        Suppression of cyclooxygenase-2 expression induced by Toll-like receptor 2 or 4 agonists by (E)-isopropyl 4-oxo-4- (2-oxopyrrolidin-1-yl)-2-butenoate

        Inflammation can be initiated by invading microbial pathogens. Toll-like receptors (TLRs) recognize molecular structures derived from microbial pathogens and regulate the activation of innate immunity. TLR signaling pathways trigger the activation of nuclear factor-κB(NF-κB) transcription factor, leading to the induction of inflammatory gene products such as cyclooxygenase-2. Here, we present biochemical evidence that the fumaryl pyrrolidinone, (E)-isopropyl 4-oxo-4-(2-oxopyrrolidin-1-yl)-2-butenoate (IPOP),previously synthesized in our laboratory inhibits NF-κB activation induced by TLR agonists and overexpression of two downstream signaling components of TLRs. IPOP also inhibits TLR agonist-induced expression of cyclooxygenase-2. Our results suggest that IPOP can modulate TLR-mediated inflammatory responses and, potentially, the risk of many chronic inflammatory diseases.

        Parthenolide Suppresses the Expression of Cyclooxygenase-2 and Inducible Nitric Oxide Synthase Induced by Toll-Like Receptor 2 and 4 Agonists

        Toll-like receptors (TLRs), which are pattern recognition receptors (PRRs), recognize pathogen-associated molecular patterns (PAMPs) and regulate the activation of innate immunity. All TLR signaling pathways culminate in the activation of NF-κB, leading to the induction of inflammatory gene products such as cyclooxygenase-2 (COX-2) and inducible nitric oxide synthase (iNOS). Parthenolide, a sesquiterpene lactone isolated from the herb feverfew (Tanacetum parthenium), has been used as folk remedies to treat many chronic diseases for many years. In the present report, we present biochemical evidence that parthenolide inhibits the NF-κB activation induced by TLR agonists and the overexpression of downstream signaling components of TLRs, MyD88, IKKβ, and p65. Parthenolide also inhibits TLR agonists-induced COX-2 and iNOS expression. These results suggest that parthenolide can modulate the immune responses regulated by TLR signaling pathways.

        음식 알러지는 만성적인 장애를 일으킬 수 있는 즉각적이며 잠재적으로 생명을 위협하는 반응이다. 이 중 계란은 알러지 위험성이 높은 물질로, 아이들에게 음식 알러지를 유발하는 흔한 물질로 알려져 있다. 계란 알러젠 중에서 계란 흰자 단백질인 OVA 이 주요한 알러젠으로 알려져 있다. 우리는 이번 연구에서 OVA 이 염증 및 면역 반응에 중요한 역할을 한다고 알려져 있는 NF-κB 활성화와 COX-2와 iNOS 발현에 어떠한 영향을 미치는지를 알아보았다. OVA은 NF-κB 활성화와 COX-2와 iNOS의 발현을 증가시켰다. 이러한 연구는 앞으로 계란 알러젠에 의한 알러지 작용기전 규명 및 알러지 치료제 개발에 중요한 역할을 할 것으로 기대한다. Egg allergies are the most prevalent food hypersensitivity in children. The major egg allergens are proteins, such as ovalbumin (OVA) and ovomucoid (OVM), which are mainly contained in egg whites. OVA is the major protein in egg white, comprising 54% of the total protein content. OVA has been widely used in experimental inhalant and dietary allergy animal models, but its mechanism has not been clearly identified. In this study, we showed that OVA induced nuclear factor-κB activation. OVA also induced the expression of cyclooxygenase-2 and inducible nitric oxide synthase. These data suggest new approaches for developing efficient anti-allergic strategies.

