        오가피(Acanthopanax sessiliflorum) 열매 추출물의 생리활성

        This study was carried out to determine the biological activity of Acanthopanax sessiliflorum fruit extracts. The phenolic compound contents of the extracts were 21.4 and 15.8 mg/g in hot water and 60% ethanol extracts. The total anti-oxidant activities of the water and the 60% ethanol extracts at a 200 μg/mL phenolic concent ration were at 92.4±0.8 and 89.2±1.1% in terms of the DPPH radical scavenging activity, 98.3±1.1 and 96.5±3.5% in terms of the ABTS radical decolorization, 2.0±0.6 and 1.2±2.8 PF in terms of the anti-oxidant protection factor, and 66.3±0.8 and 61.4±2.3% in terms of the TBARs inhibitory activity. The activities that inhibited the angiotensin-converting enzyme and xanthin oxidase were at 85.1±3.2 and 0% in the water extracts and 59.3±1.5 and 9.5±0.8% in the 60% ethanol extracts at the 200 μg/mL phenolic concentration. The tyrosinase and elastase inhibitory activities were at 56.6±1.8 and 53.1±1.1% in the water extracts and 33.7±2.2 and 22.4±3.1% in the 60% ethanol extracts. The astringent effect of the water and the 60% ethanol extracts were at 50.5±0.9 and 11.5±4.1%.

        골담초(Caragana sinica) 추출물의 생리활성 탐색

        골담초에 함유된 페놀성 물질은 70% 에탄올을 용매로 하여 18시간 추출하였을 때 가장 많이 용출되었다. 항산화 효과 중 전자공여능을 측정한 결과는 꽃과 잎 모두 70% 에탄올 추출물에서 74%로 가장 높은 항산화 활성을 나타내었으며, ABTS radical cation decolorization을 측정한 결과, 꽃의 경우 70~90% 에탄올 추출물에서 60% 이상의 항산화 활성을 나타내었다. 지용성 물질에 대한 항산화력으로 antioxidant protection factor(PF)를 측정한 결과, 70% 에탄올 추출물에서 1.2 이상의 PF를 나타내었다. 활성산소 중 지방산화를 일으키는 hydroxyl radical에 대한 골담초 추출물의 영향은 꽃의 물 추출물과 70% 에탄올 추출물에서 대조구보다 낮은 TBARS 값을 나타내었다. ACE에 대한 골담초 추출물의 저해활성은 꽃의 경우 물과 70% 에탄올 추출물 모두 80~99%의 높은 저해활성을 나타내었고, Xoase 대한 저해 활성은 꽃보다 잎 추출물의 저해활성이 높았다. 골담초 추출물의 주름개선과 미백효과를 확인한 결과 꽃과 잎 추출물 모두 페놀성 물질을 0.8 mg/mL의 농도로 처리하였을 때 50% 이상의 elastase 저해활성과 70% 에탄올 추출물에서 40.24%로 가장 높은 tyrosinase 저해활성을 나타내었고, 수렴효과는 같은 농도의 tannic acid보다 높은 수렴효과를 나타내었다. In this study, extracts from Caragana sinica flowers and leaves were tested for antioxidant and angiotensin converting enzyme inhibitory activities, along with xanthine oxidase, tyrosinase, elastase, and astringent effects. Total phenolic compounds of acetone extracts from Caragana sinica flowers and leaves were the highest at 3.42 and 2.98 mg/g, respectively, when various extraction solvents were used. Optimal conditions for extraction of phenolic compounds from Caragana sinica leaves and flowers were 70% ethanol for 18 hr. DPPH scavenging activities were the highest in 70% ethanol extracts of Caragana sinica. ABTS radical cation decolorization values of 70% ethanol extracts were higher than those 60% ethanol extracts at 74%. Antioxidant protection factor was 1.2 PF in 70% ethanol extracts from Caragana sinica flowers and leaves. TBARS was lower than that of control (0.54 μM) in all sections. Angiotensin converting enzyme inhibitory activity of Caragana sinica flower extract was 80~90% at a phenolic concentration of 0.2~1.0 mg/mL, whereas xanthin oxidase inhibitory activity of Caragana sinica leaf extract was higher than that of flower extract. Tyrosinase inhibitory activity, which is related to skin-whitening, was above 20%, whereas elastase inhibitory activity related to anti-wrinkle effect was above 50% at a phenolic concentration of 0.8 mg/mL. Astringent effects of Caragana sinica flower and leaf extracts were higher than tannic acid as a control at an equivalent concentration. This result suggests that extracts from Caragana sinica flowers and leaves are suitable as functional foods having anti-hypertension, anti-gout, and medicinal cosmetic activities, including whitening and anti-wrinkle effects.

