        Principle of Apertureless Scanning Near-Field Optical Microscopy: On the Way to the Optical Metrology of Nanostructures

        Apertureless Scanning Near-Field Optical Microscopy (A-SNOM) is a powerful tool for the observation and engineering of nanostructures in near-field optics (NFO). Nevertheless, the interpretation of the experimental recorded signals raises several difficulties due to the strong dependance of the image formation process on the shape and the optical index of the nanostructure, the properties of the probe and also on the illumination and detection conditions of the experimental configuration used. In order to perform an optical measurement, an accurate model and an inversion procedure are necessary. This last approach may offer a wide variety of applications to the SNOM (e.g. the metrology of nanostructures, the optical, thermal properties, or of optical forces...) In this contribution, we focus our attention on an A-SNOM model for metrological applications in near-field optics.

        Pooled Analysis of the Cow’s Milk-related-Symptom-Score (CoMiSSTM) as a Predictor for Cow’s Milk Related Symptoms

        Purpose: The diagnosis of cow’s milk (CM) allergy is a challenge. The Cow’s Milk-related-Symptom-Score (CoMiSSTM) was developed to offer primary health care providers a reliable diagnostic tool for CM related symptoms. The predictive prospective value of the CoMiSSTM was evaluated in three clinical trials. Methods: Pooled analyses of the three studies were conducted based on regressing the results of the month-1 chal-lenge test on the month-1 CoMiSSTM, adjusting for baseline CoMiSSTM using a logistic regression model. In addition a logistic regression model was also fitted to the month-1 challenge test result with the change in CoMiSSTM from baseline as a predictor. Results: Results suggest that infants having a low CoMiSSTM (median, 5) after 1 month dietary treatment free from intact CM protein have a significant risk of having a positive challenge test (odds ratio, 0.83; 95% confidence interval, 0.75-0.93; p=0.002). Pooled data suggest that the change in CoMiSSTM from baseline to month-1 can predict CM related symptoms as a confirmed diagnosis according to the challenge test at month-1. However, in order to validate such a tool, infants without CM related symptoms would also need to be enrolled in a validation trial. A concern is that it may not be ethical to expose healthy infants to a therapeutic formula and a challenge test.Conclusion: Pooled data analysis emphasizes that the CoMiSSTM has the potential to be of interest in infants sus-pected to have CM-related-symptoms. A prospective validation trial is needed.

      • “In the Shoes of an Algorithm”: A Media Education Game to Address Issues Related to Recommendation Algorithms

        Recommendation algorithms play an important role in the current media landscape by fostering interactions between users of digital media platforms and media content. The growing concern with recommendation algorithms raises important questions for media educators who emphasize critical analysis of media content and creative production and are required to develop media education initiatives focused on a critical understanding of digital platforms and their underlying technical infrastructures. What are teenagers’ current perception and experiences of recommendation algorithms? What should be the key issues these initiatives should address, and how should they be addressed? The In the Shoes of an Algorithm project approached these questions through design-based research (DBR). This project led to the creation of a series of pen-and-paper games designed to (1) help teenagers share their daily experiences of recommendation algorithms to identify core issues that should be addressed by media educators and (2) to foster a better understanding of key issues related to recommendation algorithms used by digital media platforms. The project was initiated in Belgium and expanded as an international program, in collaboration with South-Korean researchers and practitioners.

        Bioconjugated lanthanide luminescent helicates as multilabels for lab-on-a-chip detection of cancer biomarkers

