        카드뮴 축적 변이주인 Pseudomonas maltophilia H - 8M의 특성

        본 연구는 cadmium의 축적균주인 Pseudomonas maltophilia H-8을 MNNG로 처리하여 cadmium축적능이 가장 우수한 변이균주인 Pseudomonas maltophilia H-8M을 얻었고 그 특성을 조사하였다. 변이주 Pseudomonas maltophilia H-8M은 카드뮴농도 1,000ppm에서 36시간 배양했을 시 대조구에 비해 약 80%정도의 성장율을 나타내었다. 카드뮴 이외의 중금속의 첨가에 따른 생육상은 Pb^(2+), Zn^(2+), Hg^(2+), Cu^(2+), Cr^(2+), Co^(2+)등에는 별다른 성장 억제는 없었으나 Sn^(2+)에서는 생육억제가나타났다. 또한 50ppm의 카드뮴을 배지에 첨가 하였을 때 균체내 카드뮴 축적율은 52.92%로 가장 높게 나타났으며 축적부위는 세포벽에 80.3%의 축적율을 나타내었다. This study was carried out to investigate the characteristics of a mutant, Pseudomonas maltophilia Ho8M selected with the treatment of Pseudomonas maltophilia H-8 by N-methyl-N-nitro-N-nitrosoguanidine (MNNG). This mutant showed highest ability of cadmium accumulation. The growth rate of Pseudomonas maltophilia H-8M showed about 80% in 1000ppm Cd containing medium when compare with control for 36h at 30℃. Pseudomonas maltophilia H-8M not inhibited on the growth in addition of various heavy metal such as Hg^(2+), Zn^(2+), Pb^(2+), Cu^(2+), Cr^(2+) and Co^(2+), but inhibited in Sn^(2+) containing medium, respectively. Pseudomonas maltophilia H-8M was accumulated the highest cadmium level of 62.3% on whole cell in the medium containing 50 ppm and 80% of accumulated cadmium was distributed in the cell wall.

        The purpose of this study is to closely examine the image of physical education teachers by conducting a survey of junior high school students and high school students in Daegu sity. Followed is the result of analysis on this survey. Firstly, After examining the image of physical education teachers, the level of satisfaction and interest in the physical education classes and the level of participation in extracurricular activities according to the demography, the quality image of the teachers was relatively high and the image of teachers in performing their duties was normal. The level of satisfaction and interest in the physical education classes was normal overall according to the demography, but in case of students who have experience of physical activities, the level was somewhat high. For the type of participation in extracurricular activities, in case of students of both sexes, in any grade and whose participation in extracurricular activities is relatively low, physical extracurricular activities were the highest. For the students who have experiences of physical activities, physical extracurricular activities were the highest, and for the students who have no experience of physical activities, social extracurricular activities were the highest. Secondly, In case of the image according to the type of participation in extracurricular activities, the group of students who do artistic activities evaluated the image of teachers in professionalism and specialties more positively than any other group. Thirdly, After examining factors which influence the image of physical education teachers considering the level of satisfaction and interest in the physical education classes, the interest in the physical education classes was appeared to influence negatively on the image of attitude. And the satisfaction in the physical education classes was appeared to influence positively on the professionalism and specialties.

      • 쥐 표피의 UV-B 노출에 의한 녹차 열수 추출물의 향산화 효과

        The aims of this studies were carried out the application of antioxidant extracted with boiling water from green tea for the lipid oxidation, protecting effect of UV B exposure. Green tea was prepared by extracting 12.5g of green tea leaves twice with 500㎖ of hot water from 1.25% green tea extracts contained 4.67㎎. The polyphenol of 1.25% green tea extracts was composed the 54.12%(-) epicatechin gallate 26.21%(-)epigallocatechin 10.71% epicatechin gallate, 7.09%(-) epicatechin and 1.85% catechin. Lipid oxidation induced by ultraviolet light(UV B), and electron spin resornance(ESR) study was made on epidermal homogenates prepared from hairless mice skin to the radical mechanism. The exposure of the homogenates to UV B light resulted in the increase of times in lipid peroxide value, which was proportional to the time of UV B exposure with dimethyl-1-pyrrole-N-oxide(DMPO), spin adduct spectrum of lipid radicals was measured following UV exposure. In the presence of various concentration of GTE, the DMPO spin adduct spectrum of lipid radicals was found to be reduced markedly.

