        인슐린비의존형 당뇨병 환자에서 Lipoprotein(a)농도와 혈관합병증

        연구배경: 당뇨병 환자에서 동맥경화증의 발생위험이 정상잉인에 비해 높은 원인의 하나로써 혈액응고와 섬유소용해작용의 이상을 초래하는 Lipoprotein(a) [Lp(a)]가 논의되고 있다. 당뇨병 환자의 동맥경화증과 당뇨병성 합병증 발생위험 증가에 Lp(a)가 어느 정도 관여하는지에 대해서도 아직 확실히 알려져 있지 않다. 저자등은 인슐린비의존형 당뇨병 환자의 혈관합병증(대혈관 및 미세혈관합병증) 발생과 Lp(a)농도와의 관계 및 혈관합병증을 일으키는 다른 위험인자와 Lp(a)농도와의 관련성에 대하여 알아보고자 본 연구를 시행하였다. 방법: 인슐린비의존형 당뇨병 환자 152명(여자:남자=72:80)을 대상으로 공복상태에서 혈중 Lp(a), ApoA1, ApoB, 총 콜레스테롤, 중성지방, 고밀도지단백질-콜레스테롤 등을 측정하였다. 대혈관합병증(허혈성 심질환, 뇌혈관질환, 말초혈관폐쇄성 동맥경화증)의 진단을 위해 증상이 있는 환자들에서 심전도, 관상동맥조영술, 뇌전산호단층촬영, Doppler velocimetry, 말초혈관조영술 등을 시행하였고, 미세혈관합병증은 안저활영, 신경전달속도, BBV(beat to beat variation), VPT(vibration perception threshold), 24시간 요단백량 등을 측정하여 확인하였다. 결과: 허혈성 심질환, 뇌혈관질환등 대혈관질환 및 당뇨병성 망막증 등 미세혈관질환을 갖는 환자에서 Lp(a)농도가 대조군보다 통계적으로 유의하게 증가되어 있었다. Lp(a)농도가 30㎎/dL이상인 군에서 ApoB/ApoA1 및 저밀도지단백질-콜레스테롤이 Lp(a)농도가 30㎎/dL 이하인 군보다 높았으며, 혈관질환을 갖는 군에서 Lp(a)는 높은 ApoB/ApoA1와 저밀도지단백질-콜레스테롤과 통계적으로 의의있는 양의 상관관계를 보였다. 결론: Lp(a)는 인슐린비의존형 당뇨병 환자의 대혈관합병증과 미세혈관합병증(특히 당뇨병성 망막증)에 관련이 있는 것으로 보이며, 특히 저밀도지단백질-콜레스테롤 및 ApoB/ApoA1이 높게 나타난 인슐린비의존형 당뇨병 환자에서 Lp(a)농도 측정이 필요하리라 생각된다.

      • VRSA에 대한 항생물질을 생산하는 방선균의 분리

        본 연구가 VRSA에 대한 신규 항생물질 생산 균주를 얻기 위하여 실시하였다. 국내외에 채취한 토양시료에서 분리한 방선균 중, 임상분리 VRSA와 돌연변이 유발 VRSA에 대해 항균성을 나타내는 JY-24를 선별하였다. 선발 균주는 원통형의 포자 모양을 갖고 있으며, Bennett's 배지에 배양시 aerial mycelium color는 흰색을 나타냈으며, reverse side color는 대체로 노란색을 나타내었다. 항균물질 생산을 위한 탄소원으로는 fructose, mannose, 질소원으로는 yeast extract가 가장 우수하였으며 glucose의 첨가로 항균 물질 생산성이 증가하는 것으로 나타났다. 최적 초기 pH는 pH 7로 나타났으며, 28 ℃, 37℃, 45 ℃의 온도에서 균이 성장하는 것으로 나타났다. 또한 배양기간에 따라 항균물질 생산성을 조사한 결과, 임상분리 균주에 대해 5일 배양 시 가장 높았으나, 3일 배양과 7일 배양 시 동일한 생산성을 나타내 생산속도가 빠르며, 안정한 물질인 것이 파악되었다. 향후 균주동정 및 항균물질 정제에 관한 연구를 수행하고자 한다. An antibiotic substance producing strain JY-24, effective to the VRSA (Vancomycin-resistant Staphylococcus aureus), was isolated from soil. The spore chain of JY-24 was cylindrical, and its sulface was smooth. The aerial mass color of the strain was white, and its reverse side color was yellow. The strain did not produce soluble pigment. For the production antibiotics, fructose, mannose and yeast extract were favorable as carbon and nitrogen sources. Optimal initial pH for the production of the antibiotic was pH 7. Accumulation of the antibiotic in the culture broth reached at maximal level after 5 days cultivation in Bennett's medium.

