Since cellulose is the only organic material that is annually replenishable in very large quantities, we must explore ways to utilize it as a source of energy, food and chemicals. For the utilization of this resource, it is first enzymatic hydrolyzed to glucose, then the glucose can be used as a food, converted single cell protein by microorganism, fermented to clean burning fuel and other chemicals. Cellulolytic enzyme, cellulase, consists of two or three major components, C₁-cellulase, C_x-cellulase and β-glucosidase. C_x-cellulase are fairly common but C₁-cellulase are quite rare. Trichoderma viride is the best source of active cellulase, especially C₁-enzyme. Saccharification rate of cellulose in greatly influenced by the degree of crystallinity and extent of lignification. But by the pretreatment the substrate with cellulose swelling agent, delignifying reagent and physical treatment, the degree of saccharification is enhanced. Thus, glucose syrups of 2 to 10% concentration are realized from milled newspaper. The enzymatic hydrolysis of such energy rich material, such as cellulose, to glucose is technically feasible and practically achievable on a very large scale.

Three strains among 120 adenineless mutants of Brevibacterium ammoniagenes described in the previous paper were screened out to accumulate UV absorbing substances in the culture broth. It was analyzed to be 5'-inosinic acid and hypoxanthine by means of two dimensional paper chromatography, UV absorption spectra and periodate oxidation. 5'-IMP was isolated from the culture broth of the mutant No. 203 with anion-exchange resin amberite IRA-400 and recrystallized from ethanol. It was proved identical to authentic sample by Infra-red absorption spectrum. The growth responses of the mutant No. 2013 were demonstrated to require adenine, but not with adenosine and 5'-AMP.

As the first step in the production of nucleic acid derivatives by microorganisms, adenineless mutants were derived from Brevibacterium ammoniagenes ATCC6872. A culture of Br. ammoniagenes was exposed to ultraviolet rays for 120 second and treated with diethylsulfate in phosphate buffer for 2 hours to reach the designed death rate. The yield of mutants induced was 0.28% by the ultraviolet irradiation and 0.66% by the diethylsulfate treatment. By the diethylsulfate treatment. By the treatment of penicillin G in a hypertonic minimal medium, the yield of mutants was increased from 0.28% to 0.54% and from 0.66% to 1.5%, respectively. Thus, in was demonstrated that diethylsulfate treatment was much more efficient than UV irradiation to induce adenineless mutants of the bacteria, and total strains of 120 adenineless mutants were obtained.
Elastase Inhibitor 생성 방선균의 탐색 및 동정

Elastase inhibitor (EI) producing microorganisms were screened by microplate-assay of culture broth using Succinyl-$(Alanine)_3$-Nitroanilide as a chromogenic substrate. The portion of microorganisms showed EI activity above 90% was about 2% of all assayed. Isolated SMF-11 showed relatively high EI activity after filtration using membrane filter of NMWL 3,000. SMF-11 was identified as Streptomyces sp. according to results of morphological or physicochemical identification. SMF-11 has both rectiflexible and spiral spore chains with smooth spore surface and cell wall contained LL-DAP, iso- and anteiso-fatty acids. The amino acids in cell wall were alanine, glutamic acid and glycine. Fifty unit characters for major cluster were tested and the data were analyzed numerically using the TAXON program. The isolate SMF-11 was identified as a strain of Streptomyces lavendulae. Elastase 저해물질(EI) 생성균주로 토양분리주 SMF-11을 선별하였다. 분리주 SMF-11는 매끈한 표면의 rectiflexible 및 spiral의 포자 사슬을 동시에 형성하고 세포벽 성분으로 LL-DAP, ios- 및 anteiso-fatty acid($C_{14}-C_{17}$), 세포벽 아미노산으로 alanine, glutamic acid, glycine을 가지고 있어 방선균속으로 분류되었다. 종 수준의 동정을 위하여 화학적 시험과 50가지의 단위형질에 대한 특성을 조사하고 TAXON program으로 수리분류한 결과, SMF-11 분리주는 방선균 주군집 61의 중심균주인 Streptomyces lavendulae로 동정하였다.
