilt than control value. 3. In untreated mild essential hypertension, the changes of cardiovascular functions after head-up tilt were less than normal adult which was indicative of reduced baroreflex sensitivity. 4. In treated hypertension and diabetic patients, the changes were less marked than those of untreated hypertension. 5. The test of change of heart rate, blood pressure and systolic time interval by 70 degree head-up tilt test is considered to be a simple appropriate test for the evaluation of sensitivity of the baroreflex.

      • 폐경전후 및 폐경후 골감소에서 호르몬대체요법과 Fluocalcic Effervescent® 병합치료 후 골밀도 및 골대사의 생화학 지표의 변화

        Background: Sodium fluoride stimulates bone formation and has been used to treat osteoporosis for decades despite debate about the antifracture efficacy. For women with low bone mass, the ideal therapy should not only inhibit bone resorption but simultaneously stimulate bone formation to increase the bone mass, the ideal therapy should not only inhibit bone resorption but simultaneously stimulate bone formation to increase the bone mass above the fracture threshold. We thus performed an intervention study to investigate the effect of a low dose fluoride prospectively, in combination with HRT, on bone mineral density (BMD) and biochemical markers of bone turnover. Methods: Thirty perimenopausal or postmenopausal osteopenic women were enrolled, and were treated with Fluocalcic effervescent® (disodium monofluorophophate [MFP] 100㎎ + calcium carbonate 1,250 ㎎) 1 tablet BID and conjugated equine estrogen (HRT) 0.625㎎ QD for 12 months in Department of Internal Medicine, Kosin University Gospel Hospital from January 2001 to December 2002. BMD and chemical markers of bone turnover, serum alkaline phosphatase, serum osteocalcin, and urine deoxypyridinoline(DPD), were evaluated before and 12 months after starting treatment. Results: Serum alkaline phosphatase, serum osteocalcin, and urine DPD after 12 months were not significantly different from those of pre-treatment values. The post-treatment BMD's but they were not correlated with all the pretreatment chemical markers of bone metabolism. Changes of the BMD of L2-L4 were not correlated with the changes of chemical markers except urine DPD. Pretreatment markers, BMD of L2-L4 , serum osteocalcin, serum alkaline phosphatase, and FSH were correlated with changes in BMD of L2-L4. There was no chemical markers correlated with the change in BMD of femoral neck. Conclusion: Twelve-month treatment with MFP and HRT was not associated with improvement in BMD. Pretreatment markers, BMD of L2-L4, serum osteocalcin, serum alkaline phosphatase, and FSH may predict changes in the BMD of L2-L4.

      • Rat의 복강내에 투여한 5-fluorouracil-polyglycolic acid 제형이 장기에 미치는 영향에 관한 연구

        For more effective intraperitoneal chemotherapy, the authers made 5-fluorouracil-polyglycolic acid composite disks(FU-PGA disks) with 5-fluorouracil and polyglycolic acid, a biodegradable polymer. We inserted the FU-PGA disk into the peritoneal cavites of Sprague-Dawley male rats, the control and three experimental groups; one FU-PGA disk insert group(300mg/kg), two FU-PGA disk insert group(600mg/kg), and three FU-PGA disk insert group(900mg/kg). The control group received a similar number of PGA disk inserts. A pharmacokinetic study was performed to measure the 5-fluorouracil concentration in the peritoneal fluid, blood, and tissues(liver, kidney and heart) at 24 hours, 72hours and 168 hours after insertion of the FU-PGA disk. Light and electron microscopic studies were done. The results were as follows: 1) The number of white blood cells and platelets decreased after FU-PGA insertion. The degree was proportional to the duration and amounts inserted. The change in the number of red blood cells varied slightly. 2) Light microscopically, slight changes were noted at 168 hours in the 3 disk insert group (900mg/kg). Mild fatty change and hepatocyte degeneration around the central veins of the liver were noted, with vacuolar degeneration in the proximal convoluted tubules of the kidney. 3) Electron microscopically,the liver showed focal increases of lipid droplets in the hepatocytes, and irregular nuclear membrane with focal nucleolar segregation of the fibrillar and granular elements. Also the double membranous structure of the hepatocytes and Kupffer cells was widened with bleb formation. The kidney showed wide separation of the nuclear double membrane in the proximal convoluted tubular cells and mesangial cells, with vacuolation and myelin figure formation in the mitochondria of the proximal convoluted tubular cells, at 72 hours and 168 hours in the 2 to 3 FU-PGA insert groups. The heart showed focal loss of cristae,vacuolation and myelin figure formation in the mitochondria of the 2 to 3 FU-PGA insert groups. In conclusion, despite the large amont of FU-PGA inserted, the histopathological changes in the liver, kidney and heart were slight, and consonant with the very low amount of 5-fluorouracil concentrations detected in the liver, kidney and heart. The above results suggest that the FU-PGA composite can serve as a new device for releasing drugs in a controlled manner and easily targeted to intraperitoneal organs. This device can improve the efficacy of intraperitoneal chemotherapy for gastric cancer.