        간호사의 공감만족에 대한 국내 연구 분석

        본 연구는 간호사의공감만족에 대한 국내 연구의동향을 분석하여 향후 연구에대한 방향을 제시하고자 시도되었다. 연구조사 데이터베이스는 학술정보서비스, 국회도서관, KISS, NDSL을 이용하였다. 주요어는 간호사, 공감만족, 전문직 삶의 질 로 검색하여 최종 18편의 논문을 분석 하였다. 연구 설계는 서술적 조사연구 위주였고, 연구 대상은 병원에 근무하는 간호사로 나타났다. 공감만족과 관련이 있는 일반적 특성으로는 연령, 직위, 경력, 교육상태 등이었으며, 관련변수로는 소진, 공감피로, 직무만족, 극복력 등으로 나타났다. 공감만족과 관련이 있는 변수를 살펴본 결과 나이가 어리고 경력이 짧은 미혼인 간호사의 공감만족이 상대적으로 낮은 것으로 나타나 조직적 차원에서 지지 프로그램이 요구된다. 간호사의 공감만족은 임상실무에서 간호의 질 향상과 간호인력 관리에 기여 할 것이다. 따라서, 간호사가 간호를 하면서 높은 공감만족을 가질 수 있도록 간호 관리자의 관심과 노력이 매우 필요하다. 본 연구의 결과를 통해 국내 간호사에게 적합한 도구개발 연구, 간호사의 공감만족에 대한 본질적인 실체를 밝히는 질적 연구와 프로그램을 개발하여 효과를 검증하는 중재 연구를 제언한다. This study aimed to review and summarize the trends in research pertaining to the compassion satisfaction of nurses in Korea and to make suggestions for future research. For this study, RISS, KISS, NDSL, and National assembly library databases were searched using the key words "nurse" and "compassion satisfaction" or "professional quality of life" and "nurse". Eighteen papers on compassion satisfaction of nurses were reviewed. This was a non-experimental study (100%), and included hospital nurses (100%). The general characteristics of interest pertaining to compassion satisfaction were age, position, career, educational status, and etc. The variables with respect to compassion satisfaction were burn out, compassion fatigue, job satisfaction, resilience, and etc. The findings of this study provide preliminary data on research trends in the compassion satisfaction of nurses. Nursing managers need to maximize strategies to enhance compassion satisfaction of nurses. A qualitative study and a development of a program focused on compassion satisfaction are recommended for future research.

        TBK1-Targeted Suppression of TRIF-Dependent Signaling Pathway of Toll-like Receptor 3 by Auranofin

        Toll-like receptors (TLRs) play an important role in induction of innate immune responses. The stimulation of TLRs by microbial components triggers two branches of downstream signaling pathways: myeloid differential factor 88 (MyD88)- and toll-interleukin-1 receptor domain-containing adapter inducing interferon-β (TRIF)-dependent signaling pathways. Auranofin, a sulfur-containing gold compound (Au[I]), has been widely used for the treatment of rheumatoid arthritis. Since dysregulation of TLRs can lead to severe systemic inflammatory and joint destructive process in rheumatoid arthritis, auranofin-mediated modulation of TLR activation may have therapeutic potential against such diseases. Previously, we demonstrated that auranofin suppressed TLR4 signaling pathway by inhibiting TLR4 dimerization induced by LPS. Here, we examined the effect of auranofin on signal transduction via the TRIFdependent pathway induced by a TLR3 agonist. Auranofin inhibited nuclear factor-κB and interferon (IFN) regulatory factor 3 (IRF3) activation induced by polyinosinic-polycytidylic acid (poly[I:C]). Auranofin inhibited poly[I:C]-induced phosphorylation of IRF3 as well as IFNinducible genes such as IFN inducible protein-10. Furthermore, auranofin inhibited TBK1 kinase activity in vitro. All the results suggest that auranofin suppress TLR signaling at multiple steps.

        간호사의 간호업무성과 예측모형 - 긍정심리자본을 중심으로

        Purpose: The purpose of this study was to construct and test a structural equation model on nursing work outcomes based on Youssef and Luthans’ positive psychological capital and integrated conceptual framework of work performance. Methods: This study used a structured questionnaire administered to 340 nurses. Data were analyzed using structural equation modeling. Results: Positive psychological capital showed indirect and direct effects on job satisfaction, retention intention, organizational citizenship behavior, and nursing performance. While, the nursing work environment had direct and indirect effects on job satisfaction and nursing performance, it only had indirect effects on intention to work and organizational citizenship behavior. Additionally, a mediating effect on retention intention and organizational citizenship behavior was found between job satisfaction and nursing performance variables. Conclusion: The nursing organization needs to build a supportive work environment and reinforce positive psychological capital to improve nursing performance. Additionally, it needs to actively manage the necessary parameters involved in the stages of job satisfaction, retention intention, nursing performance, and organizational citizenship behavior of nurses. The findings propose the continuous management of nursing personnel based on nurses’ attitude outcome, behavioral intention, behavioral outcome, and stage of role performance.

