All rights reserved</P><P>Background</P><P>The presence of latent tuberculosis (TB) infection (LTBI) should be evaluated before kidney transplantation. Although a new T cell-based assay for diagnosing LTBI gave promising results, this assay has not yet been compared with the tuberculin skin test (TST) for diagnosing LTBI in renal transplant candidates before transplantation.</P><P>Patients and methods</P><P>All adult patients admitted to a single institute for renal transplantation over a 1-year period were prospectively enrolled. A clinically predictive risk of LTBI was defined as: (i) recent close contact with a person with pulmonary TB; (ii) abnormal chest radiography; (iii) a history of untreated or inadequately treated TB; or (iv) a new infection (i.e., a recent conversion of TST).</P><P>Results</P><P>Of 209 renal recipients, 47 (22%) had a positive TST≥5 mm, 21 (10%) had a positive TST≥10 mm, 65 (30%) had a positive T-SPOT.<I>TB</I> test, and 25 (12%) had an indeterminate T-SPOT.<I>TB</I> test. The induration size of TST was significantly associated with a high positivity rate on T-SPOT.<I>TB</I> (<I>P</I><0.001). Agreement between T-SPOT.<I>TB</I> test and TST≥10 mm was fair (<I>k</I>=0.24, 95% confidence interval 0.11–0.36). However, neither univariate nor multivariate analysis showed any association between the clinical risk for LTBI and positivity on T-SPOT.<I>TB</I> or TST.</P><P>Conclusion</P><P>T-SPOT.<I>TB</I> test was more frequently positive than TST in renal transplant candidates. However, further longitudinal studies are awaited to determine whether the ability of T-SPOT.<I>TB</I> assay to detect LTBI in renal transplant recipients can better predict the development of TB than can TST after transplantation.</P>

        Fatigue performance of deepwater SCR under short-term VIV considering various S-N curves

        In this study, a method for fatigue performance estimation of deepwater steel catenary riser (SCR) under short-term vortex-induced vibration was investigated for selected S-N curves. General tendency between S-N curve capacity and fatigue performance was analysed. SCRs are generally used to transport produced oil and gas or to export separated oil and gas, and are exposed to various environmental loads in terms of current, wave, wind and others. Current is closely related with VIV and it affects fatigue life of riser structures significantly. In this regards, the process of appropriate S-N curve selection was performed in the initial design stage based on the scale of fabrication-related initial imperfections such as welding, hot spot, crack, stress concentration factor, and others. To draw the general tendency, the effects of stress concentration factor (SCF), S-N curve type, current profile, and three different sizes of SCRs were considered, and the relationship between S-N curve capacity and short-term VIV fatigue performance of SCR was derived. In case of S-N curve selection, DNV (2012) guideline was adopted and four different current profiles of the Gulf of Mexico (normal condition and Hurricane condition) and Brazil (Amazon basin and Campos basin) were considered. The obtained results will be useful to select the S-N curve for deepwater SCRs and also to understand the relationship between S-N curve capacity and short-term VIV fatigue performance of deepwater SCRs.

      • Antibiotic susceptibility and resistance of Streptococcus iniae and Streptococcus parauberis isolated from olive flounder (Paralichthys olivaceus)

        The rates of antibiotic susceptibility and resistance were investigated in Streptococcus iniae and Streptococcus parauberis isolates obtained from diseased olive flounders (Paralichthys olivaceus) collected from fish farms in Jeju Island, Korea. Isolates of S. iniae (n=65) were susceptible to cefotaxime, erythromycin, ofloxacin, penicillin, tetracycline and vancomycin, as demonstrated by the minimum inhibitory concentration (MIC) test. Isolates of S. parauberis (n=86) were highly resistant to erythromycin (58% of the 86 isolates tested) and tetracycline (63% of the 86 isolates tested). Fifty-four isolates of tetracycline-resistant S. parauberis contained the tet(M/O/S) genes, of which 39 and 12 isolates contained the tet(M) and tet(S) genes, respectively, whereas 3 isolates contained both the tet(M) and tet(S) genes. Among the erythromycin-resistant isolates of S. parauberis (n=50) only 14 contained the erm(B) gene. These results suggest that the tet(S) and erm(B) genes of S. parauberis are involved in the acquisition of high-level resistance to erythromycin and tetracycline. Our findings reveal a high rate of antibiotic resistance among strains of S. parauberis and emphasize the need to develop an appropriate vaccine to reduce the use of antibiotics.