        유근피(Ulmus pumila)로부터 분리한 페놀성물질의 미용식품활성

        Phenolic compounds of 17.9±1.0 mg/g were extracted from Ulmus pumila with 70% ethanol. The elastase inhibitory activity related with forming wrinkle was shown an excellent wrinkle improvement effect in 70% ethanol extracts as 55.5-69.5% at phenolic concentration of 50-200 μg/ml. The tyrosinase inhibitory activity related with skin-whitening was 24% in 70% ethanol extracts at phenolic concentration of 200 μg/mL. The astringent activity of 70% ethanol extracts was shown activity of 71% at phenolic concentration of 200 μg/mL therefore it is judged that there is a high effect on pores reduction of the skin. The hyaluronidase inhibitory activity of U. pumila extracts was confirmed anti-inflammation effect of 80% at phenolic concentration of 50 μg/mL. Antimicrobial activity of U. pumila water extracts was shown each 8.7, 10.0, 11.1 and 11.8 mm clear zones on Propionebacterium acnes at phenolic concentration of 50-200 μg/ mL. The stability of the multi-functional cosmetic (lotion) added U. pumila extracts was very stable for 28 days without changing of pH and viscosity also it`s stable on temperature and sun lights. As the concentration of extracts was increased, the color of lotion was getting dark, but the sensory evaluation was high at score of 8.5.

