        주암호 외남천 유역 하천수의 질소농도와 동위원소비 분석을 이용한 오염원 평가

        In an effort to investigate water pollution characteristics of Juam lake, water samples were collected from three sites (Sites A, B, and C) of Oenam stream which is a typical tributary of rural watershed in the lake and analyzed for N concentration and the corresponding isotope ratio (δ15N) of NO3-. Concentrations of NO3- were not dramatically different among the sites; 0.8±0.2 mg N L-1 (range: 0.0~4.3 mg N L-1) for Site A, 1.1±0.2 mg N L-1 (0.0~4.3 mg N L-1) for Site B, and 1.1±0.1 mg N L-1 (0.1~2.6 mg N L-1) for Site C. Meanwhile, δ15N tended to decrease with river flow; it was highest for Site A (45.5±5.3‰) followed by Site B (19.7±2.0‰) and Site C (8.7±1.5‰). Such high δ15N values of NO3- in Site A suggested that NO3- derived from livestock feedlot (specifically livestock excrete of which δ15N is higher than 10‰) is the predominant pollution sources despite mountainous area occupied the most of land-use in the watershed. Using the two-sources isotope mixing model, it was estimated that the contribution of cropping activities (i.e. fertilization) became greater in down-stream area (Sites B and C) due to the higher agricultural land-use than the up-stream area (Site A). Particularly, during the active cropping season, the low contribution of organic pollution sources indicated that domestic sewage was not the predominant pollution source. Therefore, it was suggested that agricultural sources such as livestock farming and cropping rather than mountainous and residential are the dominant sources of water pollution in the study area. These results could be effectively utilized in elucidating water pollution sources in rural areas and selecting water management practices.

        몬테카를로 시뮬레이션을 이용한 일식메뉴 중 비가열 처리 식품의 정량적 위해평가

        본 연구는 식중독을 일으킬 수 있는 황색포도상구균(Staphylococcus aureus)의 검출을 이용하여 일식당의 식자재, 조리공정, 조리 기구, 조리 종사자의 오염수준을 예측하고 오염에 영향을 미치는 인자를 확인하고자 하였다. 시료는 서울시에 위치한 일식당에서 초밥과 생선회를 수거하였다. 몬테카를로 시뮬레이션을 통해 초기 오염수준으로부터 각 공정별 위해 인자를 고려하여 최종오염수준을 예측한 결과, 새우 및 문어초밥의 경우, 모든 초밥이 저장기간 동안 S. aureus가 증식되는 것으로 나타났으며 특히, 문어초밥이 가장 높은 오염도를 보였다. 이는 생선회가 온도변화에 따른 균의 증식이 활발하고, 일식당에서 초밥을 섭취하기 직전에 초밥 중 생선회의 급속한 온도 변화가 나타나기 때문으로 판단된다. 초밥의 민감도 분석 결과, 완성된 초밥의 섭취 시 온도의 민감도 값이 0.419로 최종오염수준을 증가시키는 것으로 예측되었다. 두 종류의 초밥에서 동일하게 보관 시간과 섭취 시의 온도가 최종 오염수준에 가장 크게 영향을 주는 것으로 나타났다. 이는 일정 시간이 지난 후 초밥을 섭취하는 것이 균의 농도 증가에 가장 크게 영향을 미친다는 것으로 초밥을 조리 후 즉시 섭취하는 것이 균의 증식을 크게 예방할 수 있음을 알 수 있다. 연어, 도미, 방어회의 경우 섭취 시간에서 민감도 값이 0.71로 가장 큰 민감도를 나타내었고, 섭취 시 온도 또한 민감도가 크게 나타났다. 초밥의 시나리오 분석 결과, 실온 보관 시, 섭취시간이 2시간 이내일 때 식중독을 유발하지 않는 안전한 수준임을 알 수 있었다. The aim of this study was to control the outbreak of food pathogen through quantitative microbial risk assessment (QMRA). We used Monte Calro Simulation (MCS) to predict contamination levels of Staphylococcus aureus on the raw materials, equipments and cook in Japanese restaurant located in Seoul. The result of sensitivity analysis showed that the most significant factor for the outbreak of food pathogen was consumption temperature and storage time. In shrimp and octopus sushi, `consumption temperature` was the highest sensitivity value of 0.419 followed by `storage time` of 0.374. To increase safety of sushi, consumers should have sushi as soon as possible after cooking. In sushi `storage time after cooking` was determined as Critical Control Point (CCP). To determine Control Limit (CL), Scenario Analysis (SA) was carried out. In sushi, SA was carried out using storage time as a unit condition. Safety level of S. aureus was set lower than 5 log CFU/g. After 2 hr `storage time` the number of S. aureus increased to 3.908 log CFU/g. Therefore, `storage time` of sushi was set as CL in case of room temperature storage.