      • S nutrition alleviates salt stress by maintaining the assemblage of photosynthetic organelles in Kentucky bluegrass (Poa pratensis L.)

        <P>To assess the roles of sulfur (S) nutrition in salt stress tolerance in Kentucky bluegrass (Poa pratensis L.). The plants grown in S-supplied or S-deprived condition for 4 weeks were exposed to salt stress with 100 mM NaCl or non-salt stress, respectively, for 21 days. Osmotic potential was significantly decreased by salt stress from day 14. Photosynthetic pigments such as chlorophyll and carotenoid were decreased by salt stress which was more severe in the absence of S, but their content was largely recovered in the presence of S-nutrition. The proteomic analysis of multi-protein complexes in the thylakoid by BN-PAGE showed that the expression of PSI, PSII and RuBisCo level was repressed under salt stress in the absence of S, whereas their expression was largely recovered by S supply. Enzymatic activity confirmed the responses of RuBisCo, estimated by the BN-PAGE, showing a decreased activity in S-deprived and/or salt stressed levels. The decreased RuBisCo activity was significantly related to S content as affected by S nutrition and/or salt stress. Significant relationship between S content and Na, K, Fe content was also observed. These results indicate that S-nutrition modulates the negative responses to salt stress tolerance in photosynthetic organelles of P. pratensis.</P>

        Reactivities of organic isothiocyanates and thiocyanates toward dialkyl bis(phosphine) complexes of palladium(II) and platinum(II)

        Room-temperature reactions of trans-[PdEt<SUB>2</SUB>L<SUB>2</SUB>] (L=PMe<SUB>3</SUB>, PEt<SUB>3</SUB>, PMe<SUB>2</SUB>Ph) with organic isothiocyanates [R-NCS; R=benzyl; CH(CH<SUB>3</SUB>)Ph, R-(-) and S-(+); indanyl, S-(+)] afforded the S,S-coordinated Pd(II) complexes [Pd(S<SUB>2</SUB>C?N-R)L<SUB>2</SUB>] containing a dithiocarbonimidato (S<SUB>2</SUB>C?N-R) group. Similar reactions involving allyl isothiocyanates produced the cationic η<SUP>3</SUP>-allyl Pd complex [Pd(η<SUP>3</SUP>-allyl)(PMe<SUB>3</SUB>)<SUB>2</SUB>]<SUP>+</SUP>(NCS)<SUP>-</SUP>. When [Pd(S<SUB>2</SUB>C?N-R)(PMe<SUB>3</SUB>)<SUB>2</SUB>] was treated with 1equiv of a chelating phosphine [L-L=depe (1,2-bis(diethylphosphino)ethane) and dmpe (1,2-bis(dimethylphosphino)ethane)], the corresponding complexes [Pd(S<SUB>2</SUB>C?N-R)(L-L)] were produced. Reactions of trans-[PdEt<SUB>2</SUB>L<SUB>2</SUB>] (L=PMe<SUB>3</SUB>, PMe<SUB>2</SUB>Ph) with organic thiocyanates (R-SCN; R=benzyl, Et) resulted in the formation of [Pd(CN)<SUB>2</SUB>L<SUB>2</SUB>] and an organic disulfide by S-C bond cleavage of R-SCN. However, similar reactions of the dimethyl analogs, trans-[PdMe<SUB>2</SUB>L<SUB>2</SUB>] (L=PMe<SUB>3</SUB>, PEt<SUB>3</SUB>), with benzyl thiocyanate afforded different products, [Pd(NCS)<SUB>2</SUB>L<SUB>2</SUB>] or [PdMe(NCS)L<SUB>2</SUB>]. Treating [Pt(styrene)(PMe<SUB>3</SUB>)<SUB>2</SUB>] with benzyl isothiocyanate gave the S-coordinated dithiocarbonimidato Pt(II) complex, [Pt(S<SUB>2</SUB>C?N-R)(Me<SUB>3</SUB>P)<SUB>2</SUB>] (R=benzyl). In contrast, cis-[PtEt<SUB>2</SUB>(PMe<SUB>3</SUB>)<SUB>2</SUB>] reacted with the isothiocyanate to afford the trialkyl Pt(IV) complex [PtEt<SUB>2</SUB>(SCN)(CH<SUB>2</SUB>Ph)(PMe<SUB>3</SUB>)<SUB>2</SUB>].

