        Selection and evaluation of reference genes for gene expression using quantitative real‐time PCR in Mythimna separata walker (Lepidoptera: Noctuidae)

        In order to precisely assess gene expression levels, the suitable internal reference genes must be served to quantify real‐time reverse transcription polymerase chain reaction (RT‐qPCR) data. For armyworm, Mythimna separata, which reference genes are suitable for assessing the level of transcriptional expression of target genes have yet to be explored. In this study, eight common reference genes, including β‐actin (β‐ACT), 18 s ribosomal (18S), 28S ribosomal (28S), glyceraldehyde‐3‐phosphate (GAPDH), elongation fator‐alpha (EF1α), TATA box binding protein (TBP), ribosomal protein L7 (RPL7), and alpha‐tubulin (α‐TUB) that in different developmental stages, tissues and insecticide treatments of M. separata were evaluated. To further explore whether these genes were suitable to serve as endogenous controls, three software‐based approaches (geNorm, BestKeeper, and NormFinder), the delta Ct method, and one web‐based comprehensive tool (RefFinder) were employed to analyze and rank the tested genes. The optimal number of reference genes was determined using the geNorm program, and the suitability of particular reference genes was empirically validated according to normalized HSP70, and MsepCYP321A10 gene expression data. We found that the most suitable reference genes for the different experimental conditions. For developmental stages, 28S/RPL7 were the optimal reference genes, both RPL7/EF1α were suitable for experiments of different tissues, whereas for insecticide treatments, 28S/α‐TUB were suitable for normalizations of expression data. In addition, 28S/α‐TUB were the suitable reference genes because they have the most stable expression among different developmental stages, tissues and insecticide treatments. Our work is the first report on reference gene selection in M. separata, and might serve as a precedent for future gene expression studies.

        The Effect of Chinese Martial Arts SANDA on the Physical Self-Reliance and Life Satisfaction of University Students

        본 연구의 목적은 중국 무술 싼다가 지역 대학생들의 신체자존 및 삶의 만족도에 미치는 영향을 규명하기 위해 수행되었다. 연구대상은 산동체육대학(山東體育學院) 2학년 3개 반 비체육전공 여대생반 26명, 체육전공 여대생반 22 명, 체육전공 남대생반 24명, 총 72명의 대학생을 대상으로 비교 실험을 진행하였다. 실험 전, 중, 후 세 번 조사하고, 그 중 실험 대상자는 216명의 학생으로 구성되어 있고, 전체 연구는 중등 단련 강도로 16주간의 무술 싼다 연습을 진행하 였다. 이상과 같은 연구방법을 통해 도출된 연구결과는 다음과 같다. 무술 싼다 단련이 대학생들의 심리건강증진에 도움이 된다는 것을 밝혔다. 조사데이터를 SPSS 통계 분석을 통해 본 결과, 실험 시간에 따라 대학생들의 신체자존감과 생활만족 도에 미친 영향이 다르다는 결과를 보였다. 16주 훈련을 한 여대생의 단련 효과가 8주 단련효과 보다 더 낮았으며, 남학생 의 경우신체자존감 면에서 8주의 단련 효과가 16주보다 뛰어났다. 본 연구를 통하여 무술 싼다 운동이 심리건강증진에 대한 기초자료로 활용될 것으로 사료된다. The purpose of this study was carried out to establish the effect of Chinese martial arts SANDA on the physical self-reliance and life satisfaction of university students. The subjects of the study were 26 non-athletic female college students in three classes of the second year of the Shandong Sports University, 22 female college students majoring in physical education, 24 male college students majoring in physical education, and 72 university students in total. Three surveys before, after, and after the experiment consisted of 216 students, and the whole study conducted 16 weeks of martial arts SANDA practice with secondary training intensity. The results of the studies produced by the above methods are as follows. It revealed that training in martial arts, "SANDA," helps college students improve their psychological health. According to the analysis of SPSS statistics, the impact of the survey data on self-esteem and satisfaction of college students varies depending on the time of the experiment. The 16-week training for female college students was lower than the eight-week training, and the eight-week training for male students was better than the 16th week in terms of physical self-esteem. Through this research, it is believed that the martial art "SANDA Movement" will be used as basic data for the promotion of psychological health.

        PS-CONNECTEDNESS OF L-SUBSETS

Fan's Theorem holds for PS-connectedness in L-topological spaces.

