        Caffeic acid phenethyl ester의 처리에 의한 NSAID activated gene-1의 과대발현

        파이토케미칼의 일종인 CAPE가 암세포 생장에 미치는 영향과 유전자 발현을 연구하기 위하여, 인간 대장암 세포주 HCT116에 CAPE를 처리하였다. CAPE의 처리는 농도 의존적으로 암 세포 생존율을 감소시키고, 세포사멸을 유도함을 확인하였다. CAPE에 의해 차별적으로 발현되는 유전자를 분석하기 위하여, oligo DNA microarray 실험을 수행하였다. 그 결과, 20 μM CAPE를 24시간 동안 처리한 경우, 2배 이상 발현이 증가되는 유전자 266개, 2배 이상 발현이 감소되는 유전자 143개를 확인하였다. 발현이 증가되는 유전자중 3개(NAG-1, p21, GADD45A)를 선택하여, RT-PCR을 수행하였다. 그 결과, 모든 유전자의 발현이 마이크로어레이 실험결과와 일치하였다. 또한, CAPE를 농도 의존적으로 처리한 후, NAG-1 유전자와 단백질의 발현을 확인한 결과, mRNA 수준과 단백질 수준에서의 발현양상이 동일함을 확인하였다. 게다가, CAPE를 포함한 5개의 다른 종류의 파이토케미칼(resveratrol, genistein, daidzein, capsaicin)을 처리한 경우, 처리한 모든 파이토케미칼에 의해 NAG-1 유전자의 발현이 증가됨을 확인하였다. 이중 CAPE가 가장 낮은 농도의 처리임에도 불구하고 NAG-1의 발현을 가장 강하게 유도하였다. 결론적으로 이러한 연구결과는 CAPE에 의한 세포사멸은 항암유전자인 NAG-1의 과대발현과 밀접한 관련이 있음을 의미한다. To investigate whether caffeic acid phenethyl ester (CAPE) could affect cancer cell viabilities and gene expression, human colorectal HCT116 cells were incubated with CAPE. CAPE decreased cancer cell viabilities and induced apoptosis in a dose-dependent manner. To analyse differently expressed genes by CAPE, we performed oligo DNA microarray analysis. We found that 266 genes were up-regulated more than twofold, whereas 143 genes were down-regulated more than twofold by 24 hr of treatment with 20 μM CAPE. Among the up-regulated genes, we selected 3 genes (NSAID activated gene-1 [NAG-1], cyclin-dependent kinase inhibitor 1A [CDKN1A, p21] and growth arrest and DNA-damage-inducible alpha [GADD45A]) and performed reverse-transcription PCR to confirm microarray data. In addition, we found that CAPE increased NAG-1 gene and NAG-1 protein expression in a dose-dependent manner. And, several other phytochemicals (resveratrol, genistein, daidzein and capsaicin) also could induce NAG-1 expression in human colorectal HCT116 cells. However, CAPE was the highest inducer of NAG-1, even in low concentrations. Overall, these results imply that cancer cell death by CAPE is closely related with over-expression of NAG-1.

        5 alpha-reductase deficiency is a rare autosomal recessive disorder caused by mutations in the SRD5A2-gene, resulting in absent or diminished dihydrotestosterone (DHT) formation and, hence, in an underdevelopment of the external genitalia in patients with 46,XY karyotype. Recently we experienced a 17 years old patient with chief complaint of primary amenorrhea, who showed 46,XY karyotype, enlarged clitoris, virilization, undeveloped breast and palpable bilateral inguinal mass. We diagnosed it as 5 alpha-reductase deficiency and removed the bilateral gonads, so we report it with brief review of literature.

        화장료 소재로서 뜰보리수 과실 추출물의 항산화, 항염증 및 항노화 활성

        Elaeagnus multiflora, commonly called cherry silver berry, is used as a medicinal material in China and Korea. The purpose of this study was to investigate the antioxidant and anti-aging properties of the fruit extract of E. multiflora using different solvents, such as hot water extract (HWE), butylene glycol with water and ethanol extract (BGE), and propylene glycol with water and ethanol extract (PGE). In the measurement of the radical scavenging of 2,2’-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) and 2,2-diphenyl-1-picryl hydrazyl (DPPH), PGE showed the highest scavenging rates of 84.27% and 95.30%, respectively at a concentration of 2 mg/mL. The SOD-like activity was the highest, 39.98%, in HWE at 2.0 mg/mL, xanthine oxidase inhibition showed 44.98% in HWE. In the analysis of anti-aging effect at 2.0 mg/mL, collagenase and elastase showed the highest inhibition activities, viz. 37.83% and 19.78% in HWE, respectively. In the astringent effect analysis, HWE showed about 90% activity at a concentration of 5.0 mg/mL and about 50% at 25 mg/mL. Tyrosinase inhibition activity for whitening effect was the highest (44.24%) in PGE, followed by HWE, 40.07% at 2.0 mg/mL. Results revealed that PGE had excellent antioxidant activity and HWE showed better anti-inflammatory, anti-aging and astringent effects than BGE and PGE. Therefore, E. multiflora fruits seems to be a natural resource that can be used as a cosmetic material.