        Background: The 2009 Influenza A virus, H1N1 subtype, was identified as the cause of outbreaks of febrile respiratory infection. The World Health Organization and Centers for Disease Control and Prevention considered addressing pandemic H1N1 vaccination in an effort to control future outbreaks. The safety of the H1N1 influenza vaccine is generally ascertained. However, adverse events may occur in a few patients, especially patients with underlying diseases. Spontaneous pneumomediastinum caused by asthma exacerbation after H1N1 vaccination has not yet been reported. Case History: A-16-years-old male presented with cough, sputum and dyspnea after H1N1 influenza vaccination. Results: Chest X-ray and chest CT showed pneumomediastinum and subcutaneous emphysema. The patient was treated with oxygen, inhalation of a bronchodilator and intravenous aminophylline. Eight days after admission, there was no further evidence of pneumomediastinum. Conclusion: We report a case of spontaneous pneumomediastinum caused by asthma exacerbation after H1N1 Influenza vaccination in stable asthma patient.       • 저탄소 도시관리를 위한 온실가스 평가지표 개발 및 활용방안

        Seoul City needs to establish the foundation for more active low-carbon urban management in consideration of local characteristics in order to contribute to low-carbon green growth of the government and participate in international efforts to respond to climate change. In general, the items related to greenhouse gases in urban management plan includes only the calculation of yearly energy consumption and yearly CO2 emission per unit building and preparation of reduction plans, but does not include specific indices for low-carbon soil utilization. Especially, the items related to greenhouse gases are examined only for the building part and little has been considered for such carbon absorption sources as vegetation and soil that can bring in the indirect effect of emission reduction. The objective of this research is to analyze greenhouse gas emission of the building part in business districts on the basis of the urban regeneration project for accomplishment of low-carbon urban management, analyze the reduction by such carbon absorption sources as vegetation and soil, and then present a plan to apply a quantified target value related to greenhouse gases to low-carbon urban management in the future. The major contents of this research are as follows: First, the status of low-carbon urban management related to greenhouse gases was analyzed. Policies and standards practiced in relation to greenhouse gases were investigated in the position of the central government and Seoul City. At the same time, the cases for management and evaluation of greenhouse gases were examined in an effort to apply effectively the quantified target value developed in this research to low-carbon policies of Seoul City. Second, the current status of greenhouse gas emission was analyzed. The current status for urban management project of Seoul City was analyzed and the urban regeneration project was targeted for this research. The target sites of the urban regeneration project were classified into the residential environment improvement project and the urban environment improvement project for selection of specific target sites for analysis and estimation of greenhouse gas emission. For selection of target sites, the environmental impact evaluation project of Seoul City which can easily provide current data of business target sites was analyzed. Out of rearranged business target sites, some target sites whose basic data related to greenhouse gas emission analysis of the building part and vegetation/soil part required in this research was not sufficient were excluded and finally 10 target sites from each of the residential environment improvement project and the urban environment improvement project were selected. Greenhouse gas emission was analyzed after classification into building and vegetation/soil parts. For greenhouse gas absorption of vegetation and soil, existing formulae were reviewed and the single-tree formula used most generally in consideration of characteristic of the downtown area was selected, and for the building part the greenhouse gas emission was estimated from building emission sources fixed before and after business execution, Third, The results of each part were combined for evaluation, and an integrated analysis was made on greenhouse gas change classified by business target sites. For 10 target sites of the residential environment improvement project, combination of the storage and absorption by vegetation/soil and the emission from buildings indicates that, on the average, the carbon change per unit area was 70.48tonC/yearㆍha and the rate of carbon change 26.65%. The target sites from the residential environment improvement project were, on the whole, general residential areas of more than 2 or 3 kinds and some include semi-industrial areas or semi-residential areas, but the floor-area-to-land ratio was around 250% and the greenhouse gas change before and after business execution was relatively small with a maximum of 70.23% and a minimum of 0.87%. The absorption and storage of greenhouse gases by vegetation ad soil was found to be equivalent to 5 - 10 % of the emission of greenhouse gases from buildings. Thus, it is judged that active securing of vegetation and soil in the business districts can contribute to some degree to the reduction of greenhouse gases. In case of 10 target sites from the urban environment improvement project, on the average, the carbon change per unit area was 2,588.71tonC/year·ha and the rate of carbon change 1,168.62%, which indicate that the greenhouse gas emission increased a great deal compared with that of the target sites from the residential environment improvement project. The rate of carbon change for a considerable number of target sites was around 500%, and some target sites such as O district and L district showed the rate of carbon change was greater than 4,000%, which indicates that the variation between districts is seriously large, N district (2,098.58% increase) and R district (5,592.00% increase) where the rate of carbon change was severe are the cases where the carbon emission prior to business execution was remarkably low compared with other target sites and the rate of change increased. Comparison of real total carbon emission of these areas after business execution with that of other target sites shows a lower emission of the areas than the average emission per unit area, Thus, since the difference between these target sites and the other ones prior to business execution was large, an exceptional application is judged to be needed in case the target value of carbon emisson is developed in the future. In case of the urban environment improvement project, the storage and absorption of greenhouse gases by vegetation and soil were analyzed to reduce about 1% of the emission from buildings and so the reduction of greenhouse gases by absorption does not appear to contribute a great deal. Fourth, a target value related to greenhouse gas emission was selected and a specific plan to use it was presented. A specific target value was selected by combining integrated analytical results of building and vegetation/soil parts. For specific application of the developed target value (index), an environmental review from the urban management planning of Seoul City and items related to greenhouse gases from the environmental impact evaluation of Seoul City were considered to understand the current status, and evaluation standards and guidelines were presented to review relevant items in the future.

