        COVID-19 상황에서 직무불안정성, 직무스트레스가 이직의도에 미치는 영향: 체육시설 종사자의 조직지원인식 조절효과를 중심으로

        Purpose: This study is to investigate the effects of job instability and job stress on the turnover intention of sports facility workers in the COVID-19 situation, considering the moderating effects of perceived organizational support. Methods: This study surveyed service workers working at public sports facilities in Seoul, Gyeonggi Province and North Chungcheong Province, South Korea. The multi-regression and hierarchical regression analysis were employed to analyze the collected data. Results: The results indicate that all of the factors constituting job instability (organizational, job, and personal factors) had a significant influence on the factors constituting job stress (job characteristic, roll-related, and human relationship factors), except for only the relationship between organizational factor and rol-related factor. Job characteristic and roll-related factors constituting job stress significantly affected turnover intention of sports facility workers. Also, it was found that the perceived organizational support moderated the relationship between job characteristic factors of job stress and turnover intention. Conclusion: Although the COVID-19 created an environment with very high job instability, the negative impacts of job instability and job stress can be minimized through consideration and support for service workers of the organization, reducing turnover intention.

        에너지전환을 위한 에너지협동조합 활성화 방안

        본 연구의 목적은 태양광발전 확대를 통한 에너지전환을 위해 시민참여형 협동조합의 실태를 분석하여 에너지협동조합의 활성화 방안을 제시하는 것이다. 문재인 정부의 ‘재생에너지 3020’ 계획 발표와 더불어 지방자치단체와 민간영역에서 주로 진행되던 태양광발전 등 에너지전환의 움직임이 한국사회에서도 활발하게 진행되고 있다. 재생에너지는 다양한 시민들의 참여 속에 소규모, 지역분산형으로 추진되어야 한다는 특징이 있다. 이러한 방식에 가장 적합한 형태 중의 하나가 시민참여형 에너지협동조합이다. 본 연구는 에너지협동조합 임직원 및 에너지협동조합에 대한 연구경험이 있는 전문가 등을 대상으로 심층인터뷰를 진행하고, 태양과바람에너지협동조합에 대한 사례분석도 병행하였다. 연구 결과 에너지협동조합의 자립을 위해 사업의 다각화와 규모의 경제 달성, 발전소 설치에 필요한 재원 충당 방법의 다양화 등이 제기되었다. 그리고 가장 시급한 제도개선 방안으로 협동조합이 어려움을 겪고 있는 부지확보 문제 해결을 위해 주차장 등에 대한 가중치 상향 조정이 제시되었다. 에너지시민성을 높이고 시민참여 활성화를 위해 다양한 조합원 참여프로그램 개발, 지역 거점 공간마련, 출자금에 대한 배당 실시 등이 제시되었다. The use of solar power has been gaining momentum and becoming more mainstream in Korea, with the recent Korean government’s Renewable Energy 2030. Renewable energy tends to operate in small scale with decentralization, which requires active citizen participation. One of the promising ways to achieve this is through Citizens’ Energy Co-operative. This study explores the possible achievements and limits of the Citizens’ Energy Cooperatives, and suggests ways to stimulate and increase the transition into using renewable energy source. In the following sections of the paper, we examine analyzed different cases of energy cooperative, conducted in-depth interviews with the members of the energy cooperatives and energy experts. Findings from this paper illustrates that diversification of business models and economics of scale, diversification of fund-raising structure for the installation of solar power generations are important for sustainability of the energy cooperatives. In order to improve “energy citizenship”, it is important to develop citizen participation programs, create local energy hub and having a contribution-based allocation system among the co-operative members.