        시공간 지각 단서에 의한 단일체(單一體)의 가변적 형태 변화

        존재하는 모든 사물과 현상은 시간의 흐름과 무한한 공간의 틀 속에서 운행한다. 사물의 변화를 느끼게 되는 것은 시간과 공간이 있기 때문이다. 시간과 공간은 존재를 증명하는 기본적 개념으로 인식의 태도는 고정된 정형성의 관점을 벗어난다. 본 연구는 시공간 지각 단서를 기초로 하여 아이덴티티 디자인의 시스템 운용 체계를 중심으로 심볼마크가 상징하고 있는 정체성의 근원을 밝힘으로써 다양하게 응용되고 있는 단일체의 가변적 형태들의 실효성을 검증하고자 했다. 시공간 지각 단서는 선행연구인 "2차원의 평면에서 작용하는 시공간의 지각적 무브망"에서 제시한 시간의 연속체-류, 수, 질, 공간의 확장세-실, 허, 장, 시공복합체의 유기-합, 초, 재로 구분한 분류 체계를 근간으로 설명했으며, 단일체의 가변적 형태변화는 본질적 대상, 즉 단일체의 확장에 의한 것으로 원래의 상태로 되돌아갈 수 있는 상태, 또는 처음의 그 상태로서 원래의 대상물을 지칭하는 것을 의미한다. 이러한 가변적 형태변화에 대한 분류와 설명을 위해 압축, 변이, 파생의 세 가지 대분류를 전제하고 각각의 개념을 재정립하였다. 또한, 단일체의 응용방법에 따라 조형적, 역동적, 조직적인 부분으로 구분하여 연구의 체계성을 확보했다. 단일체의 가변적 형태 변화는 첫째, 결합과 통합의 과정을 거치면서 핵심만을 나타내게 되는 방식으로 설명되는 조형적 형태압축, 둘째, 하나의 단일체가 변화해 가는 일련의 과정 및 변화된 형태의 역동적 형태변이, 셋째, 추가적인 개념이나 시각적 형태가 부가되거나 결합의 과정을 거쳐 새로운 형태가 나타나는 조직적 형태파생으로 설명할 수 있다. 시공간 지각 단서에 의해 지각되는 심볼들이 단순성과 조형성만을 고려한 평면적인 심볼들과 시각이미지의 지각과 인지에서 상반될 수 있는 논리적 근거는 단일체의 가변적인 형태 변화에 있으며 이는 시지각의 차원에서 더 역동적이고 진취적인 유기체로서의 생동감있는 이미지를 전달할 수 있다. 단일체의 가변적 형태 변화를 추구한 아이덴티티 디자인 시스템은 다변화하는 뉴 미디어 시대에서 커뮤니케이션의 변화된 흐름을 주도하는 새로운 시각적 정체성을 보여준다. 상징의 근원적 체계화, 시스템의 창조적 확장성과 미디어에 대응한 운용체계로 인해 기업이미지와 브랜드 이미지 체계를 구축하는 데 있어 고도의 전략적 수단으로 활용될 수 있으며 미래 아이덴티티 디자인의 가치를 한 단계 상승시킬 수 있는 새로운 개발 시스템이 될 수 있을 것이다. All existing objects run within a frame of flowing time and infinite space. Changes in objects are sensed due to the existence of time and space. Space and time are the basic concepts that prove existence, and the manner of awareness breaks away from fixed and formulated perceptions. This study aims to verify the effectiveness of diversely applied variable forms of monolithic objects by discovering the source of the identity symbolized by symbol marks, based on space time perception clues and centered around design systems` operating systems. Space time perception clues were explained on the basis of the `continuum of time - stream, numbers, origin, expansion of space-fullness, voidness, field, Organism of Time & Space agglomeration-convergence, transcendental, existence` classification system as proposed in the precedent study, "Perceptional Mouvement of Time and Space Act on Two-Dimensional Plane". Monolithic object variable form change is caused by the expansion of the fundamental object, the monolithic object, and means the designation of the original object as the condition that allows it to return to its original state, or its original condition. To classify and explain such variable form change, 3 large classification of Compression, Variation, and Derivation were assumed, and each concept was reestablished. Also, the single bodies were classified into formative, dynamic, or systematic according to application methods in order to secure the study`s systemicity. Monolithic object variable form change can be explained in the following three ways: 1. Formative form compression, explained by the method of only the cores being revealed following a process of fusion and integration 2. A series of monolithic object change processes and the dynamic form variations the changed form undergoes 3. Systematic form derivation in which a new form emerges as a result of additional concepts and visual forms being added or fusion processes. The flat symbols that only take into account the simplicity and formativeness of symbols perceived by space time perception clues, and the theoretical basis that can conflict with visual image perception and recognition, are in monolithic object variable form change. This is an organism that is more dynamic and progressive compared to that in a dimension of visual perception, and can deliver life-like images.