        유근피(Ulmus pumila)로부터 분리한 phenol성 물질의 건강기능식품 활성

        유근피에 함유된 페놀성 물질은 70% ethanol을 용매로 하여 12시간 추출하였을 때 17.9±1.0 mg/g으로 가장 많이 용출되었다. 유근피 추출물의 항산화력을 측정한 결과 전자공여능은 50 μg/mL 이상의 phenolic 농도에서 물추출물과 70% ethanol 추출물 모두에서 80% 이상의 높은 전자공여 능이 확인되었다. ABTS radical cation decolorization을 측정한 결과 유근피는 물과 70% ethanol 추출물에서 96.8±2.9%의 높은 항산화활성이 측정되었으며, antioxidant protection factor 측정에서는 물추출물과 에탄올 추출물 모두 200 μg/mL의 phenolic 농도에서 BHA 보다 높은 2.5 PF의 높은 항산화력이 확인되었다. 유근피 추출물의 thiobarbituric acid reactive substance를 측정한 결과 물 추출물과 에탄올 추출물 전 농도에서 약 80%의 높은 항산화 효과를 나타내었다. 유근피 추출물의 항고혈압 효과를 살펴보기 위하여 angiotensin converting enzyme 저해활성을 측정한 결과 물 추출물에서 약 50%, 70% ethanol 추출물에서 58.5%의 저해 활성을 나타내었으며, 200 μg/mL phenolics의 처리농도에서 물 추출물이 77.4%, 70% ethanol 추출물에서 90.6%의 저해활성을 나타내었다. 유근피 추출물의 항관절염 효과를 살펴보기 위하여 xanthin oxidase 저해 활성을 측정한 결과 물 추출물에서는 억제 효과를 관찰할 수 없었고, 70% ethanol 추출물에서 30%의 억제효과를 나타내었으며, 200 μg/mL phenolics의 처리 농도에서 48.1%의 억제력을 나타내었다. 유근피 추출물의 염증 억제효과를 측정하기 위하여 hyaluronidase 저해활성을 확인한 결과 물 추출물과 ethanol 추출물에서 70% 이상의 높은 저해 양상을 나타내었으며, 50 μg/mL phenolics의 낮은 농도로 처리하였을 경우에도 80%의 항염증효과를 나타내었고, 처리한 phenolic compound의 농도가 높아질수록 억제효과가 높아져 농도 의존적인 양상을 나타내었다. 기능성 식품으로의 활용을 위한 방안으로 유근피 추출물을 함유한 tablet(정제)를 개발하고, 개발된 제품의 관능평가를 실시한 결과 색깔의 관능평점이 8.3으로 평가되었고, 맛과 향은 8.8과 8.6의 높은 관능평가 점수를 획득하였으며, 전체적인 기호도 역시 8.7점으로 높은 평가를 받았다. The phenolic compounds which were extracted with 70% ethanol from Ulmus pumila for 12 hr were the highest as 17.9±1.0 mg/g. DPPH scavenging activity of 70% ethanol extracts was also the highest as 89.5±1.9% and it was confirmed to be high as 80% over in both of water and 70% ethanol extracts containing 50 μg/mL over phenolic concentration. ABTS radical cation decolorization activities of water and 70% ethanol extracts were higher as 96.8±2.9%, antioxidant protection factor (PF) was 2.0 PF in 70% ethanol and showed higher activities in both of water and 70% ethanol extracts containing 200 μg/mL phenolic concentration as 2.5 PF than BHA. TBARs of 70% ethanol extracts was 86.5±4.6%, it showed high anti-oxidative activity in 50∼200 μg/mL phenolic concentrations of water and 70% ethanol extracts as 80% over. The angiotensin converting enzyme (ACE) inhibitory activity of Ulmus pumila extracts against hypertension was 77.4% and 90.6% in water and 70% ethanol extracts of 200 μg/mL phenolic concentration. Xanthine oxidase inhibitory activity of Ulmus pumila extracts for anti-gout effect was not observed in water extracts, but it showed 30% inhibitory activity in 70% ethanol extracts, and 48.1% at 200 μg/mL phenolics concentration.

        정금나무(Vaccinium oldhami) 열매의 생리활성 및 항균활성

        정금나무 열매의 생리활성 및 항균활성을 측정한 결과, ABTS radical decolorlization은 200 μg/mL에 92.7±4.1%로 positive control인 BHA의 200 μg/mL에서 52.4±1.9%보다 높은 활성을 보였다. 지용성 물질의 항산화력을 나타내는 TBARS값은 200 μg/mL에서 74.4±2.9%, antioxidant protection factor는 200 μg/mL에서 3.6±1.6%로 항산화효과가 우수한 것으로 나타났다. 정금나무 열매 추출물의 angiotensin converting enzyme 활성억제 효과는 200 μg/mL에서 28.6±0.6%의 저해율을 나타내었으며, xanthine oxidase 저해활성을 측정한 결과 200 μg/mL에서 43.3±0.8% 이상의 활성을 나타내었다. 정금나무 열매 추출물의 항균활성을 측정한 결과 Helicobacter pylori는 200 μg/0.1 mL 농도에서 24 mm의 저해환을 보였고, Staphylococcus epidermidis는 28 mm의 저해환을, Staphylococcus aureus, Eschericia coil, Propionebacterium acnes는 각각 13 mm, 26 mm, 16mm의 저해환을 나타내어 200 μg/0.1 mL의 농도 이상에서는 모두 항균활성이 나타나는 것을 확인할 수 있었다. 주름억제 효과를 나타내는 elastase 억제효과를 측정한 결과 200μg/mL에서 52.7±0.9%로 비교적 높은 활성을 나타내었다. This study was carried out to investigate the biological activity and antimicrobial activity of Vaccinium oldhami fruit extracts. ABTS radical cation decolorization and the antioxidant protection factor (PF) of extracts as 92.7±4.1% and 3.6±1.6 PF were higher than a BHT of 200 μg/mL as 52.4±1.9% and 2.0±0.8 PF, and the TBARS of extracts was 74.4±2.9% with 200 μg/mL. The hypertension inhibitory activity of extracts from Vaccinium oldhami fruit indicated the activities of 28.6±0.6% with 200 μg/mL, and anti-gout activity was 43.3 ±0.8% with 200 μg/mL. Antimicrobial activity was found in Vaccinium oldhami fruit extracts on Helicobacter pylori, Staphylococcus epidermidis, Staphylococcus aureus, Eschericia coli and Propionebacterium acne. This activity was illustrated as 24 mm, 28 mm, 13 mm, 26 mm and 16 mm clear zones with 200 μg/mL respectively, and the elastase inhibitory activity which is related to the wrinkle cause was observed in extracts as 52.7±0.9% with 200 μg/mL.

