        Genetic screening in young women diagnosed with endometrial cancer

        Objective: To evaluate the importance of Lynch syndrome associated risk screening in the patients aged less than 50 years affected from endometrial cancer. Methods: From 2007 to 2014, 41 patients affected from endometrial cancer and aged less than 50 years underwent surgery at the Complex Operative Unit of Gynecology and Obstetrics, Cannizzaro Hospital of Catania, Italy. They were selected to undergo mismatch repair gene mutation analysis using immunohistochemistry (IHC; four markers: MLH1, MSH2, MSH6, PMS2) and microsatellite instability (MSI) test. For samples that resulted negative to IHC (abnormal finding), MSI test was performed to further study the suspected mutation. Samples were classified as MSI-high (MSI-H) if more than one marker was identified as unstable; MSI-low (MSI-L) if only one marker was identified as unstable; or MSI-stable (MSI-S) if no marker was identified as unstable. Samples were subdivided into two groups: MSI-H/L and MSI-S. Statistical analysis was performed to assess differences regarding survival, tumor staging, grading, and invasion of lymphovascular space between these two groups. Results: IHC analysis showed that in 46% (19/41) of samples there was negative outcome. Forty-two percent (8/19) of these negative samples were unstable (either low or high). Of eight patients showing MSI, 75% were MSI-L, while 25% were MSI-H. Differences in survival, stage, grade, lymphovascular space invasion and Amsterdam criteria adherence were not statistically significant due to the small size of the cohort. Conclusion: IHC and MSI test results of our cohort lead us to assess the relevance of performing Lynch syndrome genetic screening in endometrial cancer patients aged less than 50 years at the time of diagnosis.

        ON THE EXPERIMENTAL ANALYSIS OF SINGLE INPUT SINGLE OUTPUT CONTROL OF YAW RATE AND SIDESLIP ANGLE

        Electric vehicles with individually controlled drivetrains allow torque-vectoring, which improves vehicle safety and drivability. This paper investigates a new approach to the concurrent control of yaw rate and sideslip angle. The proposed controller is a simple single input single output (SISO) yaw rate controller, in which the reference yaw rate depends on the vehicle handling requirements, and the actual sideslip angle. The sideslip contribution enhances safety, as it provides a corrective action in critical situations, e.g., in case of oversteer during extreme cornering on a low friction surface. The proposed controller is experimentally assessed on an electric vehicle demonstrator along two maneuvers on surfaces with significantly varying tire-road friction coefficient. Different longitudinal locations of the sideslip angle used as control variable are compared during the experiments. Results show that: i) the proposed SISO approach provides significant improvements with respect to the vehicle without torque-vectoring, and the controlled vehicle with a reference yaw rate solely based on the handling requirements for high-friction maneuvering; and ii) the control of the rear axle sideslip angle provides better performance than the control of the sideslip angle at the center of gravity.

        Postoperative Acupuncture is as Effective as Preoperative Acupuncture or Meloxicam in Dogs Undergoing Ovariohysterectomy: a Blind Randomized Study

        Background: Acupuncture has the same analgesic effect as non-steroidal antiinflammatory drugs and opioids. It is challenging to perform preoperative acupuncture in unmanageable animals, while the residual postoperative anesthetic effect facilitates the performance of acupuncture postoperatively. Objectives: To compare preoperative acupuncture or meloxicam versus postoperative acupuncture for postoperative analgesia after ovariohysterectomy. Methods: This is a horizontal prospective positive control blind randomized experimental study. Thirty-six dogs were randomly divided into three groups: GA (preemptive acupuncture), GPA (postoperative acupuncture), and GM (meloxicam 0.2 mg/kg IV preoperatively). After sedation with acepromazine (0.05 mg/kg IM), anesthesia was induced with propofol (5.3 ± 0.3 mg/kg) and maintained with isoflurane/O 2 . Fentanyl (2 µg/ kg, IV) was administered immediately before surgery. Bilateral acupuncture was performed at acupoints Large intestine 4, Spleen 6, and Stomach 36 for 20 minutes, before (GA) or immediately after surgery (GPA). Pain was evaluated by an observer blind to the treatment using the Glasgow scale before and for 24 hours after ovariohysterectomy. Dogs with a score ≥ 6 received rescue analgesia with morphine (0.5 mg/kg IM). Nonparametric data were analyzed by the Kruskal–Wallis test, followed by Dunn’s test and parametric data by ANOVA followed by Tukey’s test. Results: Two GA and one GPA dogs received rescue analgesia once. Two GM dogs received rescue analgesia and one of those was treated again twice. There were no differences in the number of dogs receiving rescue analgesia between groups (p = 0.80). Conclusion: Postoperative acupuncture was as effective as preoperative acupuncture or meloxicam in female dogs undergoing ovariohysterectomy.

