        저수태 한우 암소 자궁에서 회수된 미생물이 체외수정란 발달율에 미치는 영향

        The cause of infertility is either fertilization failure or early embryonic death. The aetiology may involve a combination of many factors, e.g. genetic factors, abnormalities in the gametes nutritional disorders, inadequate luteal function, and delayed ovulation. This study was conducted to investigate the effect of microorganisms collected from uterus of Hanwoo cattle on early embryonic development. Microorganisms isolated from the uterus of Hanwoo cattle included Bacillus cereus (Bc), Bacillus thuringiensis (Bt), Staphylococcus warneri (Sw) and Enterococcus faecalis (Ef). When cultured with Bc, Bt, Sw, and Ef, oocytes were not developed into a blastocyst in vitro. The proportion of blastocyst was dramatically increased after reducing the number of microorganisms (10⁴ CFU/ml). Interestingly, the proportion of blastocyst was decreased by adding the Sw and Ef. These results indicate that among intrauterine microorganisms, Sw and Ef strains negatively affect theearly embryonic development, thereby aggravate the rates of implantation and pregnancy. These findings will provide useful information for association studies in other pig populations.

        Effects of Total Mixed Fermentations with Bean Curd Dregs on Growth Performance of Growing Hanwoo Heifers

        본 연구는 비지박을 이용한 섬유질발효사료(TMF) 급여가 육성기 한우 암소의 증체, 사료섭취량 및 체형변화에 미치는 영향을 구명하기 위하여 수행하였다. 6개월령 한우 암소 30두(평균체중125.9±12.0 kg)를 공시하여 3처리로 각각 10마리씩 완전 임의배치하여 6개월간 사양시험을 수행하였다. 처리구는 T1 (볏짚 자유급여와 배합사료(체중1.7%/일) 제한급여구), T2(TMF 자유 급여구) 및 T3 (TMF, 볏짚 자유급여와 배합사료(체중 1.0%/일) 제한급여구)로 수행하였다. 섬유질발효사료는 비지박 60%, 옥분 16.5%, 밀기울 10.4%, 볏짚 5%, 왕겨 7%, 요소 0.5%, 고토석회 0.5%, Lactic acid bacteria 0.1%를 첨가하여 제조하였다. 섬유질발효사료의 화학적 성분과 pH 값은 저장 0, 24, 48 및 72시간에 측정하였다. 비지박을 이용한 TMF의 저장기간에 따른 결과에서 건물, 조지방, 및 조섬유의 함량은 차이가 없었다. 저장 72시간에서 ADF 및 NDF함량은 0시간에서 보다 각각 9.52 및 3.15% 유의적으로 감소하였다(p<0.05). pH 또한 저장시간에 따라 점점 감소하였다. 체중, 사료섭취량 및 일당증체량은 TMF 급여구(T2, T3)에서 T1급여구 보다 높았다(p<0.05). 체형변화에 있어서도 T1처리구 보다 TMF 급여구가 높았다(p<0.05). 따라서 본 연구의 결과 비지박을 이용한 TMF 사료의 급여는 육성기 한우 암소의 발육을 향상시키는 것으로 나타났다. This study was conducted to determine the effects of total mixed fermentation (TMF) with bean curd dregs on body weight gain, feed intake and body size of growing Hanwoo heifers. Thirty Hanwoo heifers, 6 months of age and weighing 125.9±12.0 kg, were randomly allocated to 3 experimental groups of 10 animals each for a 6-month feeding trial. The groups were T1 (rice straw offered ad libitum with restricted (1.7% BW/day) concentrate), T2 (TMF only offered ad libitum) and T3 (TMF and rice straw offered ad libitum with restricted (1.0% BW/day) concentrate). TMF feeds were composed of 60% bean curd dregs, 16.5% corn meal, 10.4% wheat bran, 5% rice straw, 7% rice hulls, 0.5% urea matter, 0.5% magnesia lime and 0.1% lactic acid bacteria. The chemical composition and pH value of TMF feeds were measured at 0, 24, 48 and 72 hr after storage. In TMF with bean curd dregs, there were no differences in concentrations of DM, EE, and CF according to storage periouds. The ADF and NDF at 72 hr of storage time showed a significant decrease (9.52 and 3.15 %, respectively) compared to 0h of storage time (p<0.05). The value of pH was gradually decreased during the storage periods. Body weight gain, feed intake and average daily gain were higher in the TMF groups (T2, T3) compared to the T1 group (p<0.05). Changes in body size were also higher in the TMF groups compared to the T1 group (p<0.05). Thus, the present results indicate that TMF with bean curd dregs may help improve growth performance in growing Hanwoo heifers.