        토마토 작물의 TYLCV 저항성 평가에 이용할 수 있는 감염성 클론 개발

        국내에서 채집한 5종의 토마토황화잎말림바이러스(Tomato yellow leaf curl virus, TYLCV) 분리주들에 대하여 PCR법을 이용하여 염기서열을 결정하였다. 전체 유전체의 1.9배 복제수를 가지는 감염성 클론을 제작하기 위하여, PCR로 증폭된 TYLCV DNA를 식물형질전환용 벡터에 각각 도입시켰다. TYLCV 저항성 평가를 위하여 저항성 토마토 시판 품종과 육성 계통을 각각 TYLCV 분리주를 가지고 있는 Agrobacterium tumefaciens로 접종하였다. 감수성 품종인 ‘Super-sunroad’ 품종은 접종 30일 후에 상엽에 황화, 위축 증상을 나타낸 반면, TYLCV에 대한 저항성유전자들로 알려진 Ty-1 또는 Ty-3 유전자를 포함하고 있는 TY12, GC9, GC171, GC173 등의 토마토 육성 계통은 전신 잎에 TYLCV 병징 발현이 없었으므로 이들 계통은 TYLCV 저항성으로 판별되었다. Agroinfiltration을 통한 접종 30일에 감수성 품종과 저항성 품종 간에 TYLCV 병징 차이가 뚜렷하였다. 특이적 프라이머들을 이용한 실시간 PCR법에 의해 TY12, GC9, GC171, GC173 육성 계통에서 TYLCV DNA가 검출되었지만, 저항성 계통에서 TYLCV DNA 축적 수준은 감수성 토마토 품종에서의 TYLCV DNA 축적 수준보다 매우 낮았다. 유사한 병징 세기 및 TYLCV DNA 축적수준이 담배가루이를 통하여 감수성과 저항성 토마토 품종 및 육성 계통에 TYLCV를 감염시켰을 때에도 관찰되었다. Agroinfiltration 시 아그로박테리움의 농도는 토마토 품종의 TYLCV 저항성 반응에 영향을 미치지 않았다. 따라서 본 연구결과들은 agroinfiltration을 통한 TYLCV 접종이 담배가루이를 통한 TYLCV 감염처럼 효과적이며 TYLCV 저항성 토마토 육성 프로그램에 활용 가능하다는 것으로 보여주었다. Five isolates of Tomato yellow leaf curl virus (TYLCV) collected from various regions of Korea were amplified using PCR and determined the sequences of full-length genome, respectively. The PCR-amplified DNA of each TYLCV isolate was introduced into a binary vector to construct infectious clone containing 1.9 copies of the corresponding viral genome. Various cultivars and breeding lines of tomato were inoculated with Agrobacterium tumefaciens harboring infectious clone of each TYLCV isolate to assess resistance against TYLCV. Susceptible cultivar ‘Super-sunread’ revealed typical yellowing and narrowing of the upper leaves. In contrast, breeding linesTY12, GC9, GC171, and GC173, which contained the TY-1 and/or TY-3 genes that confer resistance against TYLCV in nature, were completely symptomless, suggesting that the lines were resistant to challenging TYLCV isolates. Symptoms of TYLCV in susceptible tomato cultivars are significantly different from those of TYLCV in the resistant tomato cultivars at 30 days after agroinfiltration. Although genomic DNAs of TYLCV were detected from the breeding lines TY12, GC9, GC171, and GC173 using real-time PCR analysis with specific primers, levels of TYLCV DNA accumulation in the resistant breeding lines were much lower than those of TYLCV DNA accumulation in susceptible tomato cultivars. Similar symptom severity and levels of TYLCV DNA accumulation were observed from TYLCV infections mediated by Bemisia tabaci in the resistant and susceptible tomato cultivars. Concentration of agrobacterium did not affect the response of tomato cultivars against TYLCV inoculation. Taken together, these results suggest that TYLCV inoculation via agroinfiltration is as effective as inoculation through Bemisia tabaci and is useful for breeding programs of TYLCV-resistant tomato.

        야생담배에서의 동질 머리핀 RNA의 일시 발현에 의한 오이모자이크바이러스 감염 저해

        Cucumber mosaic virus (CMV), genus Cucumovirus, family Bromoviridae causes damage in many economically important horticultural and ornamental crops. Sequence alignments showed several conserved sequences sites in 3′-noncoding region (NCR) of CMV genomic RNAs in subgroup IA strains characterized so far. Based on this observation, we generated a hpRNA construct (pIRCMVNCR) harboring an inverted repeat containing a 162 bp cDNA fragment homologous to 3` NCR portion of CMV RNA3 to investigate the silencing potential for its ability to interfere with a rapidly replicating CMV. Agrobacterium-mediated transient expression of the pIR-CMVNCR had a detrimental effect on CMV infection, showing no distinct symptoms in non-inoculated leaves of the agroinfiltrated N. benthamiana plants. CMV genomic RNAs including subgenomic RNA4 were not detected by RT-PCR analysis from tissues of both the inoculated leaves and upper leaves of the agroinfiltrated plants. Accumulation of virus-derived small interfering RNAs was detected in the inoculated leaf tissues of N. benthamiana plants elicited by transient expression of pIR-CMVNCR indicating RNA silencing is responsible for the resistance to CMV.