        간호 대학생의 긍정적 정서, 셀프리더십 및 임상실습 만족도간의 관계

        Purpose: This study aimed to identify the relationship between positive affectivity, self-leadership, and satisfaction of clinical practice among nursing students. Methods: This study was a descriptive correlational study. A total of 205 participants were obtained by convenience sampling from third- and fourth-year nursing students in three nursing schools. The data were collected by using a self-reporting questionnaire. Collected data were analyzed with SPSS Win 18.0. Results: Mean scores were 3.40 (5-point scale) for positive affectivity, 3.68 (5-point scale) for self-leadership, and 3.30 (5-point scale) for satisfaction of nursing clinical practice. There was a significant positive correlation of nursing students among positive affectivity (r=.43, p<.001), self-leadership (r=.41, p<.001), and satisfaction of clinical practice. The predictors were levels of satisfaction about nursing major, grade, positive affectivity, and rehearsal, which accounted for 34% of the variance in satisfaction with clinical practice nursing students. Conclusion: It is necessary to enhance the levels of satisfaction about nursing major, the level of positive affectivity, and self-leadership in nursing students. The results of this study delineate the need for the development and application of diverse educational programs that can increase positive affectivity and self-leadership for clinical practice.

        긍정심리자본에 대한 개념분석

        Purpose: This study was done to clarify the concept analysis of positive psychological capital. Methods: Walker and Avant"s concept analysis process was used in this study. CINAHL, ProQuest, Web of science, NDSL, RISS databases from 2004 to 2016 were searched using the keywords ‘Positive psychological capital" and ‘PsyCap". Sixty-one relevant articles and three book were finally selected. Results: Analysis of identified literature indicated that positive psychological capital attributes were developable potential, positive cognition, expression of emotion, multiple psychological capability, and resources with positive effect. The antecedents were goal achievement, training, leader"s motivation, and supportive organizational environment. The consequences were overall wellbeing which is composed of individual"s physical, psychological and social wellbeing and enhancement of work outcome. Conclusion: The findings in this study provide a definition of the concept of positive psychological capital and theoretical evidence for the future research for nurses including program development.

      • Engagement of CD99 Reduces AP-1 Activity by Inducing BATF in the Human Multiple Myeloma Cell Line RPMI8226

        <P>CD99 signaling is crucial to a diverse range of biological functions including survival and proliferation. CD99 engagement is reported to augment activator protein-1 (AP-1) activity through mitogen-activated protein (MAP) kinase pathways in a T-lymphoblastic lymphoma cell line Jurkat and in breast cancer cell lines. In this study, we report that CD99 differentially regulated AP-1 activity in the human myeloma cell line RPMI8226. CD99 was highly expressed and the CD99 engagement led to activation of the MAP kinases, but suppressed AP-1 activity by inducing the expression of basic leucine zipper transcription factor, ATF-like (BATF), a negative regulator of AP-1 in RPMI8226 cells. By contrast, engagement of CD99 enhanced AP-1 activity and did not change the BATF expression in Jurkat cells. CD99 engagement reduced the proliferation of RPMI8226 cells and expression of cyclin 1 and 3. Overall, these results suggest novel CD99 functions in RPMI8226 cells.</P>

      • RGS1 expression is associated with poor prognosis in multiple myeloma

        <P>Aims Multiple myeloma (MM) is an invariably fatal disease with highly heterogeneous outcome. Because of this heterogeneity of MM, risk stratification is crucial for therapeutic decision-making. However, no immunohistochemical prognostic or predictive markers have been established yet. The expression of regulator of G-protein signalling (RGS) proteins, which desensitise G-protein-coupled receptor signalling, has been reported to be associated with the prognosis of various malignancies. Recently, our group demonstrated the importance of RGS1 in chemokine signalling in a human MM cell line and normal plasmablasts. In the present study, we explored the prognostic value of RGS1 expression in patients with MM using immunohistochemistry. Methods We evaluated RGS1 protein expression in 79 bone marrow biopsies obtained from patients with MM between 2008 and 2010 at Asan Medical Center. Correlations between RGS1 expression and clinicopathological factors were analysed. Results High RGS1 protein expression was significantly associated with poor overall survival (p=0.005). After an adjusted multivariable analysis, high RGS1 protein expression (p=0.010), high International Myeloma Working Group risk (p=0.003) and high serum lactate dehydrogenase levels (p=0.040) were significantly associated with poor outcomes. Conclusions RGS1 expression may be a prognostic marker for risk stratification and a promising target for the development of a new MM therapy.</P>