      • <i>Canavalia gladiata</i> and <i>Arctium lappa</i> extracts ameliorate dextran sulphate sodium-induced inflammatory bowel disease by enhancing immune responses

        <P><B>Abstract</B></P> <P> <I>Canavalia gladiata</I> (<I>C. gladiata</I>) and <I>Arctium lappa</I> (<I>A. lappa</I>), used in traditional medicine and food. However, their preventive effects in dextran sulphate sodium (DSS)-induced inflammatory bowel disease (IBD) remain unclear. We found that the <I>C. gladiata</I> and <I>A. lappa</I> mixture suppressed inflammation in LPS-induced RAW 264.7 cells. The mixture increased the population and activation of immune cells, up-regulation of cell cycle, and induction of IgA and IgG production in wild-type mice. Furthermore, the extract mixture prevented clinical IBD symptoms and restored IgA production in IBD mice. Similarly, a mixture of the marker compounds, chicoric acid and lupeol, ameliorated IBD-associated clinical signs, improved the population and activation of immune cells and functional defects in natural killer (NK) cells and reduced production of IgA in these mice. In conclusion, the <I>C. gladiata</I> and <I>A. lappa</I> mixture ameliorated progression of DSS-induced IBD by enhancing immune responses and recovering functional immune cell defects.</P> <P><B>Highlights</B></P> <P> <UL> <LI> <I>C. gladiata</I> and <I>A. lappa</I>, and their mixture had an anti-inflammation activity. </LI> <LI> <I>C. gladiata</I> and <I>A. lappa</I> and their mixture enhanced immune response in WT. </LI> <LI> Mixture of <I>C. gladiate</I> and <I>A. lappa</I>, can provide benefit in DSS-induced IBD models. </LI> <LI> The marker compounds can restore functional defect in DSS-induced IBD models. </LI> </UL> </P>

      • Optimization of Pasteurization Process of Beer for Avoiding Off-Flavor

        The pasteurization is employed for extending shelf-life and keeping the quality of products constant. However the pasteurization undesirably accelerates the oxidation of beer which renders volatile compounds concerning off-flavor. The pasteurization conditions was optimized to avoid off-flavor of beer during pasteurization. Under the isothermal condition, thermal destruction kinetics such as D and Z value of Lactobacillus brevis which is reported the most common beer spoilage, was determined. The binomial data (detected or non-detected of off-flavor) was treated with logistic regression to estimate off-flavor development (OFD) times. The temperature dependence of OFD times was established in terms of Arrhenius relationships. Optimized pasteurization temperature and time were found at which OFD times was not detected. The constraint for optimization was that the pasteurization degree should be larger than 5 decimal reduction time. The optimization was conducted through mathematical simulation using kinetics and temperature-dependent models for microbial death and OFD times. The optimized results were validated by the corresponding experimentations, which met the requirements that the concentration of Lactobacillus brevis was 5-Log reduction and the OFD times not detected.

        Characteristics of Language Profiles for Children with Autism Spectrum Disorder Depending on the Coexistence of Attention-Deficit Hyperactivity Disorder

        Objectives : The objective of this study was to investigate the characteristics of language profiles according to whether or not Korean children with autism spectrum disorder(ASD) also have ADHD, and to examine the relationship with executive function. Methods : Participants in the study were boys with ADHD aged 6 to 11 years who visited the clinic from January 2012 to December 2013. In this study, 25 boys with ASD were included, and completed scales included the Korean version of Autism Diagnostic Interview-Revised(K-ADI-R), Korean version of Autism Diagnostic Observation Schedule(K-ADOS), Korean ADHD Rating Scale(K-ARS), and Korean-Conners' Parent Rating Scale(K-CPRS). They also completed neuropsychological tests and assessed language profiles. Patients were categorized into two groups(with ADHD and without ADHD). T-test and Multivariate analysis of covariance (MANCOVA) was used for analysis. Results : Statistically, no difference was found in receptive and expressive language ability between the ASD groups with and without ADHD. However, a lower score in Test of Problem solving(TOPS) was observed for ASD with ADHD than for ASD without ADHD, with problem solving and finding cues showing significant differences. Conclusions : These findings suggest that language profiles in the ASD group without ADHD could be similar to those in the ASD group with ADHD, but comorbid ADHD could lead to more difficulty in linguistic ability for problem solving and could be related with executive function of the frontal lobe.