        임신말기 자궁내 발육상태와 자궁적출 시기가 특정질병 제어 형질전환 복제동물 생후 생존에 미치는 영향

        Bioorgan transgenic cloned mini pig has a problem of growth retardation in uterus during end of pregnancy so that survival rate is very low in newborn piglet. In order to support their life after birth, cesarean section of fetus with sufficient growth in uterus was tested in this study. First of all, fetus growth measured using a ultrasound scanner during pregnancy in transgenic mini pig, comparing normal pig. After 113 days for delivering, fetus was removed out of uterus. Fetus growth for normal pig was 1.8 cm at 4weeks and 14.4 cm at end of pregnancy (15 weeks). At 113 days, fetus growth was 15.9±4 cm in ultrasound scanner and real growth measurement from fetus removal out of uterus was 16.0±2 cm. It is very a similar result between measurement of ultrasound scanner and real measurement. Therefore, using ultrasound scanner for measuring fetus growth will be useful to predict fetus growth in uterus.

        소 재조합 somatotropin 단백질을 이용한 한우 유량 증진 효과

        This study was carried out to increase the breeding rate using sustained recombinant bovine somatotropin (bST) and examine the reproductive disorder and disease prevalencies of cow and calf, respectively. Sixty cows were allotted randomly to three groups in randomized complete block design; (1) No treatment, (2) 500 mg bST two times + 250 mg bST SC injection five times from 3 weeks before to 9 weeks after parturition every two weeks, (3) 500 mg bST two times before 3 weeks parturition + 250 mg bST SC injection three times from five to 9 weeks after parturition every two weeks. Developmental data of birth weight, weaning weight, total gain and average daily gain in control was 22.9±3.45 kg, 88.0±11.13 kg, 65.1±9.74 kg and 0.54±0.08 kg, respectively. There is no significant difference between Group 1 and 3 in the examined factors. However, there is significant difference between Group 1 and 2 in the birth weight (24.4±2.88 kg), weaning weight (101.0±11.77 kg), total gain (76.7±7.9 kg) and average daily gain (76.7±7.9 kg). Total gain was showed higher in Group 2 and 3 than Group 1 in the >2nd parity, and in Group 2 than Group 1 and 3 in the 1st parity. There is no significant difference in the BCS, no return days and disease occurrence of cow and calf by bST treatment. Number of service per conception was slightly increased in the Group 3 compared to Group 1 and 2. To increase the breeding ability of Hanwoo, it is recommended that 500 mg bST two times + 250 mg bST injection five times from 3 weeks before to 9 weeks after parturition every two weeks.

        Acute vascular rejection has been known as a main barrier occurring in a xenograted tissue of alpha 1,3-galactosyltransferase knock-out (GalT KO) pig into a non-human primate (NHP). Adenosine which is a final metabolite following sequential hydrolysis of nucleotide by ecto-nucleotidases such as CD39 and CD73, act as a regulator of coagulation, and inflammation. Thus xenotransplantation of CD39 and CD73 expressing pig under the GalT KO background could lead to enhanced survival of recipient NHP. We constructed a human CD39 and CD73 expression cassette designed for endothelial cell-specific expression using porcine Icam2 promoter (pIcam2-hCD39/hCD73). We performed isolation of endothelial cells (pAEC) from aorta of 4 week-old GalT KO and membrane cofactor protein expressing pig ($GalT^{-MCP/-MCP}$). We were able to verify that isolated cells were endothelial-like cells using immunofluorescence staining analysis with von Willebrand factor antibody, which is well known as an endothelial maker, and tubal formation assay. To find optimal condition for efficient transfection into pAEC, we performed transfection with GFP expression vector using four programs of nucleofection, M-003, U-023, W-023 and Y-022. We were able find that the program W-023 was optimal for pAEC with regard to viability and transfection efficiency by flow cytometry and fluorescent microscopy analyses. Finally, we were able to obtain $GalT^{-MCP/-MCP}/CD39/CD73$ pAEC expressing CD39 and CD73 at levels of 33.3% and 26.8%, respectively. We suggested that pACE isolated from $GalT^{-MCP/-MCP}$ pig might be provided as a basic resource to understand biochemical and molecular mechanisms of the rejections and as an alternative donor cells to generate $GalT^{-MCP/-MCP}/CD39/CD73$ pig expressing CD39 and CD73 at endothelial cells.