        <P>The lanthanide binuclear helicate [Eu<SUB>2</SUB>(L<SUP>C2(CO<SUB>2</SUB>H)</SUP>)<SUB>3</SUB>] is coupled to avidin to yield a luminescent bioconjugate <B>EuB1</B> (<I>Q</I> = 9.3%, <I>τ</I>(<SUP>5</SUP>D<SUB>0</SUB>) = 2.17 ms). MALDI/TOF mass spectrometry confirms the covalent binding of the Eu chelate and UV-visible spectroscopy allows one to determine a luminophore/protein ratio equal to 3.2. Bio-affinity assays involving the recognition of a mucin-like protein expressed on human breast cancer MCF-7 cells by a biotinylated monoclonal antibody 5D10 to which <B>EuB1</B> is attached <I>via</I> avidin-biotin coupling demonstrate that (i) avidin activity is little affected by the coupling reaction and (ii) detection limits obtained by time-resolved (TR) luminescence with <B>EuB1</B> and a commercial Eu-avidin conjugate are one order of magnitude lower than those of an organic conjugate (FITC-streptavidin). In the second part of the paper, conditions for growing MCF-7 cells in 100–200 µm wide microchannels engraved in PDMS are established; we demonstrate that <B>EuB1</B> can be applied as effectively on this lab-on-a-chip device for the detection of tumour-associated antigens as on MCF-7 cells grown in normal culture vials. In order to exploit the versatility of the ligand used for self-assembling [Ln<SUB>2</SUB>(L<SUP>C2(CO<SUB>2</SUB>H)</SUP>)<SUB>3</SUB>] helicates, which sensitizes the luminescence of both Eu<SUP><SMALL>III</SMALL></SUP> and Tb<SUP><SMALL>III</SMALL></SUP> ions, a dual on-chip assay is proposed in which estrogen receptors (ERs) and human epidermal growth factor receptors (Her2/<I>neu</I>) can be simultaneously detected on human breast cancer tissue sections. The Ln helicates are coupled to two secondary antibodies: ERs are visualized by red-emitting <B>EuB4</B> using goat anti-mouse IgG and Her2/<I>neu</I> receptors by green-emitting <B>TbB5</B> using goat anti-rabbit IgG.         Trade in a small-range songbird, the Javan crocias, gives insight into the Asian Songbird Crisis

        The Asian Songbird Crisisdthe recognition that the cage bird trade is the number one threat to manysongbirdsdhas focussed on either individual bird markets or specific taxa. We here show, throughsustained monitoring, that even species that were not thought to be traded are also at risk from overexploitation. The Javan crocias Laniellus albonotatus is endemic to the montane forests of western Javaethis remoteness was thought to be the reason why it had escaped from being at risk. We surveyed21 bird markets over a 44-month period; the number of crocias recorded in markets (1.29 birds/survey)was negatively related to the distance to its habitat. The size of the bird market had no effect on thenumber of crocias we recorded. Prices (US$26.70/bird), when corrected for inflation, did not change over12 years suggesting supply can keep up with demand. Turnover is high (50% sold after 8 days), and weestimate the total trade at 1,200 to 1,500 birds/year. Legal protection of Javan crocias seen in isolationthus far is proven to be ineffective. A more inclusive approach where government bodies, conservationagencies and society participate, is needed to curb the trade in this and other imperiled songbirds.

        Computed tomographic assessment of retrograde urohydropropulsion in male dogs and prediction of stone composition using hounsfield unit in dogs and cats

        Background: Persistent uroliths after a cystotomy in dogs are a common cause of surgical failure. Objectives: This study examined the following: the success rate of retrograde urohydropropulsion in male dogs using non-enhanced computed tomography (CT), whether the CT mean beam attenuation values in Hounsfield Units (mHU) measured in vivo could predict the urolithiasis composition and whether the selected reconstruction kernel may influence the measured mHU. Methods: All dogs and cats that presented with lower urinary tract uroliths and had a non-enhanced CT preceding surgery were included. In male dogs, CT was performed after retrograde urohydropropulsion to detect the remaining urethral calculi. The percentage and location of persistent calculi were recorded. The images were reconstructed using three kernels, from smooth to ultrasharp, and the calculi mHU were measured. Results: Sixty-five patients were included in the study. The success rate of retrograde urohydropropulsion in the 45 male dogs was 55.6% and 86.7% at the first and second attempts, respectively. The predominant components of the calculi were cystine (20), struvite (15), calcium oxalate (8), and urate (7). The convolution kernel influenced the mHU values (p < 0.05). The difference in mHU regarding the calculus composition was better assessed using the smoother kernel. A mHU greater than 1,000 HU was predictive of calcium oxalate calculi. Conclusions: Non-enhanced CT is useful for controlling the success of retrograde urohydropropulsion. The mHU could allow a prediction of the calculus composition, particularly for calcium oxalate, which may help determine the therapeutic strategy.