      • Antiplatelet Activity of <i>Morus alba</i> Leaves Extract, Mediated via Inhibiting Granule Secretion and Blocking the Phosphorylation of Extracellular-Signal-Regulated Kinase and Akt

        <P><I>Ethnopharmacological Relevance</I>. <I>Morus alba</I> L. leaves (MAE) have been used in fork medicine for the treatment of beriberi, edema, diabetes, hypertension, and atherosclerosis. However, underlying mechanism of MAE on cardiovascular protection remains to be elucidated. Therefore, we investigated whether MAE affect platelet aggregation and thrombosis. <I>Materials and Methods</I>. The anti-platelet activity of MAE was studied using rat platelets. The extent of anti-platelet activity of MAE was assayed in collagen-induced platelet aggregation. ATP and serotonin release was carried out. The activation of integrin <I><I>α</I></I><SUB>IIb</SUB><I><I>β</I></I><SUB>3</SUB> and phosphorylation of signaling molecules, including MAPK and Akt, were investigated with cytofluorometer and immunoblotting, respectively. The thrombus formation <I>in vivo</I> was also evaluated in arteriovenous shunt model of rats. <I>Results</I>. HPLC chromatographic analysis revealed that MAE contained rutin and isoquercetin. MAE dose-dependently inhibited collagen-induced platelet aggregation. MAE also attenuated serotonin secretion and thromboxane A<SUB>2</SUB> formation. In addition, the extract <I>in vivo</I> activity showed that MAE at 100, 200, and 400 mg/kg significantly and dose-dependently attenuated thrombus formation in rat arterio-venous shunt model by 52.3% (<I>P</I> < 0.001), 28.3% (<I>P</I> < 0.01), and 19.1% (<I>P</I> < 0.05), respectively. <I>Conclusions</I>. MAE inhibit platelet activation, TXB2 formation, serotonin secretion, aggregation, and thrombus formation. The plant extract could be considered as a candidate to anti-platelet and antithrombotic agent.</P>

      • 약지도 학습을 이용한 양파 질병 증상 자동 검출

        농업 노동력 감소 및 1인당 재배면적의 증가는 작물의 일관되고 효율적인 질병 예찰을 어렵게 하고 있으며, 이를 해결하기 위해 자동 인식 기술 등의 무인화 시스템을 이용한 기술적인 대안이 필요한 실정이다. 본 연구에서는 양파 재배 자동화를 위해 수행되었으며, 딥러닝을 이용하여 양파 재배 영상 내 질병 증상을 자동으로 검출하였다. 양파 재배지의 실시간 감시가 가능한 영상 수집 시스템을 이용하여 학습용 영상데이터를 수집하였으며, 수집된 영상은 딥러닝 모델 학습을 위한 영상크기로 샘플링 한 후 영상단위로 라벨링을 수행하였다. 양파 질병 증상 검출을 위한 기반 모델은 VGG-16을 이용하였으며, 주석작업에 필요한 노동력 절감을 위해 영상단위 라벨링으로 객체 검출이 가능한 약지도 학습 방법을 이용하였다. 딥러닝 모델의 학습결과 학습 정확도는 약 93%, 검증 정확도는 약 82%, 시험 정확도는 약 70%로 각각 관찰되었으며, 활성화 맵을 이용하여 검출된 영역을 시각화한 결과 질병 증상의 검출이 가능한 것으로 판단되며, 임계값을 이용하여 검출된 질병 영역을 이진화한 후 전체 영상에 대한 비율을 발병도로 평가할 결과 0.6 ~ 7.0% 수준으로 나타났다. 또한, 본 연구 결과를 이용하여 임계값 변화에 따른 알람 단계를 결정하는 방법을 제시하였으며, 향후 기준에 대한 선정 방법의 세분화 및 검증을 통해 양파 질병에 대한 자동 알람 정보의 제공이 가능할 것으로 판단된다.

        Antibacterial and Immuno-modulatory Activity of Ethanol Extracts from Lespedeza sp. during Helicobacter pylori Infections

        Chemical therapeutics targeted against H. pylori may lead to host toxicity and pathogen eradication failures. In this study, ethanolic extracts from five Lespedeza sp. plants were shown to inhibit the gastric-pathogen H. pylori and to modulate cytokine production. Disc agar diffusion assays showed that Lespedeza sp. ethanol extracts possess potent anti-H. pylori activity. Among the five plant extracts, the extracts from L. cyrtobotrya demonstrated the highest anti-H. pylori effect. The growth inhibitory effect against H. pylori was initiated after six h of treatment with plant extracts and the effect remained for a continuous period of 48 h. Incubation of the gastric cells infected with H. pylori with 1.25 to 50 mg/mL of Lespedeza sp. plant extracts resulted in a reduction of the production of cytokine IL-8. The plant ethanol extracts generally had little influence on AGS cell viability, indicating their safety for the treatment of bacterial infections. Three active fractions of L. cyrtobotrya also demonstrated similar anti-H. pylori and immuno-modulatory effects. Taken together, these results provide evidence that Lespedeza sp. plant extracts might be potential sources of new host friendly anti-H. pylori agents.