        The automatic-spreader of bedding materials was developed to reduce labor cost, and to achieve successful biosecurity in duck houses. Algorithm of the device was designed to realize a concept of the automatic unmanned operation including entire processes such as loading and spreading of the bedding materials. From the field measurement, the relationship between the expected water content reduction and weight of bedding materials per unit area according to the operation condition of the devices was induced. In the case of the measurement of particulate matters during the process of spreading works, the results of using both conventional manual-spreader and automatic-spreader were exceeded the allowable limit of inhalable and respirable dust, respectively. But, workers using automatic-spreader could be free from harmful aero-condition because they did not stay inside the facility during the spreading works. In addition, from the Duck hepatitis virus PCR test, influence on pulmonary tissue of the duck was not found. It could be expected that the development of the automatic-spreader of bedding materials for duck house can contribute to the advancement of duck breeding facilities.

        개 parthenote in vitro culture시 EDTA 첨가에 의한 발달율 향상

        Somatic cell nuclear transfer (SCNT) is a useful biotechnological tool for animal cloning. Until now, SCNT has been inefficient, especially in dog. It is believed that an embryo developmental block in SCNT embryos is cause of low production efficiency. However, no studies have been performed on canines for embryo developmental block. In this study, we attempted to evaluate the beneficial role of EDTA in canine parthenogenic (PA) embryos development to overcome embryo developmental block. The PA embryos were divided into 0.01 mM EDTA treated and non-treated groups. Embryo developmental efficiency was measured by activating chemically parthenote. After EDTA induction, PA embryos were evaluated for embryonic development, Reactive Oxygen Species (ROS) activity, mitochondrial integrity, ATP production and genomic activation. The EDTA treated PA embryos showed significantly higher survival rate and improved cavity formation compared to non-treated. Furthermore, cytoplasmic ROS level was mitigated and mitochondrial membrane potential was found significantly higher in EDTA treated group followed by higher ATP production. Moreover, major embryonic genomic activation specific markers/factors were also elevated in EDTA treated group. Conclusively, we elucidated that EDTA showed substantially positive effect to overcome embryo developmental block in canine.

