        Polyphasic delimitation of a filamentous marine genus, Capillus gen. nov. (Cyanobacteria, Oscillatoriaceae) with the description of two Brazilian species

        Lyngbya C. Agardh ex Gomont is a nonheterocytous cyanobacterial genus whose evolutionary history is still poorlyknown. The traditionally defined Lyngbya has been demonstrated to be polyphyletic, including at least five distinctclades, some of which have been proposed as new genera. Intraspecific diversity is also clearly underestimated in Lyngbyadue to the lack of unique morphological characters to differentiate species. In this study, we describe the new genusCapillus T. A. Caires, C. L. Sant’Anna et J. M. C. Nunes from benthic marine environments, including two new Brazilianspecies (here described as C. salinus T. A. Caires, C. L. Sant’Anna et J. M. C. Nunes, and C. tropicalis T. A. Caires, C. L. Sant’Anna et J. M. C. Nunes), and two species yet to be described, one of them from India (Capillus sp. 2.1), and the otherfrom United States of America, based on strain PCC 7419. Capillus species presented cross-wise diagonal fragmentation,assisted or not by necridic cells, which has not been previously mentioned for Lyngbya. Ultrastructural analyses showedthat C. salinus and C. tropicalis have numerous gas vesicles, which are rarely described for benthic marine species. Thenew genus formed a well-supported clade, and the D1-D1′ and Box B secondary structures of internal transcribed spaceralso supported the proposal of its new species. These findings help to clarify the diversity of species in the Lyngbya complexand the taxonomy of the group, and highlight the need of further floristic surveys in tropical coastal environments,which remain poorly studied.

        Tuning the Assembly of Bispecific Antibodies by Playing on Differential Polypeptide Chain Molar Ratios

        Bispecific antibodies (bsAb) represent the most promising class of therapeutics due to their multiple mode of action. Their development raised tremendous efforts with a special focus on the engineering of these complex molecules, especially when they adopt an asymmetrical format. Various strategies have been successful in improving heterodimerization through the introduction of point mutations to force the correct pairing of opposite chains using either steric, electrostatic steering effects or both. These great advances in protein engineering combined with the development of robust process purification methods to remove homodimers have contributed to the fast progress of bsAbs towards clinics. Nevertheless, there is still a lack of systematic and efficient production methods to fine tune the expression of correctly paired bsAbs. In this study, we have evaluated the effects of manipulating different molar ratios of three polypeptide chains constituting a model bsAb, co-transfected, expressed and assembled in a single Chinese Hamster Ovary cell line. Two parental clones showing different heterodimerization and productivity profiles were chosen. We demonstrated that addition of gene copies at specific ratios to these clones could bias towards the generation of sub-clones with either reduced proper assembly or tune expression for enhanced integral bsAb. Our results demonstrate that expression of bsAb can be influenced beyond the initial engineering stage.

        Full-time RVing is a mobile lifestyle consisting of living year-round in a recreational vehicle (RV). Called full-time RVers or vanlifers, their RV becomes their only dwelling and their only mode of transportation. By being constantly mobile, their conception of home changes and brings a new relation to the world in neo-liberal times. It differentiates them from other mobile people, such as tourists, who still have a located place to come back to, or migrants, who want to settle in their new place of residence. This lifestyle on the road sheds new light on the concept of lifestyle mobilities. Based on research material from several fieldworks in North America and online, this paper focuses on this relationship to place developed by full-time RVers, their idea of home, their ways of making it a home, their ideas of roots, their capacities to be both at home and away, and the challenge they all face with respect to the structural rules of state operation: their need for a home address.

        Role of a Micronized Purified Flavonoid Fraction as an Adjuvant Treatment to Rubber Band Ligation for the Treatment of Patients With Hemorrhoidal Disease: A Longitudinal Cohort Study

        Purpose: Nonsurgical treatment of hemorrhoidal disease (HD) includes medical and instrumental techniques. We aimed to compare the efficacy of the most frequently used nonsurgical strategies, either alone or in combination, applied in an ambulatory setting. Methods: Patients who received nonsurgical treatment for HD by proctology appointment at the Gastroenterology Department of Braga Hospital were evaluated. Isolated rubber band ligation (RBL) and a combination of RBL with a micronized purified flavonoid fraction (MPFF) were the 2 most frequently used strategies. Symptoms of HD (bleeding, pruritus, pain at rest, pain at defecation and prolapse) were assessed at days 0, 7, and 28 by using a severity grading scale (0 to 4/5). A Global Symptom score was constructed to assess the overall severity and compare the overall improvements of the HD symptoms between the 2 most frequently used strategies. Results: Nineteen patients underwent the combined treatment (RBL + MPFF group) and 25 the RBL treatment (RBL group). A comparison of the 2 treatment groups showed significant improvements in the combined treatment group in terms of bleeding at days 7 (P = 0.001) and 28 (P = 0.002) and in the pruritus intensity during the first week (P < 0.001). A trend toward clinical benefit was also verified in the combined treatment group for all other HD symptoms (pain at rest, pain at defecation and prolapse). Conclusion: A combined treatment approach with MPFF and RBL significantly reduced the intensity of bleeding during the first month and the pruritus during the first week.

        An in vitro study was conducted to investigate the effect of propionate precursor (malate or fumarate) on fermentation characteristics, production of conjugated linoleic acid (CLA) and methane (CH₄) by rumen microbes when incubated with linoleic acid (C18:2) as a CLA precursor. Total CH₄ production for 12h incubation, however, was greatly reduced (P<0.0001) by all the supplements compared to that of control, and its production from M-LA or F-LA treatment was smaller than that from LA treatment. Supplementation of M-LA or F-LA also increased concentrations of cis9, trans11-CLA (P<0.039 - P<0.001) for all incubation times and trans10,cis 12-CLA at 1h (P<0.013), 3h(P<0.036) and 12h (P<0.025) incubation times compared to LA supplementation.