      • 사람의 머리카락 분해물로부터 신기능성 항산화제의 개발

        This study designed to elucidate development of functional antioxidants by hydrochloric acid hydrolysate obtained from human hair. Abundant composition amino acid were 14.2% cystine, 13.8% glutamic acid, 8.7% serine, 7.5% leucine, 7.0% threonine, and 6.3% aspartic acid in order, during 3hrs by 6N-HCI hydrolysis. Antioxidative activity of hydrolysate of human hair was shown at highest level in case of hydrolysis of 2hrs. Hydrolysates were fractionated by gel filteration or FPLC the fraction was higher than that of vitamin C, E and BHA. Amine acid sequences of the fraction having antioxidative activity were shown the threonine, alanine, serine, glutamic acid and aspartic acid. The peptides including amino acid sequences observed the chelating action of metal ion by the reaction of P.PPH. In conclusion, the peptides found antioxidative activity of the free radical as binding action.

      • 다섯가지 기초 식품군의 변이원성과 아질산 처리에 의한 변이원성의 변화

        다섯가지 기초 식품의 메타놀 추출물에 대한 돌연변이원성에 대하여 Salmonella typhimurium TA 100에 S-9mix의 첨가없이 실험하였다. 다섯가지 기초 식품군 중에서 Ⅰ group의 His?? revertant colony수가 가장 높았고 그 다음이 Ⅳ,Ⅱ, Ⅲ, Ⅴ group의 순이었다. 한편 각 식품군의 메타놀 추출물에 아질산을 처리한 후 돌연변이원성 colony수의 변화는 Ⅱ group의 김치가 S-9mix의 첨가없이 질산염 처리후의 His?? revertant colony수가 가장 높았다. 아질산 처리에 의한 is?? revertant colony수의 하루 섭취량은 아질산을 처리하지않은 식품의 것에 비하여 약 1.6배 높았다. Dairly intake basic five foodstuffs results in the formation of mutagens by Salmonella typhimurium TA 100 without metabolic activation, S-9 mix. Each foodstuffs were dried and then extracted with methanol using a Soxhlet apparatus. I group foodstuffs were appeared large number of His??revertant colonies, and in order of Ⅳ,Ⅱ,Ⅲ and Ⅴgroup. In the other hand, studies were also carried out to test the changes of the number of His??revertant colonies induced by nitrite treatment of each group. Ⅱgroup showed particularly highest increase of His?? revertant colonies after nitrite treatment without S-9 mix. Total daily intake of His?? revertant colonies by nitrite treatment were calculated to be approximately 1.6 times than those by nitrite untreatment.       • 직업전문학생의 여가활동 참여정도와 가치관 형성의 관계

        The purpose of this study is to re-examine the relationship between leisure-time activities and formation of value system in vocational school. For this purpose, this study has surveyed the participation of the students in leisure-time activities in vocational schools, where they take vocational training under the rubric of "First human, Second function". 1. The man has more active than the woman in terms of the frequency of participation as well as in terms of the period of participation. As for the period of participation, the university graduate is the longest. In terms of the attitude of participation, the middle school graduate is most active. 2. The group who participates in leisure-time activities less than three times a week has more positive point of view toward nation, occupation, marriage, family, livelihood and human life than the other groups. 3. The more frequent the group participates in leisure-time activities, the more positive point of view toward occupation. On the contrary, the less frequently participating group has a more positive point of view toward livelihood. 4. The group who is organizational in participation has a positive point of view toward nation and marriage. Meanwhile, the group who is not organizational has a positive point of view toward life, occupation and human life. 5. The group who takes an active attitude to participating in leisure-time activities has a positive point of view toward livelihood and human life. 6. The group who participates in the physical leisure-time activities has a positive point of view toward marriage, while the group who participates in the social leisure-time activities has a positive point of view toward family and life. 7. The frequency of participation is inversely proportional to the point of view toward family and life, while the period of participation is directly proportional to the point o view toward occupation and human life.

        고정화 시스템을 이용한 용균효소의 생산

        해양에서 분리된 용균활성이 우수한 균주의 균체고정화에 의한 연속생산을 위한 조건을 검토하였다. Bacillus subtilis SH-1의 균체를 고정화하여 column형 reactor에 의하여 연속적으로 효소생산을 실험하였다. 연속발효의 최적조건은 담체는 3.0% sodium alginate로 직경 2.4mm의 bead를 제조하여 기질의 주입속도는 20ml/h 공기의 주입속도는 2.0ml/min이였다. Column형 reactor(용량 2.8ℓ)에 의한 연속발효시 25일까지 76.5~88.0 units/ml의 최적의 효소활성을 갖는 용균효소를 생산할 수 있었다. Bacillus subtilis SH-1 screened from coastal sea water of South Korea was used to produce bacteriolytic enzyme. The production of bacteriolytic enzyme by immobilized cells was investigated. The optimum conditions for the continuous production of the bacteriolytic enzyme using immobilized cells were 2.4 mm diameter of 0.3% alginate beads, 20 ml/h of substrate feeding rate and 20 ℓ/min of aeration rate. A productivity of 76.5 to 88.0 units/ml could be obtained for 25 days by continuous column reactor under the optimum conditions.