        채진목(Amelanchier asiatica) 열매 에탄올 추출물의 생리활성

        본 연구에서는 채진목(Amelanchier asiatica)의 항산화 활성과 xanthine oxidase, angiotensin converting enzyme 그리고 elastase 저해활성 등의 생리활성 효과를 조사해 보았다. 채진목열매추출물에는 17.6 mg/g의 페놀성 물질이 함유되어 있으며, 추출물의 항산화 활성 실험에서 전자공여능은 채진목 열매 추출물을 50 μg/mL 이상으로 처리했을 때, 50% 이상의 효과를 보였으며, 200 μg/mL 이상 처리 하였을 때는 90% 이상의 전자공여능을 보였다. 또한 ABTS [2,2`-azinobis(3-ethylbenzothiazoline-6-sulfonic acid] radical cation decolorization 측정과 thiobarbituric acid reaction substance 측정에서도 각각 200 μg/mL 이상 처리하였을 때, 98%와 73%의 우수한 항산화력을 보였으며, antioxidant protection factor 측정에서는 1.03 PF의 효과를 보였다. XOase 저해활성 측정에서는 200 μg/mL 이상 처리하였을 때, 15% 이상의 저해활성을 나타냈으며, ACE 저해활성에서는 150 μg/mL에서 50% 이상, 200 μg/mL에서 82% 이상의 우수한 저해활성을 확인할 수 있었다. Elastase 저해활성에서는 50, 100, 150, 200 μg/mL 첨가 시 각각 3.9, 18.8, 32.1, 41.3%의 저해율을 보여 농도가 증가함에 따라 그 효과 또한 증가함을 알 수 있었다. 또한 대조구인 Vit C가 200 μg/mL 첨가 시 12.6%의 낮은 저해율을 보여 상대적으로 그 효과의 우수함을 알 수 있었다. 본 연구의 결과로 채진목의 항산화활성 및 통풍과 고혈압, 주름개선에 도움을 줄 수 있는 기능성식품 및 기능성 화장품의 개발 가능성이 확인되었으며, 안전한천연 기능성소재로서의 활용을 기대 할 수 있게 하였다. Amelanchier asiatica fruits have been used as a traditional medical food. This research was investigated to assess angiotensin converting enzyme, xanthine oxidase (XOase) and elastase inhibitory activity and antioxidant activities. The content of total phenolic compounds in A. asiatica fruits extracts was 17.6mg/mL. In extracts, the electron donating ability by 1,1-diphenyl-2-picrylhydrazyl free radical scavenging test of A. asiatica fruits extracts was 90.18% at 200 μg/mL. The 2,2`- azinobis(3-ethylbenzothiazoline-6-sulfonic acid radical decolorization of A. asiatica fruits extracts was 98.81% at 200 μg/mL. The inhibition rate of the antioxidant protection factor was 1.03, and thiobarbituric acid reactive substance was 73.27% at 200 μg/mL. The XOase inhibition activity of A. asiatica fruits extracts of showed to be 13.19% at 200 μg/mL. The angiotensin converting enzyme activity was significantly inhibited by A. asiatica fruits extracts as 82.52% inhibitory rate at 200 μg/ mL. Elastase inhibitory activity in the A. asiatica fruits extracts (41.48% at 200 μg/mL) was higher than vitamin C (12.8% at 200 μg/mL). These results suggests that A. asiatica fruits extracts have the greatest property as a functional food and functional cosmetic source.