      • DMNQ S-64 Induces Apoptosis via Caspase Activation and Cyclooxygenase-2 Inhibition in Human Nonsmall Lung Cancer Cells

        <P>Shikonin has been reported to induce apoptosis and inhibit angiogenesis in vivo and in vitro. 6-(1-propoxyiminoalkyl)-5,8-dimethoxyoxy 1,4-naphtoquinone S-64 (DMNQ S-64) was synthesized as a shikonin derivative. In this article, the underlying apoptotic mechanism of DMNQ S-64 was examined. DMNQ S-64 exerted cytotoxicity against A549 lung carcinoma cells with IC(50) of 27.3 microM. Apoptotic bodies were observed in DMNQ S-64-treated A549 cells by 4'-6-diamidino-2-phenylindole (DAPI) staining assay. DMNQ S-64 also increased sub-G1 DNA portion in a concentration-dependent manner by flow cytometric analysis. Western blotting has revealed that DMNQ S-64 effectively activates the expression of caspase 8, 9, and 3, cleaves poly (ADP-ribose) polymerase, and increases the ratio of Bax/Bcl-2. Furthermore, cytochrome c was released in a concentration-dependent manner by DMNQ S-64. Similarly, DMNQ S-64 significantly increased caspase 3 activity by enzyme-linked immunosorbent assay (ELISA). It also significantly inhibited the level of prostaglandin E2 (PGE(2)) by ELISA and downregulated the expression of cyclooxygenase-2 (COX-2) in a concentration-dependent manner. Taken together, DMNQ S-64 may exhibit cytotoxicity against A549 cells via caspase activation and COX-2 inhibition.</P>

        Complete Sequence of the Mitochondrial Genome of Spirometra ranarum: Comparison with S. erinaceieuropaei and S. decipiens

        This study was undertaken to determine the complete mitochondrial DNA sequence and structure of the mito- chondrial genome of Spirometra ranarum, and to compare it with those of S. erinaceieuropaei and S. decipiens. The aim of this study was to provide information of the species level taxonomy of Spirometra spp. using the mitochondrial ge- nomes of 3 Spirometra tapeworms. The S. ranarum isolate originated from Myanmar. The mitochondrial genome se- quence of S. ranarum was compared with that of S. erinaceieuropaei (GenBank no. KJ599680) and S. decipiens (Gen- Bank no. KJ599679). The complete mtDNA sequence of S. ranarum comprised 13,644 bp. The S. ranarum mt genome contained 36 genes comprising 12 protein-coding genes, 22 tRNAs and 2 rRNAs. The mt genome lacked the atp8 gene, as found for other cestodes. All genes in the S. ranarum mitochondrial genome are transcribed in the same direction and arranged in the same relative position with respect to gene loci as found for S. erinaceieuropaei and S. decipiens mt ge- nomes. The overall nucleotide sequence divergence of 12 protein-coding genes between S. ranarum and S. decipiens differed by 1.5%, and 100% sequence similarity was found in the cox2 and nad6 genes, while the DNA sequence diver- gence of the cox1, nad1, and nad4 genes of S. ranarum and S. decipiens was 2.2%, 2.1%, and 2.6%, respectively.