        Short-term exposure to gold nanoparticle suspension impairs swimming behavior in a widespread calanoid copepod

        <P><B>Abstract</B></P> <P>Calanoid copepods play an important role in the functioning of marine and brackish ecosystems. Information is scarce on the behavioral toxicity of engineered nanoparticles to these abundant planktonic organisms. We assessed the effects of short-term exposure to nonfunctionalized gold nanoparticles on the swimming behavior of the widespread estuarine copepod <I>Eurytemora affinis</I>. By means of three-dimensional particle tracking velocimetry, we reconstructed the trajectories of males, ovigerous and non-ovigerous females. We quantified changes in their swimming activity and in the kinematics and geometrical properties of their motion, three important descriptors of the motility patterns of zooplankters. In females, exposure to gold nanoparticles in suspension (11.4 <I>μ</I>g L<SUP>−1</SUP>) for 30 min caused depressed activity and lower velocity and acceleration, whereas the same exposure caused minimal effects in males. This response differs clearly from the hyperactive behavior that is commonly observed in zooplankters exposed to pollutants, and from the generally lower sensitivity of female copepods to toxicants. Accumulation of gold nanoparticles on the external appendages was not observed, precluding mechanical effects. Only very few nanoparticles appeared sporadically in the inner part of the gut in some samples, either as aggregates or as isolated nanoparticles, which does not suggest systemic toxicity resulting from pronounced ingestion. Hence, the precise mechanisms underlying the behavioral toxicity observed here remain to be elucidated. These results demonstrate that gold nanoparticles can induce marked behavioral alterations at very low concentration and short exposure duration. They illustrate the applicability of swimming behavior as a suitable and sensitive endpoint for investigating the toxicity of nanomaterials present in estuarine and marine environments.         Perioperative morbidity of radical trachelectomy with lymphadenectomy in early-stage cervical cancer: a French prospective multicentric cohort

        Objective: The aim of this study was to determine the predictive factors of postoperative morbidity of patients who have undergone a radical trachelectomy (RT) for early-stage cervical cancer and to assess the oncologic outcomes. Methods: We retrospectively analysed the data of 2 prospective trials on sentinel node biopsy for cervical cancer (SENTICOL I and II). Patients having a RT for early-stage cervical cancer with negative sentinel lymph node and safe margins, were included. Results: Forty-nine patients met the inclusion criteria. Forty-five patients had a laparoscopicassisted vaginal technique and 4 patients a total laparoscopic technique. The median age was 32 years (range, 22–46 years). 83.7% of patients had a stage IB1 disease. There were 63.3% squamous cell carcinomas and 34.7% adenocarcinomas. The median follow-up was 46 months (range, 1–85 months). Two patients (3.3%) had a severe postoperative complication (Clavien-Dindo ≥III and/or CTCAE ≥3). The main postoperative complications were urinary (28.6%), lymphovascular (26.5%) and neurologic (14.3%). On a multivariate analysis, postoperative complications were significantly associated with history of pelvic surgery and IB1 International Federation of Gynecology and Obstetrics stage. Inclusion in high surgical skills centers decrease the risk of postoperative complications. During the follow-up, 3 patients (6.1%) had a local recurrence and one patient died from a breast cancer. Between group with complications and group without any complications, overall survival and recurrence-free survival did not significantly differ at 5-year of follow-up. Conclusion: RT has few severe postoperative complications and appears as a safe alternative to spare fertility of young patients. To guarantee best surgical outcomes, patients should be referred to expert center.