        Cough Inhibition Activity of Schisandra chinensis in Guinea Pigs

        Chronic cough is very common in respiratory clinics, and no effective drugs are available. Schisandra chinensis (Turcz.) Baill. (S. chinensis), an important traditional Chinese medicine, has been extensively prescribed for patients with a persistent cough. Preliminary research indicated that 95% ethanol extracts (EE) of S. chinensis showed remarkable antitussive activity in guinea pigs exposed to cigarette smoke (CS). To find out the antitussive ingredients of S. chinensis, EE was divided into four fractions according to the polarity: petroleum ether extract (PEE), ethyl acetate extract (ECE), n-butyl alcohol extract, and residue extract. The antitussive, antioxidant, and anti-inflammatory effects of the four fractions were evaluated in a guinea pig model of cough hypersensitivity induced by CS exposure. Eighteen main constituents of the two effective fractions, PEE and ECE, were identified using ultra-high-pressure liquid chromatography electronic spray ion time-of-flight mass spectrometry. The cough inhibition activities of compound 1, 3, 9, 10, 17 were evaluated on citric acid induced acute cough guinea pigs. The results showed that the antitussive activity of EE was almost all contained in PEE and ECE. The 16 major peaks in PEE were identified as 15 lignans (1–12 and 14–16) and 1 triterpene (compound 13), and 3 major peaks (1, 17, and 18) in ECE were also identified as lignans. Three doses of five compounds brought about a significant decrease in number of cough efforts (P < .01), and the cough inhibition rates were between 40.9% and 85.1%. Therefore, lignans are the antitussive ingredients of S. chinensis.

      • 触变与持守:越南瑶族的黑齿习俗

        Dyeing teeth is a kind of ancient body decoration culture, which has historically been widespread in the Asia Pacific region. There used to be no custom of dyeing teeth in the Yao, but some branches ,like the Bai Ku Yao(He Yao) of Bao Teng County in Vietnam, followed the custom by contact with more black-tooth Yue or Pu people. Influenced by modern aesthetic ideas, the Vietnamese society no longer holds "Black Is Beauty", the Yao’s dyeing teeth custom has also changed. However, it becomes the last holder of the custom in reality thanks to relatively closed lifestyle with relatively conservative ethnic cultures. In essence, the acquiring, holding, or abandoning the custom of dyeing teeth in the Yao of Vietnam are local cultural strategies to meet their life needs by the environmental and social changes.

      • Predictive Impact of Common Variations in DNA Repair Genes on Clinical Outcome of Osteosarcoma

        We aimed to assess the role of XPG, XPC and MMS19L polymorphisms on response to chemotherapy in osteosarcomas, and the clinical outcomes. One hundred and eighty five osteosarcoma patients who were histologically confirmed were enrolled in our study between January 2007 and December 2009. Genotyping of XPG, XPC and MMS19L was performed in a 384-well plate format on the MassARRAY$^{(R)}$ platform. Individuals with XPG TT genotype and T allele were more likely to be better response to chemotherapy than CC genotype, with the OR (95% CI) of 4.17 (1.64-11.54) and 2.66 (1.39-5.11), respectively. Those carrying MMS19L TT genotype and T allele showed better response to chemotherapy, with ORs (95% CI) of 4.8 (1.56-17.7) and 2.3 (1.22-4.36), respectively. Patients carrying TT genotype of XPG and MMS19L showed a significantly longer overall survival than CC genotype, with a 0.47 and 0.30-fold risk of death when compared with the wild-type of the gene. XPG and MMS19L are correlated with response to chemotherapy and prognosis of osteosarcoma, so that they could be used as predictive markers for prognosis.

      • Identification of a Novel Human Zinc Finger Gene, ZNF438, with Transcription Inhibition Activity

        There were many different families of zinc finger proteins that contained multiple cysteine and/or histidine residues and used zinc to stabilize their folds. The classical C2H2 zinc finger proteins were the founding members of this superfamily and were among the most abundant proteins in eukaryotic genomes. C2H2 proteins typically contained several C2H2 fingers that made tandem contacts along the DNA. Here we reported a novel C2H2 type zinc finger gene, ZNF438, which encoded 828 amino acids that formed five zinc finger domains. Bioinformatics analysis revealed that the ZNF438 was mapped to human chromosome 10p11.2 and shared 62% identity with rat and mouse homologues. RT-PCR analysis indicated that it was ubiquitously expressed in 18 human adult tissues. With immunofluorescence assay, it was shown that the exogenous Flag-tagged ZNF438 was located in nucleus of COS-7 cells. To further explore the function of ZNF438, we examined the transcriptional activity of ZNF438 protein by transfecting recombinant pM-ZNF438 into mammalian cells. The subsequent analysis based on the duel luciferase assay system showed that ZNF438 was a transcriptional repressor.

        Wing weight estimation considering constraints of structural strength and stiffness in aircraft conceptual design

        According to the requirement of wing weight estimation and frequent adjustments during aircraft conceptual design, a wing weight estimation method considering the constraints of structural strength and stiffness is proposed to help designers make wing weight estimations rapidly and accurately. This method implements weight predictions on the basis of structure weight optimization with stiffness constraints and strength constraints, which include achievement of wing shape parametric modeling, rapid structure layout, finite element (FE) model automated generation, load calculation, structure analysis, weight optimization, and weight computed based on modeling. A software tool is developed with this wing weight estimation method. This software can realize the whole process of wing weight estimation with the method and the workload of wing weight estimation is reduced because much of the work can be completed by the software. Finally, an example is given to illustrate that this weight estimation method is effective.