        저근백피 추출물의 항노화 활성에 관한 연구

        This study was conducted to investigate the anti-aging effects and cell toxicity of water extracts obtained from Ailanthi radicis cortex at 95℃. The amount of total flavonoid and polyphenol compounds was 4.80 mg/g and 136.89 mg/g, respectively. The astringent effect as a measure of pore tightening activity of the Ailanthi radicis cortex extract was 36.34% at 100 μg/mL. The tyrosinase inhibition as a measure of whitening activity was 53.20% at a concentration of 1,000 μg/mL. The collagenase and elastase inhibitory activities as a measure of anti-wrinkle activity were 48.97% and 18.02% at 1,000 μg/mL, respectively. Cell viability, as determined using the MTT assay, Ailanthi radicis cortex water extract showed no toxicity toward RAW264.7 macrophages and HaCaT keratinocytes. The Ailanthi radicis cortex water extract promoted the synthesis of procollagen type-1 and collagen type-1 at concentration dependent manner. These results suggested that the water extract from the Ailanthi radicis cortex have anti-aging properties and could be used as a natural source for development of functional cosmetics.

        화장료 소재개발을 위한 5종 약용식물 열수추출 혼합물의 항산화성 연구

        The present study aimed to examine the antioxidant activity and cytotoxities of cosmetic material development mixture extracts (C. chinensis, P. amurense, H. cordata, S. glabrae and L. bicolor) with different extraction mixing ratios (ME1, 1:1:1:1:1; ME2, 1:1:2:1:1; ME3, 1:1:4:1:2; ME4, 1:1:6:1:2). The results showed that superoxide dismutase (SOD)-like activity was the highest, and 19.24% for the ME4 at 1,000 ㎍/mL. In the measurement of the radical scavenging of ABTS and DPPH, ME3 showed the highest scavenging rates of 97.34% and 99.81%, respectively, at a concentration of 1,000 ㎍/mL. The collagenase and elastase inhibition abilities of ME3 and ME2 mixtures were found to be 47.09% and 17.19% at 1,000 ㎍/mL, and at concentration of 1,000 ㎍/mL, tyrosinase showed the highest inhibition activities 57.3% in ME3. Furthermore, cytotoxicity test of the extract mixture on RAW264.7 macrophages and HaCaT keratinocytes was conducted using the MTT assay. The RAW264.7 macrophages and HaCaT keratinocytes cell toxicity revealed that cell survival did not reduce at 25-100 ㎍/mL concentration. The inhibition of NO production of 5 medicinal herbs mixture decreased by about 13%-15% in the treatment of 100 ㎍ /mL. These results indicated that both ME3 and ME4 have excellent anti-aging and antioxidant activity. Therefore, it can be concluded that ME3 and ME4 with 5 medicinal herbs mixture have the potential to be used as functional ingredient for beauty food and skin health.

        인간 대장암 세포주에서 capsaicin 처리에 의한 차별적인 유전자 발현의 p53 의존성 분석

        본 연구에서는 대장암 세포주 모델에서 파이토케미칼 capsaicin에 의한 항 생장 활성과 유전체 수준에서의 유전자 발현 변화를 연구하였다. 그 결과, 처리한 capsaicin 농도 의존적으로 세포 생존율이 감소함을 확인하였고, capsaicin은 다양한 유전자의 발현 변화를 유도하였다. DNA microarray 실험결과 100 μM capsaicin의 처리에 의해 2배 이상 증가되는 유전자 103개가 확인된 반면, 2배 이상 발현이 감소되는 유전자 153개가 확인되었다. 발현이 증가되는 유전자 중 4개(NAG-1, DDIT3, GADD45A 그리고 PCK2)를 선택하여 RT-PCR을 수행한 결과, DNA micorarray 실험과 일치함을 확인하였다. 또한 100 μM capsaicin의 처리에 의해 암 억제유전자인 p53의 발현이 증가됨을 RT-PCR과 real-time PCR 방법으로 확인하였다. 게다가, NAG-1, DDIT3 그리고 GADD45A 유전자는 p53의 존재에 관계없이 발현이 증가되는 반면, PCK2 유전자는 반드시 p53에 의해 발현이 유도됨을 확인할 수 있었다. 이러한 연구는 대장암 세포주에서 capsaicin에 의한 항암 기전을 이해하는데 도움을 줄 것으로 기대된다. In the present study, we investigated anti-proliferative activities of capsaicin and gene expression changes in response to capsaicin treatment in human colorectal HCT116 cells. The results showed that capsaicin decreased cell viabilities in a dose dependent manner and induced global gene expression changes. We found that 103 genes were up-regulated more than twofold, whereas 153 genes were down-regulated more than twofold by 100 μM capsaicin treatment. Among the up-regulated genes, we selected 4 genes (NAG-1, DDIT3, GADD45A and PCK2) and performed RT-PCR to confirm the microarray data. We found that 100 μM of capsaicin increased tumor suppressor p53 gene expression. In addition, the results showed that NAG-1, DDIT3 and GADD45A expressions were not dependent on p53 presence, whereas PCK2 expression. The results of this study may help to increase our understandings of the molecular mechanism of anti-proliferative activity mediated by capsaicin in human colorectal cancer cells.