        항종양 면역반응 유도를 위한 수지상세포의 최적 활성화 조건

        수지상세포는 종양면역에서 필수적인 강력한 CTL 반응을 개시할 수 있는 유일한 세포이다 . 특히 외인성 종양항원에 대한 CTL 반응 유도는 활성화된 수지상세포의 IL-12 분비를 통한 CD4+ helper T세포의 cross-priming을 필요로 한다. 그러나 최근에 활성화된 수지상세포는 $Th_1$ 면역반응을 유도하지만 활성화 시간이 경과함에 따라 오히려 $Th_2$반응을 유도 할 수 있다. 따라서 본 연구에서는 OVA를 종양항원 모델로 설정하여 종양특이적인 CTL 반응을 형성하기 위한 최적의 수지상세포 활성화 조건을 조사하였다. 마우스 골수세포에 서 수지상세포로의 분화는 항원제시 기능을 위한 표면분자의 발현 측면에서 볼 때 배양 6일-7일 정도가 적합하였다. 수지상세포의 IL-12 생성능은 배양 6일 이상, OVA 항원 탑재 8시간 이상의 경우에 연이은 LPS 성숙자극으로 오히려 감소하는 경향을 보였다. 즉 배양 6일의 수지상세포에 OVA 항원 탑재를 8시간 수행한 경우(8-h DC)가 in vitro에서의 IL-12생성능, ex vivo에서의 세포내 $IFN-{\gamma}$를 발현하는 CD8+ T세포의 증가 및 OVA 특이적인 세포독성효과 등에서 가장 좋은 결과를 보였다. 또한 in vivo에서 종양 치료 및 예방효과에서도 8-h DC로 면역한 경우에 가장 우수한 종양형성 억제 효과와 생존기간 연장효과를 보였다. 현재 대부분의 수지상 세포를 이용한 항종양 백신에서 항원 탑재반응을 24시간 동안 수행하고 있으나, 본 실험의 결과로 볼 때, 8시간의 in vitro 항원 탑재가 보다 효과적인 종양특이적 CTL 반응과 항종양 면역반응을 유도함을 알 수 있다. 결론적으로 본 연구를 통하여 8시간 이상의 항원접촉은 수지상세포의 기능적 활성능력을 오히려 고갈시킬 수 있음을 제시한다. Dendritic cells (DCs) are the only antigen presenting cells (APCs) capable of initiating immune responses, which is crucial for priming the specific cytotoxic T lymphocyte (CTL) response and tumor immunity. Upon activation by DCs, CD4+ helper T cells can cross-prime CD8+ CTLs via IL-12. However, recently activated DCs were described to prime in vitro strong T helper cell type 1 $(Th_1)$ responses, whereas at later time points, they preferentially prime $Th_2$ cells. Therfore, we examined in this study the optimum kinetic state of DCs activation impacted on in vivo priming of tumor-specific CTLs by using ovalbumin (OVA) tumor antigen model. Bone-marrow-derived DCs showed an appropriate expression of surface MHC and costimulatory molecules after 6 or 7-day differentiation. The 6-day differentiated DCs pulsed with OVA antigen for 8 h (8-h DC) and followed by restimulation with LPS for 24 h maintained high interleukin (IL)-12 production potential, accompanying the decreased level in their secretion by delayed re-exposure time to LPS. Furthermore, immunization with 8-h DC induced higher intracellular $interferon(IFN)-{\gamma}+/CD8+T$ cells and elicited more powerful cytotoxicity of splenocytes to EG7 cells, a clone of EL4 cells transfected with an OVA cDNA, than immunization with 24-h DC. In the animal study for the evaluation of therapeutic or protective antitumor immunity, immunization with 8-h DC induced an effective antitumor immunity against tumor of EG7 cells and completely protected mice from tumor formation and prolonged survival, respectively. The most commonly used and clinically applied DC-based vaccine is based on in vitro antigen loading for 24 h. However, our data indicated that antigen stimulation over 8 h decreased antitumor immunity with functional exhaustion of DCs, and that the 8-h DC would be an optimum activation state impacted on in vivo priming of tumor-specific CTLs and subsequently lead to induction of strong antitumor immunity.