      • 엉겅퀴로부터 분리한 Silymarin 및 Silybin이 Macrophages에 의한 사람 Low Density Lipoprotein의 산화에 대한 항산화 효과

        This study was undertaken to evaluate an antioxidative activity of silymarin and silybin obtained from Silybum marianum against oxidation of human low density lipoprotein (LDL). The electrophoretic mobility observed apparently was higher phase for LDL oxidized by macrophages compared to native LDL. Silymarin and silybin inhibited the copper-catalysed oxidation of human LDL in a dose-dependent manner. Silymarin and silybin at the concentration of 50μM/ml also inhibited the copper catalysed oxidation of LDL induced by the cell J774 and macrophages. LDL reisolated from the cell incubation in the presence of silymarin or silybin was degraded at rates similar with native LDL. Silymarin or silybin found to be potential inhibitors against oxidation of ^125I-LDL by macrophages and endothelial cells.

      • 단백질 염산 가수분해 조미액의 제조중 chloropropanols 생성 억제 공정 개발

        Protein hydrolyzates by hydrochloric acid widely used as seasonings and sensory flavours. Protein hydrolyzates contained mutagens of chloropanols such as monochloropropanol, dichloropropanols. This study have now been developed for the chloropropanols reduction using modified bydrolyzed processing. The Ist was performed to adjust the range of 0.5∼1.0 in mole rate of hydrochloric acid/protein-N at 95∼98℃ for 5 days, 2nd process hydrolyzed the ranges of 0.5∼1.0 in mole ratio HCl/protein-N and adjust with pH 8.8 for 2 day. The final 3rd process filterated and adjusted pH 5.2. The modified process maintained to reduce the chlorophenols of protein hydrolyzates prepared by hydrochloric acid.

      • 대두배아 사포닌의 유리기 생성 억제 및 세포독성

        대두 배아로부터 saponin을 분리·정제하여 유리기 억제효과 및 동물의 암세포에 대한 세포 독성을 조사하였다. 유리기 억제활성에 대한 결과를 보면 대두 배아 saponin을 0.5 및 1.0% 씩 첨가한 그여 식에서 4주 동안 사육한 후 간장을 절취하여 간장 획분의 활성산소 및 항산화 관련 효소에 대하여 평가하였다. 간장의 mitochondria 및 microsome 획분의 hydroxy radical (·OH)의 생성에 미치는 영향에서 급여식에 사포닌을 첨가한 급여군에서 ·OH기의 생성억제 효과를 나타내었다. 간장의 microsome 획분에서도 0.5 및 1.0% 투여군에서 H_2O_2 생성억제 효과가 인정되었다. 또한 간장의 cytosol획분에서 O_2· 의 생성은 대조군에 비하여 현저한 억제 효과를 나타내었다. 대두 배아 사포닌의 세포독성을 P338 (mouse lyoupoid neoplasm) 과 L1210 (mouse leukemia) 세포주를 사용하여 실시한 결과 200 ㎍/mL 농도에서 높은 성장억제효과를 나타내었다. 그리고 처리 농도에 따른 대두 배아 추출물이 P338과 L1210에 대한 성장 효과 실험에서도 200㎍/mL에서 높은 성장 억제 효과가 나타났으며 농도가 높을수록 완전히 억제되는 것을 볼 수 있었다. This study was carried out to investigate functional activities of the free radical scavenging and germ of Glycin max. Merrill for cytotoxicity toward P338 and L1210 cells derived from mouse. Effect of crude saponin were examined to oxygen radicals and their scavenger enzymes in liver fractions of Spragae-Dawley(SD) rats. Male rats were fed basic diets of control and experiment diets of 0.5 ~ 1.0 % crude saponin. there were no significant differences in hydroxy radical (·OH) formation of liver mitochondria and microsomes in 1.0% group, while ·OH formations were significantly decrease in 0.5% and 1.0% saponin compared with control group. Their oxygen radical (O_2) scavenging activities were significantly decrease in liver cytosol of 0.5% and 1.0% saponin group compared with control group. Soybean germ saponin was isolated purified by the method of HPLC to investigate the cytotoxicity of mouse cells by using the MTT assay. SA-I less than 50% cytotoxicity. These results suggest that the saponin may play a effective role in attenuating a oxygen radical formations and increasing a scavenger enzyme activities.
