        Cynara scolymus L. Inhibits the LPS-induced Inflammatory Reaction via Suppression of NF-κB Activity in RAW 264.7 Cells

        Cynara scolymus L. (artichoke) has been traditionally used in the treatment of digestive-related disease, severe hyperlipidemia and liver disease. Recently, anti-inflammatory effect of artichoke has been reported by several studies, but its mechanisms are still unclear. In this study, the anti-inflammatory mechanism of artichoke was studied using an in vitro acute inflammation model. The effect of artichoke on the expression of pro-inflammatory cytokines, such as interleukin- 1beta (IL-1β), IL-6, and tumor necrosis factor-alpha (TNF-α) in murine macrophage cell line, RAW264.7, was examined by using ELISA and RT-PCR. As a result, artichoke inhibited the expression of IL-1β, IL-6 and TNF-α in macrophages activated by lipopolysaccharide (LPS) at a dose dependent manner. The expression of inflammatory mediators such as NO and PGE2 was next investigated in RAW264.7 cells stimulated with LPS. Artichoke also inhibited the production of NO and PGE2 in the cells at a dose dependent manner. Artichoke suppressed the mRNA expression of iNOS and COX-2 in macrophages by LPS. The effect of artichoke on the activation of NF-κB was examined and LPS-induced NF-κB activation was decreased by treatment of artichoke, suggesting that artichoke has anti-inflammatory effect by inhibiting NF- κB activation.

        Inhibitory Effect of Farfarae Flos Water Extract on COX-2, iNOS Expression and Nitric Oxide Production in lipopolysaccharide - activated RAW 264.7 cells

        Farfrae Flos has been clinically used for the treatment of asthma in traditional oriental medicine. There is lack of studies regarding the effects of Farfrae Flos on the immunological activities. The present study was conducted to evaluate the effect of Farfrae Flos on the regulatory mechanism of cytokines and nitric oxide (NO) for the immunological activities in Raw 264.7 cells. In Raw 264.7 cells stimulated with lipopolysaccharide (LPS) to mimic inflammation, Farfrae Flos water extract inhibited nitric oxide production in a dose-dependent manner and abrogated inducible nitric oxide synthase (iNOS) and cyclooxygenase (COX-2). Farfrae Flos water extract did not affect on cell viability. To investigate the mechanism by which Farfrae Flos water extract inhibits iNOS and COX-2 gene expression, we examined the on the phospholylation of inhibitor κBα and production of TNF-α, IL-1β and IL-6. Results provided evidence that Farfrae Flos inhibited the production of interleukin-1β (IL-1β) and the activation of phospholylation of inhibitor κBα in Raw 264.7 cells activated with LPS. These findings suggest that Farfrae Flos can produce anti-inflammatory effect, which may play a role in adjunctive therapy in Gram-negative bacterial infections.

      • Interleukin-2와 결핵균 30 kDa 항원이 구개편도 및 말초혈액 T 세포 증식에 미치는 상승효과

        Widespread use of BCG has not controlled tuberculosis, and more effective vaccines are clearly needed. Although chemotherapy will remain the mainstay of antituberculosis treatment, the use of adjunctive immunotherapeutic modalitites is attractive, particularly in persons with drug-resistant tuberculosis. Administration of IL-2 or IFN-γto tuberculosis patients enhance bacillary elimination. Cell-mediated immunity is the critical protective immune response in tuberculosis. Mycobacterial antigens are recognized by T cells and that elicit production of protective cytokines are potentially important vaccine antigens. The 30 kDa antigen is secreted in large quantities by growing mycobacteria. That antigen elicits greater proliferation in lymphocytes from healthy tuberculin reactors than healthy tuberculin nonreactors. In this study, the T lymphocyte proliferative responses to 30 kDa antigen from Mycobactrium tuberculosis H37Rv were examined by using tonsilar and peripheral blood lymphocytes from PPD(+) and PPD(-) tonsilectomized persons. When cultured with 30 kD antigen, tonsilar mononuclear leukocytes and T cells of PPD(+) demonstrated more ^3H-thymidine incorporation than PPD(-) persons (stimulation index was 2.5 and 1.9, 0.8 and 1.0, repectively). Peripheral blood mononuclear cells (PBMC) and peripheral blood T lymphocytes were shown the similar responses to this antigen. The combination of IL-2 and 30 kDa antigen elicited a significant proliferative responsiveness in tonsilar mononuclear leukocytes and T cells of PPD(+) persons (SI was 20 and 14.1). PBMC and peripheral blood T cells of PPD(+) persons were also shown a significant responsiveness, but PPD(-) persons did not show. These results demonstrate that the 30 kDa antigen and IL-2 have a synergistic stimulatory property in mycobacteria sensitizing lymphocytes.

        Post-traumatic cholesteatoma can be originated in the middle ear or mastoid cavity after head trauma such as temporal bone fracture, penetrating injury of the temporal bone or blast injury of the tympanic membrane. It usually takes a long time to develope cholesteatoma after injury, and physicians may overlook its association with previous, trauma. Post-traumatic cholesteatoma can be suspected in patients with head trauma history and atypical cholesteatoma findings. We experienced two cases of cholesteatoma which found behind the intact tympanic membrane in 7 and 13 years old boys with history of head trauma, and we are reporting this with a discussion and literatural review.