        다수성 버터헤드 상추 ‘썬레드버터’ 육성

        A new red butterhead lettuce (Lactuca sativa L.) with transverse broad elliptic, red leaves, ‘Sun Red Butter’, with late bolting and high yield, was developed from a cross between ‘Jajusangchu’ (red leaf color and half crisp) and ‘Dukseomjeokchukmyeon’ (high yield). The crossing and selection for advanced lines was previously carried out using the pedigree method during the period 2001-2009. The advanced lines were evaluated for yield and adaptability at several locations in Korea (Gangwon-do, Gyeonggi-do, Chungcheongbuk-do, Jeollabuk-do, Gyeongsangnam-do, and Jeju-do) in 2011. The type of matured stage is early and medium butterhead lettuce. The shelf life of ‘Sun Red Butter’ was four weeks longer than that of ‘Germania’ at 4°C. The anthocyanin content of ‘Sun Red Butter’ was lower than that of ‘Germania’ at 11.2 mg/100 g. The BSL (latucin+8-deoxylactucin+lactucopicrin) content of ‘Sun Red Butter’ was higher than that of ‘Germania’ at 32.8 μg/g, DW. The marketable yield of ‘Sun Red Butter’ was higher than that of ‘Germania’ by 67% (at 621.4 g per plant), and ‘Sun Red Butter’ showed a particularly improved yield under high-temperature cultivation in the field. Furthermore, ‘Sun Red Butter’ had better taste characteristics and was crisper and sweeter than ‘Germania’. Therefore, we recommend that the new cultivar ‘Sun Red Butter’ would be suitable for cultivation in spring and fall. Furthermore, this new cultivar could be harvested in a single harvest, as in a head lettuce, and could therefore be managed more efficiently and economically by farmers.

      • 비대칭적인 소속 함수를 갖는 퍼지 교통 제어기

        Nowadays the traffic conditions have been getting worse due to continuous increase in the number of vehicles. So it has become more important to manage traffic signal lights efficiently. Recently fuzzy logic is introduced to control the cycle time of traffic lights adaptively. Conventional fuzzy logic controller adjusts the extension time of current green phase by using the fuzzy input variables such as the number of entering vehicles at the green light and the number of waiting vehicle during the red light. However this scheme is inadequate for an intersection with variable traffic densities. In this paper, a new FLC with asymmetric membership functions that reflects more exactly traffic flows than other FLCs with symmetric ones regardless of few control rules is propsed. The effectiveness of the proposed method was shown through simulation of a single intersection. The experimental results yielded the superior performance of the proposed FLC in terms of the average delay time, the number of passed vehicles, and the degree of saturation.

        순무모자이크바이러스에 대한 무 육종 계통 저항성 평가

        무(Raphanus sativus L.) 육종 계통들에 대한 순무모자이크 바이러스(Turnip mosaic virus, TuMV) 저항성을 평가하기 위하여, 20개 무 육종계통들의 잎에 순무모자이크바이러스 BR 병원성을 가지는 국내 분리 계통을 즙액 접종하였다. 순무모자이크바이러스를 접종한 무 개체들은 $22^{\circ}C{\pm}3^{\circ}C$에서 재배하였으며 4주 동안 바이러스 병징 발현을 육안으로 조사하여 저항성을 평가하였다. 순무모자이크바이러스 감염에 의해서 무 육종계통들의 다른 병징 발현에 의해 분석한 결과, 16개 무 계통은 약한 모자이크, 모틀링에서 심한 모자이크 전신 병징을 보였으며 감수성으로 판별되었다. 이러한 결과는 순무모자이크바이러스 외피단백질 유전자에 대한 특이적 역전사중합효소연쇄반응에 의하여 확인되어, 순무모자이크바이러스가 병징을 발현시킨 원인이었다. 이와는 다르게 4개 무 육종계통들에서는 모자이크 등 전신 감염 병징이 발현되지 않았으며, 동일한 무 육종 계통들의 개체들에서 8주 동안 병징이 관찰되지 않았다. 역전사중합효소연쇄반응으로 조사한 결과 4개 무 육종 계통들의 상엽들에서 순무모자이크바이러스가 검출되지 않았다. 이런 결과는 4개 무 육종계통들이 순무모자이크바이러스에 대한 강한 저항성을 가지고 있음을 예시해준다. Leaves of twenties radish (Raphanus sativus L.) inbred lines were mechanically inoculated with Turnip mosaic virus (TuMV) strain HY to evaluate TuMV resistance of the radish inbred lines. The inoculated radish plants were incubated at $22^{\circ}C{\pm}3^{\circ}C$ and resistance assessment was examined using symptom development for 4 weeks. Based on the reactions of differential radish inbred lines, 16 radish lines were produced mild mosaic, mottling, mosaic and severe mosaic symptoms by TuMV infection. These results were confirmed by RT-PCR analysis of TuMV coat protein gene, suggesting that TuMV is responsible for the disease symptoms. Four resistant radish lines did not induce systemic mosaic symptoms on upper leaves and chlorosis in stem tissues for 4 weeks, showing they were symptomless by 8 weeks. Further examination of TuMV infection in the 4 radish lines showed no TuMV infection in all systemic leaves. These results suggest that the 4 radish lines are highly resistant to TuMV.