        호두의 화장품 기능성

        호두과육으로부터 용매별로 추출한 추출액중의 phenolic compound 함량은 열수, 에탄올, 메탄올, 아세톤 추출물 순서로 24.3, 34.4, 32.5, 15.1 ㎎/g의 함량을 나타내어 에탄올 추출물의 phenolic compound 함량이 가장 높았으며, 인체에 무해한 추출 용매인 열수 및 에탄올 농도별 추출물의 phenolic compound 비교한 결과 60% 에탄올 추출물에서 34.7 ㎎/g과 열수 추출물에서 24.6 ㎎/g 함량의 phenolic compound를 나타내었다. 호두추출물의 항산화 활성 중 전자공여능의 경우 열수추출물에서 78.1%, 60% 에탄올추출물에서 80.6%의 활성을 나타내었으며, ABTS radical cation decolorization 측정에서는 열수추출물과 60% 에탄올추출물 각각 98.1%와 98.3%의 높은 항산화 활성을 나타내었다. Antioxidant protection factor (PF)의 경우는 열수추출물과 60% 에탄올추출물에서 각각 1.1±0.2 PF와 1.1±0.4 PF로 높지는 않았으며, TBARs inhibitory activity 측정에서는 열수추출물과 60% 에탄올추출물에서 각각 60%와 75%로 비교적 높은 항산화력을 가졌음이 확인되었다. 호두추출물의 항염증효과를 살펴본 결과 100 ㎍/㎖의 농도에서 50%의 NO발현이 억제되었고, western blot에 의한 iNOS 및 COX-2의 억제효과는 대조군에 비해 100 ㎍/㎖의 농도에서 10% 정도의 낮은 저해 효과를 나타내었다. Tyrosinase의 저해 활성의 경우 60% 에탄올추출물에서 43%의 저해율을 보였으며, 수렴효과의 경우 60% 에탄올추출물에서 55%의 억제효과를 나타내어, 호두추출물은 미백 및 주름개선용 화장품소재로서 개발이 가능하다고 판단되었다. The phenolic compounds of walnut extracts by various solvents were shown to be 24.3 ㎎/g in hot water, 34.4 ㎎/g in ethanol, 32.5 ㎎/g in methanol and 15.1 ㎎/g in acetone. In a comparison of phenolic compounds from hot water and different concentrations of ethanol, which are harmless, 60% ethanol extract and hot water extract were 34.7 ㎎/g, 24.6 ㎎/g. The electron donating ability (EDA) of walnut extracts in hot water and 60% ethanol were 78.1% and 80.6%. According to ABTS radical cation decolorization for antioxidant activity, hot water and 60% ethanol extract showed high antioxidant activities of 98.1% and 98.3%. Antioxidant protection factor (PF) were 1.1±0.2 PF and 1.1±0.4 PF in hot water and 60% ethanol extract. In TBARs inhibitory activity, each extract showed high antioxidant activities at 60% and 75%. Anti-inflammation effects of walnut extract were tested, and inhibition of NO was 50% in 100 ㎍/㎖ phenolics. Inhibitory activity against iNOS and COX-2 were shown, through Western blot, to be 10% in 100 ㎍/㎖ phenolics. Tyrosine inhibitory activity of 60% ethanol extract was 43%, and astringent effect of 60% ethanol extract was 55%. These results suggest that walnut extracts are suitable for functional cosmetics requiring skin-whitening and anti-wrinkle activity.