        암세포 증식에 대한 YB-1 안티센스 올리고핵산염의 영향

        목적: YB-1은 PCNA, DNA 중합효소 및 MDR 유전자 등의 전사인자로 작용한다. YB-1 유전자는 정상 어른의 간에서는 발현되지 않지만 태아의 간이나 재생 중인 간에서는 발현이 현저히 증가되어서 세포의 증식과 밀접한 관련이 있음이 보고되었다. 본 연구에서는 YB-1 유전자의 안티센스 올리고핵산염을 이용하여 YB-1의 발현을 억제함으로써 암세포 증식을 차단 할 수 있는지를 실험하고 암치료를 위한 유전자 요법으로서의 가능성을 알아보았다. 방법: 세포주로는 Chang liver, HepG2, CT-26 세포를 사용하였고, 사람의 정상세포로는 섬유아세포와 내피세포가 혼재된 조직을 사용하였다. YB-1 유전자의 안티센스 올리고핵산염으로는 YB-1 cDNA의 변역 시작 부위에 상보적인 21mer 올리고핵산염을 제작하여 사용하였다. 세포의 성장은 MTT assay를 이용하였고, 유전자 발현은 Northern blot으로 분석하였으며, 세포주기 변화는 propidium iodide로 염색하여 유식세포분석기로 분석하였다. 동물실험에서는 CT-26 세포를 1.0×10(5)개씩 Balb/c 생쥐의 피하에 접종하여 종양을 유도하였다. 종양이 유도된 Balb/c 생쥐에 YB-1 안티센스 올리고핵산염을 꼬리정맥이나 종양조직에 주사한 후 종양의 크기를 측정하여 종양억제 효과를 관찰하였다. 결과: YB-1 안티센스 올리고핵산염은 CT-26 세포에서는 50 nM 이상 농도에서 Chang liver와 HepG2 세포에서는 10 nM 이상 농도에서 세포주의 성장을 강하게 억제하였지만 정상 조직세포의 성장에는 아무런 영향을 미치지 않았다. 안티센스 올리고핵산염을 DOTAP에 담지한 경우 세포성장 억제에 미치는 효과가 안티센스 올리고핵산염 단독 처치 보다 강하였다. 이때 YB-1의 발현은 증식이 억제된 세포주(Chang liver 및 CT-26)에서는 감소하였으나 정상조직세포에서는 변화가 없었다. 증식이 억제된 세포주에서 세포주기를 살펴보면 초기에 S phase가 감소함을 관찰 할 수 있었다. 꼬리정맥이나 종양조직에 YB-1 안티센스 올리고핵산염을 주입 시 종양의 크기가 유의하게 감소하였다. 결론: 이상의 실험 결과 YB-1 안티센스 올리고핵산염은 암세포의 성장을 저해하며 종양 동물모델에서 종양의 성장을 억제할 수 있음을 시사하였다. Background: Human YB-1 is a transcription factor that binds to the inverted CCAAT box in the promoter region of a variety of genes such as PCNA, DNA polymerase and MDR. In this study we evaluated the effect of YB-1 antisense oligonucleotides on tumor cell growth. Methods: Chang liver, HepG2 and CT-26 cells were cultured as immortalized cell lines. The MTT (3-[4,5-dimethylthiazol-2-yl] 2,5-diphenyltetrazolium bromide) assay, Northern blot and flow cytometric analyses were used to determine cell growth, gene expression and cell cycle changes. In an animal model, CT-26 cells were injected into Balb/c mice to induce tumor; YB-1 antisense oligonucleotides were injected into the tail vein or tumor tissue of the mice; change of tumor size was then measured. Results: Phosphorothioated YB-1 antisense oligonucleotides suppressed the proliferation of the immortalized liver cells (Chang liver cells) and a variety of cancer cells (HepG2 and CT-26 cells); however, it did not inhibit normal cell growth. The DOTAP/antisense oligonucleotide mixture showed stronger effects on cell proliferation than did the antisense oligonucleotide alone. The YB-1 antisense oligonucleotide decreased specific expression of the YB-1 mRNA in the immortalized cancer cell lines. Flow cytometric analysis revealed that the inhibition of cell proliferation might have been due to a decrease in the S phase of the cell cycle. We found that in an animal tumor model, the administration of the YB-1 antisense oligonucleotide, in the vein or tumor tissues, decreased the tumor size significantly. Conclusions: These results suggest that the YB-1 antisense oligonucleotide may inhibit growth of a variety of cancer cells.(Korean J Med 71:293-301, 2006)