        ON THE EXPERIMENTAL ANALYSIS OF INTEGRAL SLIDING MODES FOR YAW RATE AND SIDESLIP CONTROL OF AN ELECTRIC VEHICLE WITH MULTIPLE MOTORS

        With the advent of electric vehicles with multiple motors, the steady-state and transient cornering responses can be designed and implemented through the continuous torque control of the individual wheels, i.e., torque-vectoring or direct yaw moment control. The literature includes several papers on sliding mode control theory for torque-vectoring, but the experimental investigation is so far limited. More importantly, to the knowledge of the authors, the experimental comparison of direct yaw moment control based on sliding modes and typical controllers used for stability control in production vehicles is missing. This paper aims to reduce this gap by presenting and analyzing an integral sliding mode controller for concurrent yaw rate and sideslip control. A new driving mode, the Enhanced Sport mode, is proposed, inducing sustained high values of sideslip angle, which can be limited to a specified threshold. The system is experimentally assessed on a four-wheel-drive electric vehicle. The performance of the integral sliding mode controller is compared with that of a linear quadratic regulator during step steer tests. The results show that the integral sliding mode controller significantly enhances the tracking performance and yaw damping compared to the more conventional linear quadratic regulator based on an augmented singletrack vehicle model formulation.

        Stabilization Region of PD Controller for Unstable First Order Process with Time Delay

        Time delays affect considerably the performance of the process. Moreover the stabilizing regions of the controller parameters can be difficult to compute. This work provides the stability conditions for unstable first order plus time delay (UFOPTD) systems when a PD controller is used. Also, the parametric stabilization region is obtained by means of an analysis of the characteristic quasi-polynomial. The results presented in this work show that a derivative action at the controller minimizes the effects of the time-delay achieving the stabilization of UFOPTD systems with large time-delay, in contrast with a process considering controller without derivative action. The behavior of the control strategy is illustrated with some numerical examples.

        Prediction-observer Scheme for Linear Systems with Input-output Time-delay

        This work deals with the prediction and control problems associated with a class of linear systems with time-delay at the input-output path. A general prediction observer scheme that estimates the future value of the delayed system from the output is proposed. Later, a full-information predictor-observer is introduced and the convergence of the estimated future values is formally proven for a time-delay τ of any size by increasing the dimension of the proposed predictor-observer. The estimated future state is used to design a feedback law that compensates the delay effects on the original system. The performance of the prediction-observation control strategy is shown by means of numerical simulations. Thus, it is illustrated that the performance of the proposed full-information predictor-observer is improved with respect to the sequential sub-predictors previously presented in the literature.

        Routing Protocols for VANETs: An Approach based on Genetic Algorithms

        Vehicular Ad Hoc Networks (VANETs) are self-configuring networks where the nodes are vehicles equipped with wireless communication technologies. In such networks, limitation of signal coverage and fast topology changes impose difficulties to the proper functioning of the routing protocols. Traditional Mobile Ad Hoc Networks (MANET) routing protocols lose their performance, when communicating between vehicles, compromising information exchange. Obviously, most applications critically rely on routing protocols. Thus, in this work, we propose a methodology for investigating the performance of well-established protocols for MANETs in the VANET arena and, at the same time, we introduce a routing protocol, called Genetic Network Protocol (G-NET). It is based in part on Dynamic Source Routing Protocol (DSR) and on the use of Genetic Algorithms (GAs) for maintenance and route optimization. As G-NET update routes periodically, this work investigates its performance compared to DSR and Ad Hoc on demand Distance Vector (AODV). For more realistic simulation of vehicle movement in urban environments, an analysis was performed by using the VanetMobiSim mobility generator and the Network Simulator (NS-3). Experiments were conducted with different number of vehicles and the results show that, despite the increased routing overhead with respect to DSR, G-NET is better than AODV and provides comparable data delivery rate to the other protocols in the analyzed scenarios.

        Altered platelet proteome in lupus anticoagulant (LA)-positive patients—protein disulfide isomerase and NETosis as new players in LA-related thrombosis

        Patients with antiphospholipid syndrome (APS) are at high risk of developing venous and arterial thromboembolism (TE). The role of platelets in the pathogenesis of these prothrombotic conditions is not yet fully understood. The aim of this study was to gain mechanistic insights into the role of platelets in APS by comparing the platelet proteome between lupus anticoagulant (LA)-positive patients with (LA+TE+) and without a history of TE (LA+TE−) and healthy controls. The platelet proteome of 47 patients with LA, 31 with a history of TE and 16 without thrombotic history, and 47 healthy controls was analyzed by two-dimensional differential in-gel electrophoresis and mass spectrometry to identify disease-related proteins. Afterward, selected LA-related platelet proteins were validated by western blot and ELISA. Alterations of 25 proteins were observed between the study groups. STRING pathway analysis showed that LArelated protein profiles were involved in platelet activation, aggregation, and degranulation. For example, protein disulfide isomerase family members, enzymes that promote thrombosis, were upregulated in platelets and plasma of LA+TE+patients. Leukocyte elastase inhibitor (SERPINB1), an antagonist of neutrophil extracellular trap (NET) formation, was decreased in platelets of LA+TE+patients compared to healthy controls. Additionally, citrullinated histone H3, a NET-specific marker, was increased in plasma of LA+TE+patients. These findings suggest that decreased platelet SERPINB1 levels favor prothrombotic NETosis, especially in LA+TE+patients. Our findings reveal protein abundance changes connected to altered platelet function in LA-positive patients, thus suggesting a pathogenic role of platelets in thrombotic complications in APS.