        V79 배양 세포에 있어 인삼추출물이 DNA복제 및 회복 합성에 미치는 분자생물학적 연구

        본 연구는 인삼추출물이 DNA 복제 및 회복 합성에 미치는 영향을 조사하기 위해 수행되었다. DNA 복제 및 회복 합성도의 측정은 V79 chinese hamster lung cell의 핵 내 DNA에 [3H]-tymidine이 유입되는 양으로 측정하였다. 그 결과 인삼추출물의 농도(0~1ug/㎖)가 높을수록 점점 더 크게 감소하였다. 이와 같이 인삼추출물에 의하여 DNA 복제 합성도가 감소하는 현상은 농도가 더 높을수록 효과적인 것으로 보아 농도에 의존한다는 것을 알 수 있었다. 한편 DNA 회복 합성도에 인삼추출물이 미치는 영향을 알아보기 위해 우선 DNA를 손상시켰다. DNA를 손상시키는 물질은 여러 가지 있으나, 본 실험에서는 자외선과 methylmethane sulfonate(MMS)를 사용하였다. 이렇게 손상된 DNA가 인삼추출물(0~1ug/㎖)에 의하여 얼마나 회복되었는가를 알아보기 위하여 인삼추출물의 농도를 각각 달리하여 V79 cell에 첨가하였다. 그 결과 인삼추출물의 농도가 높을수록 DNA 회복합성도는 증가하였다. 이와 같이 인삼추출물이 DNA 복제 합성은 억제하고, DNA 회복 합성은 증가시키는 상반된 효과를 보여 주었다. 이러한 상반된 효과는 복제합성과 회복합성의 두 과정에서 서로 다른 효소체계가 관여하기 때문인 것으로 보인다. 인삼추출물이 DNA복제 및 회복합성에 미치는 영향에 대하여 보다 더 구체적인 분자생물학적 메커니즘 연구가 이루어져야 할 것이다.

      • 소아 삼출성중이염에서 중이환기관삽입의 임상적 고찰

        A clinical course following tympanostomy tube on 50 ears in 26 children with secretory otitis media, who were examined every one or two months for at least 24 months postoperatively, was observed. The mean duration of functioning tube was 8.9 months after initial procedure and repeated procedures were required in 32%. And postoperative otorrhea was noted in 15.5%. Atrophy of the ear drum was observed in 30%, tympanosclerosis in 24%, and adhesive otitis media in 6%. At postoperative 24th month, there was no abnormality on the ear drum in 38% and normal hearing in 66%.

        고지방식이를 섭취시킨 흰쥐의 혈청 지질성분에 도라지가 미치는 영향

        Objects : This study was carried out to investigate the effects of Platycodi Radix powder on improvement of lipid composition in high fat-fed rats. Methods : Male sprague-Dawley rats of 7 weeks old, weighing 318.43±3.09g, were divided into two groups; the control group(C group: high fat(30%, Lard)) and platycodi Radix group(P group: 20% Platycodi Radix powder), they were fed experimental diets for 8 weeks. Results : Food intake and body weight gains had differences between the groups. Serum cholesterol levels were significantly lower in P group than C group. The level of serum HDL- Cholesterol was higher in P group than C group. The level of Serum LDL-C was significantly lower in P group than in C group. The ratio of HDC-Cholesterol to total Cholesterol were higher in P group than in the C group, too. The Value of atherogenic index(AI) was determined to be low in P group.

      • 기니픽에서 절단된 외반규관의 골유합에 관한 형태학적 연구

        There has been a little hiistopathologic study concerning the bony healing of injured labyrinth. Authors obserbed a seriease of morphologic change of guinea pig temporal bone after transection of the lateral semicircular canal(LSCC). In the 1st. postoperative week there was a proliferation of perilymphatic connective tissue meshwork along with aggregation of fibroblast around the necrotic material occluding transected bony canal lumen. In the 2nd, postoperative week there was a fibroblastic ingrowth with callous bone formation over the surgical defect of LSCC. In the 4th. postoperative week the fibrous tissue over the surgical defect completed callus formation, and the necrotic material in the canal lumen showed on going new bone formation. These findings suggested that the periosteal layer conduct early calus formation, and the endosteal layer paly a role in forming enchondral bone in late stage.

      • 경유양동상미로접근법에 의한 안면신경감압술 16례

        With increase of traffic accidents, the otologic problems including facial palsy with skull fracture are more concerned. This paper is report our experiences with 16 cases of traumatic facial nerve palsy which have been employed transmastoid supralabyrinthine facial nerve decompression during last four years. The results are as follows: 1. Of the 16 patients, 13 patients(81.3%) were men and 3 patients (18.8%) were women. And sex ratio was 1.4:1 (male predominent). 2. In fracture type, longitudinal fracture was 10 cases (62.5%), transverse fracture was 1 case(6.3%) and mixed fracture was 2 cases(12.5%). 3. Cerebral contusion was combined in 8 cases(50%) and facial bone fracture was combined in 6 cases(37.55). Because of these combined lesions, otologic diagnoses and treatment was delayed. 4. The most common physical finding on clinical examination were a bloody otorrhea(75%) and hearing impairment(75%). 5. In frequency of lesion in facial nerve, tympanic segment(35.7%) was the most common site, followed by geniculate ganglion(14.3%). Suggested mechanisms in facial palsy were nerve compression(57.1%), granulation tissue and congestion. 6. There were improvment of House-Brackmann grades in 13 cases after decompression. 7. Incudostapedial separation was found in 4 cases and incus fracture was found in 2 cases. Hearing improvement was found in 3 cases being ossiculoplasty.
