        밀 ( Triticum spp. ) 의 미성숙배로부터 유도한 현탁 배양세포에서의 염색체 변이

        Suspension cell lines have been newly established from the calli derived from the immature embryo culture of hexapolid(Triticum aestivum var.sicco), tetrapolid(T.durum) and diploid (T. tauchii or Aegilops squarrosa) wheat species. The chromosomal variation in suspension cultured cell lines was examined and old cell line, C82d, established from T. aestivum var. copain was also used. New method using 1-bromonaphthalene for metaphase trapping of suspension cells was developed. Variation in chromosome number was observed amongst all the suspension lines. Cells with doubled chromosome number and deleted chromosome were also observed. Extensive structural changes in chromosomes were found in C82d line. Chromosome aberrations showed loss of chromosome arms and chromosome segment. The mean chromosome number in suspension cells of T. aestivum var. sicco was 40, in C82d line 33, in T.durum 28 and in T. tauchii 14. The stability of chromosomes in suspension cells of diploid and tetrapolid wheats was higher than that of hexaploid wheat.

        한국산 무릇 ( Scilla scilloides complex ) 의 세포유전학적 연구 - 2. 제주도 집단에서 게놈의 분포

        Cytogenetic analyses were done in thirteen natural populations of Scilla scilloides complex collected from Chejudo. Six genotypes, BB (2n=18), ABB (2n=26), AABB (2n=34), ABBB (2n=35), AABBB (2n=43) and AAABBB (2n=51) were observed, where A and B denote genomes with x=8 and x=9, respectively. Genotypes BB, ABB and AAABBB were found for the first time in Chejudo populations. Genotypes BB and ABB were never reported from the Korean populations. Six of the populations investigated were mixed populations of two or three cytogenetic types, and others were pure populations consisting of BB (1), AABB (4) and AABBB (2). AABB genotypes were predominant over other genotypes and AABBB was the next. AA genotypes growing in the mainland of Korea was not found. It was suggested that the Chejudo populations of S. scilloides complex were closely related to the Japanese populations than mainland populations of Korea. The number of B-chromosome ranged from one to four. The B-chromosomes were all isochromosomes (F) and no chromosome fragment (f) was found. The frequencies of B-chromosome were 75% in BB and AABBB genotypes, respectively. The frequency of B-chromosome in AABB plants was similar to that of AABB plants reported in the mainland populations.

        A karyotype of cytotype BB plant in Scilla scilloides complex was established and the frequency of B-chromosomes were investigated. Chromosome complements of BB genome were composed of five pairs of subtelocentric and four pairs of metacentric chromosomes. Chromosome 1 has satellite with nucleolar organizer. Polymorphism was found in chromosome 2. The karyotype of cytotype BB will be available for analysis of genome composition in various cytotypes of S. scilloides complex. The frequency of B-chromosome was 38.6%. Numbers of B-chromosome ranged from 1 to 4 and plants with 2B-chromosomes were predominant (57.2%). Two types of B-chromosomes, F and F`, were found; F is large iso-chromosome and F` a small one.

        리기다소나무 임상에 있어서 Seedling 및 Sapling 의 현재량에 관한 연구

        Distribution and standing crops of seedling and sapling as undergrowing tree were investigated in a Pinus rigida forest. The relative light intensity (RLI) on the ground level was 3.9%∼59.3% which indicates a wide range. Under this range of RLI seedlings and saplings distribute well in the stand. Eight trees were selected from the stand, then measured dry weight of stem, branches and leaves using the stratified clip technique. A possible relationship between the diameter at the bottom of the tree (DO) and standing crops was investigated. As Do increased from 0.8㎝ (T_1) to 7.2㎝ (T_2), the standing crops also increased from 42.6g to7,410.4g. Since semi-Iog values between Do and standing crops clearly indicates a straight line, there is close relationship between these two factors. Therefore it is possible to estimate the standing crops of seedling and sapling by measuring Do. Productive structure shows that photosynthetic layer is located above the 1/2 height of the tree and maximum photosynthetic layer apperars on 2/3 level in height.

        한국 호밀 ( Scale cereale L. ) 의 B 염색체 출현밀도와 지리적 분포

        The frequencies and geographical distribution of B chromosomes on 15 strains of rye(Secale cereale L.) collected from various localities in Korea were investigated. All of the 15 strains of rye investigated were found to have B chromosomes, and the frequencies of B chromosomes ranged from 6% to 51% with 20.1% average. Plants with 2Bs seem to be the most stable in populations with B chromosomes. Of 1400 plants examined, one plant was observed to have a deficient-B chromosome in Buyo rye.