        자색 고구마추출물의 기능성 화장품 활성

        The optimum condition for phenolics contents extraction from purple sweet potato was 12 hrs. in 50% ethanol. The electron donating scavenging activities (DPPH), ABTS radical cation decolorization (ABTS) and antioxidant protection factor (PF) of Jami, Yeonjami and Shinjami were higher than general sweet potato, and thiobarbituric acid reaction substance (TBARs) was below 30%. The minimum inhibitory concentration (MIC) against Staphylococcus aureus and Escherichia coli on skin were each 5,000 and 2,500 ppm in all purple sweet potatoes, and MIC of Jami was the lowest as 2,500 ppm against Staphlococcus epidermidis. The whitening (tyrosinase inhibitory) activity of purple sweet potatoes was the highest as 62.5% and 48.7% in Jami water and ethanol extracts. The anti-inflammation (hyaluronidase inhibitory) activity of purple sweet potatoes was the highest as 25.3% and 94.4% in Jami water and ethanol extracts. The safety of cosmetic with Jami extracts was assessed by various safety profiles. pH and viscosity change of essence for 90 days was not detected. Cosmetic was stable to temperature and light for 90 days. The result to measure changes of skin color and pore size of the skin was that an aged skin was more effective than young skin.

        영실에 존재하는 페놀성 화합물의 추출 최적 조건을 알아보고자 추출용매를 달리하여 phenol성 물질의 함량을 비교한 결과, 70% ethanol을 용매로 하여 24시간 교반 추출하였을 때 phenol성 물질의 용출량이 13.8 ㎎/g 이상으로 가장 높았다. Helicobacter pylori에 대한 항균효과는 영실추출물을 150 μg/㎖의 농도로 첨가하였을 때 생육 저해환이 12 ㎜, 200 μg/㎖ 일 때 14 ㎜의 생육저해환이 나타나 항균활성이 우수한 것을 알 수 있었다. H. pylori에 대한 저해물질을 정제하기 위하여 영실을 70% 에탄올로 추출한 후 Sephadex LH-20 column과 C₁? cartridge column을 이용하여 항균활성을 가지는 물질을 순수 정제하고 구조를 동정한 결과 protocatechuic acid, chlorogenic acid 및 quercetin으로 동정하였으며, 단일물질에 의한 H. pylori에 대한 저해효과 보다 혼합물질에 의한 synergy 효과에 의해 저해 효과가 높아지는 것을 알 수 있었다. The antimicrobial activity of 70% ethanol extracts from Rosa multiflora Thunberg fruit against Helicobacter pylori was examined. The inhibitory activity of Rosa multiflora Thunberg fruit extracts against H. pylori was determined to clear a zone of 14 mm with 70% ethanol extracts. Purification of inhibitory compounds was carried on Sephadex LH-20 and C₁? cartridge column chromatography using a gradient procedure, with increasing ethanol (0→100%) in H₂O. The chemical structure of the purified inhibitory compounds on H. pylori was identified to be protocatechuic acid, chlorogenic acid and quercetin by FAB-MS, NMR and IR spectrum.

        Rosmarinus officinalis extracts had a significant antimicrobial activity against Helicobacter pyori. Total phenolic contents and inhibition zone of rosemary extracts were estimated to be 25.7 mg/g and 14 mm at 200 μg/mL of phenolic contents, respectively. The Sephadex LH-20 and MCI-gel CHP-20 column chromatographic separations for the phenolic extracts from R. officinalis leaves led to isolation of five acids, whose structures were determined as protocatechuic acid (A), coumaric acid (B), caffeic acid (C), chlorogenic acid (D), and rosmarinic acid (E), from interpretation of spectroscopic data including nagative fast atom bombardment (FAB)-mass, 1H-NMR, 13C-NMR, and IR. All isolated compounds were tested for antimicrobial activity against H. pyori. The purified single compound showed less antimicrobial activity against H. pylori than the mixed purified compounds, which generate A+B, A+E, C+D, C+E (each 200 μg/disc) excellent as large clear zone by synergy effect. These results indicate rosemary extracts are preventive agents against H. pyori.