        캄보디안 상황버섯 추출물의 주름개선 효과 연구

        사람의 피부는 자외선, 오염된 공기, 화학 제품 등 환경에 끊임없이 노출된다. 이들로 인한 자유라디칼과 활성 산소는 피부세포에 큰 손상을 끼치게 된다. 캄보디안 상황버섯 열수, 에탄올 추출물의 기능성 화장품 소재로 활용하기 위하여 지방 산패 억제능 측정과 주름개선 효과 측정을 하였다. 캄보디안 상황버섯 열수, 에탄올 추출물에 금속이온을 첨가하여 지방산패 억제능을 측정한 결과 캄보디안 상황버섯 열수 추출물 보다 에탄올 추출물이 Fe²?와 Cu²?chelating 능력이 우수하였다. 캄보디안 상황버섯 에탄올 추출물의 경우 0.1 ㎎/㎖, 0.5 ㎎/㎖, 1 ㎎/㎖의 농도에서 대조군인 butylated hydroxytoluene (BHT)보다 높은 지방산패 억제를 나타내었다. 캄보디안 상황버섯 열수, 에탄올 추출물은 산화 촉매제인 Fe²를 함유한 지방 산패 억제능이 Cu²?를 이용한 지방산패 억제능 보다 우수하였다. 주름개선 효과 측정으로 elastase 저해활성을 측정한 결과 캄보디안 상황버섯 에탄올 추출물은 0.01 ㎎/㎖의 농도에서 50.7%의 저해활성을 나타내어, 대조군인 urosolic acid보다 높은 elastase 저해활성을 타나내었다. 또한 에탄올 추출물의 경우 0.1 ㎎/㎖의 농도에서 약 50%의 저해활성을 나타내어 캄보디안 상황버섯 열수 추출물 보다 에탄올 추출물이 collagenase에 대해 높은 저해활성을 나타내었다. 이상의 결과에서 캄보디안 상황버섯의 에탄올 추출물은 지방산패 억제능이 우수할 뿐만 아니라 주름 개선 효과가 있음을 확인할 수 있었고, 주름 개선 화장품 소재로서의 가능성을 확인할 수 있었다. The skin of human is constantly being exposed to environmental irritants such as ultraviolet, smoke and chemicals. These irritants cause free radicals and reactive oxygen species which leave serious damages on the cells of skin. The water and ethanol extracts of Cambodian Phellinus linteus were investigated for the activities of anti-lipid peroxidation and anti-wrinkle effects to apply as a functional ingredient for cosmetic products. As the result of evaluation of liquid oxidation rate by add Fe²? and Cu²? to Cambodian Phellinus linteus extracts, Cambodian Phellinus linteus ethanol extracts were higher than Cambodian Phellinus linteus water extracts in the chealting ability of Fe²? and Cu²?. The Cambodian Phellinus linteus ethanol extracts exhibited that anti-lipid peroxidation higher than butylated hydroxytoluene (BHT) at the concentration of 0.1 ㎎/㎖, 0.5 ㎎/㎖ and 1 ㎎/㎖. Cambodian Phellinus linteus water and ethanol extracts showed a higher inhibitory effect on Fe²?-induced lipid peroxidation compared to Cu²?-induced lipid peroxidation. In the case of anti-wrinkle effect, the elastase inhibition activity of Cambodian Phellinus linteus ethanol extracts was 50.7%, and it is higher than urosolic acid at the concentration of 0.01 ㎎/㎖. Also, in collagenase inhibition activity, Cambodian Phellinus linteus water extract showed low effect, but Cambodian Phellinus linteus ethanol extract was about 50% at a 0.1 ㎎/㎖. concentration. These results proved that Cambodian Phellinus linteus had anti-lipid peroxidation and anti-wrinkle effect. Therefore, Cambodian Phellinus linteus could be useful as an anti-wrinkle cosmetic ingredient.