        토마토덤불위축바이러스 현장진단용 급속 면역 금 나노입자 막대 키트 개발

        A rapid, user-friendly and simple immune-chromatographic dipstick kit named ‘rapid immuno-gold strip’ (RIGS) kit was developed in a novel single strip format to detect on-site detection of Tomato bushy stunt virus (TBSV). Immunoglobulin G (IgG) from polyclonal antisera raised in rabbits against TBSV was purified through protein-A affinity chromatography and then the purified TBSV-IgG was conjugated to colloidal gold nano-particles which served as a test line on nitrocellulose membrane. Protein A that non-specifically binds to TBSV antibody was used as a control line on the same strip. The diagnosis process with the RIGS-TBSV involves simply grinding the suspect plant sample in a bag that contains the extraction buffer and inserting the strip the bag. Results can be seen in 5-10 minutes. The flow of the complexes of gold particles coated with TBSV-IgG and a crude sap from TBSV infected tomato plants resulted in intensive color formed on the test lines proportional to the concentrations of TBSV. The RIGS-TBSV kit did not show any cross-reactions against Tomato spotted wilt virus and Tomato yellow leaf curl virus that are major causal viruses. These results indicate that the RIGS-TBSV kit is highly sensitive and is not required for laboratory training and experience prior to testing. The RIGS-TBSV kit is suitable for on-site determination of suspect TBSV-infected plants as well as for laboratory diagnosis.

        연구보문 : 자연과학 ; 오이모자이크바이러스 현장진단용 급속면역금나노입자 막대 키트 개발

        ‘급속면역금나노입자막대 (RIGS) 키트’라 명명된 빠르며 사용자 편의 및 간단한 면역흡착막대 방식의 키트가 오이모자이크바이러스 (CMV)의 현장진단을 하기 위하여 개발되었다. 토끼의 CMV항혈청에서 정제된 면역글로블린G (IgG)는 단백질 A 크로마토그래피법으로 정제되었으며 이후 금나노입자와 결합하여 니트로셀룰로스막에서 진단선 표시하도록 고안되었다. CMV항체와 비특이적으로 결합하는 단백질A를 같은 진단막대에서 대조선으로 이용되었다. RIGS-CMV 키트를 이용한 진단은 의심 식물 시료를 완충액이 들어간 플라스틱 봉지에 넣은 후 착즙 후 진단막대기를 넣으면 된다. 결과는 5-10분 후 알 수 있다. CMV가 감염된 고추, 오이 및 멜론의 즙액에 RIGS 막대기를 넣고 진단한 결과 CMV 농도에 비례하여 진단선이 형성됨이 관찰되었으며, 이들 키트들은 CMV와 연관 되지 않은 다른 고추바이러스들에서 비특이적 반응이 형성되지 않았다. 이런 결과들은 RIGS-CMV 키트가 매우 민감하며, 진단에 별 다른 실험실 기술이나 경험이 필요하지 않는 다는 것을 의미한다. 그러므로, RIGS-CMV 키트는 CMV 감염이 의심되는 식물체들의 현장 진단 뿐만 아니라 실험실에서의 CMV 진단에 효과적이다.         저온에서도 결구가 잘 형성되는 배가 반수체 배추 ‘원교20051호’ 개발

        Kimchi cabbage (Chinese cabbage) is a very common Korean traditional vegetable, cultivated across 32,000 ha, which accounts for about 13% of the total vegetable cultivation area in Korea. Since 2010, frequent occurrences of extreme weather conditions caused a shortage of summer, winter, and autumn Kimchi cabbage. Therefore, the National Institute of Horticultural and Herbal Science (NIHHS) selected a haploid breeding method of microspore culture to develop inbred lines that show resistance under extremely low or high temperature for Kimchi cabbage production. ‘Wonkyo20051ho’ is a double-haploid (DH) inbred line developed using the microspore culture method. Eleven heat resistant parental plants were selected through summer cultivation of 37 resources collected in 2014. Microspore culture was efficient in regenerating four resources, from which 13 DH inbred lines were developed. After artificially inoculating ‘Seosan’ clubroot with low concentration inoculum, 7 inbred lines showing moderate resistance were selected. As a result of the autumn cultivation of 83 breeding plants, including 6 selected inbred lines, the ‘18-FH98’ inbred line forming tight heads with yellow inner leaf under low temperature was finally selected during the 2018 autumn field trial. The selected DH inbred line was named ‘Wonkyo20051ho’ and is expected to be a valuable breeding material possessing tolerance to low temperature and clubroot.