        감염 조절용 차단막의 두께가 광중합기의 중합광에 미치는 영향

        연구목적: 본 연구는 감염 조절용 차단막을 여러 겹으로 사용했을 때 광중합기의 광강도와 파장, light diffusion 등에 미치는 영향에 대해 조사하였다. 연구 재료 및 방법: 감염 조절용 차단막은 투명 랩 (크린랩)을 사용하였고 광중합기는 할로겐 광중합기 (Optilux 360)와 LED 광중합기 (Elipar FreeLight 2)를 사용하였다. 차단막을 1겹, 2겹, 4겹, 8겹으로 광중합기의 광섬유말단을 감싸고 휴대용 광강도 측정기 (Cure Rite)로 광중합기의 광강도를 측정하였다. 광중합기를 주문제작한 optical breadboard에 고정시킨 후 휴대용 spectroradiometer (CS-1000)를 이용하여 광중합기의 파장을 측정하였고, DSLR (Nikon D70s)을 이용하여 광중합기의 light diffusion을 사진 촬영하였다. 결과: 광강도 측정 결과는 차단막의 두께가 증가할수록 광강도가 유의하게 감소하였으나 할로겐 광중합기에서 1겹과 2겹 사이에는 유의차가 없었으며, 4겹 이상의 차단막을 투과할 때 광강도가 더 많이 감소하였다. 여러 겹의 차단막을 투과한 광중합기의 전체적인 파장 형태와 peak wavelength의 변화는 관찰되지 않았다. Light diffusion 사진 촬영 시, LED 광중합기에서는 차단막의 두께가 미치는 영향이 없었으나 할로겐 광중합기에서는 차단막을 4겹 사용했을 때부터 중합광이 조사되는 각도가 감소하기 시작하여 8겹 사용했을 때 통계적으로 유의하게 감소하는 것을 볼 수 있었다 (p < 0.05). 결론: 광중합형 복합레진을 광중합할 경우 감염 조절용 차단막이 찢어지는 경우를 대비하여 1겹으로 사용하기 보다는 2겹으로 사용하는 것이 환자간의 교차감염을 예방하는데 유리할 것으로 사료된다. Objectives: This study investigated the effect of infection control barrier thickness on power density, wavelength, and light diffusion of light curing units. Materials and Methods: Infection control barrier (Cleanwrap) in one-fold, two-fold, four-fold, and eightfold, and a halogen light curing unit (Optilux 360) and a light emitting diode (LED) light curing unit (Elipar FreeLight 2) were used in this study. Power density of light curing units with infection control barriers covering the fiberoptic bundle was measured with a hand held dental radiometer (Cure Rite). Wavelength of light curing units fixed on a custom made optical breadboard was measured with a portable spectroradiometer (CS-1000). Light diffusion of light curing units was photographed with DSLR (Nikon D70s) as above. Results: Power density decreased significantly as the layer thickness of the infection control barrier increased, except the one-fold and two-fold in halogen light curing unit. Especially, when the barrier was four-fold and more in the halogen light curing unit, the decrease of power density was more prominent. The wavelength of light curing units was not affected by the barriers and almost no change was detected in the peak wavelength. Light diffusion of LED light curing unit was not affected by barriers, however, halogen light curing unit showed decrease in light diffusion angle when the barrier was four-fold and statistically different decrease when the barrier was eight-fold (p < 0.05). Conclusions: It could be assumed that the infection control barriers should be used as two-fold rather than one-fold to prevent tearing of the barriers and subsequent cross contamination between the patients.