        더덕, 도라지 표고균사발효물의 유용성분 및 생리활성

        한약재로 사용되는 더덕과 도라지에 표고의 유용성분을 함유한 소재를 개발하고자 더덕표고균사발효물(FCLM)과 더덕도라지균사발효물(FPLM)을 제조하였다. 두 가지 발효물의 일반성분, 유리당, 유기산, 𝛽-glucan, ergothioneine, ergosterol, vitamin D<sub>2</sub> 함량 및 3T3-L1 지방전구세포의 분화에 미치는 영향을 연구하였다. 일반성분 분석결과, FCLM에서 조섬유와 조지방함량이 FPLM에 비하여 높게 나타났으며, FPLM은 조단백질과 가용성무질소물의 함량이 FCLM에 비하여 높게 나타났다. 유리당은 FCLM과 FPLM 모두 arabinose, glucose, sucrose 3종이 검출되었으며, 총 유리당 함량은 FPLM에서 높게 나타났다. 유기산 함량은 발효 전 더덕과 도라지에 비하여 발효 후 FCLM과 FPLM에서 낮게 나타났다. 𝛽-Glucan 함량은 FCLM과 FPLM 두 발효물에서 대조구로 사용한 표고보다 높은 함량을 보였다. 항산화 물질로 알려진 ergothioneine의 함량은 FCLM이 FPLM보다 높게 나타났다. Ergosterol 함량은 대조구로 사용한 표고에서 가장 높게 나타났으며, 발효물들은 비슷한 함량을 나타내었다. Vitamin D<sub>2</sub>는 발효물들에서만 검출되었으며, FPLM (0.58±0.01 mg%)가 FCLM (0.47±0.01 mg%)보다 높은 함량을 보였다. 더덕과 도라지 표고균사발효물인 FCLM과 FPLM는 3T3-L1 지방전구세포에 대하여 발효 전 더덕과 도라지에 비하여 높은 수준의 세포 생존율을 나타내었다. 또한 FCLM과 FPLM은 3T3-L1 지방전구세포 분화를 발효 전 더덕과 도라지에 비하여 높은 수준으로 억제하여, 비만 억제 효과가 있는 것으로 기대된다. This study was conducted to develop food and medicinal products containing useful components of Lentinula edodes in Codonopsis lanceolata and Platycodon grandiflorus for use as herbal medicine. We manufactured C. lanceolata (FCLM) and P. grandiflorus (FPLM) extract fermented with L. edodes mycelium. The effect of the two fermented products on proximate composition, free sugar, organic acid, 𝛽-glucan, ergothioneine, ergosterol, and vitamin D<sub>2</sub> levels, and 3T3-L1 preadipocyte cell growth were studied. The proximate composition analysis results showed that the crude fiber and crude fat content in FCLM was higher than that in FPLM, and the crude protein and soluble nitrogen content in FPLM was higher than that in FCLM. Free sugar analysis detected arabinose, glucose, and sucrose in both FCLM and FPLM, and the total free sugar content was high in FPLM. The organic acid content was lower in FCLM and FPLM compared to C. lanceolata and P. grandiflorus before fermentation. The 𝛽-glucan content was higher than that of L. edodes used as a control in both fermented products, FCLM and FPLM. The content of ergothioneine, an antioxidant, was higher in FCLM than in FPLM. Ergosterol content was highest in L. edodes which was used as a control, and the two fermented products showed similar content. Vitamin D<sub>2</sub> was detected only in FCLM and FPLM, and FPLM (0.58±0.01 mg%) showed a higher vitamin D<sub>2</sub> content than FCLM (0.47±0.01). FCLM and FPLM showed a higher level of cell viability for 3T3-L1 pre-adipocytes compared to non-fermented C. lanceolata and P. grandiflorus. In addition, FCLM and FPLM inhibited 3T3-L1 preadipocyte differentiation more than C. lanceolata and P. grandiflorus before fermentation, which may exert an anti-obesity effect.