        TGF- ${\beta}$ 첨가가 한우 난포란의 체외성숙에 미치는 영향

        소 체외성숙시 난포란에 영향을 주는 과립막세포와 난자의 핵성숙을 촉진하는 물질이 확인되지 않은 혈청내의 물질 뿐만 아니라 호르몬이나 생리활성인자 등에 의해 촉진됨이 밝혀졌다. 이에 따라 체외성숙 및 체외발달에 사용되는 배양액의 조성도 복합배양액에서 단순배양액으로 전환을 시도하고 있으며, 체내의 조건에 보다 더 접근하고자 하는 시도들이 수행되고있다. 본 연구는 한우 난포란의 체외성숙율 및 체외수정 후 배발달율을 향상시키기 위하여 TGF-${\beta}$의 첨가가 미치는 영향에 대하여 조사하였다. 한우 난포란의 체외성숙시 TGF-${\beta}$ 0.1, 1. 10ng/ml를 첨가하였을 때 24시간에 MetaphaseⅡ 도달율은 95.8%, 95.8%, 100%를 나타내었으며, 농도간의 유의적인 차이는 없었다. TGF-${\beta}$로 체외성숙된 난포란을 체외수정 후 TGF-${\beta}$가 체외성숙 배양액을 동일하게 체외발달을 유도하였으나, 0~0.8%의 배반포율을 보였으며, TGF-${\beta}$로 체외성숙된 난포란을 체외수정 후 TCM 199_10% FBS, 0.8% BSA, 0.1% PVA에 배양한 결과 12.4%, 12.8%, 8.5%의 배반포 발달율을 보였고, 무혈청 배양액인 IVMD, IVD에 배양한 결과 38.4%, 34.8%의 배반포 발달율을 보였으며, 유의적인 차이를 나타냈다(P<0.005). 결론적으로 TGF-${\beta}$는 한우 난포란의 체외성숙시 상당히 유용한 물질이나, 체외발달에는 만족할 수 없어, 이외의 생리활성 인자들간의 상호관계에 대하여 더 많은 요구가 요구된다. It is well known that unidentified factors in sera, hormones and growth factors promote the proliferation of granulosa cells and nuclear maturation of bovine COCs in vitro. Attemps had been developed the simple composition of culture media and similar system to in vivo conditions has been applied. In the present study, we investigated the effect of TGF-${\beta}$ on in vitro maturation and in vitro development of Hanwoo COCs. When the COCs were matured in TCM 199 containing 0.1, 1 or 10 ng/ml TGF-${\beta}$ for 24 hrs, metaphaseⅡ of COCs were obtained 95.8%, 100% of matured COCs, respectively and there were no differences among the concentrations of TGF-${\beta}$. Matured COCs with TGF-${\beta}$ cultured in maturation medium after in vitro fertilization, developmental rate to blastocyst were 0~0.8%. Matured COCs with TGF-${\beta}$ were cultured in TCM 199+10% FBS, 0.8% BSA, 0.1% PVA, blastocyst formation were showed in 12.4%, 12.8%, 8.5% of those and cultured in IVMD or IVD without serum were 38.4%, 34.8%, respectively. There were significant differences among the media (P<0.05). TGF-${\beta}$ is available for i vitro maturation of bovine COCs, but further investigation would be need for finding the synergistic autocrine/paracrine fashion of other growth factors in early bovine development.

        오징어 부산물의 급여가 한우암소 산육 및 도체특성에 미치는 영향

        본 연구는 한우 암소의 산육 및 도체특성에 있어 오징어 부산물의 급여 효과를 알아보기 위해 수행되었다. 한우 16두(나이 7세, 체중484.1±63.6 ㎏)를 대조구 및 처리구(오징어 부산물 첨가), 등 2그룹(각각 8두)으로 완전임의 배치하였다. 사료급여는 두 그룹 모두 자유채식토록 하여 150일간 시험을 수행하였다. 일당증체량은 처리구가대조구 보다 유의적으로 높았다(p<0.05). 시험기간 내 사료섭취량 및 사료요구율은 처리구가 대조구 보다 유의적으로 낮았다(p<0.05). 육량특성에 있어서, 등지방두께, 등심단면적 및 육량지수는 두 처리구간 비슷한 경향을 나타내었고, 도체중은 처리구가 363.4 ㎏ 대조구 353.1 ㎏으로 처리구간 유의적 차이는 없었다. 육질특성에서는 육색, 지방색, 조직감 및 성숙도는 두 그룹간 유의적 차이가 없었다. 그러나 근내지방도 및 육질 1등급 이상 출현율은 대조구 보다 처리구가 높았다. 그러므로 본 연구의 결과 오징어부산물의 급여는 한우 암소의 산육 및 도체특성을 향상시키는 것으로 생각된다. This study was conducted to evaluate the effect of feeding squid by-products on the growth performance and carcass characteristics of Hanwoo cows. Sixteen Hanwoo cows, aged 7 years and weighing 484.1±63.6 ㎏, were randomly allocated to 2 feeding groups (8 animals per group): control and treatment (fed squid by-products). Both groups were fed ad libitum for 150d. Average daily gain was significantly higher in the treatment group than in the control group (p<0.05). During the experimental period, DMI and feed conversion ratios were significantly lower in the treatment group than in the control group (p<0.05). The results of yield traits, back fat thickness, rib eye area, and yield index were similar between the two groups. Carcass weights were 363.4 ㎏ and 353.1 ㎏ for treatment and control, respectively, but these differences were not statistically significant. Quality traits such as meat color, fat color, texture, and maturity were also not significantly different between the two groups. However, marbling score and rate of appearance of high quality grade (1??, 1?, and 1) were higher in the treatment group than in the control group. These results suggest that feeding squid by-products may improve the growth performance and carcass characteristics of Hanwoo cows.