        7-Epiclusianone, the Natural Prenylated Benzophenone, Inhibits Superoxide Anions in the Neutrophil Respiratory Burst

        Despite defenses by polymorphonuclear neutrophils in the host against invading agents, overproduction of oxidant species by phagocytes can lead to damage in the surrounding tissues. Several benzophenones have been shown to possess anti-inflammatory properties. The effect of the natural benzophenone 7-epiclusianone isolated from leaves of Garcinia brasiliensis was investigated by using in vitro antioxidant and ex vivo anti-inflammatory assays, focusing on the neutrophil respiratory burst and on the biochemical pathways involved. The bioactive extract, 7-epiclusianone, showed low in vitro antioxidant activity as evaluated by the 2,2-diphenyl-1-picrylhydrazyl free radical scavenging assay, the reducing power test,or the chelating power assay. However, the benzophenone displayed potent activity in the ex vivo model of the neutrophil respiratory burst, inhibiting the generation of superoxide anions in a dose-dependent manner. When the respiratory burst was triggered by N-formyl-methionyl-leucyl-phenylalanine, a chemotactic peptide, the 50% effective concentration (EC50) was 41.18 lg/107 cells. When phagocytes were stimulated directly through protein kinase C via phorbol, the EC50 was 34.3 lg/106cells. The results indicated that 7-epiclusianone was able to down-regulate inflammatory phagocyte superoxide anion release through a mechanism controlled by tyrosine protein phosphorylation and by a direct stimulation of protein kinase C. These findings could lead to new therapeutic approaches for inflammation management and the development of new drugs.

        Characterization of the Phenol Monooxygenase Gene from Chromobacterium violaceum: Potential Use for Phenol Biodegradation

        In this work, the biodegradation mechanism of phenol and sub products (such as catechol and hydroquinone) in Chromobacterium coli was investigated by cloning and molecular characterization of a phenol monooxygenase gene in Escherichia coli. This gene (Cvmp) is very similar (74 and 59% of similarity and identity, respectively) to the ortholog from Ralstomia eutropha, bacteria capable of utilizing phenol as the sole carbon source. The phenol biodegradation ability of E.coli recombinant strains was tested by cell-growth in a minimal medium containing phenol as the sole source of carbon and release of intermediary metabolites (catechol and hydroquinone). Interestingly, during the growth of these strains on phenol, catechol, and hydroquinone accumulated transiently in the medium. These metabolites were further analyzed by HPLC. These results indicated that phenol can be initially orto or para hydroxylated to produce cathecol or hydroquinone, respectively, followed by meta-cleavage of aromatic rings.         Therapeutic Feasibility of the Natural Products in the Heart Complaints: An Overview

        Heart pain is the most frequent complaint leading patients to seek medical help. Functional heart symptoms, especially chest pain, are prevalent and, according to the International Classification of Diseases (ICD-10), are described as “somatoform autonomous functional disorders of the cardiovascular system.” The problem lies in the fact that pain does not always have a somatic background, that is, it may be related to crucial underlying heart disease. The population does not know how to differentiate somatic pain from significant ischemic symptoms, and based on the patient's complaints, traditional medicine ends up treating other underlying cardiac diseases. Many unsuccessful unconventional therapies have been proposed in recent years, including herbal medicines that seek to disrupt the disease's pathogenesis. The present review summarizes research carried out in the last 5 years on natural products' heart complaints, including myocardial ischemia, arrhythmia, and heart failure. Several herbal medicines may be used as a replacement or complementary treatment strategy. A total of 17 medicinal plants have shown promising results in preclinical studies. However, human clinical trials are scarce; only two have been presented. Generally, the data are bland, and many issues have been raised about herbal therapies' safety, efficacy, and mode of action. Besides, relevant clinical trials, future perspectives, and possible clinical applications are discussed.

        Partial denture metal framework may harbor potentially pathogenic bacteria

        PURPOSE. The aim of this study was to characterize and compare bacterial diversity on the removable partial denture (RPD) framework over time. MATERIALS AND METHODS. This descriptive pilot study included five women who were rehabilitated with free-end mandibular RPD. The biofilm on T-bar clasps were collected 1 week (t1) and 4 months (t2) after the RPD was inserted (t0). Bacterial 16S rDNA was extracted and PCR amplified. Amplicons were cloned; clones were submitted to cycle sequencing, and sequences were compared with GenBank (98% similarity). RESULTS. A total of 180 sequences with more than 499 bp were obtained. Two phylogenetic trees with 84 (t1) and 96 (t2) clones represented the bacteria biofilm at the RPD. About 93% of the obtained phylotypes fell into 25 known species for t1 and 17 for t2, which were grouped in 5 phyla: Firmicutes (t1=82%; t2=60%), Actinobacteria (t1=5%; t2=10%), Bacteroidetes (t1=2%; t2=6%), Proteobacteria (t1=10%; t2=15%) and Fusobacteria (t1=1%; t2=8%). The libraries also include 3 novel phylotypes for t1 and 11 for t2. Library t2 differs from t1 (P=.004); t1 is a subset of the t2 (P=.052). Periodontal pathogens, such as F. nucleatum, were more prevalent in t2. CONCLUSION. The biofilm composition of the RPD metal clasps changed along time after RPD wearing. The RPD framework may act as a reservoir for potentially pathogenic bacteria and the RPD wearers may benefit from regular follow-up visits and strategies on prosthesis-related oral health instructions.
