Degranulation in human mast cells contributes to provocation of allergic inflammation and innate immunity to parasites. Trichomonas vaginalis is transmitted by sexual intercourse and causes the acute and chronic allergic inflammation. Although a recent report has shown that mast cells are degranulated in response to T. vaginalis-derived secretory products (TvSP), detailed signaling mechanisms of TvSP- induced mast cell degranulation are not fully understood. We report that N-glycosylation of NOX2 play an important role in surface trafficking of NOX2, which can regulate ROS-dependent degranulation in TvSP- stimulated human mast cells. Stimulation with TvSPinduced NOX2 activation such as p47phox phosphorylation and NOX2 glycosylation, production of intracellular ROS, and release of granular proteins such as histamine and -hexosaminidase via BLT1. Inhibition of ROS generation with NOX2 inhibitors also reduced TvSP-triggered degranulation. Moreover, TvSP promoted trafficking of intracellular NOX2 to the cell surface in a BLT1-dependent manner. Such BLT1-mediated surface migration of NOX2 was dependent upon N-glycosyation status of NOX2. Inhibiting N-glycosylation of NOX2 by N-glycosylation inhibitor tunicamycin reduced surface trafficking of NOX2, ROS generation, and degranulation induced by TvSP. PKC inhibitor prevented TvSP-triggered glycosylation of NOX2, its surface trafficking, ROS generation, degranulation. Finally, disruption of lipid rafts with M □CD prevented glycosylation-dependent surface trafficking of NOX2, ROS generation, and degranulation. These results suggest that PKC-mediated N- glycosylation regulates its trafficking of NOX2 to the lipid rafts, which is required for ROS-dependent degranulation in mast cells induced by T. vaginalis-derived secretory products.
Transcriptional repression of ANGPT1 by histone H3K9 demethylase KDM3B

Here we report that the H3K9 demethylase KDM3B represses transcription of the angiogenesis regulatory gene, ANGPT1. Negative regulation of ANGPT1 by KDM3B is independent of its Jumonji (JmjC) domain-mediated H3K9 demethylase activity. We demonstrate that KDM3B downregulates ANGPT1 via interaction with SMRT, and suggest that the repressor complex is formed at the promoter area of ANGPT1. Using MTT and wound healing assays, depletion of KDM3B was found to increase cell proliferation and cell motility, indicating that KDM3B has a role in angiogenesis. [BMB Reports 2015; 48(7): 401-406]
Computed Tomographic and Magnetic Resonance Imaging Features of Oral Melanoma in a Dog

Oral melanoma is the most common type of oral tumor in dogs. In this report, computed tomography (CT) and magnetic resonance imaging (MRI) were performed to diagnose a right oral pigmented mass in an 8-year-old dog. The oral mass appeared as a homogeneous soft tissue density parenchyma on pre-contrast CT images, and with heterogeneous enhancement on post-contrast images. Bone destruction of the right mandibular body around the mass and mild enlargement of the right mandibular lymph node were also found. On MRI, the bulky oral mass showed mixed hyperintensity and isointensity compared to the adjacent muscle, where irregular hyperintensity on T1-weighted images corresponded to hypointensity on the T2-weighted images. Based on the physical examinations and imaging results, melanoma was suspected and confirmed via fine-needle aspiration. These unique MRI signals were due to the high paramagnetic melanin content in the tumor, therefore MRI examination could be useful for diagnosis of melanoma.
The burden of acute myocardial infarction after a regional cardiovascular center project in Korea

<P><B>Objective</B></P><P>The aim of this study was to examine the impact of a government-directed regional cardiovascular center (RCVC) project on the length of stay (LOS) and medical costs due to acute myocardial infarction (AMI).</P><P><B>Design</B></P><P>A retrospective claim data review.</P><P><B>Setting</B></P><P>Forty hospitals including four RCVCs in Korea.</P><P><B>Participants</B></P><P>A total of 1469 AMI patients who visited a RCVC in two regions between February 2009 and December 2011.</P><P><B>Intervention(s)</B></P><P>RCVC project has been fostering specialized center by region for management of cardiovascular disease. It has built a system that could receive intensive care quickly within 3 h when disease occurred.</P><P><B>Main Outcome Measure(s)</B></P><P>Changes in the LOS and cost were evaluated using the difference-in-differences (DIDs) method combined with propensity score matching (1:1) and multilevel analysis with adjustment for patient's and institutional factors.</P><P><B>Results</B></P><P>The net effect of RCVC project implementation showed decline of LOS (−0.71 days) and total medical costs (−797 US dollars) by DID. After the RCVC project, the LOS for patients with AMI hospitalized in a RCVC was decreased by 8.9% (<I>β</I> = −0.094, <I>P</I> = 0.041) compared with patients hospitalized in a hospital not designed as a RCVC. Compared with costs before the RCVC project, they were decreased by 11.5% (<I>β</I> = −0.122, <I>P</I> = 0.004).</P><P><B>Conclusions</B></P><P>We provided evidence regarding the change in the societal burden due to AMI after regionalization. Although there was a reduction of LOS and direct medical costs reported in limited number of regionalized hospitals, in the long term we can anticipate an expanding impact in all regionalized hospitals.</P>