        CuZnSOD와 APX를 엽록체에 발현시킨 담배식물체의 Highlight와 Chilling 스트레스에 대한 광합성 효율

        환경스트레스에 대한 항산화효소의 보호효과를 이해하고 효과적인 스트레스내성 식물을 개발하기 위하여 CuZnSOD와 APX를 엽록체에 발현시킨 형질전환식물체 (CA식물체)에 강한 빛 (highlight, 1,100$\mu$mol m$^{-2}$ sec$^{-1}$)과 4$^{\circ}C$ chilling을 처리하였다. CA식물체 잎절편에서의 보호효과를 CuZnSOD을 도입한 SOD식물체, APX을 도입된 APX식물체 및 비형질전환식물체 (NT식물체)의 것과 비교하였다. CA식물체의 광계2에서의 광합성 효율 (Fv/Fm)은 highlight와 4$^{\circ}C$ chilling복합처리 1시간째에서 NT식물체에 비해 약 15%의 보호효과를 나타내었고, 광계1에서의 P700 redox state는 처리 후 3시간째에 약 23%의 보호효과를 나타내었다. SOD식물체와 APX식물체의 복합처리에 대한 보호효과는 CA식물체와 NT식물체의 중간 효과를 나타내었다. 이러한 결과는 엽록체에서 CuZnSOD와 APX의 동시발현이 highlight와 chilling에 의한 산화스트레스를 극복하는 데 매우 중요하게 관여함을 직접적으로 제시하는 것이다. In order to understand the protection effects of antioxidant enzymes against oxidative stress caused by various environmental stresses, transgenic tobacco (Nicotiana tabacum cv, Xanthi) plants expressing both copper/zinc superoxide dismutase (CuZnSOD) and ascorbate peroxidase (APX) in chloroplasts (referred to as CA plants) were subjected to highlight (1,100$\mu$mol m$^{-2}$ sec$^{-1}$) and chilling at 4$^{\circ}C$. The protection effects of CA plants using leaf discs were compared with those of transgenic plants expressing either CuZnSOD or APX in chloroplasts (SOD plants or APX plants, respectively) and non-transgenic (NT) plants. CA plants showed about 15% protection in the photosynthetic efficiency (Fv/Fm) of photosystem II relative to NT plants 1 hr after treatment of both highlight and chilling, whereas they showed about 23% protection in the redox state of P700 in photosystem I at 3 hr after treatment. SOD plants or APX plants showed an intermediate protection effect between CA plants and NT plants. These results demonstrated that the coexpression of CuZnSOD and APX in chloroplasts importantly involves in the protection effects against oxidative stress caused by various environmental stresses.

        Giemsa C - band 에 의한 호밀의 핵형분석

        Giemsa C-banding analysis with Korean rye strain revealed that individual chromosomes had characteristic banding patterns making it possible to distinguish the seven rye chromosomes one from another. It seems to be evident that B-chromosomes in rye contain proportionally more heterochromatin than autosomes.

        Multicolor FISH와 Feulgen 염색법을 이용한 Angelica속 식물의 세포유전학적 분석

        Karyotype analysis and chromosomal localization of 5S and 45S rDNAs using multi-color fluorescence in situ hybridization (McFISH) technique were carried out in two Angelica species. The numbers of diploid chromosomes were the same in two same in two species as 2n=22, however the lengths of chromosomes were varied from 4.25 to 6.50 ${\mu}{\textrm}{m}$ in A gigas and 4.95 to 8.50 ${\mu}{\textrm}{m}$ in A acutiloba. The chromosomes of A. gigas were composed of five metacentric and six submetacentric pairs, while those of A. acutiloba were six metacentic, one submetacentric and four subtelocentric paris. In FISH experiments, the numbers and size of 45S rDNA signals were varied between two species, however dach signal of the 5S rDNA was observed in two species.

        Geographical distribution of diploid plants with AA genome (2n=16)and allotetraploid with AABB genome (2n=34) of Scilla scilloides Complex from Korea has been studied. The composition and frequencies of B chromosomes were also investigated. Plants with AABB genome were predominant over AA genome plants. A mixed population of AA and AABB genome plants was found for the first time. Aneuploid plants have not been found. Chromosomes of AA genome were composed of three pairs of metacentric, two pairs of submetacentric, two pairs of subtelocentric and one pair of telocentric chromosomes, whereas BB genome was four pairs of metacentric and five pairs of subtelocentric chromosomes. B chromosomes were classified into two categories, isochromosome (F) and chromosome fragment (f). The frequencies of B chromosomes were 43% in AA genome plants and 44% in AABB genome plants. The number of B chromosome ranged from 1 to 3 and 1 to 7 in AA and AABB genome plants, respectively. B chromosome combinations were F and F+f in AA genome plants and F,F+f and f in AABB genome plants.

        2배체 담배 Nicotiana plumbaginifolia의 핵형 분석과 rDNAs의 Physical Mapping

        2배체 담배인 Nicotiana plumbaginifolia를 대상으로 상염색법과 FISH 기법을 통한 염색체 분석을 수행하여 다음과 같은 결과를 얻었다. N. plumbaginifolia의 체세포 염색체 수는 2n=20이며, arm ratio 비교를 통한 핵형 분석에서 중기 염색체 조성은 3쌍의 중부 염색체 (염색체 1,2 및 7)와 7쌍의 차중부 염색체 (염색체 3, 4, 5, 6, 7, 9 및 10)로 관찰되었다. 염색체의 길이는 2.29~4.50 $\mu\textrm{m}$로 나타났으며, 염색체 1번과 2번은 부수체 염색체로 관찰되었다. 5S와 45S rDNA를 탐침으로 FISH를 수행한 결과 2번 염색체의 동원체 부위에서 한 쌍의 5S signal이 확인되었고, 1번 염색체의 부수체에서 한 쌍의 45S signal이 관찰되었다. Karyotype analysis and chromosomal localization of 5S and 45S rDNAs using FISH (fluorescence in situ hybridization) technique were carried out in Nicotiana plumbaginifolia. Somatic chromosome number of N. plumbaginifolia was 2n=20 and kyarotype was composed of three pairs of metacentric chromosomes (1,2 and 7) and seven pairs of submetacentric chromosomes (3, 4, 5, 6, 8, 9 and 10). Length of chromosomes was ranged from 2.29 to 4.50 ${\mu}{\textrm}{m}$. Chromosome 1 and 2 were identified as satellite chromosomes. In FISH experiment, one pair of 5S rDNA signals was detected on the centromeric region of chromosome 2 and one pair of 45S rDNA signals was detected on the terminal region of chromosome 1.