        무증상 갑상선기능저하증 환자에서 Thyroxine 치료에 따른 혈청 지질 및 아포지단백의 변화

        Background: Subclinical hypothyroidism(SCH) is a common biochemical abnormality which can be found in routine screening tests of thyroid function. We are increasingly faced with the question of whether its an indication for thyroxine replacement therapy. The effect of thyroxine replacement on lipid profile in SCH has aroused a great interest because of an association of overt hypothyroidism(OVH) with hyperlipidemia and increased risk of coronary artery disease. Method: We prospectively evaluated the changes in lipids and apoproteins before and after thyroxine replacement therapy in 23 patients with SCH and in 37 patients with OVH. We measured serum total cholesterol and triglyceride using autoanalyzer, high density lipoprotein(HDL) chole-sterol by dextran sulfate method, Apo A1 and Apo B by immunonephelometric assay. Results: Thyroxine replacement therapy significantly decreased total cholesterol, low density lipoprotein(LDL) cholesterol and apo B levels, but did not affect the level of triglyceride, HDL cholesterol or apo AI in patients with OVH. In SCH, thyroxine replacement therapy with the doses to normalize serum TSH concentrations also decreased significantly the level of cholesterol and LDL cholesterol albeit apo B levels did not change. Moreover, in most of patients with OVH (11 of 12) and in all of patients with SCH(5 of 5) who had had hyperchlesterolemia before treatment, thyroxine replament normalized their cholesterol and LDL cholesterol levels. Conclusion: In regard to the beneficial changes in blood lipid levels, patients with SCH should be treated, especially in cases who have other risk factors for the development of atherosclerosis. If thyroxine replacement only will reduce the incidence of coronary artery disease in SCH remains to be elucidated by long-term prospective studies(J Kor Soc Endocrinol 11:41-51, 1996).

        Post mortem insect colonization and body weight loss in rabbit carcasses

        This study considers the impact of various treatments of rabbit carcasses on post mortem insect colonization and weight loss. A statistically accurate time for the change in carcass weight and insect access time was determined using time series analysis depending on different carcass treatments that determined insect access. The decomposition stage was relatively short. Fifteen freshly killed white rabbits (Oryctolagus cuniculus) weighing between 2.5 kg and 4.5 kg were used as animal models in this study. Three carcasses were treated in each of five ways: normal, burnt, wrapped, hung, and immersed. A total of thirteen insect species were attracted to normal, burnt, and hung carcasses. Insects were not collected from sealed and immersed carcasses. The normal and burnt carcasses had a higher number of species and abundance compared to hung carcasses. The present study found no difference in insect colonization in normal and burnt carcasses. The determination of the weight of remaining carcass was clearly divided into the insect‐accessible (i.e., normal, burnt, and hung carcasses) and insect‐inaccessible carcasses (i.e., sealed and immersed carcasses), although no difference was found in the decrease in the rate of the remaining carcass in insect‐inaccessible carcasses.
